Conception de Ligands Protéiques par Bioinformatique et Modélisation Moléculaire
The increasing structural and functional knowledge of proteins gives us a clearer idea of how they interact. This information could be used to design new compounds, able to interact with a wide variety of interesting targets, as well as improve our comprehension of this phenomenon. This work presents the development of a new method enabling the design of protein ligands based on the transfer of a set of amino acids of a known ligand, contributing to the binding of a chosen target on a host protein, containing less than 100 residues (mini-proteins). These host proteins, able to reproduce the interaction after motif insertion, are identified in a systematic manner from the PDB. This approach has been applied to the development of new ligands of the Kv1.2 channel using the structural and functional knowledge of the interaction with the BgK toxin. Three ligands have been designed with inhibition constants in the micro molar range. Those results demonstrate the possibility of designing new ligands with a method based on the transfer of a functional motif of residues on a new scaffold which is sterically and electrostatically compatible with the interaction targeted.L'accroissement des connaissances, structurales et fonctionnelles, des protéines nous donne désormais une vision plus précise des phénomènes d'interaction. L'utilisation de ces informations pour le développement de ligands permettrait d'obtenir de nouveaux composés, capables d'interagir avec diverses cibles d'intérêt, et d'améliorer notre compréhension de ces interactions. Ce travail présente le développement d'une nouvelle méthode de conception de ligands protéiques, laquelle repose sur le transfert d'un groupe de résidus, appartenant à un ligand connu et contribuant de façon importante à la liaison avec une cible d'intérêt, sur une protéine hôte, de moins de 100 résidus (mini-protéines). L'identification de protéines hôtes, aptes à reproduire l'interaction après transfert du motif, est réalisée de manière systématique à partir des structures présentes dans la PDB. L'approche a été appliquée pour le développement de ligands du canal Kv1.2, à partir de connaissances structurales et fonctionnelles de l'interaction de ce même canal avec la toxine BgK. Trois ligands, possédant des constantes d'inhibition micro molaires, ont été ainsi conçus. Ces résultats démontrent la possibilité de mettre en application une méthode de conception de ligands, basée sur le transfert de motifs de « hotspots », sur une plateforme structurale de nature protéique, dont les aspects stérique et électrostatique sont compatibles avec une interaction donnée
REALISME MAGIS DALAM NOVEL O KARYA EKA KURNIAWAN
Tujuan penelitian ini ialah mengetahui bentuk dari realisme magis pada novel O
dengan rumusan masalah bagaimana realisme magis dalam novel O karya Eka
Kurniawan. Metode yang digunakan ialah metode Close Reading dengan teknik
analisis isi melalui pendekatan postmodernisme. Metode close reading yang
digunakan hanya mengambil diksi-diksi dari kutipan yang membentuk realisme
magis dan tema-tema kecil, serta tema utama untuk menemukan pesan dalam
novel. Penelitian ini dilakukan di Jakarta, selama semester genap tahun
2016/2017. Penelitian ini difokuskan pada bentuk realisme magis dalam novel O.
Adapun subfokus penelitian ini ialah menganalisis realisme magis dengan lima
karakteristik yang dirumuskan oleh Wendy.B Faris. Untuk menganalisis
penelitian dibantu oleh tabel analisis lima karakteristik realisme magis dengan
kriteria analisis yang meliputi (1) irreducible element (elemen tak tereduksi), (2)
phenomenal world (dunia fenomenal), (3) unsettling doubt (keraguan yang
meresahkan), (4) merging realms (penggabungan dua dunia/wilayah) dan (5)
Disruption of time, space, and identity (gangguan waktu, ruang, dan identitas).
Berdasarkan hasil analisis menggunakan lima karakteristik realisme magis, novel
O karya Eka Kurniawan memenuhi lima karakteristik realisme magis dengan
karakteristik yang dominan adalah karakteristik phenomenal world (dunia
fenomenal). Hal ini didasari oleh permainan narasi oleh pengarang terhadap
peleburan hal fantasi dan realitas yang tidak ingin terlempar secara jauh ke dalam
sebuah realisme. Dari hasil analisis itulah terbentuk sebuah pesan secara implisit
mengenai hakikat hidup. Jadi, dapat dikatan lima karakteristik realisme magis
tersebut memiliki keterkaitan satu sama lain dalam mewujudkan bentuk dari
realisme magis pada novel yang dianalisis. (M.Y.M).
The purpose of this study is to know the form of magical realism in novel O with
the formulation of the problem is how the magical realism in the novel O by Eka
Kurniawan. The method used is the method of Close Reading with content
analysis techniques through postmodernism approach. The method of close
reading that is used only takes the dictions from the citation which forms magical
realism and small themes, as well as the main theme for finding the message in
the novel. This research was conducted in Jakarta, during the even semester of
2016/2017. This research is focused on the form of magical realism in novel O.
The subfokus of this research is to analyze the magical realism with five
characteristics formulated by Wendy.B Faris. To analyze the research is assisted
by the analysis table of the five characteristics of magical realism with the
analysis criteria that include (1) irreducible element, (2) phenomenal world, (3)
unsettling doubt, (4) merging realms, and (5) Disruption of time, space, and
identity. Based on the results of the analysis using the five characteristics of
magical realism, novel O works by Eka Kurniawan meet the five characteristics of
magical realism with the dominant characteristic is phenomenal world
characteristic. It is based on a narrative game by the author of the fusion of
fantasy and reality that does not want to be thrown into a realism. From the results
of that analysis formed an implicit message about the nature of life. Thus, the five
characteristics of magical realism are related to each other in realizing the form of

Computerized adaptive and multistage testing with R: using packages catR and mstR
The goal of this guide and manual is to provide a practical and brief overview of the theory on computerized adaptive testing (CAT) and multistage testing (MST) and to illustrate the methodologies and applications using R open source language and several data examples. Implementation relies on the R packages catR and mstR that have been already or are being developed by the first author (with the team) and that include some of the newest research algorithms on the topic. The book covers many topics along with the R-code: the basics of R, theoretical overview of CAT and MST, CAT designs, CAT assembly methodologies, CAT simulations, catR package, CAT applications, MST designs, IRT-based MST methodologies, tree-based MST methodologies, mstR package, and MST applications. CAT has been used in many large-scale assessments over recent decades, and MST has become very popular in recent years. R open source language also has become one of the most useful tools for applications in almost all fields, including business and education. Though very useful and popular, R is a difficult language to learn, with a steep learning curve. Given the obvious need for but with the complex implementation of CAT and MST, it is very difficult for users to simulate or implement CAT and MST. Until this manual, there has been no book for users to design and use CAT and MST easily and without expense; i.e., by using the free R software. All examples and illustrations are generated using predefined scripts in R language, available for free download from the book's website. His research interests include both theoretical and methodological development as well as open source implementation and dissemination in R. He is the main developer and maintainer of the packages catR and mstR, among others. Duanli Yan, PhD, is Manager of Data Analysis and Computational Research for Automated Scoring group in the Research and Development division at the Educational Testing Service (ETS). She is also an Adjunct Professor at Rutgers University. Dr. Yan has been the statistical coordinator for the EXADEP™ test, and the TOEIC® Institutional programs, a Development Scientist for innovative research applications, and a Psychometrician for several operational programs. Dr. Yan received many awards, including the 2011 ETS Presidential Award, the 2013 NCME Brenda Lyod award, and the 2015 IACAT Early Career Award. She is a co-editor for Computerized Multistage Testing: Theory and Applications and a co-author for Bayesian Networks in Educational Assessment. Alina A. von Davier, PhD, is Senior Research Director of the Computational Psychometrics Research Center at Educational Testing Service (ETS) and an Adjunct Professor at Fordham University. At ETS she leads the Computational Psychometrics Research Center, where she is responsible for developing a team of experts and a psychometric research agenda in support of next generation assessments. Computational psychometrics, which include machine learning and data mining techniques, Bayesian inference methods, stochastic processes and psychometric models are the main set of tools employed in her current work. She also works with psychometric models applied to educational testing: test score equating methods, item response theory models, and adaptive testing.
