An Experimentally Tested Scenario for the Structural Evolution of Eukaryotic Cys(2)His(2) Zinc Fingers from Eubacterial Ros Homologs
The exact evolutionary origin of the zinc finger (ZF) domain is unknown, as it is still not clear from which organisms it was first derived. However, the unique features of the ZF domains have made it very easy for evolution to tinker with them in a number of different manners, including their combination, variation of their number by unequal crossing-over or tandem duplication and tuning of their affinity for specific DNA sequence motifs through point substitutions. Classical Cys2His2 ZF domains as structurally autonomous motifs arranged in multiple copies are known only in eukaryotes. Nonetheless, a single prokaryotic Cys2His2 ZF domain has been identified in the transcriptional regulator Ros from Agrobacterium tumefaciens and recently characterized. The present work focuses on the evolution of the classical ZF domains with the goal of trying to determine whether eukaryotic ZFs have evolved from the prokaryotic Ros-like proteins. Our results, based on computational and experimental data, indicate that a single insertion of three amino acids in the short loop that separates the β-sheet from the α-helix of the Ros protein is sufficient to induce a structural transition from a Ros like to an eukaryotic-ZF like structure. This observation provides evidence for a structurally plausible and parsimonious scenario of fold evolution, giving a structural basis to the hypothesis of a horizontal gene transfer (HGT) from bacteria to eukaryotes. © 2013 The Author
The "Petakopadesa" with critical apparatus and commentary.
In editing the Petakopadesa I have made use of the following MSS:- B1. ) B2. ) B3. ) - Palm-leaf MSS in Burmese characters. S. - Paper MS in Sinhalese characters. Unsatisfactory as the MSS are from the point of view of an editor because of the abundance of corrupt readings, B3, however is comparatively more reliable than the others. B2. very closely follows B3, discrepancies being due to the scribe. Bl. and S. on the other hand differ materially from B2. and B3, both in arrangement and in the readings. I have taken the reading that is common to a larger number of the MSS, with a partiality however, for B3. The Petakopadesa is reputed to be the work of MahakaccSyana, the famous disciple of the Buddha. Every chapter of the Petaka end with affirming the authorship of MahakaccSyana. Gandha-Vamsa supports this and tradition in Burma confirms this view. This, however, cannot be accepted as true. Prof. Hardy is of opinion that the author's name was probably Kaooayana which was subsequently changed Into Mahakaooayana. The exact date of the Petaka is as yet undetermined. All that can be said at present is that the work was written sometime between the 5th century A.D. and the date of the later all Pali Canonical works. The Petakopadesa is divided into eight chapters or bhumis. These are the following (1) AriyasacoapakasanS-pathamabhllmi (2) Sasanapatth Snadutiyabhumi. (3) Sattadhitthana -tatlyabhumi. (4) Suttavieaya-oatutthabhuml. (5) HaraTlbhanga-pancamabhllml. (6) Suttatthasamuccayabhumi (7) Harasampatabhumi. (8) Suttarebhangiya. Of these eight bhumis Nos. (2) (5) and (7) hare got their corresponding counterpart in the Netti which is also the work of the same author. The same line of thought, the same outlook of the author can be observed in both the Netti and the Petaka
Complementary therapeutic effects of dual delivery of insulin-like growth factor-1 and vascular endothelial growth factor by gelatin microspheres in experimental heart failure.
Aims Strategies to prevent adverse left ventricular (LV) remodelling after myocardial infarction have included several traditional approaches and novel cell-based or gene therapies. Delivery of growth factors in post-infarction heart failure has emerged as a valuable alternative strategy. Our aim was to investigate the effects of sequential release of vascular endothelial growth factor (VEGF) and insulin-like growth factor-1 (IGF-1) from biodegradable gelatin microspheres in experimental heart failure. Methods and resultsGelatin hydrogel microspheres were known to guarantee a sustained release of encapsulated growth factors, characterized by an initial burst followed by a slower release. Rats with moderate myocardial infarction were randomized to receive empty microspheres (MI), microspheres loaded with IGF-1 or VEGF, or a combination thereof (DUAL). Myocardial injections of microspheres were performed at the time of surgery, and treatment lasted 4 weeks. Echocardiography, LV catheterization, morphometric histology and immunohistochemistry, and molecular assessment of downstream mediators [e.g. Akt, endothelial nitric oxide synthase (eNOS), and sarco/endoplasmic reticulum calcium ATPase-2 (SERCA-2)] were assessed at the end of the treatment period. Infarct sizes were 33 ± 2, 28 ± 4, 24 ± 3, and 16 ± 3 in the MI, IGF-1, VEGF, and DUAL groups, respectively. IGF-1 attenuated LV remodelling, improved LV systolic and diastolic function, increased myocyte size, and reduced apoptotic deaths, capillary loss, and indexes of inflammation. VEGF-treated animals displayed a marked myocardial neoangiogenesis that led to the formation of mature vessels if combined with IGF-1 delivery. Downstream effects of IGF-1 were principally mediated by the AktmTOR (mammalian target of rapamycin)-dependent pathway, and both growth factors, particularly VEGF, induced a robust and sustained increase of eNOS. ConclusionIGF-1 and VEGF exerted complementary therapeutic effects in post-infarction heart failure. Biodegradable gelatin microspheres provide sustained and controlled growth factor release locally, exposing myocardial tissue without the side effects of systemic administration. © 2011 The Author
The representation of the self and one's own story by witnesses who survived the horrors of history leads to an outcome in which the borders between history and story, subjective memory and archival documents, real events and individual perception of them, are always highly blurred and far from clear and well defined. Through Vittore Bocchetta's (b. 1918) own oral and written account of his life -in which history and story, memory and myth are intertwined- and its rewriting by Giuliana Adamo in her volume Una vita contro (2012), the author has tried to legitimise the individual account with an accurate verification against the counter history offered by archival documents. Thus the witness subjective voice becomes part of a polyphony which historians must try to comprehend
Ceija Stojka, a Roma author who was born and lived in Austria and survived the porrajmos, began publishing her writings some decades after her experience of the concentration camps to express the horrors she experienced first-hand. In her autobiographical writings, Stojka does not only talk about her two years as an inmate in Auschwitz, Ravensbrück and Bergen-Belsen, but also about her life before and after the war. While there is a strong need to write in order to bear witness to the experience and to process her trauma, her texts are also a journey into Roma history and into the most hidden spaces of her identity, that of her family and her people. His reflections often dwell on the contact with the majority society and the society’s perception of the Roma. The confrontation with the so-called gagé (non-Roma) helps to add extra pieces of identity and to highlight not only the differences, but also – beyond all stereotypes and categorizations – the similarities between human beings.

In applying any of the aforementioned models to the learning process, it is important to consider the specific characteristics of each learner and the abilities of the instructor
