From the railroad station to the tube-shaped bus stops; from Ukraine to downtown Curitiba; from Paisagem interior to Reika; from the nineteenth century to the twenty-first century, Helena Kolody's literature guarantees her presence in the consolidation of the art/life binomial. The portrait of the author ends up by also being made up of an abundant iconographic apparatus, of the over five hundred critical texts listed, and of her complete work.     Esta pesquisa analisa atuação de Helena de Magalhães Castro (1902-1995), declamadora, violonista, propagadora do folclore nacional, fundadora e presidenta da Instrução Artística do Brasil (IAB). O núcleo documental central para o desenvolvimento dos estudos é o acervo pessoal, até então inédito, de Helena de Magalhães Castro, constituído por cartas e postais, Revistas da IAB, álbuns de viagens pelo Brasil e exterior. A análise contempla sua produção artística entre os anos de 1924 a 1931, com ênfase em sua apresentação oficial como declamadora e violonista, ou seja, do início de sua trajetória profissional até o ano da fundação da IAB. Pretende-se também contextualizar a prática da declamação no Brasil como um nicho comercial relevante e, concomitantemente, compreender a atuação de Helena como mediadora e estilizadora de canções populares. Serão levantadas reflexões sobre a utilização de fontes populares na construção de seu repertório artístico, buscando lançar luz sobre sua trajetória e dar visibilidade ao seu papel de divulgadora da arte nacional.This research analyzes the performance of Helena de Magalhães Castro (1902- 1995), reciter, guitarist, propagator of national folklore, founder and president of the Brazilian Artistic Instruction (IAB). The central documentary nucleus for the development of the studies is the personal collection, hitherto unprecedented, by Helena de Magalhães Castro, consisting of letters and postcards, IAB Magazines, travel albums in Brazil and abroad. The analysis contemplates his artistic production between the years 1924 to 1931, with an emphasis on his official presentation as a reciter and guitarist, that is, from the beginning of his professional career until the year of the foundation of the IAB. It is also intended to contextualize the practice of reclamation in Brazil as a relevant commercial niche and, at the same time, to understand Helena\'s performance as a mediator and stylist of popular songs. Reflections will be raised on the use of popular sources in the construction of his artistic repertoire, seeking to shed light on his trajectory and give visibility to his role as a promoter of national art

    Transient observations : the textualizing of St Helena through five hundred years of colonial discourse

    This thesis explores the textualizing of the South Atlantic island of St Helena (a British Overseas Territory) through an analysis of the relationship between colonizing practices and the changing representations of the island and its inhabitants in a range of colonial 'texts', including historiography, travel writing, government papers, creative writing, and the fine arts. Part I situates this thesis within a critical engagement with post-colonial theory and colonial discourse analysis primarily, as well as with the recent 'linguistic turn' in anthropology and history. In place of post-colonialism's rather monolithic approach to colonial experiences, I argue for a localised approach to colonisation, which takes greater account of colonial praxis and of the continuous re-negotiation and re-constitution of particular colonial situations. Part II focuses on a number of literary issues by reviewing St Helena's historiography and literature, and by investigating the range of narrative tropes employed (largely by travellers) in the textualizing of St Helena, in particular with respect to recurrent imaginings of the island in terms of an earthly Eden. Part III examines the nature of colonial 'possession' by tracing the island's gradual appropriation by the Portuguese, Dutch and English in the sixteenth and early seventeenth century and the settlement policies pursued by the English East India Company in the late seventeenth and early eighteenth century. Part IV provides an account of the changing perceptions, by visitors and colonial officials alike, of the character of the island's inhabitants (from the late eighteenth to the early twentieth century) and assesses the influence that these perceptions have had on the administration of the island and the political status of its inhabitants (in the mid- to late twentieth century). Part V, the conclusion, reviews the principal arguments of my thesis by addressing the political implications of post-colonial theory and of my own research, while also indicating avenues for further research. A localised and detailed exploration of colonial discourse over a period of nearly five hundred years, and a close analysis of a consequently wide range of colonial 'texts', has confirmed that although colonising practices and representations are far from monolithic, in the case of St Helena their continuities are of as much significance as their discontinuities

    Profesor Helena Więckowska (1897–1984) była z pewnością jedną z najbardziej znaczących postaci w polskim bibliotekarstwie, w okresie powojennym. Wniosła także istotny wkład w rozwój polskiej bibliologii i wydatnie przyczyniła się do ukształtowania systemu akademickiego kształcenia bibliotekarzy. Bibliografia Jej dorobku naukowego liczy 221 pozycji. Są w nim rozprawy, artykuły, recenzje, hasła w słownikach i encyklopediach. Jest to również dorobek zróżnicowany tematycznie, co potwierdza trafność określenia „bibliolog praktyk”, użytego wobec dokonań Profesor Więckowskiej przez Krzysztofa Migonia.The author of the article discusses the activities of Helena Więckowska – the second in the order director of the Library of the Łódź University. In the text you can find information about the problems that she as a director had to face: first of all with the facilities but also with the development of collections. The author mentiones that Helena Więckowska cared very much about the vocational training of employees: improving the librarians’ qualifications, gaining degrees. In 1954 she decided to train students in the field of using the library.     ACME and SPLiT-seq: Quantitative and evolutionary single-cell transcriptomics in planarians

    The last decade has seen an exponential development of scRNA-seq technologies. This thesis begins by recapitulating the history of transcriptomics, from its early years to the development of modern single-cell protocols, in order to understand the evolution and current state of the field. During the experimental part, I focus on the description and validation of a versatile pipeline for scRNA-seq. This combines a novel sample preparation strategy (ACME) with a powerful in situ barcoding platform (SPLiT-seq) to overcome some of the restrains of current single-cell protocols. ACME provides simultaneous tissue dissociation, fixation and permeabilization, resulting in cells that can be cryopreserved, sorted by FACS and used in multiple platforms. As proof of concept, ACME-cells are combined with SPLiT-seq (Rosenberg et al., 2018), obtaining over 32,000 cell transcriptomes and profiling two comprehensive single-cell atlases for the planarian species Schmidtea mediterranea and Dugesia japonica. Later, I use the same pipeline in a single-cell RNA interference study, in S. mediterranea, to describe and quantify the effects of the hnf4 knockdown at tissue-resolution. Our results reveal cellular and genetic changes in the gut and parenchymal populations of knockdown animals, proving that our transcriptomic approach can finely dissect tissue specific effects in knockdown conditions. In the last part of the thesis, I present the single-cell atlases of multiple planarians (Schmidtea mediterranea, Schmidtea polychroa, Dugesia japonica, Girardia tigrina and Polycelis nigra), and compare the differences between sexual and asexual strains and life-history stages of the same species. This data includes over 116,000 cell transcriptomes, and sets the bases for a future cross-species comparison that will explore the diversity of cell types and gene expression in planarians at single-cell resolution. Overall, these examples show how single-cell transcriptomics is moving toward the end of the era of descriptive cell type atlases into more complex quantitative and evolutionary studies

Programa de Pós-Graduação em Literatura.No presente trabalho realiza-se uma leitura intertextual entre a Bíblia e Os escravos, coletânea de poemas de teor abolicionista do poeta romântico Antônio Frederico de Castro Alves (1847-1871), objetivando demonstrar que os textos poéticos arquitetam-se na desconstrução e reconstrução dos textos bíblicos. A leitura dos poemas centra-se nos personagens: Deus, Eros e Mãe, os quais conformam uma trindade poética/sagrada. A pesquisa divide-se em três movimentos: Prelúdios do sagrado no Romantismo, Tríade melódica e À guisa de coda: trindade poética. No primeiro efetuam-se algumas aproximações ao conceito do sagrado e aos Romantismos francês e brasileiro. O seguinte corresponde à leitura das composições, através das linhas melódicas: A dualidade de Deus, A ambivalência de Eros e O duplo calvário da Mãe escrava. E no último movimento amalgamam-se as inter-relações entre a trindade cristã e poética e os dramas bíblico e poético
