    Advocates of the use of genetic engineering techniques in agriculture contend that this new biotechnology promises increased productivity, better use of natural resources and more nutritious foods.     Employee performance has a very important role because it will affect the employee output so that will affect the performance of a company. This study analyzes employee performance at PT Central Batam Raya are reviewed through the variables of emotional intelligence and human resource development. This research is a quantitative research with the method used is descriptive analysis and multiple regression analysis and data collection techniques through questionnaires. Questionnaires distributed amounted to 102 respondents. The method used in this study was chosen mainly because of the limited time, cost and energy that the author has, so that in this case the writer hopes to be able to examine specific aspects of a social situation in depth. Data collection is done by primary data and secondary data. The results showed that the variable emotional intelligence and human resource development had a significant effect simultaneously on employee performance with t count> t table (4,860 > 1,98397) and (4,935 > 1,98397), significant value <0.05 and F count> F table (70,337 > 3,09) with a significant value <0.05

    This paper has been prepared as part of the Zambia country study of the Macroeconomic and Regional Integration in Southern Africa (MERRISA) project and serves as a background paper for modeling exercises. The paper focuses on analyzing institutional constraints on the development of the agricultural sector in Zambia. It argues that by changing some of the rules and neglecting to integrate these changes into the complete institutional setting, policymakers have been unable to achieve their goals. Other constraints on Zambia's agricultural development are of a more technical nature. There are problems with Zambia's infrastructure. This paper argues that these are problems of the provision of public and merit goods mainly occurring in rural areas. Farmers emphasize their limited access to resources like credit, fertilizer, and draft animals.     Indonesia is well known for its diversity which scattered from east to west. The diversity itself can be considered as the intangible aspects which relates directly with the aspect of tourism. Tourism provides profitable sectors in many countries because tourism has three major functions. First, to support economic aspect by developed local society. Second, to maintain culture and heritage. Third, to preserve the environment, those three aspects will support the strength of the nation. The purpose of this study is to analyze the reason why the tourism aspect cannot be established by following Indonesian tourism strategic planning. This research is using qualitative method to describe the situation from the problem arise within the society and applying descriptive method in analyzing the data particularly for the tourism development. In my writing, I would like to provide argument why the tourism in several areas cannot be fully developed by following the program of sustainable tourism. There are several major factors which can be described as the findings of this research which takes part in Murung Raya, Central Kalimantan such as lack of local empowerment. This is because the local government do not pay attention to the characteristic of particular culture in nearby area as a great cultural heritage, there is no involvement of local society, the allocation of budgeting do not focus on the tourism development, less priority of cultural and tourism aspects from the program of regional strategic development, the tourism development pay less attention to the social regulations and it creates less awareness of the local community about how important tourism to support livelihood as source of alternative income

    Optimizing Genetic Manipulation of Methanogens through Faster Cloning Techniques

    Methanogenesis is the biological production of methane. Only anaerobic archaea known as methanogens are capable of such a metabolic feat. They have strict living conditions and substrate sources which determine their rate of metabolism. This is of particular importance from a greenhouse gas reduction perspective or biogas capturing perspective. One of the best ways to optimize methanogen methane production is via genetic manipulation. The current procedures are timely though, therefore a faster cloning processes should be developed. The objective of this study was to optimize a premade genetic transformation kit known as the Gibson Kit. The Gibson Kit was supposed to ease the work of genetic manipulation by combining several linear chunks of DNA together at once via the use of sequence overhangs. The resulting plasmid from the Gibson could then be transformed into E. coli where E. coli is supposed to replicate the plasmids extremely quickly. Afterwards, that plasmid could then transform methanogens. Several baseline PCR (polymerase chain reaction) transformations were performed to linearize and amplify the desired plasmid of pNB72.3. PCR amplifications of the desired gene segments which would be added to the plasmid were also performed. The desired gene segments being assembled were supposed to take out the production of Cytochrome C within the methanogen electron transport system by deleting the ccmf gene. Several PCR experiments were carried through without success. The cause of the failures included the primers “hair pinning” at the recommended annealing temperatures, primers being too nonspecific, and primers unable to perform efficiently at the recommend annealing temperatures. After several tries of no success, the Gibson Kit was tested without PCR linearization of the circular plasmid. Instead, standard digestion was relied on. With standard electro competent cell transformation and antibiotic screening, the Gibson product was then tested for successful transmission of the plasmid to the E. coli. Results were negative; therefore, the optimization of faster cloning techniques was unsuccessful, but it will help guide future efforts

    The Building Transmission Capacity grant activities focused on educating both policy makers (primarily at Public Utility Commissions) and utilities across the West. Western Grid Group (WGG), the grant recipient, chose three methods to reach these audiences - direct outreach, a website that contains information on policies and strategies to integrate more variable generation resources, and a report - The Best of the West, Policies and Practices to Support Transition to a Lower-Carbon Electric Sector in the Western Interconnection and that highlights what is working in the West. While all avenues for education are effective the Best of the West report is the first west-wide assessment of its kind. The report details incremental changes that are working to integrate variable generation but it also expounds on what fundamental or transformative changes are needed to get to the 20% wind penetration and beyond

    Nowadays, every business transaction and many other activities in this world are used to create a credible contract to secure their legal rights and obligations. Even though a contract is well-established, sometimes there is a condition which makes the contract undesirable anymore, such as increasing in price material, changes in environment regulation or a better offer from other company. Therefore, conducting efficient breach is one of the solutions to obtain a higher surplus or even to prevent from a further loss. To perform efficient breach, the breaching party may breach the original contract only if the victim of the breach receive full compensation, thus there is no party which become worse off. Besides, the other reason by paying full compensation to the victim of the breach is to gain efficiency which is introduced by Richard Coase. Efficient breach is one of the legal doctrines in contract law and closely related with economic. However, the doctrine of efficient breach has created controversy, because there are many criticism regarding morality and inefficiency relatives to not breaching the contract. Therefore, this research will be conducted to observe and analyse the differences of efficient breach from law and economic perspective and the impact of efficient breach to the breaching party and the victim of the breach as a result of efficient breach.
