Diurnal régulation of the RNA polymerase III and the role of MAFi in the liver
No full textL'ARN polymérase III (Pol III) synthétise des petits ARNs non-codants dont la plupart sont essentiels pour la traduction de l'ARN en protéine. L'activité de la Pol III est en conséquence très étroitement régulée par des facteurs de croissance et de prolifération comme c-MYC, RBi, TRP53 et MAFi. MAFi est un répresseur de la transcription par la Pol III qui est régulé par le complexe TORCi, lequel va inactiver MAFi en le modifiant par phosphorylation en fonction de l'état nutritionnel de la cellule. La régulation de la Pol III est donc sensible aux changements environnementaux comme la disponibilité de nutriments, mais lorsque j'ai commencé ma thèse, personne ne s'était attelé à étudier la transcription de la Pol III au cours d'une journée. La première partie de ma thèse a donc été dédiée à étudier la régulation de la Pol III au cours de la journée. Pour ce faire, j'ai utilisé (1) des souris de type sauvage (Ctrl), (2) des souris avec une délétion du gène de l'horloge circadienne Arntl (Arntl/-), (3) des souris nourries toutes les trois heures et (4) des souris avec une délétion de Mafi dans tout l'organisme (Ma.fr >-). En comparant la localisation de la Pol III sur ses gènes cibles dans mes différents modèles de souris, j'ai pu montrer que la transcription de la Pol III augmente pendant la journée sous le contrôle de l'horloge circadienne, puis durant la nuit, la transcription reste élevée de par une régulation de MAFi.
La deuxième partie de ma thèse avait pour but de comprendre le rôle de MAFi dans le foie d'une souris, autre que son rôle de répresseur de la Pol III. En effet, MAFi a été précédemment identifié comme étant un facteur important dans le métabolisme des lipides, et les souris dont le gène Mafi a été supprimé dans tout l'organisme sont résistantes à l'obésité provoquée par une nourriture riche en matière grasse. Le rôle précis de MAFi dans le métabolisme des lipides reste cependant encore controversé et les conséquences d'une suppression du gène Mafi dans le foie seulement restent encore méconnues. Pour comprendre le rôle de Mafi dans le foie, j'ai utilisé des souris ayant une délétion du gène Mafi soit dans tout le corps (Mafr/-) soit seulement dans le foie (MafrL/"L). Etonnamment, les souris MafrL/-L étaient plus grosses que les souris Ctrl, phénotype inverse de celui des souris Mafi'/'. Pour comprendre ces différences, j'ai étudié le niveau d'accumulation d'ARN et de protéines dans le foie de souris Mafr/- et Mafr L/ L et dans celui des souris contrôles correspondantes. Les résultats indiquent que dans le foie de souris MafrV-L, les différences phénotypiques sont principalement expliquées par des changements au niveau protéique et non au niveau transcriptionnel, et que Mafi est impliqué dans le métabolisme des lipides, peut-être par un effet sur la traduction
Going Beyond Counting First Authors in Author Co-citation Analysis

counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

Appropriate Similarity Measures for Author Cocitation Analysis
No full textWe provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. No full textWe conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
