    This dataset comprises experimental data gathered from 17 scientific articles, including different strains of this lactic acid bacteria, manufacturing scale (laboratory or pilot), fermentation conditions (medium composition, temperature, pH value and neutralizing solution for controlled pH, harvesting time), cooling and concentration conditions (temperature, speed, time,), composition of protective solutions, stabilization operating conditions (freezing and or drying), storage conditions (temperature and time). Survival rates (and functionality when available) at different steps of the production process are also included.     This dataset contains experimental data on the on the biological, physical and biochemical characterization of Lactobacillus delbrueckii subsp. bulgaricus CFL1 in the presence of three protective formulations: a galacto-oligosaccharides-contining extract from chickpeas cooking wastewater (GOS), produced in the frame of PREMIUM project (777657) (Martins et al 2023), a commercial fructo-oligosaccharides product from inulin (FOS), and sucrose, and at different steps of the stabilization process (freezing, freeze-drying and two weeks of storage at 28°C). The data set includes i) biological activity (culturability, acidifying activity, viability), ii) physical properties of the matrix (glass transition temperature and water content), iii) biochemical composition (Fourier transform infrared features), iv) biophysical (membrane integrity and phospholipid organization) and fermentation data (optical density and acidification rate as a function of time)

    Evaluation of the protein content of small extracellular vesicles derived from human melanoma cells treated with Doxorubicin

    ilustraciones, diagramasLas Vesículas Extracelulares (EV) son estructuras membranosas liberadas al espacio extracelular, que transportan biomoléculas y permiten la modulación de microambientes locales y distantes. En particular las células cancerígenas liberan EV, incluyendo vesículas extracelulares pequeñas (sEV) como exosomas, que participan en la comunicación intercelular. Estudios in vitro han mostrado que el tratamiento con Doxorrubicina (Doxo) incrementa el nivel de secreción de sEV. Este agente quimioterapéutico puede inducir respuestas de estrés celular que alteran el microambiente tumoral y, además, se ha asociado con efectos secundarios como la cardiotoxicidad. Este estudio evaluó el efecto de Doxo (10 nM) sobre la biogénesis, producción y contenido de sEV derivadas de células de melanoma humano A375, realizando un análisis multidimensional. A nivel intracelular, la inmunocitoquímica mostró un leve aumento en los marcadores característicos de sEV, destacándose especialmente CD81 a las 96 horas (p = 0,0083). Asimismo, se observó un incremento en las estructuras positivas relacionadas con como cuerpos multivesiculares y exosomas a las tinciones con naranja de acridina (A.O) y Bodipy-TR en células vivas a las 24 y 96 horas, respectivamente. Las sEV fueron aisladas del medio tras 48 horas de condicionamiento, utilizando cromatografía de exclusión por tamaño (SEC). El análisis por seguimiento de nanopartículas evidenció un aumento significativo en la concentración de sEV (13,6 veces; p = 0,000014), acompañado de cambios en la distribución de tamaños en la población tratada (Doxo: 135,4± 5,60 nm). Mientras, la microscopía electrónica de barrido (SEM) mostró morfologías compatibles con un proceso de vesiculación activa. Por otro lado, la detección de citoquinas mediante un arreglo de anticuerpos indicó un aumento en los niveles de KITLG, CXCL1, CXCL12, CCL5, VEGF, IL-3, IL-4 e IL-10, siendo TGF-β la más destacada (p = 0,0134). El análisis bioinformático sugirió que algunas de las citoquinas encontradas en estas sEV podrían estar implicadas en la progresión tumoral y en efectos cardiotóxicos inducidos por el tratamiento. En conjunto, los resultados evidenciaron que la exposición de células A375 a Doxo 10nM potencian la biogénesis y liberación de sEV, sugiriendo que la diseminación de sEV con elevado contenido de TGF-β puede contribuir a los efectos a nivel paracrino y autocrino en su microambiente, y a efectos secundarios atribuidos a la Doxo (Texto tomado de la fuente).Extracellular Vesicles (EV) are membrane-bound structures released into the extracellular space that transport biomolecules and enable the modulation of local and distant microenvironments. Cancer cells actively secrete EV, including small extracellular vesicles (sEV) such as exosomes, which play a key role in intercellular communication. In vitro, studies have demonstrated that treatment with Doxorubicin (Doxo) leads to an increased secretion of sEV. This chemotherapeutic agent can induce cellular stress responses that alter the tumor microenvironment and has also been associated with side effects such as cardiotoxicity. This study evaluated the effect of Doxo (10 nM) on biogenesis, production, and content of sEV derived from human melanoma A375 cells through multidimensional analysis. At the intracellular level, immunocytochemistry revealed a slight increase in characteristic sEV markers, with a notable rise in CD81 expression at 96 hours (p = 0.0083). Furthermore, an increased presence of structures associated with multivesicular bodies and exosomes was detected through acridine orange (A.O.) and Bodipy-TR staining in live cells at 24 and 96 hours, respectively. sEV were isolated from the conditioned medium after 48 hours using size-exclusion chromatography (SEC). Nanoparticle tracking analysis revealed a significant increase in sEV concentration (13.6 F.C; p = 0.000014), accompanied by changes in size distribution within the treated population (Doxo: 135.4 ± 5.60 nm). Meanwhile, scanning electron microscopy (SEM) showed morphologies consistent with active vesiculation. On the other hand, cytokine detection using an antibody array indicated increased levels of KITLG, CXCL1, CXCL12, CCL5, VEGF, IL-3, IL-4, and IL-10, with TGF-β being the most prominent (p = 0.0134). Bioinformatic analysis suggested that some of the cytokines in these sEV might be involved in tumor progression and Doxo-induced cardiotoxic effects.     Fernanda Fonseca disp Numéro National de Thèse : 2001INAP0003Ce travail a pour objectif d'evaluer l'effet des conditions de cryoprotection, de congelation et de stockage, sur l'activite acidifiante de ferments lactiques concentres congeles, afin de mieux maitriser leur qualite. Il vise egalement a mieux comprendre les mecanismes impliques dans la degradation de l'activite acidifiante, subie par les cellules pendant leur production et leur conservation. Les relations entre les proprietes thermiques des suspensions bacteriennes et leur activite biologique sont egalement recherchees. Une methode nouvelle, a ete developpee pour quantifier la resistance a la congelation et au stockage des bacteries lactiques. La methode repose sur une relation lineaire entre l'activite acidifiante et la duree de stockage a l'etat congele des suspensions bacteriennes. Elle permet de determiner la perte d'activite lors de la congelation et la vitesse de perte lors du stockage. La resistance de l. Bulgaricus cfl1 a la congelation et au stockage a ete evaluee en presence de 12 solutions de cryoprotecteurs. Les effets mesures, avant et apres congelation et lors du stockage, different selon la molecule consideree, ne sont pas lies entre eux et ne s'additionnent pas en melange. Les cellules bacteriennes seules ne sont pas caracterisees par une temperature de transition vitreuse (tg) determinee par dsc. C'est le milieu de resuspension qui determine la valeur de la tg des suspensions bacteriennes. L'influence de la vitesse de congelation et de la temperature de stockage sur la resistance des cellules depend du cryoprotecteur ajoute. Les molecules qui preservent le mieux les cellules pendant la congelation sont celles qui abaissent fortement le point cryoscopique. Pendant le stockage, la vitesse de perte d'activite acidifiante depend de l'ecart entre la temperature de stockage et la temperature de transition vitreuse (ts - tg). Lorsque ts devient superieure a tg - 20\ c, la vitesse augmente significativement, mais sa valeur depend du cryoprotecteur utilise

    The characters in O Cobrador, by Rubem Fonseca, either by vengeance, or as a response to a threat or to the dissatisfaction with the social place imposed on them, live situations of challenge towards the contrary power. Considering the corpus of the present work, which studies the violence and eroticism in the short stories in O Cobrador, as well as its links to power and resistance, according to philosopher Michel Foucault, such opposition will be found in the violent attitude of the main character in the title short story; in Mandrake s decision to pursue the elucidation of the crime, against the will of his powerful client; in the riot broken out by the convicted interns of the Onze de Maio Home; and in the socially condemned and forbidden passion of the Pierrô da Caverna. The establishment of parallels between the heroes in O Cobrador and other heroes found in comparable narratives by the same author and others, highlight this confrontation. Although they confront and win powerful forces, these fictional men and women are damned to frustration, given the fungibility of their triumphs.Os personagens de O Cobrador, de Rubem Fonseca, ou pela vingança, ou como resposta à ameaça sofrida ou à insatisfação com o lugar social que lhes foi imposto, desafiam o poder estabelecido e violam lei e ordem em busca de bens e corpos inacessíveis de outra maneira. Este trabalho analisa a violência e o erotismo em quatro contos de O cobrador, bem como sua inscrição na relação entre poder e resistência, com base nas reflexões do filósofo Michel Foucault.     This book entitled Vegetation Dynamics – Ecosystem Management, Conservation, and Protection has eight chapters, four related to vegetation dynamics and four related to monitoring and management. In the introductory chapter, Ana Cristina Gonçalves and Teresa Fidalgo Fonseca discussed the importance of species traits and sites in the sustainability of the forest systems as well as the importance of monitoring, modelling and management. Mariana Niculescu discussed the effects of the changes in management practices, invasive species, climate change and mining on meadows and forest systems in Romania. The author pinpointed the effects of unsuitable management on the sustainability of the agroforestry systems. Overall, maintaining agroforestry sustainability, with suitable management practices, would enable regenerating vegetation. Arvid Odland evaluated the effect of water reduction on forest floodplain wetlands. The species proportion and cover were influenced by its capacity for flowering and fruiting, which were also related to the temperature and water level. This review highlighted the importance of long-term studies as vegetation dynamics evolve slowly over time. Mallem Hamida evaluated the effect of vegetation on soil rehabilitation in pre-desert conditions, particularly in wind erosion mitigation and soil properties improvement in Algeria. Vegetation enhanced dune stabilisation due to its impact on wind erosion, and vegetation improved soil properties. However, variations were found when different species were compared. António L. Crespí et al. explored the concept of vegetation robustness and proposed a methodological approach to characterise the functional traits of vegetation and their potential correlation with environmental factors. The study indicated that the resilience of these areas was influenced by their resistance and was linked to environmental variability, particularly geomorphological and climatic factors. Damien Hicks reviewed the influence of sampling vegetation and a set of quantitative tools to analyse data. It included the optimisation protocols to enable the use of data for different analyses, from multivariate analysis to complex statistical analysis of data with network structure, machine learning methods and Bayesian methods. Fonseca et al. conducted a comprehensive review of the traits, silviculture and management of Pseudotsuga menziesii in Portugal. The authors analysed the area potentially suitable for establishing stands of this species in Portugal, as well as its potential growth and wood properties. Iglesias et al. discussed the effects of pure, even-aged plantations of Eucalyptus globulus and invasive species on biodiversity and using an invasive species (Phytolacca americana) to produce pulp and paper. The physical, mechanical, and chemical properties of Phytolacca americana confirmed that the species is suitable for producing paper in a pulp blend of the two species

    Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Ciências da Saúde, Programa de Pós-Graduação em Enfermagem, Florianópolis, 2013.Pesquisa qualitativa com abordagem sócio-histórica, com objetivo de historicizar o poder que as enfermeiras exerceram no processo de organização e implementação do Serviço de Enfermagem no Hospital Infantil Joana de Gusmão, no período de 1977 a 1991. Foi utilizada a técnica da História Oral Temática. Os sujeitos do estudo foram dez enfermeiras e um médico. A coleta de dados foi realizada através das fontes orais (entrevistas semiestruturadas) e fontes documentais (fotografias, jornais entre outros). A análise e interpretação dos dados foram realizadas através da Análise de Conteúdo Temática de Minayo e com base no referencial teórico de Michel Foucault. Este estudo foi aprovado pelo Comitê de Ética em Pesquisa do HIJG sob o parecer consubstanciado número 243.006, relatado dia 08/04/2013 e norteado também pela Portaria 466/12 do Conselho Nacional de Saúde. Os resultados obtidos são apresentados na forma de dois artigos. O primeiro artigo, "Hospital Infantil Joana de Gusmão de Florianópolis (SC): Os fazeres das enfermeiras no processo de organização do Serviço de Enfermagem a partir dos seus saberes (1979-1991)", teve como objetivo historicizar os fazeres das enfermeiras no processo de organização do Serviço de Enfermagem do HIJG a partir de seus saberes (1979-1991). Os depoimentos dos entrevistados desvelam o desafio enfrentado por estas enfermeiras para organizar e implantar um Serviço de Enfermagem de qualidade no HIJG e revelam as estratégias de saber-fazer utilizadas para alcançar seus objetivos. Conclui-se que a organização e implementação do Serviço de Enfermagem foi permeado por lutas e resistências. As enfermeiras através do exercício do seu poder garantiram seu espaço dentro do hospital, desencadeando muitas conquistas, em virtude de seus saberes. Pode-se inferir que aorganização do Serviço de Enfermagem do HIJG, trouxe grandes transformações para a saúde da criança catarinense. Os seus depoimentos expressam a todo o momento a busca pela autonomia da profissão e da especificidade da enfermagem, e ao final deste estudo, verificamos que estas enfermeiras alcançaram a sua autonomia, o seu poder, através da implantação de uma assistência de enfermagem qualificada e humanizada. E o segundo artigo, ?As relações de poder e suas repercussões no cotidiano da enfermagem no Hospital Infantil Joana de Gusmão de Florianópolis (SC) (1979-1991)?, objetivou historicizar o poder que as enfermeiras exerceram no processo de organização e implementação do Serviço de Enfermagem no Hospital Infantil Joana de Gusmão (1979-1991). Este estudo possibilitou identificar as lutas e as resistências enfrentadas por estas profissionais para organizar o Serviço de Enfermagem no HIJG e revelam o poder exercido pelas enfermeiras no processo disciplinar da assistência de enfermagem. Os resultados expressam que as enfermeiras do HIJG reinventaram a sua prática assistencial, demonstraram que as resistências encontradas no processo de organização do Serviço de Enfermagem, serviram de aprendizado e desconstrução de conceitos relacionados à profissão. Ficou claro que as relações de poder existem em qualquer instância, independente do nível de conhecimento dos profissionais e do lugar que ocupam na instituição, mas que este poder circula em rede e é dinâmico, e pode ser exercido por qualquer indivíduo. Os resultados apontam que além de respeito e reconhecimento profissional, estas enfermeiras desenvolveram através de seus saberes uma rede poder e as questões emergidas referentes à resolução de problemas, tiveram relação direta com a liderança e esta característica importante, permitiu a visibilidade delas no cumprimento de seu papel no cotidiano do trabalho.Abstract : Qualitative research of socio- historical approach, in order to historicize the power that nurses exerted on the organization and implementation of the Nursing Service at Hospital Infantil Joana de Gusmão process in the period 1977-1991. The technique of thematic oral history was used. The subjects were ten nurses and a doctor. Data collection was conducted through oral sources (structured interviews) and documentary sources (photographs, newspapers etc.). The analysis and interpretation of the data was performed using the qualitative analysis of Minayo and based on the theoretical framework of Michel Foucault. This study was approved by the Ethics Committee in Research of the opinion embodied HIJG under number 243 006, reported on 08/04/2013 , and also guided by Ministerial Order 466/12 of the National Health Council The results are presented in the form of two articles. The first article, "Hospital Infantil Joana de Gusmão de Florianópolis (SC): The doings of the nurses in the organization of the Nursing Service from their knowledge (1979-1991) process,"aimed to historicize the doings of the nurses in the process organization of the Nursing Service of HIJG from their knowledge (1979-1991). The interviewees' statements unveil the challenge faced by these nurses to organize and implement a quality nursing service in HIJG reveal the strategies and know-how used to achieve their goals. We conclude that the organization and implementation of the Nursing Service was permeated by struggles and resistances. Nurses through the exercise of its secured her place in the hospital, triggering many achievements, by virtue of their knowledge. It can be inferred that the organization of the Nursing Service of HIJG brought major changes to the health of Santa Catarina child. Their statements express all the time to search for autonomy and specificity of the profession of nursing, and by the end of this study, we found that these nurses have achieved its autonomy, its power through the deployment of a skilled nursing care and humane. And the second article,"Power relations and their impact on the practice of nursing in the Hospital Infantil Joana de Gusmão Florianópolis (SC) (1979-1991)", aimed to historicize the power that nurses exerted on the organization and implementation of the process Nursing Service Hospital Infantil Joana de Gusmão (1979-1991). This study identified the struggles and resistances faced by these professionals to organize the Nursing Service in HIJG and reveal the power exercised by nurses in the disciplinary process of nursing care. The results show that nurses HIJG reinvented their care practice showed that the resistance encountered in the organization of the Nursing Service of process served learning and deconstructing concepts related to the profession. It was clear that power relations exist in any instance, regardless of the level of knowledge among professionals and the place they occupy in the institution, but that this power flows and network is dynamic, and may be exercised by any individual. The results indicate that in addition to respect and professional recognition , these nurses have developed their knowledge through a network and power issues emerged related to problem solving, were directly related to the leadership and this important feature, enabled the visibility of them in fulfilling their role in daily work

    Jovens em devir: invenção de novas possibilidades de vida para além da institucionalização

    Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Filosofia e Ciências Humanas, Programa de Pós-Graduação em Antropologia Social, Florianópolis, 2014.A presente tese tem como temática a desinstitucionalização de jovens "egressas" de serviços de acolhimento institucional (abrigo e casa-lar), habitantes de Santa Catarina (Palhoça e Florianópolis) e Rio Grande do Sul (Porto Alegre). Para tanto, a desinstitucionalização é tomada como um processo amplo e complexo que implica mudanças, não só de lugar (desacolhimento), mas de estado, em especial em termos daquilo que as jovens podem vir a ser (devir), e também como contexto privilegiado para a emergência da dimensão vivencial dos sujeitos. Partindo dessa temática mais geral, a pesquisa buscou investigar as maneiras pelas quais estas jovens inventam/criam novas possibilidades de vida, a partir de condições que a princípio reforçariam sua vulnerabilidade e falta de potência, e também aquilo que elas podem vir a ser (devir) para além da institucionalização. A etnografia, realizada junto a oito jovens "egressas" de serviços de acolhimento (Isabelle, Nicole, Virgínia, Estella, Nina, Clarissa, Alice, Olívia), permitiu destacar fundamentalmente três modos pelos quais as jovens inventam/criam novas possibilidades de vida. Uma primeira via para a invenção estaria na prática de "circulação de crianças", a qual, ao invés de ser uma iniciativa da família ou mesmo do Estado, teve início pela ação das próprias jovens que, durante a infância, se desvencilharam, através da fuga, de diferentes situações de maus-tratos e exploração. A segunda está relacionada às maneiras pelas quais as jovens vivenciam a experiência da maternidade e de cuidado com os filhos, tendo em vista a relação que estabelecem com as políticas públicas assistenciais, com as novas tecnologias reprodutivas, com os seus companheiros e o trabalho, etc. E a terceira refere-se a uma espécie de "virada no jogo das relações de parentesco", em especial , quanto às relações que as jovens estabeleciam com os parentes (principalmente mãe, pai e irmãos) previamente ao acolhimento institucional. A análise dos modos de vida dessas jovens permitiu observar que estes têm como característica uma "espécie de improvisação contínua" que, ao contrário de revelar uma postura simplesmente reativa (marcada pela busca da sobrevivência) diante das adversidades, está relacionada àquilo que elas souberam criar, ou seja, à descoberta do que gostam e sabem fazer. Para contemplar essa diversidade de experiências, foi preciso conduzir a análise e a própria escrita etnográfica tendo em vista o desafio de compreender o sujeito de uma maneira que não estamos acostumados a ver. Ou seja, a improvisação contínua foi revelando não um sujeito acabado ou precário, mas sim configurações de sujeito que vão sendo constituídas contingencialmente e que apontam para novas possibilidades de devir.Abstract : The present thesis dwells upon the deinstitutionalizat ion of young women from foster care services in the states, habitants of Santa Catarina (Palhoça and Florianopolis) and Rio Grande do Sul (Porto Alegre), Brazil. Deinstitutionalizat ion is understood as a broad and complex process which involves changes, not only from one place to another (since they leave the institution), but also as a change in their state of being, especially when we consider what these young women could become (becoming). Likewise, deinstitutionalizat ion is understood as a fruitful context for the emergence of the experiential dimension of the subjects. Taking this general theme as a starting point, the research seeks to investigate the ways through which these young women invent/create new life possibilit ies based on conditions which, at first could reinforce their vulnerability and lack of potency, and it also explores what they could become beyond the boundaries of their institutionalizat ion. The ethnography, which took place with eight young women (Isabelle, Nicole, Virgínia, Estella, Nina, Clarissa, Alice, Olívia) who had been through the foster care system, enabled me to highlight three ways where they invent/create new life possibilit ies. The first path to invent ion can be found in the "children circulation" practice, a practice that, instead of being an init iat ive of the family or even the state, was an init iat ive of the young women themselves, who, during their childhood, ran away leaving behind different situations of abuse and exploitat ion. The second, is related to the way they experience motherhood and caring for their children. Here the idea is to think about these experiences in relation to the relat ionships they establish with welfare policies, with the new reproductive technologies, with their partners, with work, etc. The third path to invention deals with what one could describe as a ?turning tables on kinship?, notably reggarding the relat ionships they had with their kin (mainly mother, father, brothers and sisters) before their institutionalizat ion. The analyses of the ways of life of these young women, allowed me to observe that these ways of life have, as a prime characteristic, something like a "continuous improvisat ion". However, contrary to common belief, this "continuous improvisat ion" does not only mean a reactive attitude (given by a survival drive when faced with hardship), but it is related to what they knew and wanted to create. In other words, it is linked to their process of finding out what they knew and wished to do. In order to be able to contemplate the diversity of experiences it was necessary to build the analyses, and even the ethnographic text, taking into consideration the challenge of understanding the subject in a way not familiar to us. Thus, the notion of continuous improvisation helped to unfold not a final or precarious subject, but configurations of subjects, which are being constituted contingently and that signal to new possibilit ies of becoming
