Translational studies in X-linked adrenoleukodystrophy
De stofwisselingsziekte X-gebonden adrenoleukodystrofie (X-ALD) wordt gekenmerkt door gestoorde afbraak van zeer lange-keten vetzuren (ZLKV). Marc Engelen onderzocht mogelijk nieuwe medicijnen. Engelen concentreerde zich op twee stoffen: bezafibraat (dat de hoeveelheid vet in het bloed vermindert) en lovastine (een cholesterolverlagend medicijn). Bezafibraat leek veelbelovend, maar blijkt de concentratie ZLKV niet omlaag te kunnen brengen bij de mens, omdat zelfs bij de maximaal toegestane dosis de benodigde concentratie in plasma niet wordt bereikt. Lange tijd werden vrouwen met X-ALD beschouwd als draagsters, niet als patiënten. Een groot deel van de draagsters boven de 60 jaar ontwikkelt echter neurologische klachten en/of symptomen, zo blijkt uit een cohortstudie
From scientific communication to public knowledge : the scientific article web published as a knowledge base
Linking Electronic published scientific articles to Web ontologies is a cognitive tool of which its impacts and possibilities are far from being evaluated. The objective of this research is to investigate the potential of Web published scientific articles, conceived not only as texts, but also as a machine readable knowledge base, explicitly and formally related to Web-based public ontologies, that represent the assented knowledge of a specific domain. A prospective survey is developed to identify similar proposals and innovative experiences in electronically publishing scientific articles, authoring tools and citation analysis. Scientific methodology is also reviewed looking for structural characteristics of the scientific method presented in the written text of scientific articles. Experiences in developing Markup Language for some specific areas of knowledge, such as Chemical Markup Language, Mathematics Markup Language and Biology Markup Language, are also reviewed. An electronic publishing process is outlined which would permit the electronic publishing of not only scientific articles as full-texts, but would also enables an author to formalize the “deep structure” of a scientific article, containing assumptions, hypotheses, methodology, citations, datasets used, conclusions and contributions. All these elements are published as a knowledge base, using XML language, thus outlining a Sm-ML, a Scientific methodology Markup Language. Concepts expressed in the different parts of a Scientific article are to be linked to public Web ontologies, thus enabling the establishment of a formal relationship between the Scientific article specific knowledge base to ontologies like the UMLS – the Unified Medical Language System (http://www.nlm.nih.gov/pubs/factsheet/umls.html). The citations of an article are also be linked to the cited Web published scientific articles as qualified citations, in which the reasons to cite and the relationship between this specific scientific article and its citations are made explicit. The proposed model can enhances the scientific communication process, permitting semantic retrieval, critical inquiring, semantic citation, comparison, coherence verification and validating of a scientific article against public Web ontologies, which express the assented knowledge of a scientific area. The model was also conceived as the base for developing enhanced authoring and retrieval tools
De kanarie in de kolenmijn
Marianne Thieme en Ewald Engelen waarschuwen tegen de onhoudbaarheid van de huidige politieke en financiële keuzes. Als een kanarie in de kolenmijn signaleren zij gevaren die onze toekomst bedreigen. Dat het fossiele denken gedurende lange tijd zo'n beetje alle verzamelde reserves aan fossiele brandstoffen erdoor heeft gejaagd. Dat honger niet het gevolg is van te weinig voedselproductie, maar dat het vooral gaat over voedselverdeling en -verspilling. Dat de strijd om water er niet een is van de verre toekomst. De meeste politici en wetenschappers kiezen voor marginale veranderingen binnen de status quo waarbinnen we ons richten op meer economische groei en meer vrijhandel. Marianne Thieme en Ewald Engelen pleiten voor een radicale koerswijziging. Thieme vanuit ecologisch oogpunt, Engelen vanuit economisch perspectief. Ze komen tot een gemeenschappelijke conclusie: het financiële systeem en het politieke stelsel zijn volledig vermolmd. Plan B is onontkoombaar

The Canary in the Coal Mine
Ewald Engelen and Marianne Thieme warn against the unsustainability of today's politics and financial choices. Like a canary in a coal mine, they identify the dangers that jeopardise our future. Such as the fossil perspective, which has led to the near-complete depletion of the collected reserves of fossil fuels. Or the fact that famine is not the result of insufficient food production, but predominantly due to food distribution and waste issues. And that conflicts over water are not a thing of the distant future, but are already manifest in the present. The majority of politicians and scientists opt for marginal changes within the status quo, focused on more economic growth and increased free trade. Ewald Engelen and Marianne Thieme advocate a radical change of direction. Engelen from an economic perspective, Thieme from an ecological standpoint. They reach a common conclusion: the financial system and the political structure are rotten to the core. Plan B is unavoidable. Ewald Engelen (1963) is professor of financial geography at the University of Amsterdam, where he studies the global financial system. As a publicist and activist, he is well-known for his criticism of the euro and TTIP, the trade agreement between the United States and the European Union. Marianne Thieme (1972) is group leader for the Party for the Animals in the House of Representatives in the Netherlands, lawyer, animal rights and environmental activist and publicist. In 2002 she was one of the co-founders of the Party for the Animals
