    Evaluation of protease and exopolysaccharide production by Pleurotus ostreatus and Pleurotus eryngii

    Os cogumelos comestíveis despertam o interesse por apresentar propriedades nutricionais e farmacêuticas, dentre os quais se destacam representantes do gênero Pleurotus, osquais são capazes de secretar uma gama de metabólitos primários e secundários. Desta forma, o principal objetivo desta pesquisa visou avaliar a produção de proteases e exopolissacarídeos (EPS) deP. ostreatus e P.eryngii utilizando planejamento fatorial por fermentação submersa considerando a influência de diferentes concentrações de fontes nutricionais. Para isso, a produção de proteases e exopolissacarídeos foi verificada por fermentação submersa conduzida por 15 dias, com meio contendo os seguintes componentes: glicose, peptona, extrato de levedura, com pH inicial de 6,2. Foi utilizado um planejamento fatorial 23 com 4 pontos centrais, totalizando 12 experimentos, para cada espécie. A partir do meio líquido recuperado do processo se obteve os exopolissacarídeos e proteases. Nas condições experimentais a produção de EPS não foi significante para as duas espécies, por conta de influências dos componentes do meio em determinadas concentrações. Em contrapartida, a atividade proteolítica determinada se mostrou bastante significativa com máxima atividade proteolítica (5,28 U/mL) na espécie de P.eryngii em 8 g/L de extrato de levedura, 10 g/L de glicose e 15 g/L de peptona. Estacondição foi a mais eficiente para a produção das proteases com atividade ótima em pH 6,0 e temperatura ótima a 50˚C. As características destas enzimas como pH e temperatura ótima, juntamente com outros testes podem permitir sua utilização futura na produção como aditivos em vários setores industriais.The edible mushrooms arouse the interest for their nutritional and pharmaceutical properties, among which the representatives of the genus Pleurotus stand out, which are able to secrete a range of primary and secondary metabolites. Then, the main objective of the research was the production of proteases and exopolysaccharides of P. ostreatus and P.eryngii using factorial planning by submerged fermentation considering the various sources of nutritional foods. For this, a protease and exopolysaccharide production was verified by submerged fermentation for 15 days, with the following components: glucose, peptone, yeast extract, with initial pH of 6.2. It was for factorial planning 23 with 4 central points, totaling 12 experiments, for each species. From the liquid medium recovered the process of production of proteases and exopolysaccharides. Experimental experiments on the production of EPS were not significant for both species, due to influences of the components of the medium in certain concentrations. In contrast, an age proteolytic activity in a proteolytic activity (5.28 U/mL), as P.eryngii in 8 g/L of yeast extract, 10 g/L of glucose and 15 g/L of peptone. This condition was more efficient for a protease production with the highest pH 6.0 and an optimum temperature of 50 ° C.     Professor Pimenta Claro (1957-2018): Pioneer in dynamics of mechanical systems at the University of Minho

    This work highlights the importance of Professor Pimenta Claro in the genesis and development of a new scientific area at the Department of Mechanical Engineering of the University of Minho, namely Dynamics of Mechanical Systems. Professor Pimenta Claro initiated his academic career in October 1980, coming from industry, where he was a well-recognized engineer in the field of mechanical design. Professor Pimenta Claro concluded his Pedagogical Aptitude and Scientific Capacity Tests (PAPCC) – MSc equivalent – in 1987, with dissertation title “Estudo Comparativo das Previsões Teóricas do Desempenho de Chumaceiras Radiais Hidrodinâmicas com Resultados Experimentais”. Professor Pimenta Claro received his doctorate degree in 1994 with thesis “Reformulação de Método de Cálculo de Chumaceiras Radiais Hidrodinâmicas – Análise do Desempenho Considerando Condições de Alimentação” under the mentorship of Professor Sousa Miranda, which was in fact the first PhD in Mechanical Engineering defended at the University of Minho. In 1997, Professor Pimenta Claro broken with his past background – classical tribology – to open a new research domain – Dynamics of Mechanical Systems. Since then, Professor Pimenta Claro has coordinated and participated in several scientific projects both with national and international partners, as well as projects with industrial partners. Professor Pimenta Claro was author of numerous publications, including scientific papers, books, conference papers, etc., and supervised PhD and MSc students. From 2007 to 2013 he coordinated the research group called Dynamics of Mechanical Systems. Professor Pimenta Claro was also pioneer and responsible for the creation of new courses on dynamics of mechanical systems offered in different degrees at the School of Engineering of the University of Minho. Thus, the main purpose of this work is to highlight Pimenta Claro’s contributions to the vast scientific area of Dynamics of Mechanical Systems at the Department of Mechanical Engineering of the University of Minho

In the positive potential case, we exhibit situations where the existence of solutions depends on the strength of the coupling, being this one modulated by a parameter.     From the city of heroes to the city of Ilhas: the Ilhíada by Alberto Pimenta

    This paper reflects on the latest poem published by the Portuguese author Alberto Pimenta, Ilhíada. Even if it creates an explicit dialogue with the poem attributed to Homer, the Iliad, already from the title, it can be seen that Ilhíada is the polar opposite of the Homeric text. The story Alberto Pimenta introduces us to is a tale of the ilhas, an idiosyncratic urban organisation from the city of Porto (Portugal), usually linked to the poorest sectors of the population. As soon as the city became an important tourist destination and “the imaginal engineering” (Hiernaux, 2012) started to create a tourist hotspot narration, successive administrations began to find these ilhas a problem that had to be dealt with. In the poem, the lyrical voice remembers the everyday life that existed before their inhabitants began to be besieged by the building companies. We move, then, from the tales of Troy, an important urban centre of ancient times where heroes fight in a war, to the margins of Porto, where common people just try to live their everyday lives. In this essay, I will argue that the location in which Pimenta chooses to set this story – with the excluded ones, the “subalterns” (Spivak 1985) – challenges the categories of “city”, “hero”, “History” and even “poetry”. To that effect, I will use a comparative perspective that will allow me to uncover the main differences between the two texts

    An engineer at the shadow of great politicians: Dermeval Pimenta and the search for economic development in Minas Gerais (1937-1951)

    O objetivo desta tese é compreender a importância de Dermeval José Pimenta para o desenvolvimento econômico mineiro entre 1937 e 1951. Para tanto foi pesquisado a sua atividade, principalmente, durante sua atuação na administração da Rede Mineira de Viação (1937-1945), como secretário da Viação de Obras Públicas (1943-1945) e na Companhia Vale do Rio Doce (1946-1951). A justificativa desse recorte temporal devese ao fato de verificar-se a convergência de grande presença do Estado na economia do país e a maior atuação prática e de reflexões teóricas (com publicações posteriores) de Pimenta em Minas Gerais. Ademais, foi traçado o percurso acadêmico de Dermeval Pimenta a fim de entender como ele teve acesso a algumas ideias e quais foram elas, durante sua formação. Em seguida, busca-se apreender quais os vínculos entre estas influências sobre os escritos de Pimenta. Tais informações são mobilizadas com o objetivo de entender a compreensão de Pimenta sobre a questão do desenvolvimento econômico, bem como a relação entre ela e seu exercício profissional. Neste sentido, a importância desse trabalho é pelo caráter inédito da forma de pesquisar a expertise técnica do autor, resgatando seu valor para a temática do desenvolvimento econômico de Minas Gerais. A metodologia da pesquisa foi realizada, primordialmente, a partir de fontes históricas. Entre elas, incluem-se jornais do período, documentos do arquivo público mineiro, de arquivos pessoais, do arquivo permanente da Escola de Minas, da biblioteca pública de Belo Horizonte, bem como os próprios livros de autoria de Pimenta, dentre outras. Com base na pesquisa empreendida, buscou-se esclarecer a forma específica de enquadramento do autor no entrelaçamento entre a política e a economia, bem como seu ideal do desenvolvimento econômico. No que diz respeito a sua atuação pública, a estratégia de Pimenta para promover o desenvolvimento econômico, foi por meio da reconstituição da trajetória histórica de Minas Gerais, utilizada para embasar suas ideias e análise sobre os fatos econômicos. Como em seus próprios trabalhos, Pimenta foi capaz de estabelecer diálogos com alguns dos principais políticos do Brasil e demonstrou-se a favor da interferência estatal na época que presidiu a antiga CVDR. Em especial, o que fica aqui demonstrado é o diferencial de Pimenta em sua atuação; pois demonstrou seu ideal de desenvolvimento econômico em sua atuação prática; mostrou sua reflexão nacionalista, regionalista e as assegurou-as em diálogos políticos e econômicos, e implementou alguma delas, principalmente, no Vale do Rio DoceThe main purpose of this thesis is to understand the importance of Dermeval José Pimenta for the economic development of Minas Gerais Brazilian State in the years from 1937 to 1951. Having this in mind a research about his activities was carry out mainly during his performance in the administration of Rede Mineira Viação (1937-1945), as Secretary of Public Works Roads (1943-1945) and at Vale do Rio Doce company (1946-1951). This timing parenthesis is due the fact of the convergence in this period of a larger presence of the State in the economy and a greater practical performance and theoretical reflections followed by publications by Pimenta in Minas Gerais. Furthermore, Dermeval Pimenta\'s academic path was draw in order to understand how he had access to some ideas and what they were during his training. Then, this research seeks to understand the links between these academic influences on Pimenta\'s writings. Such information is useful in order to get deeper insight in the author\'s understanding of the issue economic development and its relationship with his professional practice as well. In this sense, the importance of this research is due to the unprecedented nature of the way to research a author\'s technical expertise, recovering its value for the topic of economic development in Minas Gerais. Methodologically the research was carried out primarily from historical sources, which include newspapers from the period, documents from the Minas Gerais public archive, personal archives, the permanent archive of the Escola de Minas, the public library of Belo Horizonte, books written by Pimenta as well, among others. Launching from this research, the Pimentas specific way of framing the intertwining between politics and economics was at stake as well as his ideal of economic development. Regarding Pimentas public performance strategy to promote economic development, this was done through the reconstruction of the historical trajectory of Minas Gerais State, that for him is the grounding of his ideas and analysis of economic facts. As in his own work, Pimenta was able to establish dialogues with some of Brazil\'s leading politicians and was in favor of state interference at the time he was the president of the former Companhia Vale do Rio Doce - CVDR. In particular, what is argued here is Pimenta\'s differential in its performance, because he demonstrated his ideal of economic development in his practical performance. He maintain in his reflections that he was nationalist and regionalist and ensured this in political and economic dialogues, and performed some of them, mainly in the region of Vale do Rio Doc

    Proteases hydrolyze peptide bonds between proteins and amino acid chains, and have industrial importance. These biocatalysts from microorganisms are more used industrially due to their desirable characteristics in biotechnology. Of the fungi producing proteases, there is a lack of studies in relation to mushroom proteases belonging to the genus Auricularia. The objective of this work was to investigate the production of proteases from edible mushrooms of the genus Auricularia to select a protease-producing species for future biotechnological application. The species A. delicata and A. fuscosuccinea were cultivated in Glucose-Yeast Extract-Peptone (GYP) liquid medium, and the bioprocess was carried out at 150 rpm, at 30°C. After 72 hours the crude extract was recovered by vacuum filtration and, after determining the qualitative activity of proteases in milk agar, the quantitative activity was defined using azocasein 1.0% (w/v) as substrate. In the crude extract obtained from the cultivation of the species that showed the highest proteolytic activity, partial characterization was carried out regarding the effect of pH and temperature on enzyme activity. The results showed that, in solid medium, Auricularia species were shown to produce proteolytic enzymes and, in submerged culture, statistically, the significant value of proteolytic activity (44.33 U/mL) was determined in the crude extract of A. delicata. These enzymes showed optimal activity at pH 5, at 50 and 60 °C, characterizing them as acid proteases, biocatalysts that have potential for application in the food industry.Proteases hidrolisam ligações peptídicas entre proteínas e cadeias de aminoácidos e são de grande importância industrial. Estes biocatalizadores oriundos de microrganismos são mais utilizados industrialmente por suas características desejáveis em biotecnologia. Dos fungos produtores de proteases, há carência de estudos em relação às proteases de cogumelos pertencentes ao gênero Auricularia. O objetivo deste trabalho foi investigar a produção de proteases de cogumelos comestíveis do gênero Auricularia para selecionar uma espécie produtora de proteases para futura aplicação biotecnológica. As espécies A. delicata e A. fuscosuccinea foram cultivadas em meio líquido Glicose-Extrato de Levedura-Peptona (GYP), e o bioprocesso foi conduzido a 150 rpm, a 30°C. Após 72 horas o extrato bruto foi recuperado por filtração a vácuo e, determinada a atividade qualitativa de proteases em Ágar leite, a atividade quantitativa foi definida usando como substrato azocaseína 1,0% (p/v). No extrato bruto obtido do cultivo da espécie que apresentou maior atividade proteolítica foi realizada a caracterização parcial quanto ao efeito do pH e temperatura na atividade das enzimas. Os resultados mostraram que, em meio sólido, as espécies de Auricularia se mostraram produtoras de enzimas proteolíticas e, em cultivo submerso, estatisticamente, o valor significativo da atividade proteolítica (44,33 U/mL) foi determinada no extrato bruto de A. delicata.
