No full textMechanical Processes of a Single Synthetic Molecular Machine Studied by AFM-based Force Spectroscopy
No full textSome biomolecules are able to generate directional forces by rectifying random thermal motions. This allows these molecular machines to perform mechanical tasks such as intracellular cargo transport or muscle contraction in plants and animals. Although some artificial molecular machines have been synthesized and used collectively to perform mechanical tasks, so far there have been no direct measurements of mechanical processes at the single-molecule level. Here we report measurements of the mechanical work performed by a synthetic molecule less than 5 nm long. We show that biased Brownian motion of the submolecular components in a hydrogen-bonded [2]rotaxane -a molecular ring threaded onto a molecular axle- can be harnessed to generate significant directional forces. We used the cantilever of an atomic force microscope to apply a mechanical load to the ring during single-molecule pulling-relaxing cycles. The ring was pulled along the axle, away from the thermodynamically favoured binding site, and was then found to travel back to this site against an external load of 30 pN. Using fluctuation theorems, we were able to relate the measurements of the work done at the level of individual molecules to the free energy change measured previously by ensemble measurements. Finally, we used dynamic single-molecule force spectroscopy to probe kinetic information of the interaction between the molecular ring and the preferred binding site. The results also demonstrate that AFM-based single-molecule force spectroscopy, which has been widely used to investigate the mechanochemical behaviour of (bio)macromolecules, can be applied to a molecule that is less than 5 nm in its extended form

Educational aspirations among ethnic minority youth in Brussels: Does the perception of ethnic discrimination in the labour market matter? A mixed-method approach
No full textEthnic disparities in educational aspirations and choices are important to comprehend ethnic education inequality. Based on a mixed-method approach (3121 questionnaires and 40 interviews of pupils), this article investigates ethnic differences among nine ethnic minority groups of pupils in Brussels with regard to their educational aspirations. The multivariate analysis of the questionnaires shows that pupils from only four out of the nine ethnic minorities hold significantly higher aspirations than the majority group. In addition, our mixed-method results did not support the hypothesis on perceived ethnic discrimination in the labour market in explaining the higher educational aspirations of ethnic minority youth. Nevertheless, personal experience of discrimination at school is significantly associated with higher educational aspirations. We conclude by highlighting the relevance of the parental transmission of the intergenerational mobility project in explaining ethnic differences in youth's educational aspirations. © The Author(s) 2013.SCOPUS: ar.jinfo:eu-repo/semantics/publishe
Representation of an “Angel in the House” and “Fallen Woman” in Elizabeth Gaskell’ s Ruth
No full textSince a woman in Victorian England was expected to be a manager of household, there was a stereotype of Victorian woman called the “angel in the house”. There was also another stereotype of Victorian woman called “fallen woman”. As literature represents life, woman’s roles and stereotypes in Victorian are also reflected in Elizabeth Gaskell’s Ruth. The objectives of this study are to describe the representation of angel in the house and fallen woman in this novel and to describe social perspective and how the fallen character struggle to face and overcome the social construction surrounding her. The study employs library research and sociological approach to analyze Victorian woman’s roles and the stereotypes. It analyzes the character, conflict and setting that brings the study to the analysis of Victorian woman’s roles in this novel. From the analysis, it can be concluded that there are woman’s roles performed by female characters in this novel that is “angel in the house” and “fallen woman”. Ruth, as the main character, performs both images. She performs “angel in the house” when she becomes a governess. Beside that, she falls into fallen woman because of her naivete and her innocence. However, as fallen woman, she succeeds to struggle to get her position in society and becomes an honorable woman even though she has to die in the end of the novel
