The role of phosphorylation of ISGF3 components in the regulation of ISG expression and viral protection
Wydział BiologiiInterferony pełnią kluczową rolę we wzbudzaniu przeciwwirusowej odpowiedzi immunologicznej. W swojej klasycznej formie odpowiedź zależna od IFNα jest oparta na kompleksie ISGF3, składającym się z ufosforylowanych białek STAT1 i STAT2 oraz IRF9. Oprócz kanonicznej sygnalizacji w zapewnieniu ochrony przeciw wirusom zaangażowane są również inne kompleksy w tym U-ISGF3 (składający się z niefosforylowanych form STAT1 i STAT2 oraz IRF9) czy STAT2/IRF9 w swojej nie- lub ufosforylowanej formie. Stosując techniki molekularne, w tym także całogenomowe takie jak RNA- i ChIP-Seq, oraz przy użyciu JAK Inhibitor I, wyjaśniliśmy rolę jaką w odpowiedzi konstytutywnej, a także zależnej od interferonu i czasu, odgrywa proces fosforylacji. Zbadaliśmy dwa rodzaje linii komórkowych: typu dzikiego oraz pozbawione STAT1 i na tej podstawie wyciągnęliśmy wnioski dotyczące zależności ochrony przeciwwirusowej od ilości białek STAT1, STAT2 i IRF9 w komórce. Ponadto, eksperymenty porównawcze na komórkach ze stabilną nadprodukcją STAT1, STAT2 i IRF9 wykazały potencjalną rolę U-ISGF3 (i prawdopodobnie też U-STAT2/IRF9) w regulacji konstytutywnej oraz indukowanej IFNα przedłużonej ekspresji ISG i ochronie przeciw wirusom. To sugeruje, że musi zostać przekroczony pewien minimalny poziom białek STAT1, STAT2 i IRF9 w komórce (a co za tym idzie kompleksów U-ISGF3 i/lub U-STAT2/IRF9), aby doszło do zapoczątkowania ekspresji ISG. W konsekwencji, wraz z klasycznym kompleksem ISGF3, także U-ISGF3 i U-STAT2/IRF9 mogą odgrywać zasadniczą rolę w zależnej i niezależnej od IFNα transkrypcji ISG oraz w zwalczaniu infekcji wirusowych.Interferons play crucial role in the triggering of antiviral response, that in its most classical form is based on STAT1 and STAT2 that become phosphorylated and with IRF9 form ISGF3. Besides canonical ISGF3-dependent signalling, also other complexes function and may cooperate to provide effective viral protection. The examples are U-ISGF3 (that consists of unphosphorylated STAT1 and STAT2 together with IRF9) and STAT2/IRF9 in both unphosphorylated or phosphorylated form. Using different molecular techniques, among them whole-genome approaches, such as RNA-Seq and ChIP-Seq, in combination with JAK Inhibitor I, we provide further evidence on the role of phosphorylation in both, basal as well as IFN- and -time dependent transcriptional responses under wild-type and STAT1-deficiency conditions, we as well considered the dependence of viral protection on the amount of the native STAT1, STAT2 and IRF9. Additionally, comparative experiments in cells overexpressing STAT1, STAT2 and IRF9, revealed a potential role of U-ISFG3, and possibly U-STAT2/IRF9, in the regulation of constitutive and long-term IFNα-treated ISG expression and viral protection. This strongly suggests that a certain threshold of STAT1, STAT2 and IRF9 expression and levels of U-ISGF3 (and/or U-STAT2/IRF9) has to be reached to be able to trigger ISG transcription. Po kładce nad urwiskiem, przymrużając oko. O tłumaczeniu poezji humorystycznej na przykładzie Rymowanek Szymborskiej
The article presents a practical-theoretical reflection on translating humoristic poetry on the basis of the Author’s experience as a translator of Wisława Szymborska’s Rymowanki dla dużych dzieci. Since humor is the dominant function of the text, and laugh is the expected reaction, functional equivalence - rather than semantic equivalence - is here to be pursued. The goal of any translator of humoristic texts is to produce an equivalent effect on the reader. In the case of Szymborska’s Rymowanki, the task is made more difficult by the poetic form, which plays a primary role in generating the comic effect. The Author notices that the three categories of joke – universal, cultural and linguistic - proposed by Debra Raphaelson-West do not take into account such highly formalized utterings as humoristic verses, in which the form is not a mere decoration aiming at potentiating the aesthetic effect, but definitely contributes to generate the humoristic effect. The article shows how the mechanisms of laugh/smile proper of these poetic texts were reproduced in translation following some simple principles, and ends with the proposal to add to Raphaelson-West’s classification a fourth category: the poetic joke

Summary. Edge detection is an important step in image analysis. Using mathematical morphology and individual structural elements can detect certain types of edges. But much remains undetected. This paper proposes edge detection algorithm based on the structural element made up of many individual structure elements. The experimental results are showing.

Drowning in the blue, a vision of romantic love in Juliusz Słowacki's and Zygmunt Krasiński's letters
The epistolary consciousness of the romantics was exceeding the boundaries of the applied letter, romantic improvisation or the existential experiment - it was becoming an attempt to create a new language and new, extremely emotional form of artistic form of expression. The concept of the romantic open text adopted the idea of putting a special emphasis on the interactions between the letters (especially the poetic ones) - on their complementarity and permeation. What was the way in which Juliusz Słowacki and Zygmunt Krasiński were defining themselves and their relation as they were corresponding with each other during 9 years of
turbulent relationship? What do the letters say about those two authors and their real feelings? How do the letters of philomaths differ from the correspondence of poets who were "drowning in the blue"? In this presentation the author will try to refer to these and other elements of romantic language, to an emotion as a component of life-writing, and also to the similarities and differences which can be found in letters of Polish romantics. Neuroimaging methods for analysing connectivity in the presence of white matter lesions
White Matter Hyperintensities (WMHs) are often observed in the MRI scans of the ageing brain. Previous studies show that the WMH load correlates with cognitive decline, as well as with an increased risk of stroke and dementia. Most of the studies use a global WMH load across the whole brain as the only metric. This thesis proposes new insight into such data by introducing a methodology to analyse the WMH impact on the structural and functional brain connectivity in a localised manner, using white matter tracts. The thesis also supports other studies by exploring the potential impact of WMHs presence on tractography modelling, as well as adapts the method to Multiple Sclerosis (MS) lesion data.
In the first part of the thesis, we presented and evaluated two possible measures of the WMHs impact on the WM tracts from the structural perspective. We showed that despite the different distributions of the two measures, both show a similar relationship with the functional connectivity measure. We explored several possible options for quantifying the resting state functional connectivity between the endpoints of the WM tract in the presence of WMHs. We showed that the choice of connectivity measure (full or partial correlation), as well as gray matter parcellation, made a substantial difference in the results. We also observed variability in the results among the tracts and compared our findings to the results from the literature.
The majority of this work depends on the correct definition of the white matter tracts. Therefore, the second part of the thesis focuses on evaluating the impact of the presence of WMHs on tractography modelling. Despite changes in the microstructural parameters within the WMHs and their proximity, we found no meaningful alterations in the shape of the WM tracts in the presence of WMHs on the tract. Finally, we explored the possibility to apply some of this methodology to data from patients with Multiple Sclerosis, which is the most common cause of neurological disability in young adults. MS lesions have different aetiology to the WMHs but may have a similar appearance. We showed that directly applying the method between those two conditions may not be possible, and we proposed an alternative approach, suited to the MS data

The article is based on anthropological fieldwork basing on participant observation andinterviews conducted in 2014 during Program „Sigma” Erazmus Mundus 2 in Tuzla,Lukavac, Sarayevoand Mostar. The interviewees were the members of Polish minority inBosnia and Herzegovina engaged in Polish cultural organisations. The study aimed atunderstanding various types of bonds with Poland as a state or the culture and even familyhistories, as presented in interviews and behaviours. The author poses the question: whyintence development of Polish organisations flourished in the period of war in 1991-1995,inspite of the fact that Polish emigration to Bosnia started in the second half of 19th century. Presently the organizations are fading and even dissapearing. Cloud Computing in Sustainable Mobility
AbstractSustainable mobility takes into account a broader approach than sustainable transport as it is a concept integrating realization of the needs of stakeholders by using remote access to the properties of desired goods and services. It realizes the need to minimize the negative impact on the environment. Sustainable mobility remains one of the most important subject and challenge influencing European competitiveness. Transport activities apply to micro and macroeconomic issues, moving between and across various sectors, is still the highest of global logistics costs. By using cloud computing model in mobility and transportation management systems managers can gain reduction of the total cost of provided services for residents; more agility, flexibility, and elasticity; quick and cost-efficient reaction to less-predictable events and changing customer (or other stakeholders) requirements; risk reduction, globally accessible services, easy and fast implementation and strong support for sustainable development. The use of cloud computing in sustainable mobility is than mobility of people and cargos with respect to energy, environment, safety and security as well as socio-economic issues by giving access to real-time data, elasticized, fast reaction to the events and their negative consequences in available, efficient and affordable manner to all interested stakeholders. Cloud computing has already been noticed by European Union and exists in different acts and regulations’ recommendations, but it is definitely insufficiently analyzed and, most important, implemented in the mobility and transportation aspects
