    Aquaporin-4 mediates blood brain barrier permeability during endotoxemia

    Aquaporin-4 (AQP4) is a water channel expressed on astrocytes known to regulate the exchange of fluids and solutes between the parenchyma of the central nervous system (CNS) and perivascular spaces. We hypothesized that activation of AQP4 may mediate permeability of the CNS during systemic inflammation. Here, mice were i.v. injected with fluorescent tracers of low (1 kd) and intermediate (10 kd) molecular weights 24 hours after induction of endotoxemia with i.p. LPS. Fluorescent microscopy using tissue sections and cleared half-brains showed that endotoxemia increased tracer penetrance into the brain parenchyma. Temporal and spatial distribution of tracers suggested that penetrance occurred across neurovasculature, choroid plexus, and circumventricular organs (CVOs). Treatment with the AQP4 inhibitor, AER-271, blocked tracer passage across the glia limitans and caused tracer accumulation within perivascular spaces. These data show that AQP4 mediates the flux of solutes from the perivascular space to the brain parenchyma during systemic inflammation and highlight a novel strategy to neuroprotection during sepsis.Submission original under an indefinite embargo labeled 'Open Access'.     En la presente investigación se planteó el siguiente problema ¿Cuál es la relación entre el mensaje publicitario y la identidad de la marca del spot Huawei P8 del 2015 en los estudiantes del V ciclo de comunicaciones turno tarde de la Universidad Jaime Bausate y Meza en el 2016? Asimismo se determinó el siguiente objetivo: Identificar la relación entre el mensaje publicitario y la identidad de la marca del spot Huawei P8 del 2015 en los estudiantes del V ciclo de comunicaciones turno tarde de la Universidad Jaime Bausate y Meza en el 2016. Para ello se utilizó la técnica de las encuestas y el instrumento utilizado en este proyecto fue el cuestionario con un factor de validación del 91 % y confiabilidad de 0.90. Llegando a la siguiente conclusión, que efectivamente existe relación entre el mensaje publicitario y la identidad de la marca en base al spot “Huawei p8”, donde los alumnos encuestados manifiestan que se sienten atraídos con el mensaje del spot publicitario, que transmite los diferentes atributos del producto el cual genera una mejor identidad de marca

    En la presente investigación se planteó el siguiente problema ¿Cuál es la relación entre el mensaje publicitario y la decisión de compra en base al spot “Habla ilimitado a todos los claros del Perú”, en alumnos del V ciclo de periodismo turno tarde de la Universidad Jaime Bausate y Meza, 2016? Asimismo, se determinó el siguiente objetivo: Identificar la relación entre el mensaje publicitario y la decisión de compra en base al spot “Habla ilimitado a todos los Claros del Perú”, en base a los alumnos del V ciclo de periodismo turno tarde de la Universidad Jaime Bausate y Meza, 2016. Para ello se utilizó la técnica de las encuestas y el instrumento realizado en este proyecto fue el cuestionario con un factor de validación del 94% y confiabilidad de 0.85, llegando a la conclusión que efectivamente existe relación entre el mensaje publicitario y la decisión de compra en base al spot “Habla ilimitado a todos los claros del Perú”, donde los resultados al principio no fueron los esperados, sin embargo en el trabajo se manifestó que todo mensaje publicitario pasará por una decisión de compra para evaluar los atributos por más que esta sea una buena opción a simple vista, logrando así que se tome la mejor decisión para satisfacer la necesidad del comprador

Curso de Engenharia de Aquicultura.A ostra perlífera nativa Pteria hirundo possui um grande potencial para a aquicultura nacional devido a sua capacidade de produzir pérolas. Apesar do seu interesse econômico, nada ainda se conhece a respeito do seu sistema imune. Nesse contexto, o objetivo deste trabalho foi realizar um estudo preliminar das células imunes ou hemócitos (células circulantes da hemolinfa) dessa espécie, a fim de se conhecer melhor o seu sistema imunológico. Através de microscopia de luz, foi possível identificar, caracterizar e quantificar as populações de hemócitos presentes na hemolinfa. Os hemócitos de P. hirundo são semelhantes aos encontrados em outros moluscos bivalves. Foram identificados dois tipos principais de células: hemócitos com grânulos citoplasmáticos (hemócitos granulares ou HG) e hemócitos sem grânulos (hemócitos hialinos ou HH). Através de ensaios in vitro com microesferas fluorescentes de látex (2 µm de diâmetro), foi possível confirmar que os hemócitos apresentam capacidade fagocítica. Por meio da técnica de imunoflourescência indireta, não foi possível detectar a presença do peptídeo antimicrobiano mitilina nas células circulantes de P. hirundo. A participação dos hemócitos nas respostas de defesa imune foi avaliada 24 h após um estímulo microbiano com a bactéria marinha Vibrio harveyi. Apesar de não terem sido encontradas diferenças estatísticas na concentração total de hemócitos, a proporção de HG circulantes nos animais estimulados com V. harveyi e com salina foi 15,5 % maior do que em animais não estimulados. O presente estudo é de grande importância para o conhecimento de alguns aspectos do sistema imunológico da ostra perlífera nativa P. hirundo, podendo contribuir para a prevenção e controle de enfermidades nos cultivos.The indegenous pearl oyster Pteria hirundo has a great potential in aquaculture due to its capacity to produce pearls. Despite of its economic importance, little is known about its immune system. In this context, the aim of this study was to basically characterize the immune cells or hemocytes (circulating cells of the hemolymph) of this species, in order to increase the knowledge of its immune system. By using light microscopy, it was possible to identify, characterize and quantify the hemocyte populations found in the hemolymph. The pearl oyster hemocytes are similar to those found in other bivalve molluscs. Two main types of hemocytes were identified: cells with cytoplasmatic granules (granular hemocytes or HG) and agranular cells (hyaline hemocytes or HH). Through in vitro assays with fluorescent latex microspheres (2 µm diameter), it was possible to confirm the phagocytic capacity of these cells. Using immunofluorescence, it was not possible to detect the presence of the antimicrobial peptide mytilin in the circulating cells of P. hirundo. The involvement of hemocytes in the immune defense responses was evaluated at 24 hours after a microbial challenge with the marine Gram-negative bacterium Vibrio harveyi. Although no differences were found in the concentration of hemocytes, the proportion of circulating HG in animals stimulated with V. harveyi or saline was 15,5 % higher than in non-stimulated animals. The present study is of particular importance for the knowledge of some aspects of the immune system of the indigenous pearl oyster P. hirundo, which can contributes to prevention and control of diseases in cultured animals

En estas obras domésticas, el arquitecto se centra en construir, en los diversos contextos analizados, y atendiendo los requerimientos de los propietarios, el proyecto de una casa. Así pues, el trabajo se centrará en analizar las casas que Moneo concibe durante su trayectoria profesional. La investigación aborda las intenciones del arquitecto, las estrategias compositivas, las técnicas y el anhelo intelectual, tratando de indagar en las referencias utilizadas tanto en su investigación como en la docencia. A través de las casas construidas a lo largo de su trayectoria, esta investigación trata de aproximarse a la noción de casa de Rafael Moneo.Rafael Moneo’s architecture (1937) is known worldwide, being considered one of the most influential and renowned architects of his generation, leaving a noticeable mark on contemporary architecture. Even so, there is a branch little documented and investigated about this author: the houses. That is why the study focuses and delves into the house in the career of Rafael Moneo. In these domestic works, the architect focuses on building, in the various contexts analyzed, and meeting the requirements of the owners, the project of a house. Thus, the work will focus on analyzing the houses that Moneo conceives during his professional career. The research addresses the architect’s intentions, compositional strategies, techniques and intellectual desire, trying to investigate the references used both in his research and in teaching.     Leakage of the blood-borne neuroinflammatory molecule fibrinogen correlates with microglial immunofluorescence in the fetal brain during maternal immune activation

    Background: Influenza A virus (IAV) infection during gestation is associated with an increased risk of neurodevelopmental disorders (NDDs) in offspring, like schizophrenia and autism spectrum disorders. The pathophysiological mechanisms underpinning this association remain a subject of investigation. We posit that maternal immune activation (MIA) precipitated by IAV infection may disrupt selective permeability at the maternal-fetal interface, consequently compromising the integrity of the fetal blood-brain barrier (BBB). This compromised barrier function could lead to aberrant trafficking of neuroinflammatory blood-borne molecules into the fetal compartment, predisposing offspring to neuroinflammatory insults implicated in NDD etiology. Our study aims to: (1) investigate whether fibrinogen, a neuroinflammatory mediator, breaches the placental and fetal brain barriers (vascular or ventricular) during MIA, and (2) determine if this correlates with microglial immunofluorescence. Methods: C57Bl/6NTac pregnant mice were inoculated with mouse-adapted IAV (103 or 104 TCID50) or saline at gestational day 9.5. Expression of tight junction proteins (TJPs) and CD31+ endothelial cells in placentas and fetal brains was assessed via qPCR and immunohistochemistry. Co-staining of fibrinogen and Iba1+ cells (microglia/macrophages) was assessed in the subventricular zone (SVZ) of the lateral ventricle in fetal brains. Results: No statistical differences were seen regarding TJPs and CD31+ quantification in placentas or fetal brains. A significant increase in fibrinogen levels was noted in fetal brains exposed to 104 TCID50 IAV versus other groups. Moreover, a positive correlation between fibrinogen and Iba1+ staining at the SVZ was noted. Conclusion: Large neuroinflammatory glycoprotein fibrinogen can reach the fetal brain during IAV infection. We detected increased correlation in fibrinogen leakage and Iba1+ cell immunofluorescence mainly at the SVZ. Ongoing experiments will determine potential origin and trafficking of other blood-borne molecules that might be implicated in fetal neuroinflammation upon MIA

    Epidemiological studies link neurodevelopmental disorders (NDDs) with exposure to maternal viral infection in utero. The hypothesized mechanism governing this link comes from animal models that initiate maternal inflammation with a non-pathogenic viral mimic to induce an acute immune response. These studies indicate that maternal intestinal T helper 17 (TH17) cells are activated to produce interleukin (IL)-17, and this pro-inflammatory cytokine is implicated as a major driver of fetal brain abnormalities. Priming of TH17 cells is also observed following respiratory influenza A virus (IAV) infection; however, whether these TH17 cells might be driving fetal brain abnormalities during gestational IAV infection has never been examined. We aim to determine how IAV infection during pregnancy impacts maternal intestinal immune cells and if IAV-mediated activation of these cells is sufficient to result in aberrant brain development. To test our hypothesis, we inoculated pregnant C57BL/6NTac mice on gestational day (GD)9.5 with H3N2 IAV strain X31. Maternal serum, lungs, and intestine, placentas and fetal brains were collected on GD11.5 and 16.5, two- and seven-days post inoculation (dpi) to evaluate peak innate and adaptive immunity, respectively. Pregnant dams received 104 TCID50 X31 (X31hi; n=12, n=10), 103 TCID50 X31 (X31mod; n=14, n=9), or a mock-inoculation with saline (control; n=13, n=10) across three identical replicates per end point. Lung histopathology scores, viral expression, and inflammatory genes were upregulated in a dose- and time-dependent manner. Respiratory IAV infection led to colonic shortening at both time points despite no detection of virus in the intestine. Morphological changes were accompanied by upregulation of TH17 cell gene markers in the intestine at the high viral titer only. This includes genes encoding RORγt and IL-6, indicating priming of naïve T cells into TH17 cells. Flow cytometric analyses of intestinal TH17 cells at 2 and 7 dpi revealed a complex dose-dependent phenotypic shift. Placental immune transcripts were altered in a dose-dependent manner, demonstrating possible sequestering of adaptive immune cells and recruitment of neutrophils at the maternal-fetal interface. Immunohistochemistry revealed no changes in fetal microglial colonization patterns or proliferative capacities at GD11.5. Ongoing studies will analyze the fetal brain at GD16.5. So far, our data show that a higher infectious dose of gestational IAV is necessary to induce downstream immune changes, confirming the use of live pathogens in NDD modeling to evaluate the complete immune response and improve translation to the clinic

    Raza is, beyond its literary merit, an unusual text, since it is the first time that a chief of government justifies his victory in a fiction where himself is represented. In addition, the novel was destined to turn into cinematographic document, which is offered as example in the social context of an autarkic state. By means of the fiction, this first literary work establishes the main points of the moral commitment of the author, according to the concepts of mother land, nation, loyalty, etc.Raza, más allá de su valoración literaria, es un texto insólito por cuanto ningún jefe de estado o de gobierno en pleno ejercicio de su autoridad ha justificado su victoria por medio de una ficción donde, además, él mismo está visiblemente representado; menos aún que la novela sea algo provisional por estar destinada a convertirse en documento cinematográfico que, al tiempo, se ofrece como paradigma ejemplar en el contexto social de un estado autárquico. Este primerizo trabajo literario, entre sublimaciones argumentales e idealizaciones de personajes, establece, por medio de la ficción y con evidente huida de una realidad nociva para su honor, las líneas dramáticas de su «compromiso moral» según los conceptos de patria, nación, lealtad, etc., los cuales sustentan las bases de su personal ideograma vital. Lejos de formas literarias cercanas a la deshumanización del arte, como las generaciones de escritores coetáneos iban a establecer, el novelista se orienta hacia la expresión realista, si bien adornada con efectos de una retórica florida que aspira a funcionar como prosa de inspiración culterana
