    New targets for therapy in polyglutamine (polyQ) expansion diseases

    The polyglutamine (polyQ) repeat disorders are a family of inherited disorders characterized by progressive neurodegeneration, as well as the formation of intracellular protein aggregates. Huntington's disease (HD) is the most prevalent disorder in the family of polyQ diseases. This family includes nine other neurodegenerative disorders: Dentatorubral- pallidoluysian atrophy (DRPLA), Bulbo-Spinal Muscular Atrophy (BSMA) and Spinocerebellar ataxia (SCA) types 1-3, 6, 7, 12 and 17. Each disease is caused by the expansion of a tract of repeated CAG triplet in a distinct gene, causing transcription of proteins with lengthened polyQ repeats. Although mutations occur frequently in a ubiquitously expressed gene, neurodegeneration occurs in a specific cell type. The mutant proteins involved in polyQ disease are unrelated and they share only the glutamine extension. As for the pathogenic mechanism by wich the repeat expansion leads to the disease, major models include a loss of function of the gene and a gain of function by the mutant RNA transcript or protein product. Indeed, altered function in the ubiquitin-proteasome system (UPS), endoplasmic reticulum (ER) and mitochondria function, as well as a primary failure in autophagy has been demonstrated. However, the pathogenic mechanism varies from one disease to another, depending on the motif, length, and intragenic location of the repeat. Understanding the molecular mechanisms of repeat instability and pathogenic process is not only of scientific interest but also essential for the development of rational treatment of these diseases

    Through the analysis of some significant passages of Athanasius' apology «Against the Pagans on the Incarnation of the Word», and also thanks to their comparison with other selected texts by the same author, P. F. Beatrice tries to date this still problematic work to around the middle of the fourth century. In particular, he intends to show that Athanasius did know the Neoplatonic philosophy of his time sufficiently well and that the target of his polemic was especially Porphyry. But anti-pagan criticism also supplies Athanasius with a substantial support in his struggle with the Arian heresy which had been shared by his great precursor and adversary Eusebius of Caesarea

    In the field of linguistic studies on Italian song lyrics, the genre of progressive rock remains one of the most marginal and least appreciated from the point of view of linguistic innovations. Furthermore, the subalternity that is generally recognized in textual composition compared to musical composition (which involves limitations that devalue the literary component of the text) is considered particularly marked in the manifestations of art music, like prog is. Through a linguistic-literary analysis of the texts contained in the first two albums of one of the most representative bands of the genre, Banco del Mutuo Soccorso, the discussion aims to highlight the themes and stylistic features that characterize the language of the authorial couple Francesco Di Giacomo-Vittorio Nocenzi, in order to underline its literariness and originality and define its poetics.Nel panorama degli studi linguistici sulla canzone italiana, il genere del rock progressive resta uno dei più marginali e meno apprezzati dal punto di vista delle innovazioni linguistiche. Inoltre, la subalternità che genericamente si riconosce al testo rispetto alla musica (e che comporta limitazioni che ne svalutano la componente letteraria) è considerata particolarmente spiccata nelle manifestazioni della musica colta come il prog.     Nel quadro trasformativo e di aggiornamento infrastrutturale che sta attraversando la città di Genova da almeno due decenni, l’episodio del Parco Scientifico e Tecnologico di Erzelli rappresenta una sorta di unicum per la posizione geografica, per il programma funzionale e il processo attuativo e gli attori coinvolti. Posizionato sulla collina omonima che domina la Val Polcevera sul fronte occidentale e in posizione strategica rispetto alle principali infrastrutture ferroviarie e carrabile del Ponente genovese, il Parco risponde ad un’esigenza di delocalizzazione e, in un certo senso, di aggregazione territoriale di grandi attività legate alla tecnologia, alla produzione, al terziario e alla ricerca scientifica. In questo processo di riposizionamento, la realizzazione del Parco affronta criticità legate, in primis, all’accessibilità e conseguentemente all’attrattività dei territori, declinando il tema degli epicentri di sviluppo in chiave tecnologica, produttiva e terziaria. Il Parco diviene inoltre rappresentativo della graduale transizione della Val Polcevera da valle industriale “pesante” a valle produttiva “leggera e pulita”. Obiettivo del contributo è, anche in questo episodio di realizzazione di un importante epicentro di sviluppo urbano e territoriale, quello di approfondire il ruolo dell’amministrazione pubblica, osservando il progressivo passaggio dalla dimensione del regolatore/pianificatore, a quello di investitore per finire a quello sempre più rilevante di proprietario/possessore

    Auf dem Hintergrund der seit mehreren Jahren anhaltenden Flüchtlingskrise wird in diesem Beitrag korpusbasiert, qualitativ und kontrastiv untersucht, wie in den Massenmedien in Deutschland und Italien, insbesondere in der politischen Berichterstattung, das komplexe Thema der Migration und der damit zusammenhängenden aktuellen Flüchtlingskrise metaphorisch konzeptualisiert wird. Ziel ist herauszustellen, ob sich im sprachlichen Repertoire politisch meinungsbildender Periodika bereits eine Ikonografie bestimmter migrationsproblematischer Figuren herausgebildet hat, welchen semantischen Bereichen sich diese zuordnen lässt und was sie über die Migrationsprozesse aussagt. Die kontrastive Herangehensweise soll außerdem herausstellen, ob die Wirtschaftskrise, die in Italien größere Ausmaße annimmt als in Deutschland, eventuell auch eine unterschiedliche Wahrnehmung der Migrationsproblematik bedingt

    Bioactive Phenolic Compounds in the Modulation of Central and Peripheral Nervous System Cancers: Facts and Misdeeds.

    Efficacious therapies are not available for the cure of both gliomas and glioneuronal tumors, which represent the most numerous and heterogeneous primary cancers of the central nervous system (CNS), and for neoplasms of the peripheral nervous system (PNS), which can be divided into benign tumors, mainly represented by schwannomas and neurofibromas, and malignant tumors of the peripheral nerve sheath (MPNST). Increased cellular oxidative stress and other metabolic aspects have been reported as potential etiologies in the nervous system tumors. Thus polyphenols have been tested as effective natural compounds likely useful for the prevention and therapy of this group of neoplasms, because of their antioxidant and anti-inflammatory activity. However, polyphenols show poor intestinal absorption due to individual intestinal microbiota content, poor bioavailability, and difficulty in passing the blood–brain barrier (BBB). Recently, polymeric nanoparticle-based polyphenol delivery improved their gastrointestinal absorption, their bioavailability, and entry into defined target organs. Herein, we summarize recent findings about the primary polyphenols employed for nervous system tumor prevention and treatment. We describe the limitations of their application in clinical practice and the new strategies aimed at enhancing their bioavailability and targeted delivery. © 2020 by the authors
