Programa de Pós-Graduação em Literatura.Pretende-se, ao longo desta dissertação, destacar a noção de ruptura a alimentar a obra de Lima Barreto. Para tanto, investiga-se primeiramente a fortuna crítica existente sobre Lima Barreto, tanto aquela vigente a sua época, como a posterior. Busca-se, a seguir, na contramão da crítica instituída, indicar aquelas que seriam as rupturas perpetradas por Lima Barreto, tanto em termos estéticos com ideológicos, momento em que se inverte, de certa maneira, o sinal da crítica: aquilo que ela apontava como defeito bem poderia ser mérito de Lima Barreto. Testamos esse ponto de vista na leitura mais detida de um dos romances de Lima Barreto, o Triste Fim de Policarpo Quaresma, e, depois, em observações esparsas em torno de sua obra e de suas posturas estéticas e ideológicas. Trilhado esse caminho, restava uma questão, entre outras: a necessidade de situar a obra de Lima Barreto no contexto da literatura brasileira, em que Lima Barreto aparece, normalmente, como um autor "pré-modernista". Contesta-se no corpo do texto, tal enquadramento, indicando-se, no contraponto, a modernidade e atualidade de Lima Barreto, ou a do que se poderia denominar de "bloco-popular" da sua época
DNA fusion gene vaccination mobilizes effective anti-leukemic cytotoxic T lymphocytes from a tolerized repertoire
No full textThe majority of known human tumor-associated antigens derive from non-mutated self proteins. T cell tolerance, essential to prevent autoimmunity, must therefore be cautiously circumvented to generate cytotoxic T cell responses against these targets. Our strategy uses DNA fusion vaccines to activate high levels of peptide-specific CTL. Key foreign sequences from tetanus toxin activate tolerance-breaking CD4+ T cell help. Candidate MHC class Ibinding tumor peptide sequences are fused to the C terminus for optimal processing and presentation. To model performance against a leukemia-associated antigen in a tolerized setting, we constructed a fusion vaccine encoding an immunodominant CTL epitopederived from Friend murine leukemia virus gag protein (FMuLVgag) and vaccinated tolerant FMuLVgag-transgenic (gag-Tg) mice. Vaccination with the construct induced epitopespecificIFN-c-producing CD8+ T cells in normal and gag-Tg mice. The frequency and avidity of activated cells were reduced in gag-Tg mice, and no autoimmune injury resulted. However, these CD8+ T cells did exhibit gag-specific cytotoxicity in vitro and in vivo. Also, epitope-specific CTL killed FBL-3 leukemia cells expressing endogenous FMuLVgag antigen and protected against leukemia challenge in vivo. These results demonstrate a simple strategy to engage anti-microbial T cell help to activate epitope-specific polyclonal CD8+ T cell responses from a residual tolerized repertoire
The maternal immune system during pregnancy and its influence on fetal development
No full textThe maternal immune system plays a critical role in the establishment, maintenance, and completion of a healthy pregnancy. However, the specific mechanisms utilized to achieve these goals are not well understood. Various cells and molecules of the immune system are key players in the development and function of the placenta and the fetus. Effector cells of the immune system act to promote and yet limit placental development. The T helper 1 (Th1)/T helper 2 (Th2) immune shift during pregnancy is well established. A fine balance between proinflammatory and anti-inflammatory influences is required. We herein review the evidence regarding maternal tolerance of fetal tissues and the underlying cell-mediated immune and humoral (hormones and cytokines) mechanisms. We also note the many unanswered questions in our understanding of these mechanisms. In addition, we summarize the clinical manifestations of an altered maternal immune system during pregnancy related to susceptibility to common viral, bacterial, and parasitic infections, as well as to autoimmune diseases.Peer reviewe
Heterogeneous and tissue-specific regulation of effector T cell responses by IFN-gamma during Plasmodium berghei ANKA infection.
No full textIFN-γ and T cells are both required for the development of experimental cerebral malaria during Plasmodium berghei ANKA infection. Surprisingly, however, the role of IFN-γ in shaping the effector CD4(+) and CD8(+) T cell response during this infection has not been examined in detail. To address this, we have compared the effector T cell responses in wild-type and IFN-γ(-/-) mice during P. berghei ANKA infection. The expansion of splenic CD4(+) and CD8(+) T cells during P. berghei ANKA infection was unaffected by the absence of IFN-γ, but the contraction phase of the T cell response was significantly attenuated. Splenic T cell activation and effector function were essentially normal in IFN-γ(-/-) mice; however, the migration to, and accumulation of, effector CD4(+) and CD8(+) T cells in the lung, liver, and brain was altered in IFN-γ(-/-) mice. Interestingly, activation and accumulation of T cells in various nonlymphoid organs was differently affected by lack of IFN-γ, suggesting that IFN-γ influences T cell effector function to varying levels in different anatomical locations. Importantly, control of splenic T cell numbers during P. berghei ANKA infection depended on active IFN-γ-dependent environmental signals--leading to T cell apoptosis--rather than upon intrinsic alterations in T cell programming. To our knowledge, this is the first study to fully investigate the role of IFN-γ in modulating T cell function during P. berghei ANKA infection and reveals that IFN-γ is required for efficient contraction of the pool of activated T cells
Políticas do primitivo: as estéticas modernistas de Mario de Andrade e Lezama Lima
No full textDissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Comunicação e ExpressãoEste trabalho visa, fundamentalmente, interpretar as tensões entre as estéticas de Mário de Andrade e de Lezama Lima na medida em que elas instauram discursividades alternativas que situadas no confronto primitivo/moderno, configuram identidades culturais diferenciadas. Enquanto problema da modernidade, a presença do primitivo coloca em crise a identidade cultural do Ocidente ao provocar o surgimento do Outro, vale dizer, a irrupção da diferença no âmbito do Mesmo. Neutralizada na abstração conceitual de "primitivismo", essa diferença é incorporada à identidade ocidental, ora a partir de concepções evolucionistas, ora a partir de operações dialéticas. Numa tentativa de interferir nesse processo incorporativo, postulamos a ressemantização do conceito de primitivo, com o propósito de recuperá-lo enquanto elemento perturbador dos efeitos padronizadores da modernidade. Nesse sentido, os primitivos latinos-americanos podem ser pensados como formas de apropriação trangressivas que, entre outras coisas, quebram com uma lógica causalista, fragmentam a linearidade das perspectivas evolucionárias e reelaboram, assim, estagnadas tradições estéticas e históricas. Consideramos, portanto, que as estéticas de Mário de Andrade e de Lezama Lima, inseridas nos conflitos de uma modernidade periférica, que se contrapõe aos efeitos globalizadores da racionalidade moderna

Hypersingular integrals arise in many contexts, such as singular and hypersingular boundary integral
equations, which are tools for modeling many phenomena in different branches of the applied sciences.
Here we derive an extended product rule and by a mixed combination with the one weight product
rule introduced in [9], we propose a compound scheme of quadrature rules which allows a significant
reduction in the number of evaluations of the density function f . Conditions assuring the stability and
the convergence of the the mixed scheme in weighted uniform form are deduced. Some numerical
experiments are also given, in order to highlight the efficiency of the mixed approach

Lima Barreto / Roberto Arlt: a comunidade em exílio
No full textTese (doutorado) - Universidade Federal de Santa Catarina, Centro de Comunicação e Expressão, Programa de Pós-Graduação em Literatura, Florianópolis, 2009.A presente tese opera no limiar entre a literatura brasileira e a argentina, mais especificamente com dois de seus escritores bastante admirados, mas não raro censurados pela crítica em função de supostos deslizes de linguagem e estilo. Lima Barreto (1981-1922) e Roberto Arlt (1900-1942), estes "homens do subsolo" no espaço da literatura nacional, são foco deste estudo que procura, através da leitura dos romances - Recordações do Escrivão Isaías Caminha, Triste Fim de Policarpo Quaresma, Vida e Morte de M. J. Gonzaga de Sá, de Lima Barreto, e El juguete rabioso, Los siete locos e Los lanzallamas,de Roberto Arlt -, acompanhar o exílio e a relação das personagens com a comunidade. A partir da definição do conceito de exílio como negatividade, tal como estabelecem Maurice Blanchot, Jean-Luc Nancy, Giorgio Agamben, Franco Rella, entre outros, é possível detectar que a série de personagens solitárias e torturadas, presentes nos romances, participam de um verdadeiro projeto de exaustão do sentido da comunidade que se funda no território. Tal projeto, que exauri a comunidade territorial imaginada - seja nacional ou revolucionária - é, com efeito, fundamento de outra experiência de comunidade. Esta última não faz obra, não opera limite, mas se dá como exílio, como abertura, como acolhimento do ser inqualificável, tal como debatem os filósofos já citados. Por fim, foram tomadas as últimas experiências de escrita - de Lima Barreto, O cemitério dos vivos, e de Roberto Arlt, El desierto entra en la ciudad - que têm como tema a loucura. A partir delas é possível compreender que mesmo o "fora", ao ser entendido como louco, acaba retido nos limites de um confinamento territorial, figurando um modo do próprio Estado se imunizar, conforme Roberto Esposito, contra o "perigo" da comunidade como inclinação ao outro. Porém, a voz narrativa, esvaziada do "eu", parece ainda elaborar uma linha de fuga do território, nos fazendo vislumbrar, enfim, a comunidade em exílio
