    Analisis Potensi Genetik Padi (Oryza sativa L.) Banyuwangi Berdasarkan Ekspresi Relatif Gen Homogentisate Geranylgeranyl Transferase (HGGT)

    Kualitas gizi buah padi yang baik sangat diminati konsumen di seluruh dunia, salah satunya adalah padi dengan tokotrienol tinggi. Ekspresi relatif gen HGGT dapat digunakan untuk menemukan varietas padi yang berpotensi menghasilka ntokotrienol tinggi. Penelitian ini bertujuan untuk menemukan varietas padi yang memiliki potensi tokotrienol tinggi berdasarkan tingkat ekspresi gen HGGT diantara 8 varietas padi (Oryza sativa L.) lokal Banyuwangi. Metode penelitian ini ialah quantitative Realtime–PCR untuk mengetahui ekspresi relatif gen HGGT pada padi lokal Banyuwangi. Hasil penelitian ini menunjukkan ekspresi relatif gen HGGT pada varietas Hitam Melik (2ΔCt = 16,989542), Hitam Pekat (2ΔCt = 5,376001), Blambangan A3 (2ΔCt = 4,943469), Merah Bali (2ΔCt=3,479590), Blambangan A2 (2ΔCt = 2,599881), Berlian (2ΔCt = 1,290429), JanurKuning (2ΔCt = 1,173945), dan Sunrise of Java A3 (2ΔCt = 0,356816). Hasil uji statistik non parametrik Wilcoxon/Kruskal Wallis menunjukkan ChiSquare hitung (16,0570)>ChiSquare tabel dengan α = 0,05 (14,067) yang berarti terdapat perbedaan signifikan pada data antar sampel. Ekspresi relatif gen HGGT pada padi lokal Banyuwangi dari yang tertinggi ke terendah adalah pada varietas Hitam Melik, Hitam Pekat, Blambangan A3, Merah Bali, Blambangan A2, Berlian, Janur Kuning, dan Sunrise of Java A3. Varietas Hitam Melik memiliki ekspresi relatif gen HGGT tertinggi sehingga berpotensi menghasilkan tokotrienol lebih banyak jika dibandingkan dengan varietas lainnya

    This book will not only tell about the keys to the success of Thailand in cultivating durian, but also durian marketing. It does not only contain the author's personal experiences while in Chanthaburi Province but also contains statistical data that supports the statement, as well as other reliable sources. This book is the result of an internship program that has been carried out by the author while in Thailand, especially in Chanthaburi Province, Office of Agricultural Research and Development Region 6 Chanthaburi Province. An internship program was carried out to increase knowledge and experience of agricultural conditions in Thailand which is known to be advanced. The author would like to thank Assoc. Prof. Am On Aungsuratana as the main supervisor during the program in Thailand and Dr. Rapee Dokmaithes. In addition, thanks are also conveyed to all staff and parties involved while the author is in OARD Region 6 so the author can run and complete the internship program smoothly, as well as write about their success in developing durian in Thailand. In addition, the authors also thank the University of Jember for all the knowledge provided.     Analysis of Genetic Potential of Banyuwangi Local Rice (Oryza sativa L.) Based on Relative Expression of Homogentisate Geranylgeranyl Transferase (HGGT) and Granule-Bound Starch Synthase I (GBSSI) Gene

    Rice nutrition including vitamin and amylose contents become important aspect for many people around the world. Rice with high amylose content (low glycemic index) is good for those with Diabetes mellitus. Tocotrienol, one precursor of Vitamin-E biosynthesis is catalyzed by enzymes encoded HGGT, while amylose biosynthesis is catalyzed by enzymes encoded GBSSI. The objective of this study was to find rice varieties with high tocotrienol and/or amylose content based on the expression of HGGT and GBSSI among eight Banyuwangi local rice varieties. Relative expression of HGGT and GBSSI was measured by qRT-PCR and analyzed using 2ΔCt method. Statistical analysis resulted in the significantly different of HGGT and GBSSI relative expression among samples. Relative expression of HGGT from the highest to the lowest were demonstrated by Hitam Melik, Hitam Pekat, Blambangan A3, Merah Bali, Blambangan A2, Berlian, Janur Kuning, and SOJ A3, respectively; while relative expression of GBSSI from the highest to the lowest were demonstrated by Hitam Melik, Hitam Pekat, SOJ A3, Janur Kuning, Berlian, Merah Bali, Blambangan A3, and Blambangan A2, respectively. Based on this research we conclude that Hitam Melik potentially produces higher tocotrienol and lower glycemic index than other studied varieties
