    Molecular factors driving fibroblast growth factor receptor inhibitor resistance

    Lungenkrebs ist als kausativer Faktor in der Statistik krebs-assoziierter Todesfälle weltweit führend. Aufgrund eines meist späten Diagnosezeitpunkts in bereits fortgeschrittenem Stadium, eines oft aggressiven Wachstumsmusters und der limitierten Effizienz systemischer Chemotherapie bleibt die Prognose dieser Erkrankung äußerst schlecht. Große Fortschritte in der Erforschung der molekularbiologischen Grundlagen dieser malignen Erkrankung haben gezeigt, dass Lungenkrebs ein heterogenes Set an molekularen Subtypen umfasst, welche allesamt durch spezifische genetische und epigenetische Veränderungsmuster gekennzeichnet sind. Fibroblasten-Wachstumsfaktorrezeptoren (FGFRs) konstituieren eine Familie von Rezeptor-Tyrosinkinasen (RTKs) mit zentralen physiologischen Funktionen in der humanen Embryonalentwicklung und der Homöostase adulter Gewebe.^ ^Hyperaktivierte oder deregulierte FGFR Signalübertragung ist ein treibender Faktor der Tumorigenese bei multiplen Krebstypen, und so auch im Lungenkrebs. Ein zentrales Anliegen in der Therapieforschung ist daher die Entwicklung von Wirkstoffen, welche spezifisch auf die FGFR-Achse abzielen. Jedoch sind Resistenzmechanismen von Krebszellen gegen solche FGFR Inhibitoren bislang unzureichend erforscht. Außerdem sind die Nebenwirkungen von FGFR-gerichteter Therapie vielseitig aufgrund des breiten biologischen Spektrums der FGFR-Signalachse sowie deren Rolle in der Homöostase vieler Gewebstypen. Die hier vorliegende Dissertation hatte deswegen zum Ziel, die molekularen Mechanismen, welche der intrinsischen und erworbenen Resistenz FGFR1-getriebener Lungenkrebszellen gegen die äußerst wirksamen FGFR Inhibitoren Nintedanib und Ponatinib zugrunde liegen, zu erforschen.^ ^Molekularbiologische Analysen kombiniert mit integrativer Bioinformatik erlaubten uns, ATP-bindende Kassette (ABC)- Transporter-mediierten Efflux sowie die Induktion von MET Rezeptor als Schlüsselmechanismen im adaptiven Nichtansprechen von Lungenkrebszellen gegen diese Inhibitoren zu identifizieren. Folglich könnten ein Wechsel zu FGFR Inhibitoren, welche nicht als ABC Transporter Substrate fungieren, beziehungsweise die Applikation rationaler Kombinationsschemata mit MET-gerichteten Substanzen einer Resistenzentwicklung gegen Nintedanib und Ponatinib entgegenwirken. Untersuchungen zur intrazellulären Distribution dieser FGFR Inhibitoren zeigten überraschenderweise eine selektive Akkumulation von Ponatinib in Adiposomen, sowie von Nintedanib und PD173074, einem weiteren FGFR Inhibitor, in Lysosomen.^ ^Diese Sequestrierungseffekte stellten offensichtlich zell-intrinsische Resistenz-Mechanismen dar, die zur Reduktion zytosolischer Medikamentkonzentration -und somit zur Limitierung der Zytotoxizität- führen. Kombinationstherapie, welche die Integrität der genannten Zell-Organellen kompromitiert, repräsentiert eine mögliche Strategie, um die Arzneimittel-Target Interaktion zu erhöhen. Zuletzt wurde in dieser Arbeit im Rahmen eines Kollaborationsprojekts die biologische Aktivität liposomaler FGFR Inhibitor-Nanoformulierungen untersucht. Dieses Projekt hatte einerseits zum Ziel, die selektive intratumorale Akkumulation der Inhibitoren zu begünstigen, und andererseits, deren systemische Tolerabillität zu erhöhen. Liposomales Detargeting in vitro war gekennzeichnet durch verlangsamte zelluläre Aufnahmekinetik und durch reduzierte Zytotoxizität der nanopartikulären Form im Vergleich zu den jeweiligen freien Substanzen.^ ^Im Allograft in vivo Modell wurden verbesserte Antitumor-Effekte bei systemischer Administrierung von liposomalem Ponatinib, sowie im Gegensatz zur freien Substanz keine nennenswerten Nebenwirkungen registriert. Zusammenfassend wurden in dieser Dissertation neue Faktoren identifiziert, welche das Ansprechen von FGFR-getriebenem Lungenkrebs auf FGFR Inhibitoren beeinflussen. Außerdem wurden Strategien entwickelt, um die Effizienz dieses Therapieansatzes zu erhöhen.Lung cancer accounts for the highest number of cancer-related deaths globally. Due to the frequent diagnosis at advanced stages, aggressive growth patterns and limited efficacy of systemic chemotherapy, prognosis of this disease remains dismal. Major advances in the understanding of the molecular biology of this disease have revealed that lung cancer comprises a heterogeneous set of molecular subtypes, all characterized by specific genetic traits driving tumorigenesis. Fibroblast growth factor receptors (FGFRs) constitute a family of receptor tyrosine kinases (RTKs) that exert pivotal physiological functions in human embryonic and adult tissue. Hyperactivated FGFR signaling drives tumorigenesis in multiple cancer types, including lung cancer.^ ^Great effort has been laid on the development of molecularly-designed compounds that specifically target the FGFR axis. However, cancer cell and microenvironmental resistance mechanisms against FGFR inhibitors arise and are currently poorly understood. Furthermore, side effects of FGFR-targeted therapy are versatile, reflected by the broad biological spectrum of the FGFR signaling axis as well as its involvement in the homeostasis of many tissue types. This study, thus, aimed at dissecting the molecular mechanisms underlying acquired resistance of FGFR1-driven lung cancer cells against the promising FGFR inhibitors nintedanib and ponatinib.^ ^Molecular biological analysis in conjunction with integrated bioinformatics revealed ATP-binding cassette (ABC) transporter-mediated drug efflux and the induction of MET receptor to play key roles in the adaptive unresponsiveness of lung cancer cells against these compounds. Consequently, switching to non-ABC-substrate FGFR inhibitors or applying rational combination treatment with MET-targeting agents might circumvent acquired resistance development in case of nintedanib and ponatinib, respectively. Furthermore, characterization of the intracellular distribution of the investigated compounds revealed selective accumulation in lipid droplets for ponatinib and in lysosomes in case of nintedanib and of the experimental FGFR inhibitor PD173074. These sequestration effects represent cell-intrinsic resistance mechanisms by decreasing cytosolic drug concentrations and, thus, to limit cytotoxicity.^ ^Combination approaches compromising the integrity of these organelles represents a promising strategy to increase drug-target interaction. Lastly, in frame of a collaboration project, the biological activity of liposomal FGFR inhibitor nanoformulations was investigated in this thesis with the aim to promote selective drug accumulation at the tumor site as well as to increase systemic tolerability. Liposomal detargeting in vitro was characterized by slower cellular uptake kinetics and reduced activity of nanoparticulate as compared to free drugs. In allograft experiments, antitumor effects were achieved by systemic administration of liposomal ponatinib and, as opposed to the free drug, no adverse effects were observed.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Diese Studie hatte zum Ziel, das Zusammenspiel von MVM mit der antiviralen Aktivität in MEFs zu untersuchen, wobei der Prototyp- Stamm MVMp mit MVMCR verglichen wurde. Während für ersteres Virus eine effektive antivirale Antwort gezeigt werden konnte, welche zu einer abortiven Infektion in diesen Zellen führt, konnte letzterer Stamm propagieren, was seiner angenommenen Fähigkeit zugesprochen wird, diesen zellulären Abwehrmechanismus zu bekämpfen. Zu diesem Zweck wurde ein altes Isolat in MEFs amplifiziert und mit MVMp auf Replikationskapazität, genetische Differenzen sowie Induktion antiviraler Antworten untersucht. Potentielle zelluläre Sensoren sowie Zielpunkte viraler Bekämpfung der Wirts- Immunantwort sollten identifiziert werden. Genetische Unterschiede sollten die Dynamik aufdecken, Wachstumsfähigkeit in einer restriktiven Umgebung, bedingt durch eine angeborene Immunantwort des Wirts, wiederzuerlangen. Das Virusisolat zeigte sechs genetische Differenzen gegenüber dem gut charakterisierten MVMp- Stamm. Der Vergleich zwischen MVMp gegenüber MVMCR Infektion in MEFs zeigte interessante Befunde hinsichtlich der Suche nach Wirtszell- Sensoren und nach Angriffspunkten einer viralen Bekämpfungsstrategie: (i) SGTA, ein Tetratricopeptid- beinhaltendes Protein, wurde durch MVMp und MVMCR induziert und assoziierte mit post- nukleären Membranstrukturen. In Analogie zu den IFIT- Proteinen könnte obiges NS1- bindendes Protein als Wirtszell- Sensor für Parvoviren fungieren. (ii) Ein gemeiner Mechanismus von Viren, intrazelluläre antivirale Antworten zu unterdrücken liegt in der Identifizierung von Komplexen viraler Proteine mit Wirts- mRNA- Prozessierungsfaktoren. Unter drei NS1- interagierenden-, mRNA- modifizierenden Enzymen, DDX18, hnRNP-Q und CPSF6, wurde nur Letzteres ein spezifisches Ziel von MVMCR, jedoch nicht von MVMp zu sein. Es erscheint naheliegend, diesen mRNA- Prozessierungsfaktor als Ziel für parvovirale Bekämpfung zellulärer Immunantworten zu untersuchen

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
