Synergestic induction of NRF-2 gene by curcumin and sulforaphane and pharmacokinetics/metabolism of 13C/Dim in rats by UPLC/MS
Curcumin (CUR) and Sulforaphane (SFN) have shown remarkable cancer chemopreventive effects in numerous studies and combinations of low doses of chemopreventive agents can reduce toxicity while augmenting efficacy. The first part of the thesis investigated the chemotherapeutic effects elicited by a combination of CUR and SFN on human hepatocarcinoma
cells. The combination treatment- mediated effects on phase II/antioxidant enzymatic induction and antioxidant response element (ARE) was investigated. It was proposed that the combination of CUR and SFN could synergistically enhance the induction of ARE and the nuclear E2-factor related factor 2 (Nrf2)-mediated enzymes. Low doses of CUR and SFN significantly induced the expression of Nrf2-mediated enzymes, HO-1 and UGT1A1, promoted nuclear translocation of
Nrf2– a key regulator of phase II detoxifying /antioxidant enzymes – and synergistically induced the ARE luciferase activity. Chemical inhibitors of mRNA and protein synthesis affected the combination therapy- mediated transcriptional regulation of both HO-1 and UGT1A1. Synergism of CUR and SFN was evident at low concentrations. Such synergism in ARE-luciferase activity may partly explain the significant induction in the expression of Nrf2- mediated expression of HO-1 and UGT1A1 and the nuclear translocation of Nrf2, suggesting that a combination of low doses may be a promising strategy for cancer chemoprevention. For the second part of this thesis, a liquid chromatographic method was validated for the simultaneous analysis and pharmacokinetic evaluation of Indole-3-Carbinol (I3C), Diindolylmethane (DIM), and several I3C metabolites. I3C and DIM are naturally derived phytochemicals with promising in-vitro and
in-vivo anticarcinogenic properties. Using reversed-phase ultra performance liquid
chromatography (UPLC) coupled with mass spectrophotometry (MS), a rapid, specific, and high
throughput method was developed and validated for the quantification and identification of I3C, DIM, and other I3C metabolites in plasma. Recovery, linearity, precision, accuracy, and stability analysis fulfilled the CDER guidelines criteria. Contrary to an initial understanding of necrosis as a chaotic, non-regulated type of cell death, recent studies suggest that necrotic cell death (NCD) is a consequence of a complex and lethal cascade of genetically-encoded events. Necrosis underlies the pathology of many neurodegenerative diseases, stroke, and traumatic injury. In the Driscoll Lab, NCD mechanisms have been addressed for several years taking advantage of unique genetic and molecular biology tools developed in the model organism Caenorhabditis elegans. The necrotic paradigm we study the most involves initiation of cell death by hyperactivated ion channels expressed in six touch-sensory neurons and requires elevation of intracellular Ca2+, which activates calpain and cathepsin proteases.
I exploited the unique features of our model system to uncover novel genetic factors influencing this process. To this end, I conducted a high-throughput forward genetic screen to identify mutations that block or delay necrotic cell death induced by MEC-4(d) channel hyperactivation, and genetically mapped novel mutations capable of blocking or slowing the death process. I exploited an automated mutational screening capacity that allows sorting of individual animals based on detection of fluorescent signals that, in our particular case, had been engineered to indicate neuronal viability. I focused on the cloning of two novel mutant loci and dissected molecular mechanisms responsible for death suppression. In addition, I studied the impact of a major subset of calcium homeostasis genes in a C. elegans model of Aβ toxicity.
My research adds a new component to the current understanding of NCD, suggesting that inability to cope with endoplasmic reticulum stress (presumably induced by calcium depletion inside the ER, which affects chaperone functionality) plays an important role in progression through necrosis. I discovered that mild activation of an intact unfolded protein response (UPR), e.g., as induced by downregulation of UDP-glucose:glycoprotein glucosyltransferase (UGGT, an ER-resident enzyme involved in high-fidelity protein folding quality control) or mild increments in ambient temperature, can partially suppress necrosis in our C. elegans model, reminiscent of beneficial preconditioning effects in mammals. Additionally I found that several UPR transducers contribute to such modulation of cell death in a “tug-of-war” fashion. Our refined model of molecular mechanisms contributing and modulating necrosis suggests new strategies that could eventually limit the devastating effects of necrosis in human injury and disease.Ph.D.Includes bibliographical references (p. 154-174)by Yury Orlando Nunez Lope
Absorbing new subjects: holography as an analog of photography
I discuss the early history of holography and explore how perceptions, applications, and forecasts of the subject were shaped by prior experience. I focus on the work of Dennis Gabor (1900–1979) in England,Yury N. Denisyuk (b. 1924) in the Soviet Union, and Emmett N. Leith (1927–2005) and Juris Upatnieks (b. 1936) in the United States. I show that the evolution of holography was simultaneously promoted and constrained by its identification as an analog of photography, an association that influenced its assessment by successive audiences of practitioners, entrepreneurs, and consumers. One consequence is that holography can be seen as an example of a modern technical subject that has been shaped by cultural influences more powerfully than generally appreciated.
Conversely, the understanding of this new science and technology in terms of an older one helps
to explain why the cultural effects of holography have been more muted than anticipated by forecasters
between the 1960s and 1990s

Significance of the level of implementation of food legislation and the food safety management system in Ukraine for the dynamics of the spread of food poisoning (retro analysis)
Ensuring safe nutrition is one of the important tasks of each state for the proper state of public health. In developed countries such as the US and the EU, food safety is a public health priority and is ensured through the development of a regulatory framework, the establishment and implementation of effective food safety systems throughout the food chain. However, despite the existence of legislation and government oversight, official data show a high number of outbreaks of foodborne diseases in these countries. In this regard, WHO calls on states to constantly monitor the situation with the occurrence of food poisoning in order to improve the means of monitoring the safe nutrition of the population. In Ukraine, there is food legislation, which was formed much later than in the EU and the USA, and later the mechanism for ensuring safe food was introduced – the HACCP system (Hazard Analysis and Critical Control, English Point). The article presents retrospective analytical data related to the occurrence of food poisoning among the population of Ukraine in the pre-war period (2019−2020) and analyzes the problems that contributed to the emergence of these diseases. Comparison in dynamics allows to see not only the changes that have taken place, but also to understand the advantages and disadvantages that were applied in the previous period and develop measures based on the experience gained over the past years. The target issues of this article were the analysis of international and national food legislation as important tools for managing the level of food poisoning, a retro-analysis of the level of food poisoning in different regions of Ukraine and the establishment of factors that contributed to the occurrence of these disease

Yury Tynyanov. Energy of the Form.pdf
This book contains articles published at different times, the theme of which is being determined by the innovation of Tynyanov-theorist. Collected under one cover they express, according to the author, the essence of the book’s title. The first theoretical article serves as an introduction to the general theme. And it also is a kind of pretext to the main “Onegin” block of the book, which represents the dynamic concept of literature and the Pushkin’s novel. The article on renewal and development of the Tynyanov's concept of “Eugene Onegin” in the 1960s, which revolutionized the understanding of the poetic novel, closes the Pushkin theme. The Pushkin’s novel in poetry became the reservoir of the theoretical energy of Yu. Tynyanov. Therefore, it is obvious that the language of the novel description and the scientific text of Tynyanov could have the correlative relationships. There is an article about it, devoted to the poetics of Tynyanov’s scientific prose. In two articles completing the book, Tynyanov is mentioned in the context of the main representatives of Russian formalism – Viktor Shklovsky and Boris Eichenbaum. The involvement of the two pillars of the formal method has no opposition significance, same as significance in the similarity of their research positions. Here was important to comprehend the difference in themes, “methods” and approaches of the famous formalists – V. Shklovsky, B. Eichenbaum and Yu. Tynyanov, and also to show that the literary centrism of formalists is far from a self-evident fact. The author did not set himself the task of recalling Tynyanov by praising his ideas in the existing cultural paradigm. According to the author, the main events of the book should be both – the energetic nature of Tynyanov’s theoretical constructions and the energy of the meta text offered to the book’s readers. The book is plump with quotations and citation exchange (for which we apologize to the readers), but which the author did not neglect, so as not to violate the integrity of the narration. Also, here is a technical clarification: all emphasized citations belong to the authors of the quoted texts. The author of the book intentionally tried to avoid the interpretation scheme, following the methodological installation, which Sergei Georgievich Bocharov in the preface to his book «Subjects of Russian Literature» outlined with a notable formula: «Literary criticism is also literature and a philologist is a writer».</p
The Semantics of Animal Images in the Novel “The Monkey Comes for his Skull” by Yury Dombrovsky.
Dzīvnieku tēlu semantika Jurija Dombrovska romānā “Mērkaķis atnāk pēc sava galvaskausa”: bakalaura darbs. – Rīga, 2013. – 60 lpp.
Darbs ir veltīts dzīvnieku tēlu semantikas Jurija Dombrovska romāna „Mērkaķis atnāk pēc sava galvaskausa” izpētei. Pētījuma mērķis ir romāna dzīvnieku tēlu autora semantikas īpašību atklāšana.
Šis darbs sastāv no divām daļām. Pirmajā daļā autors veic dzīvnieku tēlu semantikas pētīšanas vēstures pārskatu literatūrzinātnē, kā arī atsevišķu dzīvnieku tēlu semantikas pētīšanas materiālu pārskatu mītoloģiskajā un tautas tradicijā. The present work is devoted to the research of semantics of the animal images in Yuri Dombrovsky’s novel “The Monkey Comes for his Skull”. The goal of the research is to reveal the specific character of author’s semantics of animal images in the novel.
The thesis consists of two parts. In the first part, the author overlooks the material regarding the history of animal image semantics’ study in the literary studies and also overlooks the material regarding the research of individual animal image semantics in mythological and folk tradition. The second part consists of analytical and comparative analyses of animal images in the novel „The Monkey Comes for his Skull”.
