    The splicing landscape is globally reprogrammed during male meiosis

    Meiosis requires conserved transcriptional changes, but it is not known whether there is a corresponding set of RNA splicing switches. Here, we used RNAseq of mouse testis to identify changes associated with the progression from mitotic spermatogonia to meiotic spermatocytes. We identified ∼150 splicing switches, most of which affect conserved protein-coding exons. The expression of many key splicing regulators changed in the course of meiosis, including downregulation of polypyrimidine tract binding protein (PTBP1) and heterogeneous nuclear RNP A1, and upregulation of nPTB, Tra2β, muscleblind, CELF proteins, Sam68 and T-STAR. The sequences near the regulated exons were significantly enriched in target sites for PTB, Tra2β and STAR proteins. Reporter minigene experiments investigating representative exons in transfected cells showed that PTB binding sites were critical for splicing of a cassette exon in the Ralgps2 mRNA and a shift in alternative 5' splice site usage in the Bptf mRNA. We speculate that nPTB might functionally replace PTBP1 during meiosis for some target exons, with changes in the expression of other splicing factors helping to establish meiotic splicing patterns. Our data suggest that there are substantial changes in the determinants and patterns of alternative splicing in the mitotic-to-meiotic transition of the germ cell cycle

    Стаття присвячена розгляду ментальної специфіки релігійного світогляду Т. Шевченка. У ній представлено основні характеристики релігійності українського народу та проаналізовано вплив етнопсихологічних чинників на їхнє формування. У процесі дослідження з’ясовано, що релігійне світобачення поета тісно пов’язане з українською ментальністю (The article is dedicated to the analysis of the mental base of T. Shevchenko’s religious world outlook. The author elucidates the main traits of the Ukrainian people’s religiousity and analyses the influence of ethnopsychological factors on their development. According to research it makes a conclusion that the poet’s religious world outlook intimately connected with Ukrainian mentality. The lines of this connection were such creations spiritual and world outlooking as: a Christian “cult of the man”, Christian “patriotism”, religious and cult’s aesthetics, “gospelіous Christianity”, religious tolerance. The reflection of deep ethnoreligious bases it watches in the poet’s understanding of God, his creation of image of God’s Mother also in his refusal of religious asceticism. In this article it was investigated that the belief in world outlook of Ukrainian’s people is an individual vital position. The personality always has a charter to choose a religion as for own internal ideal. The Ukrainians are the actors, but not the spectators in their contacts with God. The author believes, that a recognition of a value of every man and different ways of its liberty (in religious sphere also) generated a respect оf Ukrainians to spiritual achievements of other peoples. The Christian “cult of the man”, which was linked with religious tolerance, takes an important place in T. Shevchenko’s world outlook and his creative inheritance. It is an evidence of his spiritual connection with ethnoreligious views. It was elucidated in this article that strict religious asceticism and deep obedience of believers were unacceptable nor for the poet neither for Ukrainians. The God, in their religious views, loves a man. The personality feels an intimate connection in communication with Him, but not a panic fear. The God is their hope and support. He is a personification of justice and, at the same time of love to every man. Such combination permitted to Ukrainians and T. Shevchenko to address freely to God and demand of organizing the order in society. The one of personal traits of T. Shevchenko’s religious views that he stressed in life and creation on the “natural” religion, which rejects a ritual formalism, corresponds to high spiritual and mental needs of the every man. In generally, Ukrainian’s people, in spite of certain liberty in own spiritual world outlook, is very religious, analogously, the religion also took an important place in T. Shevchenko’s life

    У статті проаналізовано жанрові особливості есею-меморіуму як окремого різновиду письменницького есею. Закцентовано увагу на специфіці цього твору в порівнянні з некрологом і портретним есеєм, наголошено на його місці і ролі в сучасному письменстві. Спеціально досліджено окремі есеї in memoria у творчості В. Даниленка, зокрема присвячені Н. Зборовській та Ю. Ґудзю. Наголошено на ролі і специфіці низки мотивів у цих творах, своєрідності заголовків, образній характеристиці, функціях деталей, полідискурсивності тощо. Зроблено висновки про есей in memoria як окрему дискурсивну практику, що виникла внаслідок поєднання декількох жанрів і дискурсів, наділену особливими можливостями створення образу померлої людини і образу автора водночас.The article analyzes the genre features of the memorial essays as a separate type of essay. Emphasis is placed on the specifics of this work in comparison with the obituary and portrait essays while high lighting its place and role in modern writing. Some in memoria essays in the works of V. Danylenko, dedicated to N. Zborovska and Y. Gudz, have been specially studied. The role and specificity of a number of motives in these works, originality of titles, figurative characteristics, functions of details, polydiscursivity, etc. are emphasized. Conclusions are made about the inmemoria essayas a separate discursive practice, which arose as a result of a combination of several genres and discourses, endowed with special opportunities to create the image of the deceased and the image of the author at the same time

    Comparative study of the pharmacological effects of venarus plus, venarus, and detralex on l-name-induced endothelial dysfunction, venous tone and platelet aggregation

    In the present study we compared the pharmacological activity of Venarus Plus, Venarus and Detralex 1000 mg on the reversion of endothelial dysfunction (ED), and on the effect on venous tone, vascular permeability, and platelet aggregation. We used 150 Wistar male rats, weighing 180-220 g, and 80 adult albino rabbits weighing 2800-3200 g. Endothelial dysfunction (ED) was induced with the non-selective inhibitor of NO synthase, N-nitro-L-arginine-methyl ether (L-NAME). Functional vascular tests and biochemical markers were used to determine the reversion of the functional disorders. The anti-inflammatory effects of the drugs was evaluated in rabbits using o-xylene. The venotonic effects of the compounds was carried out on an isolated segment of the rat portal vein with Ca solutions at a concentration of 0.08-1.75 mM. Our results show that the maximum daily therapeutic dose of Venarus Plus, produces a significant decrease in the ED coefficient (СED), an increase in NO synthesis, and an extended ADP-induced platelet aggregation time. The studied drugs dose-dependently reduce vascular permeability disorders caused by the application of o-xylene, which was manifested in a profound decrease the size of spots and the extension of the time interval before their onset. To study the Ca -mediated smooth muscle response, showed that the maximum force of vein contraction occurs with a higher dosage of drugs in the presence of a lower concentration of Ca , the effects of the drugs are comparable. 2+ 2+ 2

    Increased flexibility and plasticity of the ligament-articular apparatus are among the main criteria for the qualifying of future athletes. However, high incidence of spinal injuries, early articular diseases and their complications in athletes are a major problem which often turns into a tragedy: retirement from sport at the beginning of a successful sports career. Joint hypermobility syndrome and a high risk of injuries and lesions of the musculoskeletal system are interrelated: most often they occur against the background of connective tissue dysplasia. Undifferentiated connective tissue dysplasia (UCTD) is a widespread, polygenic, hereditary, multi-syndrome state, against the background of a metabolic disturbance and self-assembly of connective tissue, particularly collagen, elastin and associated proteins and proteoglycans. Marfanoid, Ehlers-like and mixed (dysplastic) phenotypes are distinguished in the structure of UCTD. Generally, children and young people of mixed (dysplastic) phenotype have the greatest potential for sport.
