    Aerodynamic shape optimization of rotary wing aircraft components using advanced multiobjective evolutionary algorithms

    The aim of this Doctoral Thesis, sponsored by AgustaWestland, is the design and development of a multi-objective optimization procedure that involves the application of the GeDEA-II, a powerful and time-saving evolutionary algorithm recently developed by the author at the University of Padova, able to perform multi-objective optimization analyses with the general approach of the Pareto frontier search. When compared to other state-of-the-art multi-objective evolutionary algorithms, it features novel crossover and mutation operators, and demonstrated superior performance. This optimizer supervises an automatic optimization loop involving the CFD commercial and free, open source solvers, respectively Fluent® and OpenFOAM®. Altair Hyperworks package is chosen as the free-form-deformation parameterization engine. The test cases chosen to demonstrate the strength of the procedure implemented concern the aerodynamic optimization of the AgustaWestland ERICA nose region, and the optimization of the intake 1 of the AW101 helicopter, that is really challenging problems from both the engineering and the industrial point of view. Starting from the the geometry elaboration and proceeding to the results discussion, each step of the optimization procedure is described in details, with particular focus on the automatic optimization loop, directly programmed by the author in both UNIX/Linux and Windows environments. The results obtained surely demonstrate the effectiveness of the multiobjective approach chosen to carry out this work. Furthermore, some suggestions for future improvements and developments are provided, with the purpose to increase the strength of the discussed multi-objective optimization tool.Lo scopo di questa tesi di Dottorato in Energetica, finanziata da AgustaWestland, consiste nella progettazione e sviluppo di una procedura di ottimizzazione multi-obiettivo, che comprende l’applicazione del GeDEA-II, un algoritmo genetico/evolutivo recentemente sviluppato dall’autore presso l’Università di Padova. Tale algoritmo permette di effettuare analisi di ottimizzazione multiobiettivo, sfruttando l’approccio del tutto generale che va sotto il nome di “Ricerca del Fronte di Pareto”. Rispetto ad altri algoritmi evolutivi multi-obiettivo “state-of-the-art”, esso presenta operatori di crossover e mutazione innovativi, che ne migliorano in maniera significativa le performance. Questo ottimizzatore è accoppiato con i codici CFD commerciali e gratuiti, rispettivamente Fluent® and OpenFOAM®. Il pacchetto Altair Hyperworks, codice ufficiale presso AgustaWestland per il pre-processing di fusoliere di elicottero, è scelto quale software per la parametrizzazione free-form. I casi test scelti per dimostrare l’efficacia di tale procedura consistono nell’ottimizzazione aerodinamica della regione frontale del tilt rotor dimostrativo ERICA, e della presa d’aria 1 dell’elicottero AW101. Tali casi costituiscono problemi stimolanti sia da un punto di vista puramente ingegneristico, sia da un punto di vista industriale. A partire dall’elaborazione della geometria, e procedendo con la discussione dei risultati ottenuti, ogni passo della procedura di ottimizzazione è descritto in dettaglio, con particolare enfasi dedicata al ciclo di ottimizzazione, sviluppato dall’autore sia in ambiente UNIX, sia in ambiente Windows. I risultati ottenuti dimostrano l’efficacia dell’approccio di ottimizzazione basato sull’algoritmo GeDEAII, scelto per sviluppare questo lavoro. Inoltre, vengono forniti alcuni consigli inerenti lo sviluppo futuro di questa procedura di ottimizzazione, con l’obiettivo di migliorare ulteriormente le capacità e la robustezza del ciclo di ottimizzazione

    Oral and parenteral immunization with synthetic retro-inverso peptides induce antibodies that cross-react with native peptides and parent antigens

    The objective of this study was to determine whether certain retro-inverso peptides have the potential to act as synthetic vaccines in mice, when immunized by injection or orally. Immunization of mice parenterally with conjugates of three such retro-inverso peptides and orally with the unconjugated peptides elicited generally high titres of anti-peptide antibodies. Antibodies against the same three peptides cross-reacted by binding strongly in ELISA to the native peptides and vice versa, regardless of the mode of immunization. Antibodies against a retro-inverso diphtheria peptide also reacted strongly with diphtheria toxin.     AVALIAÇÃO DO EFEITO TOXICOLÓGICO DE DOSES REPETIDAS DE IVERMECTINA EM RATAS FÊMEAS

[email protected] No início de 2020, instaurou-se a pandemia de Covid-19 a qual é uma doença causada pelo vírus Sars-cov-2 que provoca diversos sintomas como febre, cansaço, tosse seca, mas principalmente afeta as vias respiratórias, comprometendo os pulmões e dificultando as trocas gasosas que consequentemente leva a casos de intubação hospitalar. Ademais, centenas de milhares de casos culminaram em óbitos pela tempestade inflamatória causada pelo novo vírus que afetava também outros órgãos, além do pulmão, explicando, assim, sua mortalidade. Além disso, notou-se efeitos depressivos em pacientes acometidos pela doença, entretanto é necessário uma análise dos fatores que desencadeiam esses efeitos, visto que podem envolver a própria enfermidade, o tempo de internação ou o tratamento envolvido para resolução do quadro clínico. A depressão é caracterizada como uma doença mental que configura-se como sentimentos de tristeza, vazio e desesperança, causados por uma alteração dos neurotransmissores de serotonina e noradrenalina. Além desses sintomas, há presença de desinteresse por atividades cotidianas, hobbies e lazeres que contribuem para o estado psíquico e emocional debilitados, consequentemente alterando todo cotidiano e atividades do dia. Devido a agressividade e gravidade dessa doença, vários medicamentos foram utilizados como possíveis tratamentos para a Covid-19. Nesse ponto, a ivermectina, a qual é um fármaco utilizado para o tratamento de diversas infestações parasitárias, foi implementada sem comprovações científicas de sua eficácia. Tendo isso em vista, a presente pesquisa tem por objetivo avaliar os efeitos toxicológicos de doses repetidas de ivermectina em ratas fêmeas, utilizando a dose administrada para tratamento da Covid-19 durante a pandemia. Relacionado a materiais e métodos, a ivermectina foi administrada diariamente por cinco dias, na dose de 12mg/kg via intragástrica. Após, vinte e quatro horas da última administração os animais foram submetidos aos testes de comportamento onde analisou-se a função locomotora pelo teste de campo aberto e o comportamento tipo depressivo pelo splash test. Além disso, com o intuito de investigar se o efeito toxicológico permaneceria, um outro grupo de animais foi exposto ao mesmo protocolo de administração de ivermectina e, quatorze dias após a última administração do fármaco, os ratos foram avaliados nos mesmos testes comportamentais. Nossos resultados demonstraram que administrações repetidas de ivermectina diminuíram a locomoção dos animais 24h após a última administração. Entretanto, 14 dias após a última administração de ivermectina, os animais apresentaram um aumento nos parâmetros de locomoção no teste do campo aberto. Além disso, as administrações repetidas de ivermectina induziram um comportamento tipo depressivo nos animais 24h após a última administração e essa alteração permaneceu 14 dias após. Baseado nos nossos resultados podemos concluir que administrações repetidas de ivermectina foram tóxicas para os animais, pois induziu alterações locomotoras e um comportamento do tipo depressivo, entretanto mais pesquisas são necessárias para explicar os mecanismos de toxicidade envolvendo doses repetidas de ivermectina, bem como possíveis outros efeitos tóxicos. Além disso, outros estudos podem ser realizados para averiguar a possibilidade da ivermectina intensificar casos depressivos e de ansiedade

    The success of Imatinib (IM) therapy in chronic myeloid leukemia (CML) is compromised by the development of IM resistance and by a limited IM effect on hematopoietic stem cells. Danusertib (formerly PHA-739358) is a potent pan-aurora and ABL kinase inhibitor with activity against known BCR-ABL mutations, including T315I. Here, the individual contribution of both signaling pathways to the therapeutic effect of Danusertib as well as mechanisms underlying the development of resistance and, as a consequence, strategies to overcome resistance to Danusertib were investigated. Starting at low concentrations, a dose-dependent inhibition of BCR-ABL activity was observed, whereas inhibition of aurora kinase activity required higher concentrations, pointing to a therapeutic window between the two effects. Interestingly, the emergence of resistant clones during Danusertib exposure in vitro occurred considerably less frequently than with comparable concentrations of IM. In addition, Danusertib-resistant clones had no mutations in BCR-ABL or aurora kinase domains and remained IM-sensitive. Overexpression of Abcg2 efflux transporter was identified and functionally validated as the predominant mechanism of acquired Danusertib resistance <i>in vitro</i>. Finally, the combined treatment with IM and Danusertib significantly reduced the emergence of drug resistance <i>in vitro</i>, raising hope that this drug combination may also achieve more durable disease control <i>in vivo</i>

    Studies on the production, characterisation and applications of monoclonal antibodies and antibody-based conjugates

    The work described in this thesis involved the production of antibodies to several defined antigens and the chemical synthesis of bispecific antibodies. Monoclonal antibodies against glial fibrillary acidic protein (anti-GFAP), and against a GFAP-like protein on the human anaplastic astrocytoma cell line, G-CCM (anti-G-CCM) were produced by electrofusion and by fusion using polyethylene glycol, respectively. These antibodies were purified with saturated ammonium sulphate (S AS) and protein A. These antibodies were fully characterised by ELISA, Isotyping, SDS-PAGE, Immunocylochemisiry, Dot Blot, Western immunoblotting, BIAcore, HPLC, Immunofluoresence and FACS An attempt was made to clone and express the variable regions of the anti-G-CCM monoclonal antibody in Escherichia colt. Iodinated Bolton-Hunter reagent (IBHR) was conjugated to bovine serum albumin (BSA), ovalbumin and keyhole limpet haemocyanin (KLH). These IBHR-protein conjugates were characterised by the iodide assay and the Bradford protein assay. The IBHR-BSA conjugate was used to immunise six mice and a rabbit to produce anti-IBHR antibodies. A monoclonal anti-IBHR antibody was not obtained. However, a polyclonal anti-IBHR antibody was produced and was purified by SAS, protein A and CNBr-activated sepharose affinity chromatography. This antibody was characterised by ELISA, SDS-PAGE and HPLC Bispecific antibodies were chemically generated using (1) anti-GFAP and ana-IBHR and (u) anti-G-CCM and anti-IBHR Ricin is a highly cytotoxic compound Bispecific antibodies were made using an acquired anti-Ricm. A chain monoclonal antibody together with anti-G-CCM and anti-GFAP. These bispecific antibodies were characterised by ELISA, SDS-PAGE, HPLC and FACS and may be used to target radioactive compounds and toxins to astrocytoma cells, in vivo. A catalytic monoclonal antibody was generated using a transition state analogue-KLH conjugate as an immunogen in six mice. This antibody was precipitated using sodium sulphate and purified by protein G affinity chromatography. This antibody catalysed the hydrolysis of the corresponding carbonate and its rate kinetics were determined

    First observation of the thermal Sunyaev-Zel'dovich effect with kinetic inductance detectors

    abstract: Context. Clusters of galaxies provide valuable information on the evolution of the Universe and large scale structures. Recent cluster observations via the thermal Sunyaev-Zel’dovich (tSZ) effect have proven to be a powerful tool to detect and study them. In this context, high resolution tSZ observations (~tens of arcsec) are of particular interest to probe intermediate and high redshift clusters. Aims. Observations of the tSZ effect will be carried out with the millimeter dual-band NIKA2 camera, based on kinetic inductance detectors (KIDs) to be installed at the IRAM 30-m telescope in 2015. To demonstrate the potential of such an instrument, we present tSZ observations with the NIKA camera prototype, consisting of two arrays of 132 and 224 detectors that observe at 140 and 240 GHz with a 18.5 and 12.5 arcsec angular resolution, respectively. Methods. The cluster RX J1347.5-1145 was observed simultaneously at 140 and 240 GHz. We used a spectral decorrelation technique to remove the atmospheric noise and obtain a map of the cluster at 140 GHz. The efficiency of this procedure has been characterized through realistic simulations of the observations. Results. The observed 140 GHz map presents a decrement at the cluster position consistent with the tSZ nature of the signal. We used this map to study the pressure distribution of the cluster by fitting a gNFW model to the data. Subtracting this model from the map, we confirm that RX J1347.5-1145 is an ongoing merger, which confirms and complements previous tSZ and X-ray observations. Conclusions. For the first time, we demonstrate the tSZ capability of KID based instruments. The NIKA2 camera with ~5000 detectors and a 6.5 arcmin field of view will be well-suited for in-depth studies of the intra cluster medium in intermediate to high redshifts, which enables the characterization of recently detected clusters by the Planck satellite

    O artigo reflete o papel das exposições curriculares na contemporaneidade, experiência singular na formação do(a) profissional museólogo(a) ao estimular o debate críticoreflexivo sobre representações e deman das sociais. Na conjuntura do processo de curricularização da extensão revela-se uma atividade com alto potencial extensionista, prática que faz parte de sua essência for madora. A partir do curso de Museologia da Universidade Federal do Rio Grande do Sul (UFRGS) será examinado o viés social que as exposições curriculares têm caracterizado nessa graduação. Uma análise qualitativa de edições de exposições curriculares - deriva da de diferentes papéis assumidos pela autora nesses processos, ora como docente, comis são avaliadora ou visitante , contribui para compreender as bases que fundamentarão a destinação de parte de sua carga horária na integralização da extensão nessa graduação. Conclui que as exposições curriculares pos suem inquestionável dimensão extensionista ao serem capazes de reforçar a interação transformadora entre universidade e outros setores da sociedade.The article reflects on the role of curricular exhibitions in contemporary times, a unique experience of the museologist's professional training by stimulating a critical-reflective de bate on representations and social demands. In the context of the extension curricular crediting process, an activity with high exten sionist potential is revealed, a practice that is part of its formative essence. From the Mu seology course at the Universidade Federal do Rio Grande do Sul (UFRGS), the social bias that curricular exhibitions have charac terized in this graduation will be examined. A qualitative analysis of editions of curricular expositions - derived from the different roles assumed by the author in these processes, sometimes as a professor, evaluation commit tee, or visitor -, contributes to understanding the bases that will base the allocation of part of her workload in the payment of the exten sion in this graduation. It has been concluded that the curricular exhibitions have an un questionable extensionist dimension as they are capable of reinforcing the transforming interaction between the university and other societal sectors
