Transcript-dependent effects of the CALCA gene on the progression of post-traumatic osteoarthritis in mice

    No full text
Osteoarthritis represents a chronic degenerative joint disease with exceptional clinical relevance.Polymorphisms of the CALCA gene, giving rise to either a procalcitonin/calcitonin (PCT/CT) or acalcitonin gene-related peptide alpha (αCGRP) transcript by alternative splicing, were reported to beassociated with the development of osteoarthritis. The objective of this study was to investigate therole of both PCT/CT and αCGRP transcripts in a mouse model of post-traumatic osteoarthritis (ptOA).WT, αCGRP−/− and CALCA−/− mice were subjected to anterior cruciate ligament transection (ACLT) toinduce ptOA of the knee. Mice were sacrificed 4 and 8 weeks post-surgery, followed by micro-CT andhistological evaluation. Here we show that the expression of both PCT/CT and αCGRP transcripts isinduced in ptOA knees. CALCA−/− mice show increased cartilage degeneration and subchondral boneloss with elevated osteoclast numbers compared to αCGRP−/− and WT mice. Osteophyte formation isreduced to the same extent in CALCA−/− and αCGRP−/− mice compared to WT controls, while areduced synovitis score is noticed exclusively in mice lacking CALCA. Our data show that expressionof the PCT/CT transcript protects from the progression of ptOA, while αCGRP promotes osteophyteformation, suggesting that CALCA-encoded peptides may represent novel targets for the treatmentof ptOA.Osteoarthritis represents a chronic degenerative joint disease with exceptional clinical relevance. Association study of the calcitonin gene-related polypeptide-alpha (CALCA) and the receptor activity modifying 1 (RAMP1) genes with migraine

Migraine is a common neurovascular brain disorder characterised by recurrent attacks of severe headache that may be accompanied by various neurological symptoms. Migraine is thought to result from activation of the trigeminovascular system followed by vasodilation of pain-producing intracranial blood vessels and activation of second-order sensory neurons in the trigeminal nucleus caudalis. Calcitonin gene-related peptide (CGRP) is a mediator of neurogenic inflammation and the most powerful vasodilating neuropeptide, and has been implicated in migraine pathophysiology. Consequently, genes involved in CGRP synthesis or CGRP receptor genes may play a role in migraine and/or increase susceptibility. This study investigates whether variants in the gene that encodes CGRP, calcitonin-related polypeptide alpha (CALCA) or in the gene that encodes a component of its receptor, receptor activity modifying protein 1 (RAMP1), are associated with migraine pathogenesis and susceptibility. The single nucleotide polymorphisms (SNPs) rs3781719 and rs145837941 in the CALCA gene, and rs3754701 and rs7590387 at the RAMP1 locus, were analysed in an Australian Caucasian population of migraineurs and matched controls. Although we find no significant association of any of the SNPs tested with migraine overall, we detected a nominally significant association (p = 0.031) of the RAMP1 rs3754701 variant in male migraine subjects, although this is non-significant after Bonferroni correction for multiple testing.Griffith Health, School of Medical ScienceNo Full Tex

Going Beyond Counting First Authors in Author Co-citation Analysis

    No full text
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

The research study studies the relationship between the tourist conditions presented by Calca district and practice of adventure tourism 2019. The population is made up of 39,210 national and foreign tourists from which those who visited the district of Calca were chosen in the month of November 2019, so the sample is made up of 90 national and foreign tourists. The investigation is descriptive and explanatory and its design is not experimental, transversal and correlational. For the data collection, a twenty-item survey was used with a five-level interpretation scale contrasted with the Cronbach's Alpha that has a value of 0.974 for the items considered in the Tourist Conditions variable and 0.953 for the items in the Practices variable of adventure tourism. The interpretation of the general results were thus “As p = 0.000 <0.05, in the Chi - square test, it can be affirmed that there are tourist conditions presented by the district of Calca for the practice of adventure tourism, 2019, where the degree of relationship through Spearman's correlation it is 53.3%, at 95% reliability ”, therefore, the Alternate Hypothesis is accepted« There are tourist conditions presented by the district of Calca for the practice of adventure tourism, 2019 »and the Hypothesis is rejected null

Appropriate Similarity Measures for Author Cocitation Analysis

    No full text
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Finally, we show by means of an example that our findings have a high practical relevance.information science;Pearson correlation;cosine;similarity measure;author cocitation analysis
