    Hha Is a Negative Modulator of Transcription of <i>hilA</i> , the <i>Salmonella enterica</i> Serovar Typhimurium Invasion Gene Transcriptional Activator

    ABSTRACT An early step in the establishment of Salmonella enterica serovar Typhimurium murine infection is the penetration of the intestinal mucosa of the small intestine. The majority of the genes responsible for the Salmonella invasive phenotype are encoded on Salmonella pathogenicity island 1, and their transcription is controlled by the hilA transcriptional activator. The expression of hilA is regulated by environmental signals including oxygen, osmolarity, pH, and growth phase such that the presence of any one suboptimal condition results in repression of hilA expression and the invasive phenotype. We have conducted a search for negative regulators of hilA by introduction of a Salmonella enterica serovar Typhimurium chromosomal DNA gene bank into a Salmonella enterica serovar Typhimurium hilA ::Tn 5lacZY reporter strain. This screen has identified the hha gene as a regulator that exerts a negative influence on hilA expression. Plasmid-encoded hha significantly reduces hilA ::Tn 5lacZY chromosomal expression, as well as expression of the invasion genes invF , prgH , and sipC . An hha null mutation results in substantial derepression of both chromosomally encoded and plasmid-encoded hilA ::Tn 5lacZY expression. Introduction of plasmid-encoded hha into strain SL1344 results in attenuation of invasion using in vitro and in vivo assays. Importantly, purified Hha protein was found to bind to a hilA DNA promoter fragment, suggesting that the regulatory activity of the Hha protein occurs at the hilA promoter. These data add detail to the developing model of the regulation of Salmonella invasion genes. </jats:p

    This book is the first comprehensive analysis of the contemporary representation of the author on screen. It does this through two main approaches: by looking at how biographies of well-known authors in Western culture have been adapted onto the film and television screen; and by examining the wider preoccupation with the idea of what the ‘author persona’ means in broader economic, cultural, industrial, and ideological terms. Drawing from current debates about the uses of the heritage industry and conventions of the Hollywood biopic and celebrity culture, this book re-frames the analysis of the author on screen in contemporary culture and theorises it under its own unique genre: the ‘literary biopic’. With case studies including adaptations of the biographies and cultural personas of William Shakespeare, Jane Austen, Oscar Wilde, Sylvia Plath, Virginia Woolf, and Allen Ginsberg—to name a few–this book examines how and why the author continues to be a prominent screen and cultural preoccupation

    Campur kode dalam film tentu menarik untuk dikaji. Salah satunya film La Hila Donggo yang berasal dari Bima, diangkat dari cerita daerah setempat dengan memadukan unsur bahasa Bima dengan bahasa Indonesia serta memiliki nilai-nilai yang patut dipelajari. Penelitian ini bertujuan untuk mengetahui bentuk campur kode dalam film La Hila Donggo serta faktor penyebab terjadinya campur kode. Penulisan artikel ini menggunakan jenis penelitian deskriptif kualitatif dengan metode penelitian yang digunakan yaitu metode simak dan menggunakan teknik catat untuk memperoleh data lisan. Adapun analisis data menggunakan metode padan dengan menggunakan teknik hubung banding menyamakan. Langkah-langkah dalam analisis data yaitu, menampilkan data bentuk campur kode, menganalisis bentuk campur kode, mengklasifikasikan bentuk campur kode, menyamakan bentuk campur kode yang sesuai dengan artinya, dan menarik kesimpulan bentuk campur kode. Adapun hasil penelitian ini ditemukan 28 bentuk campur kode ke dalam mencakup kata 17 data seperti, ori,santabe, ina, nami, nahu, mada, nggomi, ompu. Kemudian frasa 7 data seperti, dou Donggo, dana Mbojo, ana mone, lenga mada. Selanjutnya, klausa 4 data seperti, hademu nahu lenga, lao nggomi, nahu eda, lembo ade ana. Selanjutnya, adapun faktor yang menyebabkan terjadinya campur kode yang terjadi dalam film La Hila Donggo meliputi, (a) faktor kebahasaan, (b) faktor kebiasaan, dan (c) faktor sikap penutur

    Characterization of the SPI-1 and Rsp type three secretion systems in Pseudomonas fluorescens F113

    Pseudomonas fluorescens F113 is a plant growthpromoting rhizobacterium (PGPR) isolated from the sugar beet rhizosphere. The recent annotation of the F113 genome sequence has revealed that this strain encodes a wide array of secretion systems, including two complete type three secretion systems (T3SSs) belonging to the Hrp1 and SPI-1 families. While Hrp1 T3SSs are frequently encoded in other P. fluorescens strains, the presence of a SPI-1 T3SS in a plantbeneficial bacterial strain was unexpected. In this work, the genetic organization and expression of these two T3SS loci have been analysed by a combination of transcriptional reporter fusions and transcriptome analyses. Overexpression of two transcriptional activators has shown a number of genes encoding putative T3 effectors. In addition, the influence of these two T3SSs during the interaction of P. fluorescens F113 with some bacterial predators was also assessed. Our data revealed that the transcriptional activator hilA is induced by amoeba and that the SPI-1 T3SS could potentially be involved in resistance to amoeboid grazing

    Modified Open Circuit and Vacuum-assisted Venous Return Reduces Blood Usage During Cardiopulmonary Bypass

    Objectives: To determine whether vacuum-assisted venous return has clinical advantages over conventional gravity drainage apart from allowing the use of smaller cannulas, shorter tubing and reduced priming. Methods: A total of 80 CABG operations were performed at our institution between July 1999 to December 2010, using vacuum-assisted venous return with small venous cannulas connected to short tubing. These were randomized with 80 CABG operations using conventional gravity drainage. Priming volume, hematocrit value, red blood cell usage, and total blood product usage were compared by means of multivariate analysis. Results: The priming volume was 780+/-140mL for small-cannula vacuumassisted venous return, 1300+/-88mL for gravity drainage (P <.0001). Smaller priming resulted in higher hematocrit values both at the beginning of cardiopulmonary bypass (26%+/-5% compared with 21%+/-4%, respectively, P <.0001) and at the end (28%+/-4% compared with 24%+/-4%, respectively, P <.0001). Red cell transfusions were used in 12% of the patients having small-cannula vacuum-assisted venous return and 41% of the patients having gravity drainage (P =.001); total blood product usage was 15% and 61%, respectively (P =.001). Despite a postoperative blood loss, length of stay in intensive care unit was similar in both groups; the association of vacuum-assisted venous return with lowered blood product usage was confirmed also in the postoperative period. Conclusions: Modified open circuit and vacuum-assisted venous return result in 1) higher hematocrit values during cardiopulmonary bypass and 2) decreased red cell and total blood product usage

    Transplantation Pneumonia

    A clinically distinct pneumonitis occurred in six renal transplant recipients receiving azathioprine and prednisone immunosuppressive therapy. The patients ranged in age from 3 to 20 years. The onset was 42 to 102 days postoperatively, coinciding with decrease in prednisone dosage below approximately 1 mg per kilogram of body weight per day. Mild nonproductive cough, fever, and cynanosis were present. Chest x-rays demonstrated extensive hazy to nodular infiltrates usually involving both hila and lower lung fields. Cold agglutinins were present in five patients. Pulmonary function studies demonstrated an alveolar capillary block. The duration of illness was 12 to 34 days and was not influenced by antibiotic therapy. Autopsy of the single case in which death occurred revealed Pneumocystis carinii pneumonia and disseminated cytomegalic inclusion-body disease. © 1964, American Medical Association.     Queer Israelis in Berlin:Belonging, Resistance, and the Politics of Emigration

    Nathan: ‘My parents still call me, even today, asking “so, what about a boyfriend?” I tell them, “listen, I have a boyfriend every day! Every day I have a new boyfriend”. And then the question is of course, “do you want to be alone?” And this is another reason why it is better to live in this country [Germany], where being alone is not the Mark of Cain.’ Sexual orientation and left-wing political affiliation often play significant roles in decisions to leave Israel. Queer Israelis question nationalist and heteronormative expectations regarding army service, monogamy, and procreation. A general despair over the current political situation and the collapse of democracy further motivate their departures and leave many feeling that they don’t belong to the Israeli national collective, while Berlin is frequently celebrated as a cosmopolitan and unassuming city where LGTB people can live in peace. Hila Amit argues that queer Israeli emigrants, in their decision to depart, undermine Zionist ideology as well as change the obvious paths of resistance to Zionism. In stepping out of the territory of Israel, they avoid the Zionist demand to perform as strong, masculine courageous citizens. But the left-wing resistance to the regime is just as committed to a heroic ideal – the willingness to take part in often violent demonstrations and risk imprisonment or injury. Amit recognizes the vulnerability of the emigrants, who can no longer bear the hardship of the life offered to them in Israel. To be explored, then, is the political potential of passivity and unheroic conduct. As Adi, one of the participants of the study, puts it: &#8216;I’ve been an activist for 10 years. I can’t live there anymore. People come to Berlin to heal, that’s why some people call it the Berlin Sanatorium.&#8217; The discussion builds on the contributions to A Queer Way Out: The Politics of Israeli Emigration (2018), an award-winning study by Hila Amit. The evening presents an opportunity to hear both from the author and the participants of the study and hence the rare chance to discuss the product of a long-term research project with the subjects of the research themselves. Hila Amit is a writer, independent scholar, and Hebrew teacher. Her research focuses on political activism, queer kinship, national belonging, and diasporic communities. Ruth Preser teaches at the University of Haifa and is a co-founder of the Haifa Feminist Institute. She is a former ICI Fellow. Yossi Bartal was born and raised in Jerusalem and has been living in Berlin for 11 years. He studies musicology and gender studies and writes extensively in Hebrew, German, and English about politics, music, and culture. Gil Kremmer was born and raised in Israel. She fell in love with Berlin during a backpacking trip through Europe. Works as a tour guide and as a DJ and lives with her wife and young kid in Berlin
