    Doctor en Ciencias con Mención en Biología Molecular, Celular y NeurocienciasEl lupus eritematoso sistémico (LES) es una enfermedad autoinmune caracterizada por la producción de autoanticuerpos que forman complejos inmunes, los cuales son los responsables de la inflamación y daño tisular. En el LES, los linfocitos B son mediadores claves en el desarrollo de la enfermedad, dado que se diferencian a células plasmáticas secretoras de anticuerpos. Sin embargo, se ha demostrado que los linfocitos T también cumplen un papel importante en esta enfermedad ya que proveen a los linfocitos B de señales para activarse y diferenciarse a células plasmáticas secretoras de anticuerpos. El timo es un órgano linfoide primario dedicado a la generación y maduración de linfocitos T. Antecedentes recientes de la literatura han demostrado que el timo de personas adultas no sólo cumpliría una función en el desarrollo de los linfocitos T, sino que además el timo alojaría linfocitos B e incluso actuaría como nicho de células plasmáticas productoras de anticuerpos anti antígenos virales. Más aún, estudios previos de nuestro laboratorio en el modelo murino de LES NZBWF1, han demostrado que el timo de ratones enfermos de lupus presenta importantes alteraciones en las poblaciones celulares, como cambios en las proporciones de linfocitos T e interesantemente un aumento en la población de linfocitos B en comparación a animales sanos. El conjunto de estos antecedentes nos llevó a estudiar la contribución de los linfocitos T y B presentes en el timo de ratones lúpicos NZBWF1 en el desarrollo de la respuesta humoral autoinmune. 2 Nuestros resultados indican que animales enfermos presentan una involución tímica exacerbada en comparación a animales que no se han enfermado, junto con una pérdida de la estructura normal del timo determinada por las zonas de corteza y médula. Asociado a este fenómeno, observamos alteraciones en las subpoblaciones de timocitos, que son las células en diferenciación que originarán finalmente a los linfocitos T. En ratones enfermos, observamos que los timocitos CD4+ en vías de diferenciación disminuyen y aumentan los linfocitos T CD4+ de memoria y linfocitos T foliculares. Por otro lado, existe un aumento en la frecuencia de linfocitos B en el timo previo al desarrollo de la enfermedad, pero el aumento más significativo de linfocitos B en el timo ocurre una vez que se desarrolla el LES. De manera interesante, los linfocitos B cambian su distribución en el timo de ratones enfermos, formando estructuras similares a un centro germinal. Mas aún, en el timo de ratones enfermos existe un mayor número de linfocitos B que expresan Ki-67 comparado a ratones sanos, indicando que estos linfocitos se encuentran proliferando. Además demostramos que en el timo de animales enfermos hay un aumento de células que secretan anticuerpos de tipo IgG e IgM, entre las cuales una proporción de ellas producía autoanticuerpos IgG contra ADN, un autoantígeno del LES. Dado que los linfocitos T foliculares presentes en el timo del ratón lúpico activan in vitro a los linfocitos B, nuestros resultados sugieren que en el timo de los ratones enfermos existen linfocitos B que se están diferenciando en células plasmáticas productoras de autoanticuerpos. Finalmente, demostramos in vitro que los linfocitos B del timo de ratones lúpicos favorecen la diferenciación de timocitos a linfocitos T foliculares. 3 En conclusión, la acumulación de linfocitos B en el timo de ratones lúpicos participaría en la expansión de linfocitos T foliculares en este órgano, los que a su vez podrían favorecer la diferenciación de linfocitos B a células plasmáticas autorreactivas. Nuestros resultados sugieren que el timo constituye un nicho que promueve la mantención de la respuesta humoral autoinmune en el modelo murino NZBWF1 de lupus.Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by the production of antibodies that form immune complexes, which induce inflammation and tissue damage. In SLE, B cells are key mediators of the disease development, since they differentiate into antibody-secreting cells. However, it has been demonstrated that T cells also play an important role in this disease, by delivering signals that activate and differentiate B cells into antibody-secreting plasma cells. The thymus is a primary lymphoid organ dedicated to T cell generation and maturation. Recent reports in the literature have shown that the thymus of adult individuals, not only maintains its role in T cell development, but also harbors B cells and even acts as a niche of plasma cells that produce antibodies against viral antigens. Moreover, previous work in our laboratory using the SLE murine model NZBWF1, found that the thymus of lupus-diseased mice have important alterations in its cellular composition, such as changes in T cells proportions and a remarkable increase in B cells compared to healthy mice. This converging evidence led us to study the contribution of T and B cells present in the thymus of diseased NZBWF1 mice during the development of the autoimmune humoral response. Our results indicate that diseased mice show an exacerbated thymic involution compared to animals that have not yet developed the diseased, together with a loss of normal thymic structure composed of cortex and medulla. Associated to this phenomenon, we observed alterations in the thymocyte subpopulations, which are cells 5 undergoing differentiation that give rise to T cells. In diseased mice, we observed that immature CD4+ thymocytes decrease while memory CD4+ T cells and follicular T cells are increased. On the other hand, there is a higher frequency of B cells in the thymus prior to the onset of the disease, but the most significant increase occurs once mice have developed SLE. Interestingly, B cells change their distribution in the thymus of diseased mice, forming germinal center-like structures. Moreover, in the thymus of diseased mice thymus there is a higher number of B cells expressing Ki-67 compared to healthy mice, indicating that B cells are proliferating. Also, we demonstrated that in the thymus of diseased mice there is an increase of cells that secrete IgG and IgM antibodies among which a fraction produced IgG autoantibodies against DNA, an autoantigen of SLE. Follicular T cells present in the lupic mouse thymus were shown to induce B cells activation in vitro, suggesting that the thymus of diseased mice contains B cells that are differentiating in antibody-producing plasma cells. Finally, in vitro we demonstrated that thymic B cells from lupic mice favour the differentiation of follicular T cells from thymocytes. In conclusion, the accumulation of B cells in the thymus of lupic mice could participate in the expansion of follicular helper T cells in this organ, which may in turn promote the activation and differentiation of B cells into autoreactive plasma cells.     Este artículo presenta un estudio sobre la educación artística comunitaria en la Universidad de Laponia en Finlandia. En primer lugar se analizan las líneas principales de este modelo formativo y de su plan de estudios. En segundo lugar se presenta una entrevista con Timo Jokela, Decano de la Facultad de Arte y Diseño de la Universidad de Laponia y director del Departamento de Educación Artística en esa misma Facultad. En la entrevista Timo Jokela habla de las relaciones entre arte, medio ambiente, comunidad y educación partiendo de su propia experiencia como artista y centrado en el contexto finlandés. También sobre los aspectos sociales, culturales, artísticos y educativos que están implícitos en el modelo de educación artística comunitaria que se pone en práctica, como itinerario formativo, en la Universidad de Laponia. This article presents a study on community-based art education at the University of Lapland in Finland. First, the main guidelines of this training model and its curriculum are analysed. Second, the author includes an interview with Timo Jokela, Dean of the Faculty of Art and Design of The University of Lapland in Finland and Director of the Department of Art Education at the same faculty. In the interview Timo Jokela draws upon his own experience as an artist in Finland to talk about the relationships between art, the environment, community and education. He also talks about the social, cultural, artistic and educational aspects, which are central to the community-based art education scheme in place at The University of Lapland

    Ei-ladattavan hybridiauton esilämmitys : vaikutukset järjestelmän toimintakykyyn

    Tämän opinnäytetyön tarkoituksena oli tutkia ja selvittää sähköisen esilämmityksen vaikutuksia hybridijärjestelmän toimintaan. Työssä tarkasteltiin erityisesti vaikutuksia matkaan joka ajettiin pelkästään käyttämällä sähköä auton hybridiakustosta. Samalla seurattiin auton polttoaineen keskikulutuksen muutoksia. Nämä vaikutukset saatiin parhaiten näkyviin, ajamalla samoja reittejä esilämmitetyllä ja ilman esilämmitystä olevalla autolla sekä vertailemalla näistä saatuja tuloksia. Työssä käytettävä hybridiauto on tekijän omistuksessa ja omassa käytössä. Autoa käytetään pääsääntöisesti taajamaliikenteessä, mutta myös maantie- ja moottoritienopeuksissa. Ajaminen koostuu tekijän normaalista auton käytöstä. Työhön kuuluu kolme eri vaihetta. Ensimmäisessä vaiheessa autoa esilämmitettiin ennen jokaista kotona suoritettua liikkeellelähtöä, työn tekemisen aikana voimassa olleiden suosituksien mukaisesti. Toisessa vaiheessa autoa ei esilämmitetty ennen ajoon lähtöä. Kolmannessa vaiheessa tulokset kirjattiin muistiin ja analysoitiin. Hybridijärjestelmän käyttö vähentää polttoaineen kulutusta ja päästöjä verrattuna pelkästään polttomoottorilla varustettuun autoon. Auton esilämmitys parantaa sekä ajoturvallisuutta että mukavuutta, kun lämpötila on pakkasella tai lähellä nollaa. Esilämmitetty moottori saavuttaa nopeammin normaalin käyntilämpötilan verrattuna moottoriin, jota ei ole esilämmitetty ennen ajoon lähtöä. Kun moottori käy normaalilla käyntilämpötilallaan, päästöt ja polttoaineen kulutus pienentyvät verrattuna kylmänä käyvään moottoriin. Myös käynnistyksen päästöjä saadaan pienennettyä esilämmityksellä. Autolla on ajettu pääsääntöisesti lyhyitä matkoja, jolloin järjestelmän polttomoottori ei ole saavuttanut normaalia käyntilämpötilaansa. Työn tuloksena havaittiin, että esilämmitys vaikuttaa myönteisesti hybridijärjestelmän kykyyn käyttää pelkästään sähkömoottoria auton liikuttamiseen. Tämä tulos on ennakko-odotusten mukainen.The purpose of the thesis was to study the effects of electronic preheating on the operation of a hybrid system. The impact of the trip, which was driven by using only electricity from the car hybrid battery, was specifically examined in this thesis. At the same time, changes in the average fuel consumption of the car were tracked. The effects were best noticed driving the same routes with a preheated car and the car without preheating, and comparing the results. The hybrid car used in the work is owned and used by the author. The car is mainly used in urban area but also at highway speeds. Driving consists of the author´s normal car use. The work involved three different phases. In the first phase, the car was preheated before each start-up at home, in accordance with the recommendations in force at the time of the work. In the second phase, the car was not preheated before start-ups. In the third phase, the results were reported and analyzed. Using a hybrid system in a car reduces fuel consumption and emissions compared to a car with an internal combustion engine only. Car preheating improves both driving safely and comfort when the temperature is low or near zero degrees. A preheated engine reaches faster its normal operating temperature compared to an engine that has not been preheated before driving. When the engine is running at its normal operating temperature, emissions and fuel consumption are reduced compared to a cold engine. Start-up emissions can also be reduced by preheating. The car has been mainly driven for short distances, whereupon the internal combustion engine of the system has not reached its normal operating temperature. The study shows that preheating has a positive effect on the ability of the hybrid system to use only an electric motor to move the car. This result is in line with the expectations

    Analysis of monocyte cell tractions in 2.5D reveals mesoscale mechanics of podosomes during substrate-indenting cell protrusion

    Podosomes are mechanosensitive protrusive actin structures that are prominent in myeloid cells, and they have been linked to vascular extravasation. Recent studies have suggested that podosomes are hierarchically organized and have coordinated dynamics on the cell scale, which implies that the local force generation by single podosomes can be different from their global combined action. Complementary to previous studies focusing on individual podosomes, here we investigated the cell-wide force generation of podosome-bearing ER-Hoxb8 monocytes. We found that the occurrence of focal tractions accompanied by a cell-wide substrate indentation cannot be explained by summing the forces of single podosomes. Instead, our findings suggest that superimposed contraction on the cell scale gives rise to a buckling mechanism that can explain the measured cell-scale indentation. Specifically, the actomyosin network contraction causes peripheral in-plane substrate tractions, while the accumulated internal stress results in out-of-plane deformation in the central cell region via a buckling instability, producing the cell-scale indentation. Hence, we propose that contraction of the actomyosin network, which connects the podosomes, leads to a substrate indentation that acts in addition to the protrusion forces of individual podosomes.     2.5 D Tractions in monocytes reveal mesoscale mechanics of podosomes during substrate indenting cell protrusion

    Degradation and protrusion are key to cellular barrier breaching in cancer metastasis and leukocyte extravasation. Cancerous invadopodia and myelomonocytic podosomes are widely considered as structural tools facilitating these processes and are thus summarized under the term invadosomes. Despite similar behaviour on the individual scale, substantial differences have been reported to arise on the collective scale. They are considered to be a result of podosome mesoscale-connectivity. In this study, we investigated global in-plane and out-of-plane mechanical forces of podosome clusters in ER-Hoxb8 cell derived monocytes. We are able to correlate these forces with the interpodosomal connectivity. The observed traction and protrusion patterns fail to be explained by summation of single podosome mechanics. Instead, they appear to originate from superimposed mesoscale effects. Based on mechanistic and morphological similarities with epithelial monolayer mechanics, we propose a spatiotemporal model of podosome cluster mechanics capable of relating single to collective podosome mechanical behaviour. Our results suggest that network contraction-driven (in-plane) tractions lead to a buckling instability that contributes to the out-of-plane indentation into the substrate. First assigning an active mechanical role to the dorsal podosome actomyosin network, we aim at translating actomyosin hierarchy into scale dependency of podosome mechanics

    Tagging of Biomedical Articles on CiteULike: A Comparison of User, Author and Professional Indexing

    This paper examines the context of online indexing from the viewpoint of three different groups: users, authors, and professional indexers. User tags, author keywords and descriptors were collected from academic journal articles, which were both indexed in Pubmed and tagged on CiteULike, and analysed. Descriptive statistics, informetric measures, and thesaural term comparison shows that there are important differences in the use of keywords between the three groups in addition to similarities which can be used to enhance support for search and browse. While tags and author keywords were found that matched descriptors exactly, other terms which did not match but provided important expansion to the indexing lexicon were found. These additional terms could be used to enhance support for searching and browsing in article databases as well as to provide invaluable data for entry vocabulary and emergent terminology for regular updates to indexing systems. Additionally, the study suggests that tags support organisation by association to task, projects and subject while making important connections to traditional systems which classify into subject categories
