    Associação dos polimorfismos da região promotora e 3' não traduzida do gene HLA-G com doenças autoimunes e inflamatória crônica

    Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Ciências Biológicas, Programa de Pós-Graduação em Biologia Celular e do Desenvolvimento, Florianópolis, 2018.Lúpus Eritematoso Sistêmico (LES) e Artrite Reumatoide (AR) são doenças reumáticas inflamatórias crônicas de origem autoimune. Psoríase (PSO) é uma doença de pele comum, também inflamatória crônica, porém de origem desconhecida. As três doenças são multifatoriais e relacionadas ao sistema imunológico, às interações com o meio ambiente e à suscetibilidade genética. As moléculas de HLA-G são de grande importância na indução da tolerância imunológica e o estudo do gene HLA-G em doenças inflamatórias e reumatológicas é importante. O gene HLA-G possui sítios de variação (SNVs) em suas regiões regulatórias 5´URR e 3´UTR, que influenciam na expressão proteica. O presente estudo teve como principal objetivo estudar os SNVs da 5´URR em pacientes com LES, AR e PSO e indivíduos sem o histórico das doenças (466 indivíduos no total) e nove SNVs da 3´UTR em pacientes com LES e indivíduos controle (236 indivíduos no total) do Estado de Santa Catarina, visando compreender o papel desse gene no desenvolvimento das doenças e suas manifestações clínicas. Após sequenciamento das amostras e análises das sequências, foram realizadas análises de associação considerando cada locus em estudo caso-controle e caso-caso. No estudo da 3´UTR, os SNVs +3010CG, +3027AC, +3035CT, +3142CG e +3196CG foram associados ao risco de desenvolvimento de LES. As manifestações clínicas de LES também foram associadas aos SNVs da 3´UTR. A análise dos haplótipos, associou a UTR1 e as combinações UTR1+UTR1 e UTR1+UTR7 à proteção para o desenvolvimento de LES. Para a 5´URR, 17 SNVs foram associados ao risco de desenvolvimento de pelo menos uma das doenças, à LES (-725C>G>T, -483G>A, -297G>A e -284G>A), à AR (-762C>T, -725C>G>T, -540InsG a -533DelA, -509C>G, -486C>A, -483G>A e -297G>A) e à PSO (-762C>T, -725C>G>T, -716G>T, -689G>A, -646A>G, -633A>G, -509C>G, -483G>A, -443G-A, -540InsG a -533DelA, -400G>A, -391G>A, -369A>C, -355G>A e -297G>A). As manifestações clínicas das três doenças também foram associadas aos SNVs da 5´URR. Ainda, o haplótipo 5 da 5´URR foi associado ao risco de desenvolvimento de LES e AR. Concluímos que os SNVs das regiões 5´UR e 3´UT do HLA-G são associados à LES, AR e PSO bem como aos dados clínicos analisados neste trabalho. Assim, SNVs nestas regiões devem estar relacionadas com os níveis de expressão da molécula e consequentemente, com a imunorregulação, local ou sistêmica.Abstract : Systemic Lupus Erythematosus (SLE) and Rheumatoid Arthritis (RA) are chronic inflammatory rheumatic diseases of autoimmune origin. Psoriasis (PSO) is a common skin disease, also chronic inflammatory, but of unknown origin. The three diseases are multifactorial, related to the immune system, to environmental interactions and genetic susceptibility. HLA-G molecules were described as of great importance in the induction of immune tolerance and the study of HLA-G gene in inflammatory diseases and processes have become a challenge for researchers. The gene HLA-G has single nucleotide variations (SNV) in regulatory regions 5´URR and 3´UTR, described as having influence on protein expression. The present study had as main objective to study the SNV of 5´URR in SLE, AR and PSO patients and individuals without history of diseases (466 individuals in total) and nine 3´UTR SNV in SLE patients and control subjects (236 individuals in total) in the State of Santa Catarina, aiming to understand the role of this gene in the development of diseases and their clinical manifestations. After sequencing samples and analyzing the sequences, association analysis was carried out considering each locus in case-control and case-case study. The study of 3´UTR, the SNV +3010CG, +3027AC, +3035CT, +3142CG and +3196CG has been associated with the risk of development of LES. The clinical manifestations of LES were also associated to the 3´UTR SNV. The analysis of haplotypes associated UTR1 and the combinations UTR1 and UTR1 + UTR1 + UTR7 to protection against development of LES. For the 5´URR, 17 SNV were associated with risk of development for any disease; SLE (-725C>G>T, -483G>A, -297G>A and -284G>A), RA (-762C>T, -725C>G>T, -540InsG a -533DelA, -509C>G, -486C>A, -483G>A and -297G>A) and the PSO (-762C>T, -725C>G>T, -716G>T, -689G>A, -646A>G, -633A>G, -509C>G, -483G>A, -443G-A, -540InsG a -533DelA, -400G>A, -391G>A, -369A>C, -355G>A and -297G>A). The clinical manifestations of LES, RA and PSO were also associated to the 5´URR SNV. Still, the 5-haplotype of 5´URR was associated with the risk of development of LES and AR. We conclude that the SNV of the 5'UR and 3'UT regions of HLA-G are associated to SLE, RA and PSO as well as to the clinical data analyzed in this study. Thus, SNVs in these regions must be related to the expression levels of the molecule and consequently, to local or systemic immunoregulation

    Análise da fermentação de glicose e xilose por leveduras Spathaspora isoladas de madeira em decomposição

O etanol de segunda geração é dependente da fermentação dos açúcares que constituem a biomassa lignocelulósica, composta por lignina, celulose e hemicelulose. A hemicelulose é composta tanto por hexoses como por pentoses. A xilose é a pentose predominante na lignocelulose, e o segundo açúcar mais abundante na natureza. A levedura Saccharomyces cerevisiae, principal microrganismo utilizado na produção de etanol de primeira geração, é incapaz de fermentar essa pentose. Por essa razão, pesquisas para identificar novas leveduras capazes de fermentar a xilose têm enorme aplicação biotecnológica. Neste trabalho foram analisadas 42 linhagens de leveduras, isoladas de madeira em decomposição na biodiversidade brasileira, como possíveis leveduras fermentadoras de xilose. Entre essas leveduras, quatro novas espécies fermentadoras de xilose foram classificadas e denominadas Spathaspora brasiliensis, Spathaspora roraimanensis, Spathaspora suhii, e Spathaspora xylofermentans. Essas diferentes leveduras Spathaspora apresentaram diferentes rendimentos na produção de etanol quando crescidas em meios contendo xilose ou glicose como fonte de carbono, quando fermentam em sistema de batelada misturas de xilose e glicose, ou hidrolisados enzimáticos de bagaço de cana-de-açúcar. Nossos resultados indicam que as espécies S. roraimanensis, S. xylofermentans e as já conhecidas S. passalidarum e S. arborariae, apresentam os melhores rendimentos na produção de etanol durante a fermentação de xilose. Finalmente, analisamos a expressão relativa de genes, presentes no genoma da levedura S. arborariae, possivelmente envolvidos na metabolização de xilose. Nossos resultados mostraram uma maior expressão relativa de 4 genes (que codificam para as enzimas xilose redutase e xilulocinase, e dois transportadores de xilose) nas células cultivadas em xilose, quando comparado com células crescidas em glicose. Estes novos genes constituem interessantes alvos para, através de engenharia genética, otimizar a produção de etanol a partir de hidrolisados da biomassa lignocelulósica.Abstract : Growing concerns with the future depletion of fossil fuels has driven the development of new technologies for the production of alternative fuels like ethanol. Second generation ethanol depends on the fermentation of sugars from lignocellulosic biomass, composed of lignin, cellulose and hemicellulose. Hemicellulose contains both hexoses and pentoses. Xylose is the predominant pentose in lignocellulose, and the second most abundant sugar in nature. The yeast Saccharomyces cerevisiae, the major microorganism used in the production of first generation ethanol, is unable to ferment this pentose. Therefore, studies aiming the identification of new yeasts capable of fermenting xylose have several biotechnological applications. In this work we analyzed 42 yeast strains isolated from decaying wood in the Brazilian biodiversity as possible xylose-fermenting yeasts. Among these yeasts, four new xylose fermenting species were classified and named Spathaspora brasiliensis, Spathaspora roraimanensis, Spathaspora suhii, and Spathaspora xylofermentans. These different Spathaspora yeasts presented different ethanol yields when grown in media containing glucose or xylose as carbon source, during batch fermentations with glucose and xylose mixtures, or with sugarcane bagasse enzymatic hydrolysates. Our results indicate that the species S. roraimanensis, S. xylofermentans and the already known S. passalidarum and S. arborariae, show the best yields in ethanol production during xylose fermentation. Finally, we determined the relative expression of genes, present in the genome of the yeast S. arborariae, possibly involved in the metabolism of xylose. Our results showed a higher relative expression of four genes (encoding the enzymes xylose reductase and xyluloquinase, and two xylose transporters) in cells grown on xylose, as compared to cells grown in glucose. These new genes are interesting targets for optimizing, through genetic engineering, the production of ethanol from lignocellulosic biomass hydrolysates

    Estudo caso-controle do gene MTHFR (regiões 677 e 1298) em pacientes com Lúpus Eritematoso Sistêmico, em Santa Catarina - Brasil

Biologia.Introdução: Lúpus Eritematoso Sistêmico (LES) é uma doença reumática inflamatória e crônica de origem autoimune. Possui a combinação de predisposição genética e ambiental que altera a expressão de genes reguladores principalmente do sistema imune. Por se tratar de uma doença complexa, vários genes estão envolvidos na sua ação. Nesse contexto genético, o papel dos genes que codificam moléculas importantes no metabolismo tem sido alvo de interesse pela importância desses processos no organismo e no desenvolvimento de doenças. Para o metabolismo, a MTHFR (5,10-Metilenetetrahidrofolato Redutase) é uma importante enzima que está envolvida em processos de metilação e síntese de DNA e de vias metabólicas do ácido fólico (vitamina B9). Possui polimorfismos de nucleotídeo único (SNP) em diversas partes do gene MTHFR, porém os mais importantes são na região C677T [rs1801133] e A1298C [rs1801131] que diminuem a atividade dessa enzima. Nos pacientes com LES, sabe-se que as células dos leucócitos mononucleares exibem várias alterações. Com isso, ocorre a produção de citocinas anormais, diminuição da função citotóxica e aumento da resposta humoral. Portanto, variações genéticas das enzimas da via folato, que atuam em conjunto para doação de metila na região reguladora do DNA podem causar o mau funcionamento dessas células. Vários estudos comprovam que uma super expressão anormal de citocinas, no qual ocorre um aumento de autoanticorpos, está relacionada a uma baixa metilação em regiões regulatórias do DNA em células T. Objetivo: Investigar a associação do SNP do gene MTHFR (C677T e A1298C) a genótipos de risco/proteção para o desenvolvimento da doença através de um estudo casocontrole em pacientes de Santa Catarina - Brasil. Resultados: a análise dos genótipos e das frequências alélicas por PCR-RFLP para a região C677T encontra-se em Equilíbrio de Hardy- Weinberg (EHW). Porém, para a região A1298C o grupo de casos está fora do equilíbrio (p= 0,0013) e grupo dos controles encontra-se em EHW. Não foi encontrada nenhuma associação significativa entre casos e controles quando calculado o odds-ratio considerando cada alelo e cada genótipo como fator de risco/proteção para o desenvolvimento de LES na região MTHFR 677. Porém, demonstrou-se que a presença do alelo A na região 1298 do gene MTHFR aumenta em 1,618 vezes a chance de desenvolver LES (OR = 1,618; 95% IC: 1,097 – 2,388; p = 0,014), sendo que o alelo C atua como fator de proteção (OR = 0,618; 95% IC: 0,419-0,912; p = 0,014). Análises de odds-ratio foram feitas para oito manifestações clínicas da doença (Artrite, Fotossensibilidade, Eritema malar, Úlcera oral, Nefrite, Anemia, Alterações neurológicas, Serosite). Sendo que duas manifestações foram associadas com a região A1298C a de Fotossensibilidade em que o alelo A atua como fator de risco (OR = 1,947; 95% IC: 1,026 – 3,701; p = 0,041) e o alelo C como fator de proteção (OR = 0,513; 95% IC: 0,270 – 0,975; p = 0,041) e Úlcera oral em que o alelo C atua como fator de risco (OR = 1,944; 95% IC: 1,073 – 3,528; p = 0,027) e o alelo A como fator de proteção (OR = 0,514; 95% IC: 0,283 – 0,932; p = 0,027). Conclusão: Os resultados mostram uma relação dos polimorfismos do gene MTHFR com LES para pacientes do Estado de Santa Catarina - Brasil para a região A1298C, tanto na comparação estatística de Caso-Controle quanto nas manifestações clínicas para fotossensibilidade e úlcera oral.Introduction: Systemic lupus erythematosus (SLE) is an inflammatory and chronic rheumatic disease of autoimmune origin. It has the combination of genetic and environmental predisposition that alters the expression of metabolic regulatory genes. Because it is a complex disease, several genes are involved in its etiology. In this genetic context, the role of genes encoding important molecules in metabolism has been of interest in the importance of these processes in the body and in the development of diseases. For metabolism, MTHFR (5,10- Methylenetetrahydrofolate Reductase) is an important enzyme that is involved in the methylation and synthesis of DNA and metabolic pathways of folic acid (vitamin B9). It has single nucleotide polymorphisms (SNP) in several parts of the MTHFR gene, but the most important are in the C677T [rs1801133] and A1298C [rs1801131] regions that decrease the activity of this enzyme. In SLE patients, it is known that mononuclear leukocyte cells exhibit various aberrations. This leads to the production of abnormal cytokines, decreased cytotoxic function and increased humoral response. Therefore, genetic variations of the folate pathway enzymes that act together for methyl donation in the DNA regulatory region may cause malfunction of these cells. Several studies have shown that an abnormal cytokine superexpression, with increased autoantibodies production, is related to low methylation in regulatory regions of T-cell DNA. Objective: To investigate the association of SNPs in the MTHFR gene (C677T and A1298C) to genotypes of risk / protection for the development of systemic lupus erythematosus, through a case-control study in patients from Santa Catarina - Brazil. Results: analysis of genotypes and allele frequencies by PCR-RFLP for the C677T region showed a Hardy-Weinberg Equilibrium (HWE). However, for the A1298C region the control group is in HWE, but the case group is out of balance (p = 0.0013). No significant association was found between cases and controls and SLE when calculating the odds ratio considering each allele and each genotype as a risk / protective factor for the development of SLE in the MTHFR 677 region. However, the MTHFR 1298 region indicated that individuals with the A allele were more likely to develop the disease (OR = 1.68; 95% CI: 1.097-2.388; p = 0.014). Allele C acts as a protective factor (OR = 0.618, 95% CI: 0.419-0.912, p = 0.014). Odds ratios were also calculated for eight clinical manifestations of the disease (Arthritis, Photosensitivity, Eritema malar, Oral ulcer, Nephritis, Anemia, Neurological disorders, Serositis). In the A1298C region, two manifestations showed association: photosensitivity in which the A allele acts as a risk factor (OR = 1.947, 95% CI: 1.026 - 3.701, p = 0.041) and the C allele as a protection factor (OR = 0.513; 95% IC: 0.270 – 0.975; p = 0.041) and Oral ulcer in which the allele C was a risk factor OR = 1.944; 95% IC: 1.073 – 3.528; p = 0.027) and the A allele acts as a protection factor (OR = 0.514; 95% IC: 0.283 – 0.932; p = 0.027). Conclusion: The results indicate an association of the polymorphisms A1298C in the MTHFR gene with SLE in patients from the State of Santa Catarina - Brazil, as well as with the specific clinical manifestations photosensitivity and oral ulcers

    Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Ciências Biológicas, Programa de Pós-Graduação em Biologia Celular e do Desenvolvimento, Florianópolis, 2019O câncer de colo de útero (CCU), também conhecido como câncer cervical, é o terceiro tipo mais frequente na população feminina e a segunda causa de mortalidade de mulheres no mundo. O sistema imune tem um papel essencial no controle de neoplasias, que se desenvolvem através do mecanismo de escape tumoral. Um gene imunorregulador com papel nesse mecanismo é o HLA-G. Esse gene possui sítios de variação (SNV) em regiões regulatórias, 5´URR e 3´UTR. Além disso, apresenta variações em sua região codificadora, como a deleção que gera o alelo nulo G*0105N e impede a expressão de algumas isoformas proteicas. Essas SNV podem influenciar na expressão do gene. O presente estudo teve como principal objetivo avaliar associações de 20 SNV da 5?URR e 10 da 3?UTR, assim como a presença do alelo G*0105N,em mulheres que apresentavam lesão de colo de útero de alto grau (LCU) (n=28) e CCU (n=32), contrastados com dois grupos controle, mulheres sem a presença de LCU (SLU) (n=52) e um grupo populacional de mulheres sem evidência da doença (POP) (n=253), visando compreender o papel dessas SNV do HLA-G no desenvolvimento de LCU e de CCU. Após análises das sequências, foram realizadas cálculos de associação. Dez sítios da 5?URR apresentaram associação com a presença de LCU ou CCU, sendo eles: -725G>C>T, -689A>G, -646A>G, -509C>G, -477C>G, -443G>A, -400G>A, -391G>A, -369C>A e -284G>A. Os haplótipos 5'URR-6 e 5'URR-8 e as combinações haplotípicas 5'URR-1 + 5'URR-6, 5'URR-1 + 5'URR-9, 5'URR-4+5'URR-5, 5'URR-5+5'URR-7, 5'URR-5+5'URR-8, 5'URR-6+5'URR-6, 5'URR-6+5'URR-9, 5'URR-7+5'URR-8 e 5'URR-9+5'URR-9 foram associados ao risco de desenvolvimento de CCU. A combinação haplotípica 5'URR-1+5'URR-1 foi associada ao menor risco no desenvolvimento da doença. Para a 3?UTR, os sítios 14pbDel/In, +3001T/C, +3027A/C e +3196G/C apresentaram associação com suscetibilidade ou proteção ao CCU. O haplótipo UTR-1 foi associado ao menor risco para o desenvolvimento da doença e o UTR-2 como fator de risco para o CCU. O G*0105N foi detectado em três mulheres do grupo caso (CCU=2; LCU=1), sendo todas heterozigotas.Concluímos que há SNV de regiões regulatórias do HLA-G associadas ao maior ou menor risco para o desenvolvimento LCU e CCU. Assim, estudos de associação genética são importantes para identificação de SNV para futuros estudos funcionais.Abstract: Cervical cancer (CCU), also known as cervical cancer, is the third most frequent type in the female population and the second leading cause of death among women worldwide.The immune system plays an essential role in the control of neoplasias, which develop through the tumor escape mechanism.An immunoregulatory gene with role in this mechanism is HLA-G. This gene has variation sites (SNV) in regulatory regions, 5'URR and 3'UTR.In addition, there are variations in its coding region, such as the deletion that generates the null allele G * 0105N and prevents the expression of some protein isoforms.These SNVs may influence gene expression. The aim of the present study was to evaluate the associations of 20 SNV of 5'URR and 10 of 3'UTR, as well as the presence of the G * 0105N allele in women with high grade uterine cervix (LCU) lesion (n = 28) and CCU (n = 32), in contrast to two control groups, women without LCU (n = 52) and a population group of women with no evidence of disease (POP) (n = 253) aiming to understand the role of these HLA-G SNV in the development of LCU and CCU.After analysis of the sequences, association calculations were performed. Ten sites of 5'URR were associated with the presence of LCU or CCU, being: -725G> C> T, -689A> G, -646A> G, -509C> G, -477C> G, -443G> A , -400G> A, -391G> A, -369C> A and -284G. The haplotypes 5'URR-6 and 5'URR-8 and the haplotype combinations 5'URR-1 + 5'URR-6, 5'URR-1 + 5'URR-9, 5'URR-4 + 5'URR -5, 5'HRR-5 + 5'RHR-7, 5'RHR-5 + 5'RHR-8, 5'RHR-6 + 5'RHR-6, 5'RHR-6 + 5'RHR-9 , 5'URR-7 + 5'URR-8 and 5'URR-9 + 5'URR-9 were associated with the risk of developing CCU. The haplotypic combination 5'URR-1 + 5'URR-1 was associated with lower risk of disease development. For 3'UTR, the 14pbDel / In, + 3001T / C, + 3027A / C and + 3196G / C sites were associated with susceptibility or protection to CCU. The UTR-1 haplotype was associated with the lowest risk for the development of the disease and UTR-2 as a risk factor for CCU. G * 0105N was detected in three women in the case group (CCU = 2; LCU = 1), all of which were heterozygous. We conclude that there are SNV of HLA-G regulatory regions associated with greater or lesser risk of developing LCU and CCU. Thus, genetic association studies are important for the identification of SNV for future functional studies

    Efeitos do carotenoide fucoxantina na distribuição de componentes da matriz extracelular e da glicoproteína-P em células de glioblastoma multiforme humano

    Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Ciências Biológicas, Programa de Pós-Graduação em Biologia Celular e do Desenvolvimento, Florianópolis, 2022.Gliomas são tumores do sistema nervoso central (SNC) com origem nas células gliais ou em suas progenitoras e correspondem a aproximadamente 50% dos tumores primários do SNC. O glioblastoma multiforme (GBM) é o tipo de glioma mais comum e mais agressivo e sua terapia padrão é a ressecção cirúrgica seguida de rádio e quimioterapia. A temozolomida (TMZ) é o principal quimioterápico utilizado no combate ao GBM, mas apesar dos avanços nos tratamentos, a sobrevida dos pacientes continua abaixo dos 15 meses. Esse prognóstico ruim pode ser devido à presença de células-tronco tumorais (CSC) e à alta taxa de invasão do GBM, que estão envolvidas na resistência tumoral. Alterações em fibronectina (FN), tenascina-C (TNC) e glicoproteína-P (P-gp) podem estar relacionadas com a invasão tumoral e com características de células-tronco. A fucoxantina (Fx) é um composto natural que já demonstrou efeitos anticâncer, inclusive antimigratórios, se mostrando uma substância interessante para um futuro uso clínico. A distribuição e organização de FN, TNC e P-gp em células de GBM (GBM1) após tratamento com Fx 100 µM foi analisada através de imunocitoquímica. A Fx diminuiu a marcação de FN, enquanto aumentou a de TNC e de P-gp após 24 horas de tratamento. O efeito em P-gp pode ser visto após 3 e 6 horas. A superexpressão de TNC e de P-gp em tumores está associada com presença de células-tronco e piores prognósticos. Os resultados sugerem que a Fx pode estar promovendo a desdiferenciação das células GBM1.Abstract: Gliomas are central nervous system (CNS) tumors originating from glial cells or their progenitors and account for approximately 50% of primary CNS tumors. Glioblastoma multiforme (GBM) is the most common and most aggressive type of glioma and its standard therapy is surgical resection followed by radiotherapy and chemotherapy. Temozolomide (TMZ) is the main chemotherapeutic used in GBM treatment, but despite advances in treatments, patient survival remains below 15 months. This poor prognosis may be due to the presence of tumor stem cells (CSC) and the high rate of GBM invasion, which are involved in tumor resistance. Changes in fibronectin (FN), tenascin-C (TNC) and P-glycoprotein (P-gp) may be related to tumor invasion and stem cell characteristics. Fucoxanthin (Fx) is a natural compound that has already demonstrated anticancer, including antimigratory, effects, proving to be an interesting substance for future clinical use. The distribution and organization of FN, TNC and P-gp in GBM cells (GBM1) after treatment with Fx 100 µM was analyzed by immunocytochemistry. Fx decreased FN labeling, while TNC and P-gp increased after 24 hours of treatment. The effect on P-gp can be seen after 3 and 6 hours. Overexpression of TNC and P-gp in tumors is associated with the presence of stem cells and worse prognosis. The results suggest that Fx may be promoting the dedifferentiation of GBM1 cells

This chromosome is specific for studies of patrilineal ancestry. Because it is a chromosome that 95% of its composition does not undergo recombination, one of the few ways of it to suffer variations is trough bialectic mutations of a single nucleotide polymorphism (SNPs). The SNPs mutation rate is realy small, so they can be used as good markers for ancestry. Ancestry studies with SNPs for chromosome Y (Y-SNPs) collaborate for the construction of the phylogeny of the chromosome and human migration patterns. Thanks to the numerous studies in the anthropological genetics field, its already possible to know the genetic patterns of the populations and “track” the evolutionary rout that the human being did. For us to know where we came from this patterns were separated in haplogroups. An aplogroup is the genetic population of people that share a common patrilinear or matrilinear ancestor, this groups receive alphabet letters for identification, and the refinements consists in combinations of numbers and additional letters. With this studies, combined with colonization and migration ones, it’s expected that there will be a certain frequency of specific haplogroups The aim of this study was to standardize multiplex with Y-SNPs of the haplogroups R1, Q, R, F, J2 and K, relevant to the population of the State of Santa Catarina. Polymerase Chain Reaction (PCR) and 10% polyacrylamide gel electrophoresis techniques were used to reach these objectives, focusing mainly on the standardization of the PCR technique for the primers used, resulting in a large number of protocols tested, each Which with its intention of use and technical direction. The standardization of multiplex markers of these specific groups is a basic work of extreme relevance for future studies aimed at a better understanding of who we are, where we come from and where we are in the phylogenetic tree

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
