Quantification of botrytis cinerea in grapevine bunch trash by real-time PCR
Quantification of colonization of grape bunch trash by Botrytis cinerea is crucial for Botrytis bunch rot (BBR) control. A previously developed quantitative polymerase chain reaction (qPCR) method was adapted to quantify B. cinerea DNA in grape bunch trash, and a colonization coefficient (CC) was calculated as the ratio between the DNA concentrations of B. cinerea and of Vitis vinifera. CC values increased linearly with the number of conidia of B. cinerea or the quantity of mycelium of B. cinerea added to the bunch trash increased. CC values also increased linearly in bunch trash samples containing increasing percentages of B. cinerea-colonized bunch trash; in the latter samples, CC values were correlated with subsequent assessments of B. cinerea colonization of trash (as determined by plating on agar) and sporulation on the trash (as determined by spore counts after incubation in humid chambers). The qPCR assay was also validated using trash collected from bunches treated or not treated with fungicides in three vineyards in two seasons. CC values reflected the reduction in sporulation and in latent infections of mature berries caused by fungicide application. The qPCR assay enables rapid, specific, sensitive, and reliable quantification of the degree of colonization of bunch trash by B. cinerea, which makes it a useful tool for studies of the epidemiology and management of BBR
Interaction of tomato genotypes and arbuscular mycorrhizal fungi under reduced irrigation
Climate change is increasing drought events and decreasing water availability. Tomato is commonly transplanted to an open field after seedling production in a nursery, requiring large volumes of water. Arbuscular mycorrhizal (AM) fungi help plants cope with drought stress; however, their effects depend on plant genotype and environmental conditions. In this study, we assessed the interactions among different tomato seedling genotypes and two AM fungi, Funneliformis mosseae and Rhizophagus intraradices, under two water regimes, full and reduced. Our results showed that F. mosseae was more effective than R. intraradices in the mitigation of drought stress both in old and modern genotypes. However, seedlings inoculated with R. intraradices recorded the highest values of leaf area. ‘Pearson’ and ‘Everton’ genotypes inoculated with F. mosseae recorded the highest values of root, leaf, and total dry weights under full and reduced irrigation regimes, respectively. In addition, ‘Pearson’ and ‘H3402’ genotypes inoculated with F. mosseae under a reduced irrigation regime displayed high values of water use efficiency. Our results highlight the importance of using AM fungi to mitigate drought stress in nursery production of tomato seedlings. However, the development of ad hoc AM fungal formulations, which consider genotype x AM fungi interactions, is fundamental for achieving the best agronomic performances
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
In vitro antifungal activity of the tea tree (Melaleuca alternifolia) essential oil and its major components against plant pathogens
Aims: The aim of this study was to examine the effect of Melaleuca alternifolia
essential oil (TTO) and its principal components on four cereal-pathogenic
fungi.
Methods and Results: The antimycotic properties of TTO and of terpinen-4-
ol, c-terpinen and 1,8-cineole (eucalyptol) were evaluated in vitro on Fusarium
graminearum, Fusarium culmorum and Pyrenophora graminea. Moreover, barley
leaves infected with Blumeria graminis were treated with whole TTO. All the
tested fungi were susceptible to TTO and its components.
Conclusions: TTO exerted a wide spectrum of antimycotic activity. Single
TTO purified components were more active than the whole oil in reducing
in vitro growth of fungal mycelium and, among the tested compounds, terpinen-
4-ol was the most effective.
Significance and Impact of the Study: TTO and its components can be considered
potential alternative natural fungicides
DNA-based analysis for authenticity assessment of monovarietal pasta
Because of the presence on the market of high-value monovarietal pasta, the aim of this work was to evaluate the potential of DNA profiling to test its authenticity. AFLP profiles obtained from eight monovarietal pasta samples and from the corresponding varieties were compared. The possibility of identifying the variety of an unknown monovarietal pasta by referring to a durum wheat molecular markers database is proposed. © Springer-Verlag 2004
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
