    The regenerative potential of Pax3/Pax7 on skeletal muscle injury

    BACKGROUND: Skeletal muscle mishaps are the most well-known incidents in society, especially among athletes and the military population. From the various urgency, this accident needs to be cured more quickly. However, the current treatment still has some shortcomings and is less effective. In this case, Paired box 3 and Paired box 7 (Pax3/Pax7) proteins that induce stem cells could potentially be an alternative treatment for skeletal muscle injuries. This paper aimed to analyse the potential treatment of Pax3/Pax7 proteins inducing the stem cell for skeletal muscle injuries. THE MAIN BODY OF THE ABSTRACT: We did a narrative review by gathering several scientific journals from several leading platforms like PubMed and Scopus. As common accidents, skeletal muscle disease could be due to workplace and non-workplace causes. The highest risk occurs in the athlete and military environment. The treatment of current skeletal muscle injuries is protection, rest, ice, compression, and elevation (PRICE), non-steroidal anti-inflammatory drugs (NSAIDs), and mechanical stimulation. However, it is considered less effective, especially in NSAIDs, inhibiting myogenic cell proliferation. The current finding indicates that the stem cells have markers known as Pax3/Pax7. The role of both markers in muscle injury, Pax3/Pax7, as transcription factors will induce cell division by H3K4 methylation mechanisms and chromatin modifications that stimulate gene activation. CONCLUSION: Regulation by Pax3/Pax7 factors that affect stem cells and stem cell proliferation is one of the alternative treatments. This regulation can accelerate the healing of injury victims, especially injuries to the skeletal muscles. Finally, after being compared, Pax3/Pax7 induces stem cells to have the potential to be one of the skeletal muscle injury treatments.     ABSTRAK Mikrobiologi adalah ilmu yang mempelajari tentang semua mikroorganisme, seperti bakteri, fungi, alga, protozoa, dan archaea. Bakteri dapat bersifat sebagai bakteri baik dan bakteri yang dapat menyebabkan patogen. Bakteri baik merupakan sekumpulan bakteri yang mempunyai peran penting dalam membantu proses pencernaan serta penyerapan zat gizi dalam tubuh. Namun, masyarakat pada umumnya belum mengetahui perbedaan bakteri baik dan bakteri yang dapat menyebabkan pathogen. Penyuluhan tentang pemanfaatan bakteri baik untuk hidup sehat ini dilakukan untuk memberikan edukasi kepada masyarakat tentang pemanfaatan kekayaan biodiversitas mikroba Indonesia di berbagai bidang seperti kesehatan. Penyuluhan ini dilakukan melalui beberapa tahap, yaitu persiapan, pelaksanaan dan evaluasi. Penyuluhan dilakukan melalui kanal Zoom dan Youtube yang dikelola oleh Fakultas Farmasi Militer Universitas Pertahanan RI. Penyuluhan ini mengangkat tema Pemanfaatan bakteri baik untuk hidup sehat dengan mengundang pakar di bidang tersebut. Kegiatan penyuluhan dilakukan pada hari Sabtu, 14 Mei 2022 pukul 09.00-11.30 WIB dihadiri oleh 158 peserta masyarakat umum. Terjadi peningkatan pengetahuan dan pemahaman peserta penyuluhan terhadap pemanfaatan bakteri baik untuk hidup sehat. Kata Kunci: Penyuluhan, Bakteri Baik, Kesehatan, Probiotik  ABSTRACT Microbiology is the study of all microorganisms, such as bacteria, fungi, algae, protozoa, and archaea. Bacteria can act as good bacteria and bacteria that can cause pathogens. Good bacteria are a group of bacteria that have an important role in helping the digestive process and absorption of nutrients in the body. However, people in general have limited informationabout the difference between good bacteria and bacteria that can cause pathogens. This counseling on the use of good bacteria for healthy living is carried out to provide education to the public about the use of Indonesia's rich microbial biodiversity in various fields such as health. This counseling is carried out through several stages, namely preparation, implementation and evaluation. Counseling is carried out through Zoom and Youtube channels which are managed by the Faculty of Military Pharmacy, Indonesian Defense University. This counseling raised the theme of Utilizing good bacteria for a healthy life by inviting experts in the field. The counseling activity was carried out on Saturday, May 14, 2022 at 09.00-11.30 WIB and was attended by 158 participants from the general public.     Objective: Sucrose phosphorylase (SPase) is an enzyme that catalyzes the transfer of glucosyl to various acceptor molecules. Distinct types of SPaseshave been reported, and their transglycosylase activities have been shown to differ. In general, glycosylation is a process that is used to modifybioactive compounds. As such, glycosylation can increase the chemical stability of compounds and improve their characteristics such as reduce strongsmell and sour taste. We previously cloned recombinant SPase (SPaseWRS-3[1]) from Leuconostoc mesenteroides MBFWRS-3[1] in Escherichia coli.In the current study, we aimed to characterize SPaseWRS-3 and determine its transglycosylation activity using benzoic acid (BA), ascorbic acid, andkojic acid (KA).Methods: Expression analyses were conducted in lysogeny broth (LB) medium supplemented with tetracycline and expression was induced usingisopropyl-β-d-thiogalactopyranoside. The characteristics of the 56 kDa recombinant SPase (rec-SPase) were confirmed using sodium dodecyl sulfatepolyacrylamide gel electrophoresis (SDS-PAGE). Rec-SPase activity was determined spectrophotometrically using sucrose as the substrate and NADPHas the end-product at 340 nm. Transglycosylation activity was evaluated using thin-layer chromatography (TLC) on silica gel plates.Results: Our results demonstrated that the rec-SPase had an activity of 98.52% relative to the reference SPase (ref-SPase). BA and KA were determinedto undergo glucosyl transfer by rec-SPase using ref-SPase, as observed with TLC. Our findings are consistent with those reported previously for theSPase isolated from L. mesenteroides.Conclusion: Recombinant SPase activity is comparable to reference SPase activity. Our study could be the initial study to deeply observe SPase activityin other substrates as well

    CRISPR/Cas9 Genome Editing: Future Treatment of Duane Retraction Syndrome

    Duane Retraction Syndrome (DRS) is characterized by limited eye movement. One of its causes is a mutation in the CHN1, MAFB, or SALL4 gene. Nowadays, the treatment for DRS is limited to glasses, occlusion, and surgery. However, this treatment has not been able to cure the disease’s hereditary issue. Another strategy to be considered for the treatment is CRISPR/Cas9, a tool for performing gene editing with a wide range of applications, including treating genetic diseases. We made sgRNA as a first step in using CRISPR/Cas9 as a treatment for DRS in silico using the CCTop website. By computing sgRNA, conducting tests, and analyzing the results, CRISPR/Cas9 may repair genetic mutations. Currently, there are no reports on the use of CRISPR/Cas9 in DRS. Hence, this study would be very useful as a starting point for using CRISPR/Cas9 as a DRS treatment. However, it needs to be further proven through in vivo, in vitro, and clinical trials study

    Objective: The Cholecystokinin A receptor (CCK-ARs), also known as CCK1 receptor, is a type of G protein-coupled receptor that is primarily responsive to the hormone Cholecystokinin (CCK). CCK-ARs is one of the receptors characterized and validated to promote pancreatic cancer progression. Devazepide is a selective antagonist of the CCK-ARs. This study aims to find a potential ligand that has the most effective and representative interaction with cancer receptors, becoming a new therapeutic effect using molecular docking Molecular Operating Environment (MOE) with receptor code 7F8U. Methods: We conducted an in silico study by docking candidate ligands with Cholecystokinin Receptor (CCKRs) using the MOE 2015 V.10 application. The ligands of choice come from natural ingredients such as curcumin, resveratrol, berberine, baicalein, dioscin, wogonin, and piperine. Validate the receptor with the Root mean Square Deviation (RMSD) value and docking results with the GIBBS S value. Results: 6 compounds, such as curcumin, resveratrol, berberine, baicalein, wogonin, and piperine, were selected for docking as candidates to determine whether they have interactions with CCK-ARs. Based on the docking results, the Gibbs values ​​obtained were -14.9522;-12.4566;-15.5033;-12.6961;-13.4234;-11,6130 joules/kg. mol, berberine is the compound with the lowest Gibbs energy, namely -15.5033 joules/kg. mol and is one of the strongest. The interactions that occur include Methionine A121-side chain donor, Methionine B121-side chain donor, asparagine A333-amine group and nitrogen atom, B333-amine group and nitrogen atom, Arginine A336-negative oxygen atom, and B336-negative oxygen atom. Conclusion: Berberine which is a natural alkaloid, is suitable for devazepide, which is a positive control for ligand interactions when tethered to the CCKRs. This finding could be a potential new drug for pancreatic cancer. However, further studies, such as in vitro, in vivo, and clinical trials need to be conducted for ordering activity, safety, and safety of new drugs

    Chemical Properties, Biological Activities and Poisoning Treatment of Novichok: A Review

    Novichok is an organophosphate compound found as a nerve agent chemical weapon. However, the information about its chemical properties, biological activities, and molecular interactions in the body are still protected under the “top secret” security clearance. Novichok, with the codes A230, A232 and A234, is a compound whose structure has been successfully determined. The compound is synthesized from a precursor through a nucleophilic substitution reaction. Novichok agents are considered more potent than VX gas and can be applied in unitary and binary forms. This compound has ability for the binding with acetylcholinesterase (AChE) due to inability of acetylcholine metabolism. AChE catalyzes the rapid hydrolysis of acetylcholine to acetate and choline. The treatment of Novichok agent poisoning is similar to management of other nerve agents, such as atropine and pralidoxime administered intravenously. In this paper, we reviewed the Novichok component from chemical and biological perspective. Moreover, we discussed the potential molecular interaction and treatment of this compound

    Penguatan Program Promotif dan Preventif melalui Skrining Tekanan Darah, Gula Darah Sewaktu, dan Saturasi Oksigen

    ABSTRAK Penyakit tidak menular seperti obesitas, hipertensi, dan diabetes mellitus menjadi masalah kesehatan utama di Indonesia, yang memerlukan deteksi dini dan intervensi tepat waktu. Kegiatan pengabdian masyarakat ini bertujuan untuk mendapatkan gambaran indeks massa tubuh (IMT), saturasi oksigen, tekanan darah, dan kadar gula darah sewaktu (GDS) di Desa C. Penelitian menggunakan metode deskriptif kuantitatif dengan melibatkan 50 responden. Pengukuran IMT, saturasi oksigen, tekanan darah sistolik dan diastolik, serta GDS dilakukan untuk mengidentifikasi risiko penyakit tidak menular. Hasil analisis menunjukkan bahwa 56% responden berada dalam kategori obesitas, 26% tergolong overweight, dan 18% memiliki IMT normal. Tekanan darah sistolik mencatat bahwa 30% responden berada diatas normal, sementara kadar GDS yang melebihi 200 mg/dL ditemukan pada 12% responden. Hasil ini mendorong adanya pemeriksaan lebih lanjut untuk menegakkan diagnosa dan intervensi melalui edukasi kesehatan serta pentingnya gaya hidup sehat.   Kata kunci: Pengabdian Masyarakat, Deteksi Dini, Indeks Masa Tubuh, Tekanan Gula Darah, Gula Darah Sewaktu. ABSTRACT Non-communicable diseases (NCDs) such as obesity, hypertension, and diabetes mellitus are major health issues in Indonesia, requiring early detection and timely intervention. This community service activity aims to provide an overview of body mass index (BMI), oxygen saturation, blood pressure, and random blood glucose (RBG) levels in C Village, Gunung Putri District. The study employed a descriptive quantitative method involving 50 respondents. Measurements of BMI, oxygen saturation, systolic and diastolic blood pressure, and RBG were conducted to identify the risk of non-communicable diseases. The analysis revealed that 56% of respondents were classified as obese, 26% were overweight, and 18% had normal BMI. Systolic blood pressure indicated that 30% of respondents had value above normal, while random blood glucose levels exceeding 200 mg/dL were found in 12% of respondents. These findings suggest the need for further examinations to confirm diagnosis and emphasize interventions through health education and the importance of a healthy lifestyle. Keywords: Community Service, Early Detection, Body Mass Index, Blood Pressure, Random Blood Glucose
