Differential stability of 2'F-ANA*RNA and ANA*RNA hybrid duplexes: roles of structure, pseudohydrogen bonding, hydration, ion uptake and flexibility
No full text14 pags., 7 figs., 3 tabs.Hybrids of RNA with arabinonucleic acids 2′F-ANA and ANA have very similar structures but strikingly different thermal stabilities. We now present a thorough study combining NMR and other biophysical methods together with state-of-the-art theoretical calculations on a fully modified 10-mer hybrid duplex. Comparison between the solution structure of 2′F-ANA•RNA and ANA•RNA hybrids indicates that the increased binding affinity of 2′F-ANA is related to several subtle differences, most importantly a favorable pseudohydrogen bond (2′F-purine H8) which contrasts with unfavorable 2′-OH-nucleobase steric interactions in the case of ANA. While both 2′F-ANA and ANA strands maintained conformations in the southern/eastern sugar pucker range, the 2′F-ANA strand's structure was more compatible with the A-like structure of a hybrid duplex. No dramatic differences are found in terms of relative hydration for the two hybrids, but the ANA•RNA duplex showed lower uptake of counterions than its 2′F-ANA•RNA counterpart. Finally, while the two hybrid duplexes are of similar rigidities, 2′F-ANA single strands may be more suitably preorganized for duplex formation. Thus the dramatically increased stability of 2′F-ANA•RNA and ANA•RNA duplexes is caused by differencesin at least four areas, of which structure and pseudohydrogen bonding are the most important. © The Author(s) 2010. No full textO presente relatório de estágio elaborado no âmbito da unidade curricular
de Prática Educativa Supervisionada (PES), inserida no Mestrado em Educação
Pré-Escolar e Ensino do 1º Ciclo do Ensino Básico, tem como objetivo explanar
as práticas educativas desenvolvidas em ambos os contextos de estágio.
O mesmo encontra-se organizado em três capítulos, sendo que o primeiro
apresenta os referenciais teóricos e legais estudados e aprofundados ao longo
do referido ciclo de estudos que, por sua vez, sustentaram as práticas
educativas desenvolvidas. Assim, de forma a que se compreendam as práticas
educativas explanadas e refletidas ao longo do terceiro capítulo, apresentamse
as características do contexto, bem como de ambos os grupos onde se
desenvolveu a PES. Importa, ainda, referir que a aproximação à metodologia de
investigação-ação esteve por base do processo, almejando um
desenvolvimento praxiológico pelo contributo das etapas de observação,
planificação, ação, avaliação e reflexão. Este ciclo dinâmico e em espiral
fomentou a construção da identidade profissional, desenvolvendo
competências basilares para uma ação consciente, sustentada em processos
reflexivos, tendo em vista a construção de uma prática educativa apoiada em
paradigmas socioconstrutivistas.
Em suma, importa referir que as ações desenvolvidas em ambos os
contextos, bem como o apoio das supervisoras institucionais e orientadoras
cooperantes, foram essenciais para a construção de novos saberes que, desta
forma, se revelaram fundamentais para o desenvolvimento do perfil duplo de
docência.This internship report prepared within the scope of the Supervised
Educational Practice (PES) course, inserted in the Master's Degree in Preschool
Education and Primary Education, aims to explain the educational practices
developed in both internship contexts. .
It is organized into three chapters, the first of which presents the theoretical
and legal references studied and deepened throughout the study cycle, which,
in turn, supported the educational practices developed. Thus, in order to
understand the educational practices explained and reflected throughout the
third chapter, we present the characteristics of the context, as well as of both
groups where the ESP was developed. It is also important to mention that the
approach to the action research methodology was based on the process, aiming
at a praxiological development through the contribution of the observation,
planning, action, evaluation and reflection stages. This dynamic and spiral cycle
has fostered the construction of professional identity, developing basic skills for
conscious action, sustained in reflective processes, with a view to building an
educational practice based on socio-constructivist paradigms.
In short, it should be noted that the actions developed in both contexts, as
well as the support of cooperative institutional supervisors and advisors, were
essential for the construction of new knowledge that, thus, proved
fundamental for the development of the dual teaching profile
Estudo comparativo de homens e mulheres em liberdade condicional e da influência do género nas decisões judiciais
No full textAinda hoje nos deparamos com diferenças acentuadas ao nível do género, sendo a sua influência na decisão judicial um dos contextos mais abordados, verificando-se um número mais elevado de estudos referente à decisão sentencial, em comparação à concessão da liberdade condicional. Procurando observar as diferenças entre homens e mulheres ao nível da decisão judicial, foram analisadas 140 decisões de indivíduos que iniciaram a liberdade condicional com acompanhamento da Direção Geral de Reinserção e Serviços Prisionais (DGRSP). Apesar do género não se ter constituído como preditor significativo da duração da pena ou da fase de concessão da liberdade condicional, foi possível apurar diferenças entre homens e mulheres, nomeadamente, o tipo de crime que levou à condenação e a presença e especialização de antecedentes criminais. Como preditores da duração da pena, verificou-se o número de crimes cometidos no presente, para ambos os grupos, e a especialização dos antecedentes para os homens. Relativamente à concessão da liberdade condicional, não foram encontradas diferenças significativas.Nowadays we still face marked differences at gender level. Its influence on court decision is one of the most discussed contexts, where more studies have focused on sentencing decisions when compared with the granting of parole. Attempting to observe the differences between men and women at judicial decision level, 140 decisions about individuals that started parole with the supervision of the Direção Geral de Reinserção e Serviços Prisionais (DGRSP), were analyzed. Although gender was not found to be a significant predictor of sentence duration or parole granting phase, it was possible to identify differences between men and women, namely the crime type that led to conviction and the presence and specialization of criminal records. As predictors of sentence duration, the number of crimes committed was significant for both groups, as well as the criminal record specialization for men. Regarding granting parole, no differences were found between male and female parolees
Content curation in the archival group of the Círculo Operário da Bahia at the Memorial Santa Dulce dos Pobres
No full textThis study presents a proposal for content curation applied to permanent archives, with emphasis on the documentation belonging to the archival collection of the Círculo Operário da Bahia, under the custody of the Memorial Santa Dulce dos Pobres, located in Salvador, Bahia, which constitutes the object of this research. In this context, content curation is conceived as a strategic practice aimed not only at preservation, but also at the planned and selective dissemination of archival information, thereby promoting access, enhancing the value of documentary heritage, and democratizing historical knowledge. The objective of the research is to implement content curation as an archival practice to effectively disseminate information within the Memorial Santa Dulce dos Pobres archive. This is a descriptive and documentary study with a qualitative approach. The chosen methodology is a case study, using as its corpus the minute books of general assembly meetings from 1941 to 1987. The analysis was conducted through the application of archival description standards, combined with the principles of Archival Knowledge Organization, in order to identify the context of document production, the actors involved, and the possibilities for documentary mediation through curation. The results indicate a need for further studies on content curation within the field of Information Science, highlighting the expanded role of the archivist as both curator and documentary mediator of collective memory. The study also underscores the importance of archival instruments in consolidating the post-custodial paradigm and legitimizing information sources in historical contexts. These findings reinforce the need to adopt contemporary dissemination strategies and demonstrate the potential of curation to make archival collections more accessible and meaningful to society. In conclusion, it is emphasized that archival documents are symbolic constructions that express—and, if left unaddressed, may perpetuate—only a single worldview. The guidelines for content curation as an archival practice were aligned with the proposed flowchart model, encompassing the stages of source retrieval, selection and analysis, value aggregation, and sharing. Archival description served as the normative framework at all stages, respecting provenance and Milton Santos’ concept of territoriality for narrative contextualization, thereby enhancing the collection’s educational and heritage-related actions. Furthermore, it is essential that the evaluation of the entire curatorial process take the user into account, fostering the relevance of the content and the value added to his or her experience.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior – Brasil (CAPES)Este trabalho contempla uma proposta de curadoria de conteúdo aplicada a arquivos permanentes, com ênfase na documentação pertencente ao fundo arquivístico do Círculo Operário da Bahia, sob a guarda do Memorial Santa Dulce dos Pobres, localizado no município de Salvador, Bahia, objeto de estudo desta pesquisa. A curadoria de conteúdo, nesta pesquisa, configura-se como uma prática estratégica que visa não apenas a preservação, mas a difusão planejada e seletiva da informação arquivística, promovendo o acesso, a valorização do patrimônio documental e a democratização do conhecimento histórico. A presente investigação tem como objetivo a implementação da curadoria de conteúdo como uma prática arquivística para a efetiva difusão da informação no arquivo do Memorial Santa Dulce dos Pobres. Caracteriza-se como uma pesquisa de natureza descritiva e documental, com abordagem qualitativa. Adota-se, como metodologia, o estudo de caso, tendo-se como corpus cadernos de atas de assembleias referentes ao período de 1941 a 1987. A análise foi conduzida a partir da aplicação de normas arquivísticas de descrição, associadas aos pressupostos da Organização do Conhecimento Arquivístico, de modo a identificar o contexto de produção documental, os atores envolvidos e as possibilidades de mediação documentária por meio da curadoria. Os resultados obtidos apontam para a demanda de estudos sobre curadoria de conteúdo no campo da Ciência da Informação, destacando-se o papel ampliado da pessoa arquivista como curadora e mediadora documental da memória coletiva. Verificou-se, ainda, a importância dos instrumentos arquivísticos na consolidação do paradigma pós-custodial, bem como na legitimação das fontes de informação em contextos históricos. Tais achados reforçam a necessidade de adoção de estratégias contemporâneas de difusão e o potencial da curadoria para tornar os acervos mais acessíveis e significativos à sociedade. À guisa de uma conclusão, ressalta-se o entendimento de que os documentos de arquivo são construções simbólicas que expressam e podem, se não tratados, perpetuar apenas uma visão de mundo. As diretrizes para a curadoria de conteúdo como prática arquivística se alinharam ao modelo proposto em formato de fluxograma, incluindo as etapas de busca, seleção e análise de fontes, agregação de valor e compartilhamento, tendo-se em todas as fases a descrição arquivística como normativa, respeitando a proveniência e a territorialidade miltoniana para a contextualização da narrativa, de modo a potencializar ações educativas e patrimoniais do acervo. Destaca-se que avaliação de todo processo curatorial deve levar em conta o usuário, favorecendo a relevância do conteúdo e o valor agregado à sua experiência

A leitura analítica incide sobre as narrativas da autora que abordam a temática da sexualidade juvenil e as suas problemáticas, com o objetivo de estudar a forma como as mesmas contribuem para a construção da identidade pessoal e sexual do potencial recetor juvenil. Desta forma, pretende-se detetar os conflitos psicológicos dos protagonistas, os seus valores e problemas, assim como os novos valores, problemas e preocupações sociais que emergem das narrativas; compreender de que forma a autora exprime a sua capacidade em observar o quotidiano e as relações humanas; identificar os pilares em que se apoia o universo social em estudo (família, escola, grupo de amigos); enquadrar as personagens numa sociedade pós-industrial e de informação que exige ao adolescente uma superação de desafios e de novos problemas.This dissertation analyzes the different representations of sexuality in Ana Saldanha’s literary work for youth. The analytical reading focuses on the author's narratives that address the issue of teenage sexuality and its problems, aiming to study their contribution in the construction of personal and sexual identity of the potential juvenile receiver. Thus, it is intended to detect the psychological conflicts of the protagonists, their values and problems, as well as the new values, social concerns and problems that emerge from the narratives; understand how the author expresses her ability to observe daily life and human relations; identify the pillars on which relies the social universe under study (family, school, friends); framing the characters in a post-industrial and information society which requires the teenager to relentlessly surmount challenges and new problems

Neste plano, move a escrita a vibração última de encontros ocorridos no ambiente acadêmico e fora dele, atravessados pela força do tempo. Postais elaborados por artistas/professores desenham e compõem paisagens ao longo do texto e questionam modos de ensinar sobre e com arte, além de provocar uma conversa sobre a inclusão da arte na área de linguagem pela educação. Esta provocação acontece ao se observar, no ensino de arte, o esgotamento de práticas metodológicas referenciadas, especialmente na abordagem com arte contemporânea. Entre margens territoriais marcadas como arte e educação, entre tons poético e professoral, a escritura tece o conceito de arte como perceptos e afectos e, portanto, como linguagem das sensações, propondo à educação um pensamento à deriva que se abre às forças do fora, ao acaso e ao devir de seus encontros.Abstract : Events in art and education proliferate in the composition plan of this writing, composing dialogs with literature and philosophy. In this plan, the writing is moved by the ultimate vibration of encounters occurred in the academic environment and beyond, traversed by the strength of time. Postcards designed by artists/teachers draw and compose landscapes throughout the text and inquire about ways of teaching about and with art, besides provoking a conversation on the inclusion of art in the language field of education. This provocation occurs as one observes the exhaustion of referenced methodological practices in the teaching of art, especially, as regards contemporary art. Between territorial margins, labeled as art and education, between poetic and professorial tones, the writing weaves the concept art as perceptos and afectos and, therefore, as language of sensations, offering education an adrift thinking which opens itself to the strength of the outside, to random and to the infinite becoming of its encounters

O estudo realizado pela presente tese tem por objetivo analisar o tratamento dado ao tema da viagem nos romances Desmundo, O retrato do rei, Dias & Dias e Amrik, evidenciando que por intermédio das narradora-viajantes # Oribela, Mariana, Feliciana e Amina # a autora promove um diálogo entre diferentes culturas, gêneros, etnias e gerações, ao mesmo tempo em que estabelece um profícuo diálogo com o passado em sua invariante problematização concernente aos limites e cruzamentos entre o discurso ficcional e os discursos narrativos extraliterários que o cercam, sobretudo o histórico e o biográfico. In the whole of Brazilian writer Ana Miranda#s fictional narrative, the travel thematic # considered one of the most fertile themes of the western literature of all times # occupies a central space, and can even been seen as the most important structuring axis of Miranda#s works. The aim of the present thesis is to analyze the treatment given to the travel subject in the novels Desmundo, O retrato do rei, Dias & Dias and Amrik, emphasizing that, through the traveling narrators, Oribela, Mariana, Feliciana and Amina, the author promotes a connection between different cultures, genders, ethnics and generations, at the same time that establishes a profitable dialogue with the past in her invariant problematization concerning the limits and crossings between the fictional and the extra-literary narrative discourses that surrounds it, mainly the historical and the biographic ones
A responsabilidade social nas instituições de ensino superior e a promoção da inclusão na sociedade digital: um caso de estudo de capacitação digital dos estudantes PALOP do Instituto Politécnico de Santarém
No full textO contexto deste trabalho centra-se essencialmente nos estudantes oriundos do Países
Africanos de Lingua Oficial Portuguesa que frequentam o Instituto Politécnico de
Santarém, nas suas dificuldades de integração e de competências digitais, tendo como
ponto de partida o projeto JUMP TO IPSantarém. Este é um projeto constituido por
módulos de integração, implementado com estudantes de 1º ano de mestrado. No
âmbito da Responsabilidade Social e de forma a promover a inclusão destes estudantes,
este estudo apresenta uma proposta de módulos de integração/formação, que responde
às especificidades destes estudantes.
Considerando o seu contexto e objetivos esta investigação, é um estudo de caso. Sendo
o estudo aplicado em contexto real, foi escolhida uma abordagem qualitativa, em que a
recolha de dados foi feita através de um grupo focal e entrevistas semiestruturadas a
vários agentes educativos.
Os resultados demostram que a capacitação digital é uma barreira à integração destes
estudantes, mas que existem outras, como as financeiras, as culturais e as linguísticas,
que para eles são prioritárias e talvez por esta razão, não priviligiem esta questão.
A proposta de módulos de integração/formação que resulta deste estudo procura dar
resposta à barreira de capacitação digital, e assim ajudar a melhorar as condições de
integração destes estudantes.The context of this work essentially focuses on students from Portuguese-speaking
African Countries who attend the Polytechnic Institute of Santarém, on their difficulties
with integration and digital skills, taking as a starting point the JUMP TO IPSantarém
project. This is a project made up of integration modules, implemented with 1st year
master's students. Within the scope of Social Responsibility and in order to promote the
inclusion of these students, this study presents a proposal for integration/training
modules, which respond to the specificities of these students.
Considering its context and objectives, this research is a case study. As the study was
applied in a real context, a qualitative approach was chosen, in which data collection was
done through a focus group, semi-structured interviews with various educational agents.
The results show that digital training is a barrier to the integration of these students, but
that there are others, such as financial, cultural and linguistic, which are priorities for
them and perhaps for this reason, they do not prioritize this issue.
The proposal for integration/training modules that results from this study seeks to
respond to the digital training barrier, and thus help to improve the integration conditions
of these students
