Probing the nature of AFEX-pretreated corn stover derived decomposition products that inhibit cellulase activity

Sequential fractionation of AFEX-pretreated corn stover extracts was carried out using ultra-centrifugation, ultra-filtration, and solid phase extraction to isolate various classes of pretreatment products to evaluate their inhibitory effect on cellulases. Ultra-centrifugation removed dark brown precipitates that caused no appreciable enzyme inhibition. Ultra-filtration of ultra-centrifuged AFEX-pretreated corn stover extractives using a 10 kDa molecular weight cutoff (MWCO) membrane removed additional high molecular weight components that accounted for 24–28% of the total observed enzyme inhibition while a 3 kDa MWCO membrane removed 60–65%, suggesting significant inhibition is caused by oligomeric materials. Solid phase extraction (SPE) of AFEX-pretreated corn stover extractives after ultra-centrifugation removed 34–43% of the inhibition; ultra-filtration with a 5 kDa membrane removed 44–56% of the inhibition and when this ultra-filtrate was subjected to SPE a total of 69–70% of the inhibition were removed. Mass spectrometry found several phenolic compounds among the hydrophobic inhibition removed by SPE adsorption.Published version: Humpula, James F., Uppugundla, Nirmal, Vismeh, Ramin, Sousa, Leonardo, Chundawat, Shishir P. Whistle characteristics of free-ranging Indo-Pacific humpback dolphins (Sousa chinensis) in Sanniang Bay, China

Broadband recording systems were adapted to characterize the whistle characteristics of free-ranging Indo-Pacific humpback dolphins (Sousa chinensis) in Sanniang Bay, China. A total of 4630 whistles were recorded, of which 2651 with legible contours and relatively good signal-to-noise ratios were selected for statistical analysis. Of the six tonal types (i.e., flat, down, rise, convex, U-shaped, and sine), flat (N - 1426; 39.45%) was the most predominant, followed by down (N - 754; 23.35%) and rise (N - 489; 12.34%). The whistles showed a short duration (mean +/- SD: 370.19 +/- 285.61 ms; range: 29-2923 ms), a broad frequency range (fundamental contour ranged from 0.52 to 33 kHz), and two harmonics (mean +/- SD: 1.90 +/- 2.74, with the maximum frequency of harmonics beyond 96 kHz). Whistles without gaps and stairs accounted for 76.7% and 86.4%, respectively. No significant interspecies differences in frequency parameters were observed compared with S. teuszii, which is inconsistent with morphological taxonomies but confirms phylogenetic results, thus suggesting a close relation between Chinese S. chinensis and Atlantic S. teuszii. Significant intra- and interspecific differences in the genus Sousa were also observed, indicating that animal vocalization may not be limited by genetically determined traits but could also be a function of local habitat adaptation. “Assinalado" pelo trabalho poético, Cruz e Sousa encarna satanícamente o discurso e os valores da época, como um visionário que contempla sua inevitável condiçãoAnalysing som e of the prose poems by Cruz e Sousa, the author makes a point of the antagonism s and dram atic situations experienced by a black writer harassed by the prejudice in a slavocrat country. “Signalized” by the poetic art, the poet incorporates with a devilish disposition the discourse and the values of a time like a visionary who watches his inevitable conditio
