    Low-dose radiations derived from cone-beam CT induce transient DNA damage and persistent inflammatory reactions in stem cells from deciduous teeth

    OBJECTIVES: Cone-beam CT (CBCT), a radiographic tool for diagnosis, treatment, and follow-up in dental practice, was introduced also in pediatric radiology, especially orthodontics. Such patients subjected to repetitive X-rays examinations may receive substantial levels of radiation doses. Ionizing radiation (IR), a recognized carcinogenic factor causing DNA double-strand breaks (DSBs) could be harmful to undifferentiated cells such as dental pulp stem cells (DPSCs) since inaccurately repaired or unrepaired DSBs may lead to malignant transformation. The H2AX and MRE11 proteins generated following DSBs formation and pro-inflammatory cytokines (CKs) secreted after irradiation are relevant candidates to monitor the cellular responses induced by CBCT. METHODS: DPSCs were extracted from human exfoliated deciduous teeth and their phenotype was assessed by immunocytochemistry and flow-cytometry. Cells were exposed to IR doses: 5.4-107.7 mGy, corresponding to 0.5-8 consecutive skull exposures, respectively. H2AX and MRE11 were detected in whole cells, while IL-1α, IL-6, IL-8, TNFα in supernatants, using enzyme-linked immunosorbent assay (ELISA) at different time points after exposure. RESULTS: The phosphorylation level of H2AX in DPSCs increased considerably at 0.5 h after exposure (p < 0.001 for 3, 5, 8 skull exposures and p < 0.05 for 1 skull exposure, respectively). MRE11 response could only be detected for the highest IR dose (p < 0.001) in the same interval. CKs secretion increased upon CBCT exposure according to doses and time. Programa de Pós-Graduação em Letras/Inglês e Literatura CorrespondenteThis dissertation analyzes the reformulations of Brazilian gaucho identity in three contemporary Brazilian productions that retell the history of Farrapos War: the films Anahy de las Misiones (1997) and Netto Perde sua Alma (2001), as well as the television series A Casa das Sete Mulheres (2003). The importance of historical fiction in the discussion of cultural identities becomes clear in Hall#s description of identity as our continuous repositioning in relation to the narratives of the past. The revision of history in the films and the series is investigated in relation to the diverse views of postmodern fiction in the works of Fredric Jameson and Linda Hutcheon. The analysis of the television series also takes into account the different positions about the contemporary impulse of nostalgia theorized by Andrew Higson and Andreas Huyssen. Finally, the analyses draw on Àngel Rama#s concept of transculturation, as the revision of regional content in the light of a new proposal to compose a renewed heritage that can be identified with the past, for the examination of cultural identities. Based on the readings in this study, the productions discussed are regarded as loci of intercultural dialogue; and the reformulations of Brazilian gaucho identity in those productions are understood as transcultural onesEsta dissertação analisa as reformulações da identidade do gaúcho brasileiro em três produções contemporâneas que recontam a história da Guerra dos Farrapos: os filmes Anahy de las Misiones (1997) e Netto Perde sua Alma (2001), e a série de televisão A Casa das Sete Mulheres (2003). A importância da ficção histórica para a discussão das identidades culturais fica clara na descrição que Hall faz de identidade como o contínuo reposicionamento em relação às narrativas do passado. A revisão da história nestas obras é investigada em relação às discrepantes visões sobre a ficção pós-moderna no trabalho de Fredric Jameson e Linda Hutcheon. A análise da série televisiva também leva em conta o impulso nostálgico na contemporaneidade, discutido por Andrew Higson e Andreas Huyssen. Finalmente, as análises lançam mão do conceito de transculturação de Ángel Rama: a revisão de conteúdos regionais à luz de novas propostas, compondo uma nova herança cultural que ainda pode se identificar com o passado.     Mesenchymal stem cell- derived exosomes as cell-free therapeutics for sensorineural hearing loss

    Sensorineural hearing loss (SNHL) can result from various factors, including ototoxic drugs (such as aminoglycosides and chemotherapeutic agents), prolonged exposure to intense sound, and autoimmune or genetic disorders. In adult mammals, the loss of sensory cells in the cochlea is irreversible due to their lack of regenerative capacity. Current treatment options include hearing aids for mild to moderate hearing loss, which rely on residual hearing, and cochlear implants for severe cases, which provide limited auditory recovery while leading to the loss of any remaining natural hearing. Stem cell therapies, particularly those involving mesenchymal stem cells (MSCs), are being increasingly explored in regenerative medicine. MSCs are multipotent cells capable of differentiating into mesodermal lineage cells and possess immunomodulatory and regenerative properties, making them potential candidates for SNHL treatment. However, their administration carries risks, including unwanted differentiation, immune system activation, and potential tumorigenic effects. Exosomes, extracellular vesicles in the nanometer size range, are secreted by most eukaryotic cells. These vesicles, which have a double lipid membrane and contain genomic and proteomic material, play a crucial role in intercellular communication. Exosomes derived from MSCs exhibit similar biological functions to their parent cells but with significantly lower risks, as they do not trigger immune responses or pose oncological concerns. This paper aims to review current knowledge on the use of MSCs and MSC-derived exosomes for inner ear sensory cell regeneration and explore their potential for clinical applications

Para tanto, o estudo buscou compreender que, a problemática da evasão escolar e da prática do ato infracional devem ser analisadas como sendo resultantes de um contexto complexo e dinâmico, o qual tem corno base o sistema capitalista. Assim, o trabalho tem como objetivo geral identificar a relação entre a evasão escolar e o envolvimento com a prática do ato infracional, dos adolescentes participantes do PACSJ, realizado em uma unidade de ensino, de um município da Grande Florianópolis. Para isso foram realizadas pesquisas bibliográficas, documentais e a campo. Os dados encontrados apontam que direitos regulamentados são também direitos não efetivados. Relacionam-se com o agravamento da problemática da pobreza, o aumento da exclusão social, ausência do trabalho formal, mudanças de valores. Neste contexto, as políticas sociais, em especial as educacionais, não são eficazes para inserir e manter essa população na escola, como uma alternativa para o não envolvimento com o ato infraciona

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Biosynthesis of Silver Nanoparticles Using Ligustrum Ovalifolium Fruits and Their Cytotoxic Effects

    The present study reports for the first time the efficacy of bioactive compounds from Ligustrum ovalifolium L. fruit extract as reducing and capping agents of silver nanoparticles (AgNPs), developing a green, zero energetic, cost effective and simple synthesis method of AgNPs. The obtained nanoparticles were characterized by UV-Vis spectroscopy, transmission electron microscopy (TEM), X-ray diffraction (XRD) and Fourier Transform Infrared spectroscopy (FTIR), confirming that nanoparticles were crystalline in nature, spherical in shape, with an average size of 7 nm. The FTIR spectroscopy analysis demonstrated that the AgNPs were capped and stabilized by bioactive molecules from the fruit extract. The cytotoxicity of the biosynthesized AgNPs was in vitro evaluated against ovarian carcinoma cells and there were found to be effective at low concentration levels

Está constituído a partir de uma visão crítica da História, o que permite que ?outras imagens?, fragmentárias e não monumentais, também tenham espaço. Em diálogo com o princípio da montagem, este esboço apresenta-se em duas partes. Na primeira, Imagens possíveis, estão citadas as imagens elaboradas em vida e post mortem acerca do austríaco-brasileiro que nasceu em Viena em 1900, se exilou no Brasil em 1939 e morreu no Rio de Janeiro, em 1978. Na segunda, Montagens possíveis, apresentam-se duas possibilidades de exercício biográfico: pela leitura alegórica do documentário O velho e o Novo (Otto Maria Carpeaux), entendido como instrumento de intervenção no contexto ditatorial brasileiro e de uma reelaboração biográfica concernentes às suas experiências europeias; e pelo Caderno de imagens críticas, registro dos encontros em Carpeaux pelo meio de imagens críticas produzidas a partir da cesura do presente.Abstract : This biographical sketch attempts to quote some images of Otto Maria Carpeaux: various types of biographical constructions, carried out in different contexts by disparate authors under conditions just as distinct. It stems from a critical view of history, allowing for ?other images? fragmented and non-monumental ? to share the space.In dialogue with the montage principle, this sketch has two parts. The first, Possible Images, quotes the images produced during and after the life of the Austrian-Brazilian, who was born in Vienna in 1900, went to Brazil in exile in 1939 and died in Rio de Janeiro in 1978. The second part, Possible Montages, presents two possibilities of a biographical exercise: through the allegorical reading of documentary O Velho e o Novo (Otto Maria Carpeaux), understood as an instrument of intervention in the Brazilian dictatorship context and as a biographical retelling of the author?s European experiences; and through my Scrapbook of Critical Images, a record of the encounters in Carpeaux through critical images produced from the caesura of the present
