The impact of Thiosulfate sulfurtransferase (TST) on metabolic dysfunction-associated steatotic liver disease (MASLD) and the metabolic benefits of calorie restriction
Metabolic dysfunction-associated steatotic liver disease (MASLD) affects 20-30% of adults in
western countries and is closely linked to obesity and type 2 diabetes. Hydrogen sulfide (H₂S),
once solely perceived as toxic, is now recognised for its role in various physiological and
pathological processes. H₂S donors have shown promise in treating fatty liver disease and
reducing blood pressure in animal models, but their therapeutic use is hindered by challenges
in H₂S pharmacokinetics. The sulphur oxidation pathway (SOP), which regulates H₂S levels
through its disposal, has been underexplored as a potential route to therapeutic H₂S elevation.
Thiosulfate sulfurtransferase (TST), a mitochondrial enzyme, is part of the SOP and
metabolises H2S, indirectly, to prevent toxicity. Previous work leading up to this thesis showed
that TST mRNA levels were upregulated during the early steatosis stage of MASLD in humans.
Given the previously identified metabolic protective effect of adipose tissue TST elevation, I
hypothesised that elevation of hepatic TST in early MASLD was a protective mechanism,
counteracting declining liver function in MASLD. Improved metabolic health following
calorie restriction (CR) is mediated in part through increased hepatic production of H₂S. Tst⁻/⁻
mice exhibited elevated systemic H₂S levels, therefore I hypothesised they may have an
enhanced response to CR.
In chapters 3 and 4, I tested the hypothesis that elevated hepatic TST expression in MASLD
offered protection against MASLD development using a liver-specific overexpression mouse
model (Liv_hTST). Male and female C57BL/6J and Liv_hTST mice were fed either a control
diet or MASLD-inducing GAN diet for 20 weeks. Systemic and hepatic sulfide levels were
measured, fat and lean mass assessed, and glucose tolerance evaluated. In vitro, HepaRG cells
with TST overexpression were tested for lipid accumulation, oxidative stress, and
mitochondrial function. Results showed sex-specific effects on sulfide levels and glucose
tolerance, with protective effects against fibrosis in male mice in vivo, and a worsening of the
impaired lipid metabolism and mitochondrial function in vitro. This research addresses the gap
in understanding of the protective role ascribed to elevated TST expression against steatogenic
liver changes in MASLD and revealed novel sex-specific effects on systemic sulfide levels,
glucose tolerance, and fibrosis.
In chapter 5, I investigated whether Tst⁻/⁻ mice experienced enhanced metabolic benefits from
CR due to their elevated systemic sulfide. Ten-week-old male and female C57BL/6J and Tst⁻/⁻
mice underwent 4-week 30% CR. Indirect calorimetry, glucose tolerance, H₂S production, and
disposal (SOP) enzyme levels were assessed. Tst⁻/⁻ males had higher systemic but similar
hepatic sulfide levels compared to C57BL/6J males, confirming previous work. CR did not
affect sulfide levels but improved glucose tolerance in Tst⁻/⁻ males, despite their resistance to
fat mass loss. Energy expenditure and substrate utilisation were similar between genotypes.
Females were unaffected by the lack of TST and had lower levels of hepatic H₂S metabolism
enzymes. Our findings suggested mechanisms beyond hepatic sulfide modulation mediate CR
benefits. Understanding the novel role of elevated systemic H₂S and TST deficiency in
maintaining fat mass and concurrent metabolic benefits with CR may inform H₂S -targeted
therapeutic strategies in the future
Gwendolyn Brooks: Poetry and the Heroic Voice
Gwendolyn Brooks is one of the major American poets of this century and the first black woman to win the Pulitzer Prize for poetry (1950). Yet far less critical attention has focused on her work than on that of her peers. In this comprehensive biocritical study, Melhem—herself a poet and critic—traces the development of Brooks\u27s poetry over four decades, from such early works as A Street in Bronzeville, Annie Allen, and The Bean Eaters, to the more recent In the Mecca, Riot, and To Disembark. In addition to analyzing the poetic devices used, Melhem examines the biographical, historical, and literary contexts of Brooks\u27s poetry: her upbringing and education, her political involvement in the struggle for civil rights, her efforts on behalf of young black poets, her role as a teacher, and her influence on black letters. Among the many sources examined are such revealing documents as Brooks\u27s correspondence with her editor of twenty years and with other writers and critics. From Melhem\u27s illuminating study emerges a picture of the poet as prophet. Brooks\u27s work, she shows, is consciously charged with the quest for emancipation and leadership, for black unity and pride. At the same time, Brooks is seen as one of the preeminent American poets of this century, influencing both African American letters and American literature generally. This important book is an indispensable guide to the work of a consummate poet.

Heroism in the New Black Poetry: Introductions and Interviews
D.H. Melhem\u27s clear introductions and frank interviews provide insight into the contemporary social and political consciousness of six acclaimed poets: Amiri Baraka, Gwendolyn Brooks, Jayne Cortez, Haki R. Madhubuti, Dudley Randall, and Sonia Sanchez. Since the 1960s, the poet hero has characterized a significant segment of Black American poetry. The six poets interviewed here have participated in and shaped the vanguard of this movement. The use of biomaterials for stem cell therapies to prevent myocardial damage post-infarct
This thesis employed a stem cell encapsulating hydrogel patch to increase the amount of beneficial soluble factors that are delivered to the surface of damaged heart tissue following a myocardial infarction. While current medical practices to address the immediate aftermath of a myocardial infarction (MI) have evolved tremendously, there are few, if any, techniques currently administered to slow, cease, or reverse the negative side effects of an occluded artery, such as the replacement of functional myocardium with non-contractile scar tissue. Because of this scar formation, survival of the initial heart attack is commonly accompanied by a decrease in left ventricular functioning due to wall thinning and ventricular enlargement. As a result of the slow, or absent, ability of cardiomyocytes to divide and repopulate the infarcted area, the burden of heart function lies on the surrounding tissue; a load that exhausts the healthy tissue and decreases the quality of life of heart attack survivors.
Mesenchymal stem cells (MSCs) have emerged as a promising therapeutic avenue for post-MI treatment, in part due to the “survival signals” that they secrete. Previous work has shown that by increasing the amount of “survival signals” that are introduced to the damaged myocardium, the extent of cardiomyocyte cell death, and subsequent scar formation, can be decreased. While the therapeutic effects of these factors have been documented, one difficulty lies in the ability to maintain a constant flux of secreted factors to the damaged site. This project hypothesized that through the encapsulation of stem cells within an engineered hydrogel construct, the hurdle of soluble factor delivery at the site of injury could be overcome. A constant flux of paracrine factors to the heart surface would allow for cell recruitment to the site(s) of damage, prevention of tissue degradation due to inhospitable environments, and promotion of neovascularizion for sustainable tissue regeneration.
Using both a chick chorioallantoic membrane assay and a mouse model of MI, the following aims determined: 1) the vascularization potential of an MSC encapsulated patch, 2) the ability to deliver hydrogels containing pro-survival signals to the heart post-MI, and 3) the ability of these factors to decrease scar formation and improve cardiac function following a heart attack. Knowledge gained from this project will provide the basis for designing materials and strategies for similar studies in larger animal models and eventually for human clinical trials. Successful delivery of the MSC encapsulating patch, and subsequent decrease in myocardial degradation, will greatly improve the quality of life scores of individuals who have suffered heart failure. O conceito de território nos livros didáticos de geografia do ensino médio do autor Melhem Adas (1970 a 1990)
A questão do território sempre esteve presente na constituição das sociedades humanas; sua apreensão e domínio estão atreladas às mais diversas formas de poder, inclusive a ideológica, que muitas vezes tem sido utilizada como maneira de justificar as mais diferentes formas de possessão, já que sua utilização se dá por meio do domínio de um determinado grupo social. Nesse sentido, a importância do território se dá, nessa pesquisa, enquanto conceito abordado nos livros didáticos de Geografia para o Ensino Médio do autor Melhem Adas, durante o período de 1970 a 1990. Para tanto, a abordagem histórica remonta ao século XIX, com a constituição do surgimento da Escola Moderna na Europa, que a priori tinha a função de justificar a fundação do Estado-nação a partir do saber escolar instituído nas escolas. A Geografia, enquanto disciplina escolar, teve um papel fundamental para justificar a ideia do nacionalismo patriótico, tão importante para a consolidação do projeto do Estado nacional nesse período. O conceito de território foi, assim, bastante utilizado para fundamentar os ideais nacionalistas e o livro didático foi o instrumento usado para garantir tais objetivos. As mudanças políticas, econômicas e sociais transcorridas durante o processo histórico redefiniram as relações da sociedade com o território e sua concepção passou por várias transformações teóricas, as quais foram analisadas de forma diferente, por vários campos da ciência, entre eles a Geografia. O recorte histórico escolhido para essa pesquisa se deu em virtude do contexto histórico no qual foi escrito a obra didática do autor acima referenciado, que no caso do Brasil, se deu a implantação da Ditadura Militar e a abertura política, a partir de meados da década de 1980, quando ocorrem também as mudanças de paradigmas da ciência geográfica.The territory issue has always been present in the constitution of human societies; its apprehension and domain are bonded to the most several forms of power, including the ideological that has often been used as a way to justify the different forms of ownership, since its usage is through the mastery of a particular social group. In this sense, the importance of territory is presented in this research as a concept discussed in Geography textbooks for middle school author Melhem Adas, during the period from 1970 to 1990. To this purpose, the historical approach goes back to the nineteenth century (XIX) with the establishment of the emergence of Modern School in Europe, which a priori had the function to justify the founding of the nation-state from the established school knowledge in schools. Geography as school content played a key role to justify the sense of patriotic nationalism, so important to get the project of the national State in this period. The concept of territory was, thus, widely used to substantiate the nationalistic ideals and the textbook was the instrument used to ensure such goals. The political, economical and social changes that occurred during the Historical process redefined the relationship between society and territory and its conception has undergone several theoretical transformations, which were analyzed differently in a large range of science fileds, including Geography. The historical angle (selected part) chosen for this study took place because of the historical context in which it was written in the didactic work of the author mentioned above, in the case of Brazil, took the implantation of the military dictatorship and political opening, from the mid-1980, when also occur the paradigm shifts of the geographical science

bir çalışmasıdır. Hanefi mezhebinin en önemli kitaplarının çoğunun alıntılandığı fetva
koleksiyonları ve fıkıh kitapların en önemli ve alimler ve müftüler için bir referans ve

"at-Tajnis va'l-Mazid ". It is known as the most important collection of fatwas and fiqh
books, from which most of the most important books of the Hanafi sect are cited, and
a reference and source for scholars and muftis. The purpose of this study is to
contribute to the publication of the book in the form closest to its author, with the hope
of providing it as a service to scholars and Islamic libraries.
The research consists of an introduction and two parts: The introduction includes the
importance of the article, the reasons for choosing the research, its aims and plan. First
part: about evaluation: Translation of the author: her life, sheikhs, students, academic
rank, works, the name of the book and documentation of its attribution to its author
and her approach to the subject are included.
The second part: The Investigation of Her Work Named " at-Tajnis va'l-Mazid " from
the Beginning of the Book of Nikah to the End of the Chapter of Zıhâr, and a brief
translation of the symbols in the manuscript and technical indexes are also included at
the end of the research
Critical Edition and Analysis of the Ṭahārah Chapter of ʿAbd al-Malik al-Kalʿī al-Ḥanafī's Ḥall al-Ramz ʿan Matn al-Kanz
Bu çalışma, Hanefi fıkhının en alimlerinden biri olan Ebü'l-Berekât en-Nesefî'nin (ö. 710) kenzü'd dekaik adlı eserinin şeyh "Abdülmelik el-Kal‛î el-Mekkî el-Hanefî tarafından kaleme alınan hallu'r-Remz an metni'l-Kenz" adlı haşiye eserini tanıtmayı amaçlar. Eserimizin önemi yazarının ilmi bir aileden gelmesinden kaynaklanır, elimizdeki tarihi kaynaklara göre Abdülmelik'in babası ve dedesi Mekke'de Hanefî mezhebinin müftüsü idiler, Abdülmelik ise 12. Yüzyılın başında babasından sonra Mekke'de 37 yıl kadar müftülük görevini üstlendi. Bu sebepten ötürü Hall'ü-Remz adlı kitap Hicaz bölgesinde Hanefî ekolünü yansıtan çokça faydalı bilgileri ihtiva etmektedir ve kitap genelde meseleleri ta‛lîl ve tartışmalı konuları ele alarak ve kıza vede uzun zor konuları ve meseleleri ele alarak naklî delile fazla başvurulmamıştır. Bunun yanında kitapta geçen ilmi şahsiyetler, kitaplar, yazma eserin resimlerini ve fihristleri zikretmiştir.This study aims to investigate one of the most important commentaries of "Kanz al-Daka'ik" in Hanafi Fiqh by Imam Abu al-Barakat Abdullah ibn Ahmad al-Nasafi (died 710 AH) titled "Hall al-Ramz 'an Matn al-Kanz" which is written by Mufti Sheikh Abdul Malik al-Qalai al-Hanafi al-Makki (died 1228 AH). The value of this commentary stems from the fact that Sheikh Abdul Malik came from a scholarly family known for jurisprudence, as both his grandfather and father were the Hanafi Muftis in Mecca. Sheikh Abdul Malik succeeded his father as Mufti for thirty-seven years at the beginning of the twelfth Hijri century. Therefore, "Hall al-Ramz" contains unique aspects that offer it academic value, such as the influence of the Hanafi school in the Hijaz. This commentary is characterized by the relative absence of primary textual evidence in discussions, striking a balance between brevity and detail in explaining and clarifying complex issues and concerns, and focusing on reasoning and clarification in contentious areas. This thesis consists of two parts. In part one, the study section discusses the life of the author, his scholarly rank, his writings, an introduction to the manuscript, the methodology used, and its attribution to the author. In part two, the Tahkik section presents the edited text as it is in the original manuscript (comprising of the entire section of Taharah) with additional subheadings for chapters and sections. It concludes with findings and recommendations as well as general index with biographical entries of scholars, books, and manuscripts
Zandaqa et Zindīqs en islam au second siècle de l’Hégire
Dans la période d'extraordinaire effervescence sociale et intellectuelle qu'a été, en Islam, le second siècle de l'Hégire, se détache, quelque peu en marge - et de façon très informelle -, une catégorie particulière de prétendus "mal-pensants", étrangement affublés du sobriquet de zanādiqa, pluriel de zindīq. La question des zanādiqa en pays d'Islam avait déjà fait, comme le rappelle Chokr, l'objet d'un certain nombre de travaux, dont notamment - le tout premier - l'article, justement célèbre, de Georges Vajda. Mais même l'article de Vajda (issu d'un mémoire de l'École Pratique des Hautes Etudes) demeurait très incomplet ; une étude détaillée, systématique, restait à faire. Melhem Chokr a donc entrepris, dans le cadre d'une thèse de doctorat - et à son entière initiative, je tiens à le préciser - de reprendre le dossier à fond, en exploitant au maximum toute la documentation disponible. Il l'a fait avec brio, et une minutie, un souci de ne rien laisser dans l'ombre, qui le conduit même parfois à déborder un tant soit peu son propos, mais dont le lecteur exigeant, je pense, n'aura pas lieu de se plaindre. Me sera-t-il permis d'ajouter ceci ? Aujourd'hui, ce n'est pas sans quelque nostalgie que l'on se reporte à ces premiers siècles de l'Hégire, si vivants, marqués par l'extrême diversité des opinions, la hardiesse intellectuelle, une incroyable liberté de propos et de mœurs (en dépit des violences qui s'employaient à la réprimer). Un des intérêts de ce livre, et non le moindre, est de nous rappeler cette époque faste. Effect of Weight Loss by Low-Calorie Diet on Cardiovascular Health in Type 2 Diabetes: An Interventional Cohort Study
Cardiovascular disease (CVD) remains a major problem for people with type 2 diabetes (T2DM), and the leading cause of death worldwide. We aimed to determine cardiovascular benefits of weight loss with or without remission of diabetes, and to assess utility of plasma biomarkers. 29 people with T2DM were studied at baseline and after dietary weight loss. Change in plasma adipokines and lipid related markers was examined in relation to weight loss, diabetes remission, 10-year cardiovascular risk (QRISK), and duration of diabetes. QRISK decreased markedly after weight loss (18.9 ± 2.2 to 11.2 ± 1.6%, p < 0.0001) in both responders and non-responders, but non-responders remained at higher risk (15.0 ± 2.0 vs. 5.8 ± 1.6%, p < 0.0001). At baseline, plasma GDF-15 was higher in longer diabetes duration (1.19 ± 0.14 vs. 0.82 ± 0.09 ng/mL, p = 0.034), as was the QRISK (22.8 ± 2.6 vs. 15.3 ± 3.4%, p = 0.031). Leptin, GDF-15 and FGF-21 decreased whereases adiponectin increased after weight loss in responders and non-responders. However, the level of FGF-21 remained higher in non-responders (0.58 [0.28–0.71] vs. 0.25 [0.15–0.42] ng/mL, p = 0.007). QRISK change correlated with change in plasma VLDL1-TG (r = 0.489, p = 0.007). There was a positive correlation between rise in HDL cholesterol and the decrease in leptin (r = 0.57, p = 0.001), or rise in adiponectin (r = 0.58, p = 0.001) levels. In conclusion, weight loss markedly decreases cardiometabolic risk particularly when remission of diabetes is achieved. Leptin, adiponectin, GDF-15 and FGF-21 changes were related to weight loss not remission of diabetes. Normalization of 10-year cardiovascular risk and heart age is possible after substantial dietary weight loss and remission of T2DM

Bu tez üç bölüm ve bir sonuçtan oluşmaktadır. Birinci bölüm: ise giriş niteliğindedir, giriş ve tez konusu ve amaçları hakkında açıklama, tez konusunun önemi, seçilme nedenleri, önceki çalışmalar ve araştırma prosedürlerini ve planını içerir, İkinci bölüm: Kitabın yazarı İmam Burhânüddîn el-Mergînânî'nin tanıtılması ve "et-Tecnîs ve'l-Mezîd" Adlı Eserinin ve yazarın bu Eserenindeki yaklaşımının tanıtılmasını içermektedir. Üçüncü bölüm: "et-Tecnîs ve'l-Mezîd" Adlı Eserinin Büyü (Alış-veriş) Kitabının Başlangıcından Kısmet Kitabının Sonuna Kadar kısmının incelenmesidir ve sonuç bölümünde kaynakları ve önerileri yazdım. Bu tezde yaptığım araştırmalar sonucunda bu Eserinin seçkin alimlerin fetva ve görüşlerini içermesi nedeniyle fıkıh ve fetva alanında çalışanlar için en önemli kaynaklardan biri olarak kabul edildiği ve gelişmelere ve çağdaş sorunlara İslam hukukunu uygulamak ve çözmek konusunda en iyi modellerden biri olarak kabul edildiği sonucuna vardım.This thesis is a study and scientific investigation of the book "at-Tajnis va'l-Mazid" by -imām Burhān al-Dīn al-Marghīnānī, from the Beginning of the Book of "Buyû" (Shopping)" to the End of the Chapter of "Kısmet"" of Division. The thesis comprises three chapters and a conclusion. As for the first chapter, it is the introductory chapter that includes the introduction, statement of the thesis topic, its objectives, the significance of the thesis topic, reasons for its selection, previous studies, research procedures, and its structure. The second chapter, it includes an introduction to the author of the book, Imām Burhān al-Dīn al-Marghīnānī, and an overview of the book "At-tajnīs wal-mazīd" as well as an exploration of the author's methodology in this book. And third chapter investigation of the Book "at-Tajnis va'l-Mazid". In the conclusion. I have concluded through my research in this thesis that this book is considered one of the most important sources for those engaged in the fields of jurisprudence and fatwas, due to its inclusion of religious edicts and the scholarly insights of eminent scholars, furthermore, it is regarded as one of the best models for applying Islamic jurisprudence to contemporary issues, addressing challenges, and providing solutions to the problems of the present time
