Development of Drug Delivery Systems for ERAP modulators for the treatment of cancer and autoimmune diseases
No full textEndoplasmic reticulum (ER) aminopeptidases ERAP1 and ERAP2 play a key-role in antigen presentation by trimming peptides for binding to Major Histocompatibility Complex (MHC) class-I molecules and, thus, recognition by immune cells. Abnormal expression of ERAP or functional genetic variations impair this process, leading to inadequate immune responses and ultimately, contributing to the pathogenesis of cancer and autoimmune diseases. To improve current
therapeutic approaches focused on using small molecule ERAP inhibitors, two polymer-based drug delivery systems (DDS) were developed, to enhance the safety and efficacy of the compounds by promoting a controlled and targeted release and minimizing unwanted side effects. ERAP1 inhibitors were encapsulated in Poly(ethylene glycol) block-Poly(L-Histidine) - PEO-b-PHis nanoparticles, exploiting the pH-responsiveness of the PHis block for targeted release at acidic pH, similar to that present in the tumor microenvironment (TME). When encapsulated into nanoparticles, ERAP1 inhibitors reduced the stability and expression of non-classical MHC class-I molecules. Moreover, cell viability assays confirmed the non-toxicity of DDS, even when the inhibitor was administrated at high concentrations. ERAP2 inhibitors were encapsulated into polymeric electrospun nanofibers as well. The Poly(vinyl-alcohol)-Liposome - PVA_L formulation demonstrated high encapsulation efficiency and a sustained release profile, maintaining the integrity and therapeutic concentrations of ERAP2 inhibitors. Furthermore, these nanofibers loaded with ERAP2 inhibitors disrupted antigen presentation and showed no toxicity at various concentrations in the cellular environment. Overall, the designed polymer-based DDS have been shown to release ERAP inhibitors in a controlled and targeted manner, to reduce non-classical MHC class I expression and antigen presentation, and most important to exhibit no-cytotoxic effects at the
cellular level, making them promising candidates for clinical applications in the treatment of cancer and autoimmune diseases
A jurisprudência portuguesa dos tribunais superiores sobre exoneração do passivo restante (Breves notas sobre a admissão da exoneração e a cessão de rendimentos em particular)
Get PDFDesde 2004 que o Código da Insolvência e Recuperação de Empresas apresenta dois modelos insolvenciais para o tratamento da insolvência de pessoas singulares: por um lado, o modelo reeducativo, encarnado no plano de pagamentos; por outro o fresh start, consagrado com a exoneração do passivo restante. Esta última solução tem sido a opção da esmagadora maioria das pessoas singulares insolventes que, desde 2011, superam o número de processos de pessoas coletivas. Todavia, o sistema português tem sido criticado, não só pelo Memorando da Troika assinado entre o Estado Português, o BCE, a Comissão Europeia e o FMI, como pela própria doutrina e jurisprudência, pondo em evidência a necessidade de alteração do regime relativo às pessoas singulares. Aliás, tal exigência constava do referido Memorando, sem que o Governo português tenha adotado quaisquer medidas. Por outro lado, a recomendação da UE de março de 2014, relativa à recuperação e à insolvência, refere também a necessidade de redução do limite temporal de insolvências de pessoas singulares, nomeadamente no que concerne aos períodos de cessão dos rendimentos aos credores.
Nesta sede, verificamos que o regime português da insolvência, que conta já com mais de uma década de vigência, continua a apresentar fragilidades a nível da aplicação das normas, especialmente em dois pontos particulares – por um lado, a interpretação dos requisitos de acesso, que foi inicialmente feita de forma muito rígida pelos tribunais de 1.ª instância, impedindo o acesso à exoneração e, por outro, a concessão dos rendimentos necessários durante o período de cessão aos credores que, normalmente, desvaloriza as verdadeiras necessidades dos insolventes, dando um caráter punitivo, que o legislador não pretendeu atribuir, à insolvência. Studies of thiamine biosynthesis
No full textIn Escherichia coli, and other prokaryotes, thiamine (vitamin B1) is assembled by
coupling 4-amino-5-hydroxymethyl-2-methylpyrimidine pyrophosphate (Hmp-PP)
and 4-methyl-5-(?-hydroxyethyl)thiazole phosphate (Thz-P). The thiazole moiety is
biosynthesised from 1-deoxyxylulose-5-phosphate, tyrosine, and cysteine, and at least
six genes are required including thiH, thiG, thiS, and thiF. Whilst in aerobes, the C2-N3
fragment of Thz-P derives from glycine in a reaction catalysed by the flavoenzyme
ThiO, in anaerobes such as E. coli, dehydroglycine is formed from tyrosine in a ThiH
dependent reaction. This biosynthetic step requires the cleavage of the C?-C? bond of
tyrosine and a release of an aromatic side chain. ThiH shows sequence similarity with
the ‘radical S-adenosylmethionine’ (AdoMet) family of proteins, including conserved
cysteine ligands to the essential [4Fe-4S] cluster and has been shown to form a complex
with ThiG. With the purpose of studying the mechanistic enzymology by which Thz-P
is assembled it was crucial to isolate ThiH in the holo-form. Several expression systems
and purification methodologies were investigated. The optimisation of the purification
method, together with in vitro chemical reconstitution with exogenous iron and sulfide
allowed the successful isolation of holo-ThiH.To facilitate the mechanistic investigation of Thz-P biosynthesis, an in vitro assay was
developed, and the reaction products formed in vitro were elucidated and quantified.
The aromatic by-product derived from the side chain of tyrosine is p-cresol and the

No full textO contexto deste trabalho centra-se essencialmente nos estudantes oriundos do Países
Africanos de Lingua Oficial Portuguesa que frequentam o Instituto Politécnico de
Santarém, nas suas dificuldades de integração e de competências digitais, tendo como
ponto de partida o projeto JUMP TO IPSantarém. Este é um projeto constituido por
módulos de integração, implementado com estudantes de 1º ano de mestrado. No
âmbito da Responsabilidade Social e de forma a promover a inclusão destes estudantes,
este estudo apresenta uma proposta de módulos de integração/formação, que responde
às especificidades destes estudantes.
Considerando o seu contexto e objetivos esta investigação, é um estudo de caso. Sendo
o estudo aplicado em contexto real, foi escolhida uma abordagem qualitativa, em que a
recolha de dados foi feita através de um grupo focal e entrevistas semiestruturadas a
vários agentes educativos.
Os resultados demostram que a capacitação digital é uma barreira à integração destes
estudantes, mas que existem outras, como as financeiras, as culturais e as linguísticas,
que para eles são prioritárias e talvez por esta razão, não priviligiem esta questão.
A proposta de módulos de integração/formação que resulta deste estudo procura dar
resposta à barreira de capacitação digital, e assim ajudar a melhorar as condições de
integração destes estudantes.The context of this work essentially focuses on students from Portuguese-speaking
African Countries who attend the Polytechnic Institute of Santarém, on their difficulties
with integration and digital skills, taking as a starting point the JUMP TO IPSantarém
project. This is a project made up of integration modules, implemented with 1st year
master's students. Within the scope of Social Responsibility and in order to promote the
inclusion of these students, this study presents a proposal for integration/training
modules, which respond to the specificities of these students.
Considering its context and objectives, this research is a case study. As the study was
applied in a real context, a qualitative approach was chosen, in which data collection was
done through a focus group, semi-structured interviews with various educational agents.
The results show that digital training is a barrier to the integration of these students, but
that there are others, such as financial, cultural and linguistic, which are priorities for
them and perhaps for this reason, they do not prioritize this issue.
The proposal for integration/training modules that results from this study seeks to
respond to the digital training barrier, and thus help to improve the integration conditions
of these students
