Effects on Plant Growth and Reproduction of a Peach R2R3-MYB Transcription Factor Overexpressed in Tobacco

In plants, anthocyanin production is controlled by MYB and bHLH transcription factors. In peach, among the members of these families, MYB10.1 and bHLH3 have been shown to be the most important genes for production of these pigments during fruit ripening. Anthocyanins are valuable molecules, and the overexpression of regulatory genes in annual fast-growing plants has been explored for their biotechnological production. The overexpression of peach MYB10.1 in tobacco plants induced anthocyanin pigmentation, which was particularly strong in the reproductive parts. Pigment production was the result of an up-regulation of the expression level of key genes of the flavonoid biosynthetic pathway, such as NtCHS, NtCHI, NtF3H, NtDFR, NtANS, and NtUFGT, as well as of the proanthocyanidin biosynthetic pathway such as NtLAR. Nevertheless, phenotypic alterations in transgenic tobacco lines were not only limited to anthocyanin production. Lines showing a strong phenotype (type I) exhibited irregular leaf shape and size and reduced plant height. Moreover, flowers had reduced length of anther’s filament, nondehiscent anthers, reduced pistil length, aborted nectary glands, and impaired capsule development, but the reproductive parts including androecium, gynoecium, and petals were more pigmented that in wild type. Surprisingly, overexpression of peach MYB10.1 led to suppression of NtMYB305, which is required for floral development and, of one of its target genes, NECTARIN1 (NtNCE1), involved in the nectary gland formation. MYB10.1 overexpression up-regulated JA biosynthetic (NtAOS) and signaling (NtJAZd) genes, as well as 1-aminocyclopropane-1-carboxylate oxidase (NtACO) in flowers. The alteration of these hormonal pathways might be among the causes of the observed floral abnormalities with defects in both male and female gametophyte development. In particular, approximately only 30% of pollen grains of type I lines were viable, while during megaspore formation, there was a block during FG1 (St3-II). This block seemed to be associated to an excessive accumulation of callose. It can be concluded that the overexpression of peach MYB10.1 in tobacco not only regulates flavonoid biosynthesis (anthocyanin and proanthocyanidin) in the reproductive parts but also plays a role in other processes such as vegetative and reproductive development

    A guide to F-automatic sets

A self-contained introduction to the theory of F-automatic sets is given. Building on [Bell, Moosa, F-sets and finite automata, Journal de théorie des nombres de Bordeaux, 2019], contributions are made to both the foundations of this theory and to questions of a model-theoretic nature. Suppose Γ is an abelian group and F : Γ → Γ is an injective endomorphism. If (Γ, F) admits a "spanning set" then the notion of an F-automatic set can be defined. It is shown that this notion is independent of the spanning set chosen. A characterization of the existence of a spanning set is given in terms of certain functions on Γ, called "height functions". It is shown that if Γ is finitely generated then (Γ, F) admits a spanning set if and only if no eigenvalue of the matrix of F lies in the complex unit disk. A notion of sparsity among F-automatic sets, called F-sparsity, is studied. Outstanding questions from [Bell, Moosa, F-sets and finite automata, Journal de théorie des nombres de Bordeaux, 2019] are resolved, including independence from the spanning set chosen and closure under set summation. In addition, it is shown that sparsity can be characterized in terms of another natural class of functions introduced here, called "length functions". Model-theoretic tameness properties of F-automatic sets are studied. In the case where Γ is finitely generated, a combinatorial description is given of the stable F-sparse sets in terms of the F-sets introduced in [Moosa, Scanlon, F-structures and integral points on semiabelian varieties over finite fields, American Journal of Mathematics, 2004]. When Γ = ℤ, this description is extended to a characterization of the stable F-automatic sets. It is shown that if A ⊆ Γ is F-sparse then (Γ, +, A) is NIP. Automatic methods are used to show that the following structures have NIP theories: (ℤ, +, d^ℕ, ×↾d^ℕ) for d ≥ 2, (_p[t], +, t^ℕ, ×↾t^ℕ) for prime p ≥ 9, and (ℤ, +, <, d^ℕ) for d ≥ 2. (Here d^ℕ = {1, d, d^2, ...}, and likewise with t^ℕ.

Contract manufacturing service quality and its relationship with customer satisfaction and loyalty in F&B industry / Zulkefli Abdul Rahim

Business environment is highly competitive especially for the low entry barrier industry such as F&B industry, characterized by low high-tech involvement. As a result, the industry draws high number of players competing for the target markets. In order to sustain the business as well as to improve their financial performance, F&B manufacturers opt to offer contract manufacturing services (CMS) while marketing intermediaries contracted-out their products. There is a formal relationship established between the service provider and the buyer. However, the impact of service quality on retaining the business relationship particularly has not been empirically investigated. To fill this gap, this study explores the determinants of contract manufacturing service quality (CMSQ) and the interrelationship among CMSQ, customer satisfaction and loyalty of the F&B CMSs' customers in Malaysia. Four focus group interviews (28 participants) were held in this study using a semistructured guide questions and they comprised 12 contract manufacturers and 16 business customers. The participants were the key personnel, representing organizations that are actively engaged in CMS. Three new dimensions were discovered from the focus group interviews; product development capability, Halal and outcome quality in addition to the SERVQUAL dimensions

Evaluation of turbulence models for the prediction of flow properties in vegetated channels

    No full text
The performance of turbulence models was investigated to predict the flow and turbulence features of the vegetated channel using computational fluid dynamics (CFD). The Ansys Fluent, CFD software was implemented for the numerical studies. The flow was three-dimensional, incompressible, steady, and turbulent. Ten turbulence models, provided by Ansys Fluent, were implemented for the comparative study. The numerical model was validated against an experimental study conducted in the literature. The numerical studies show that the Renormalization group k-ϵ model is the most successful model for predicting the flow characteristics of the vegetated channel with a Root Mean Square Error (RMSE) value of 0.2752. At the same time, the Reynolds Stress Model gives the least successful predictive performance, indicated by an RMSE value of 0.4302. Moreover, the Spalart-Allmaras (S-A) model offers the shortest computation time with a value of 6652.393 s, whereas the Shear Stress Transport k-ω model proves to be the most time-consuming with a value of 11 952.219 s. The velocity of water flow in a channel is not uniform as it is slower at the surface of leaves and faster in the free zones. The maximum velocity is observed in the middle section of the channel, below the leaf, and between the roots with the value of u = 0.1158 m/s. Furthermore, the characteristics of turbulence in a channel are influenced by several factors such as channel geometry, flow velocity, and vegetation distribution. As a result, the presence of vegetation in a channel affects the flow and turbulence characteristics of the water significantly. © 2024 Author(s)

Three new dimensions were discovered from the focus group interviews; product development capability, Halal and outcome quality in addition to the SERVQUAL dimensions. Based on service quality literature and focus group discussions, relevant information via structured questionnaires were collected via online survey and drop-off method. 146 usable questionnaires were analysed using partial least squares. The results indicate that CMSQ consists of eight dimensions namely empathy, assurance, reliability, responsiveness, tangibles, outcome quality, Halal and product development capability. Using a one-tailed probability test with a significance level of 5%, all the three linkages are significant: CMSQ dimensions positively influenced customer satisfaction, customer satisfaction positively influenced loyalty and CMSQ positively influenced loyalty. CMSQ was found to have a small effect size on loyalty while customer satisfaction largely influenced loyalty. The result implies that contract manufacturing service quality is a complement in the formation of customer loyalty while customer satisfaction mediates partially the relationship between CMSQ and loyalty. The findings also provide empirical evidence that the contextual specific service quality is essential particularly for contract manufacturing in F&B
