Contribuição à deteção de defeitos nos rotores de turboalternadores

Programa de Pós-Graduação em Engenharia Elétrica.Este trabalho tem como objetivo desenvolver métodos de detecção, localização e caracterização de defeitos em um turboalternador em escala reduzida. Os defeitos estudados neste trabalho são o curto-circuito entre espiras do rotor e as excentricidades rotóricas. Os métodos serão, em seguida, adaptados aos estudos das máquinas de 900 e 1300 MW do parque nuclear da empresa EDF (Electricité de France). Para isto, a máquina em escala reduzida, foi simulada com o auxílio de dois modelos, um analítico e um numérico por elementos finitos. Os métodos para detectar e localizar os defeitos rotóricos foram, em seguida, desenvolvidos. Os resultados das simulações utilizando os dois modelos e os testes experimentais foram analisados e comparados entre si. Uma boa concordância foi verificada, o que confirma o bom funcionamento dos métodos propostos para a detecção dessas anomalias. Vários casos de defeitos, associados a diferentes modos de trabalho da máquina, foram estudados, comprovando a confiabilidade dos métodos de detecção desenvolvidos.The aim of this work is to develop methods to detect, locate and recognise faults in a turbogenerator of reduced scale. In the framework of this study, only short-circuit defects in the rotor windings and rotor eccentricities were investigated. The developed methods would be adapted to the machines of EDF nuclear plant of 900 and 1300 MW. First the studied machine was modelled using an analytical approach and a numerical model based on the finite element method. Then, the methods to detect and locate the rotor faults were developed. The results obtained from simulations using both approaches and experimental measurements were analysed and compared. They showed a good agreement and demonstrated a good capacity of the method proposed to detect and locate the faults. Several fault cases, at different operating points of the machine, were studied and they showed the good reliability of the method

Enhancing clustering blog documents by utilizing author/reader comments
Blogs are a new form of internet phenomenon and a vast ever-increasing information resource. Mining blog files for information is a very new research direction in data mining. Blog files are different from standard web files and may need specialized mining strategies. We propose to include the title, body, and comments of the blog pages in clustering datasets from blog documents. In particular, we argue that the author/reader comments of the blog pages may have more discriminating effect in clustering blog documents. We constructed a word-page matrix by downloading blog pages from a well-known website and experimented a k-means clustering algorithm with different weights assigned to the title, body, and comment parts. Our experimental results show that assigning a larger weight value to the blog comments helps the k-means algorithm produce better clustering solutions. The experimental results confirm our hypothesis that the author/reader comments of the blog files are very useful in discriminating blog files
Vibrationally Excited Level Identification and FIR Laser Line Prediction by the "Ritz" and "LaseRitz" Programs

Two new features have been developed. (1) A new routine has been incorporated in the Ritz program for the identification of small-amplitude vibrationally excited levels starting from the known energies of the ground-vibrational levels. We have been testing this routine in the region for . (2)A new LaseRitz program has been written for predicting FIR laser lines using known energies of the ground and the vibrationally excited levels. Smart self-assembled nanoparticles for the treatment of age-related macular degeneration
Age-related macular degeneration (AMD) is a common cause of blindness among the elderly, and is characterized by choroidal neovascularization (CNV). Traditional therapies for wet AMD with anti-vascular endothelial growth factor (anti-VEGF) agents have several limitations, mainly due to the invasiveness of intravitreal injection and its arising risks of ocular complications. However, the efficacy of drugs administered via non-invasive routes (e.g., topical and systemic administration) is severely hindered by inefficient drug delivery and adverse off-target effect. To overcome these challenges, the development of stimuli-responsive drug delivery systems has emerged as an attractive approach, enabling effective drug accumulation in CNV lesions while minimizing both localized and systemic toxicity.
This thesis is dedicated to developing stimuli-activatable nanoparticles for minimally invasive management of wet AMD. Two stimuli-responsive drug delivery strategies were explored in this work, which utilized excessive oxidative stress as an endogenous trigger and light irradiation as an exogenous trigger for precisely controlled drug release. This work also investigated the potential of combination treatment of anti-angiogenic and angio-occlusive therapies for wet AMD.
In the first project, reactive oxygen species (ROS)-responsive prodrug eyedrops (Di-DAS/P-PCD) were fabricated for precise delivery of anti-angiogenic agent dasatinib (DAS) to the oxidative microenvironment of CNV. The prodrug nanoparticles were formed by cell-penetrating peptide-modified polyethylene glycol-cyclodextrin (P-PCD) and ROS-sensitive DAS prodrug Di-DAS. The prodrug eyedrops exhibited reduced cytotoxicity to normal ocular cell lines due to the inactivity of the prodrug. Topical administration of Di-DAS/P-PCD facilitated efficient choroidal penetration and was specifically activated by elevated ROS levels in a laser-induced CNV mouse model.
In the second project, a therapeutic strategy combining anti-angiogenic and photodynamic therapies was developed for wet AMD, which was achieved by intravenous administration of photoactivatable prodrug nanoparticles (Di-DAS-VER NPs). The nanoparticles were self-assembled by ROS-sensitive anti-angiogenic prodrug Di-DAS, FDA-approved photosensitizer verteporfin (VER), and biocompatible polymer-lipid. After red-light irradiation to the diseased eyes, intraocular release of DAS was observed, together with selective occlusion of neovessels by photodynamic therapy (PDT) in CNV mice.
In the third project, a near-infrared light (NIR)-cleavable cyanine prodrug system was developed to deliver both anti-angiogenic and vascular-disrupting agents to CNV lesions. A novel cyanine prodrug of the microtubule-destabilizing agent combretastatin A-4 (CA4), IR820-CA4, was co-assembled with VEGF receptor inhibitor sorafenib (SOR) into nanoparticles (SOR/IR820-CA4 NPs) without the need for additional excipients. After intravenous administration of the nanoparticles into CNV mice, 690 nm light irradiation triggered nanoparticle decomposition, releasing SOR to inhibit angiogenesis and CA4 to devastate established CNV vasculature. The treatment did not cause any noticeable systemic side effects or localized photodamage effects in vivo.
In conclusion, this thesis presents three novel stimuli-responsive nanoparticle systems for wet AMD treatment, demonstrating improved efficacy in CNV suppression alongside favorable biosafety. This study may provide novel insights into the development of stimuli-responsive prodrug systems as minimally invasive therapeutics for wet AMD and other neovascular eye diseases.published_or_final_versionPharmacology and PharmacyDoctoralDoctor of Philosoph
