    Identification of kinetochores and DNA synthesis in micronuclei induced by mitomycin C and colchicine in Chinese hamster ovary cells.

    The presence of kinetochore and DNA synthesis in micronuclei (MN) induced in Chinese hamster ovary (CHO) cells by clastogenic and aneuploidogenic substances such as mitomycin C (MMC) and colchicine was determined by immunofluorescence technique using CREST antikinetochore antibodies and anti-bromodeoxyuridine (BrdUrd) antibodies. A cytofluorimetric analysis was also performed. Colchicine significantly increased micronucleated cells at least up to 96 h from the end of treatment. As expected, among colchicine-induced micronucleated cells the majority contained at least one CREST + MN. MMC induced a significant increase in micronucleated cells up to 120 h from the end of treatment and the great majority of MN lacked kinetochore fluorescence, indicating that MMC-induced MN were derived from acentric fragments. However, colchicine and MMC at 48 and 72 h from the end of treatment induced a significant increase of CREST- and CREST + MN, respectively, suggesting an induction of clastogenicity by colchicine and aneuploidy by MMC. The clastogenic effect of colchicine after 48 h was also confirmed by the presence of chromatid fragments in metaphase cells. A cytofluorimetric analysis indicated that, as aspected, colchicine and MMC interfere with the G2/M and S phases, respectively; however, a slight interference of colchicine with the S phase was also observed. DNA synthesis was present in MN and it was in most cases synchronous with synthesis in the main nucleus. The frequency of cells with MN in S phase observed in untreated or MMC-treated cells is in agreement with the proportion of cells without MN showing DNA synthesis. On the contrary, the frequency of cells with MN in S phase observed in colchicine-treated cells was significantly lower than that observed in control and MMC-treated cells

Campo Grande/MS / Felipe Soligo Barbosa, Luciane Coelho Rabel, Joyce Priscila Samudio da Silva, Julio Arani Pinheiro Xavier. – Campo Grande: UCDB, 2009As políticas de lazer são foco de inúmeras pesquisas e publicações no País, nos dias atuais (BARBOSA, 2008; MARCELLINO, 2002a, 2003; MARIANO, 2008; MENNEH, 2002). São eixos fundamentais de uma política de lazer: o uso do espaço urbano e a construção e animação dos equipamentos de lazer (BARBUY, 1980; MARCELLINO, 2002a; SANTOS, 1982; WILHEIM, 1976). Desse modo, é imprescindível para a democratização do lazer, mais especificamente dos conteúdos físico-esportivos, a elaboração de políticas públicas. Entretanto, não se pode deixar levar pela falácia do tarefismo, ou da política de atividades que, na maioria das vezes, acabam por constituir-se em eventos isolados e não em políticas de animação como processo. Faz-se necessário também atenção aos quatro eixos de uma política de lazer: reordenação da jornada de trabalho; reordenação do solo urbano, incluindo aí os espaços e equipamentos específicos de lazer; política de formação e desenvolvimento de pessoal para que os profissionais envolvidos trabalhem de forma coerente com os conceitos atuais e uma política de animação (MARCELLINO, 2007a).rede CEDES / M

    The role of cooling rate in the origin of high temperature phases at the chilled margin of magmatic intrusions

    Both large (i.e. from hundreds to thousands of metres thick) and small (i.e. from centimetres to a few metres thick) magmatic intrusions are characterized by mineral compositional variations proceeding from the outermost to the innermost part of the intrusive body. However, in the case of large intrusions, mineral compositions become progressively more primitive (e.g. An-rich plagioclases and En-rich pyroxenes) from the chilled margin towards the interior; whereas, the opposite occurs for small intrusive bodies. Since it is unclear to what extent variable cooling rate conditions may alter the phase compositions, we have performed isothermal and dynamic experiments within a temperature interval of 1250–1100 °C using four different cooling rates of 150, 50, 10 and 2.5 °C/h. Numerical simulations of thermal regimes in and around small and large magmatic intrusions have also been performed and compared with phase compositional variations observed in our laboratory experiments. Results indicate that, over rapid cooling rate conditions, the crystal compositions faithfully reproduce those of high-temperature formations, i.e. An-rich plagioclases, En-rich pyroxenes and Usp-poor spinels. However, such a process is limited to a maximum distance of 2–3 m from the margin of the intrusion. Moreover, in active volcanic systems, heat fluxes are released from the main regions of magma storage into host rocks; therefore, only magmas solidifying at the contact of cold wall rocks may develop chilled margins with features related to rapid cooling rate conditions. In the presence of hot host rocks, thermal gradients are significantly reduced and the role played by cooling dynamics on textural and compositional variations of minerals is practically negligible

    Questo lavoro presenta i risultati relativi ad un progetto di ricerca triennale, condotto nelle vendemmie 2000-2002, con l’obiettivo di valutare le potenzialità enologiche delle varietà autoctone venete, per la produzione di vini particolari, utilizzando la tecnica della macerazione carbonica e, per alcune varietà, della macerazione lunga. L’Azienda Regionale Veneto Agricoltura e l’Istituto Agrario di San Michele hanno collaborato alla realizzazione di una serie di analisi per la caratterizzazione dei biotipi oggetto del progetto. In questo capitolo dedicato alla discussione dei composti polifenolici verranno presentati i dati compositivi relativi alle analisi degli antociani nelle uve e nei vini, degli acidi idrossicinnamici nei vini, e dei flavonoli nelle uve. Questo studio ha permesso di creare una ampia banca dati compositiva, che è stata utilizzata per produrre delle classificazioni chemio-tassonomiche dei biotipi studiati nel progetto. I dati ottenuti potranno efficacemente supportare la descrizione ampelografica per la descrizione di ciascun biotipo e per la ricerca di possibili omonimie e sinonimie. Inoltre, si tratta di informazioni importanti dal punto di vista enologico e della qualità delle uve, in particolare rispetto alla dotazione di antociani che è essenziale per la produzione di un vino rosso, ma anche rispetto ad altri composti presenti in quantità minori ma di possibile interesse salutistico

    This study indicates the ways in which various aspects of the author(s) are brought forth in Dumb type’s performance art, the S/N production. Previous research has suggested a non-hierarchical organization of Dumb type and the absence of a “privileged author” in Dumb type’s collaborative work, S/N. However, the results that I have investigated from member’s interviews on the creative process of S/N along with my analysis of the recorded images of S/N, indicate a different aspect of the author(s). First, S/N was created through, so to speak, the collective ideas of the members of Dumb type. Further, S/N has at least nine quotations from previous performances, installations, and printed writings, besides the work-in-progress technique. Explicating one of the “author functions” as given by Michel Foucault, each text has plural subjects of the author. However, it has been revealed from members’ interviews that Teiji Furuhashi had a decision-making role in selecting the members’ ideas within the performance. Since then, S/N has had plural subjects of creation; however, Furuhashi is one of the subjects of creation along with the “privileged author.” S/N has plural authors (diffusive authors) yet at the same time, it has a “privileged author,” Teiji Furuhashi (cohesive author)

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.
