    Comparison of the binding and internalization properties of 12 DOTA-coupled and 111In-labelled CCK2/Gastrin receptor binding peptides: a collaborative project under COST Action BM0607

    Specific overexpression of cholecystokinin 2 (CCK2)/Gastrin receptors has been demonstrated in several tumors of neuroendocrine origin. In some of these cancer types, such s medullary thyroid cancer (MTC), there are no effective systemic treatments for metastatic disease. Peptide Receptor Radionuclide Therapy (PRRT) may be a viable therapeutic strategy in the management of these patients. Several CK2R targeted radiopharmaceuticals have been described in recent years. As part of the EU COST action BM0607 we studied the in vitro and in vivo characteristics of 12 DOTA‐conjugated CK2R binding peptides. In the present study, we analyzed binding and internalization characteristics. Stability, biodistribution, and imaging studies have been performed in parallel. methods. Determination of IC50 was performed using autoradiography, with peptides complexed to Indium‐115 displacing 125I‐CCK on tissue sections from human tumors. Saturation binding and internalization experiments were performed using 111In‐labelled peptides (specific activity 2‐10 MBq/nmol). The rat AR42J cell line and the human A431‐CCK2R transfected cell line were utilized for in vitro experiments. Dissociation constants (Kd) and apparent number of binding sites (Bmax) were determined with the aid of graphical analysis software. Internalization was determined in receptor expressing cells by incubating with tracer amounts of peptide at 37 and 4°C for different times up to 120 min. Surface bound peptide was then stripped either by acid wash or incubation with 1μM cold peptide at 4°C. Results. All peptides showed high and similar receptor affinity with IC50 values ranging from 0.2 to 3.4 nM. Saturation experiments also showed high affinity with Kd values in the 10‐9 ‐10‐8 M range. Bmax values estimated in A431‐CCK2R cells ranged from 1 .0‐ 4.6 E6 per cell. Slight differences in the levels of non‐specific binding to cells were observed for the different peptides. All peptides showed high levels of internalization when incubated at 37°C. Conclusion. All DOTA‐conjugated peptides showed similar receptor binding and internalization properties. Issues pertaining to in vivo stability and biodistribution will be important in determining the most promising candidates for further development and clinical application.     This paper looks at the current technologies implemented in cellular mobile telephony at this time. Technologies covered are IrDA, Bluetooth, WAP and GPRS. The author gives the fundamental ideas behind the technologies, the basic parameters and protocols for implementation, and how they are useful to cellular phone users; this is intended to promote interest in the areas, while still giving scope for further indepth research in the subject. Also covered is the potential battle between IrDA and Bluetooth for the same marketing purposes. The author also discusses the potential for 3G, and the new technologies it will bring to the mobile phone market

    ABSTRAKPerkembangan teknologi internet sangat memberikan dampak yang signifikan di dalam dunia Pendidikan, sehingga dapat mendukung proses pembelajaran di suatu sekolah. Sebagai salah satu sekolah negeri yang ada di kabupaten Malang, SD Negeri 01 Jambearjo telah memanfaatkan jaringan internet untuk mendukung proses pembelajaran di sekolah. Namun penggunaan jaringan internet di SD Negeri 01 Jambearjo saat ini masih belum optimal. Pada penelitian ini membahas mengenai analisis perbandingan kinerja Wireless Access Point (WAP) dan Virtual Access Point (VAP) berbasis Mikrotik di SD Negeri 01 Jambearjo. Pada penelitian ini dilakukan untuk mengetahui sistem manakah yang lebih baik untuk digunakan di SD Negeri 01 Jambearjo. Dalam sistem jaringan WAP dan VAP digunakan Mikrotik RB941-2nD untuk melakukan konfigurasi atau pengontrolan access point secara terpusat. Pada jaringan WAP digunakan access point yang terhubung dengan mikrotik meenggunakan kabel LAN, sedangkan jaringan VAP menggunakan mode CAPsMAN dengan menggabungkan dua mikrotik RB941-2nD menggunakan kabel LAN. Dalam melakukan pengujian, penulis menggunakan parameter berdasarkan Quality of Service yaitu throughput, packet loss, delay, dan jitter. Pada pengujian Quality of Servicedigunakan aplikasi Wireshark. Hasil pengujian pada jaringan WAP didapatkan rata-rata nilai throughput sebesar 3034 kbps, dengan packet loss bernilai 3,16%, delay sebesar 2,840 ms, dan jitter sebesar 0 ms. Sedangkan pada jaringan VAP didapatkan hasil rata-rata nilai throughput sebesar 1479 kbps, dengan packet loss bernilai 9,9%, delay sebesar 4,945 ms, dan jitter sebesar 0,0011 ms. Berdasarkan hasil pengujian didapatkan hasil Analisa bahwa jaringan WAP memiliki kualitas yang lebih baik dibandingkan dengan jaringan VAP untuk digunakan di SD Negeri 01 Jambearjo. As one of the public schools in the Malang district, SD Negeri 01 Jambearjo has utilized the internet network to support the learning process at school. However, the use of the internet network at SD Negeri 01 Jambearjo is currently still not optimal. This study discusses the comparative analysis of the performance of Mikrotik-based Wireless Access Point (WAP) and Virtual Access Point (VAP) at SD Negeri 01 Jambearjo. This research was conducted to determine which system is better for SD Negeri 01 Jambearjo. In the WAP and VAP network systems, Mikrotik RB941-2nD is used to configure or control access points centrally. In the WAP network, an access point is used that is connected to the Mikrotik using a LAN cable, while the VAP network uses the CAPsMAN mode by combining two Mikrotik RB941-2nD using a LAN cable. In conducting the test, the author uses parameters based on Quality of Service, namely throughput, packet loss, delay, and jitter. In the Quality of Service test, the Wireshark application is used. The test results on the WAP network obtained an average throughput value of 3034 kbps, with packet loss of 3.16%, a delay of 2.840 ms, and jitter of 0 ms. While on the VAP network, the average throughput value is 1479 kbps, with packet loss of 9.9%, a delay of 4.945 ms, and jitter of 0,0011 ms. Based on the test results, the analysis results show that the WAP network has better quality than the VAP network for use in SD Negeri 01 Jambearjo.     Przedstawiono budowę oraz zasadę działania unikalnego na skalę światową przyrządu umożliwiającego dokonanie pomiaru wartości sił biorących udział w procesie skrawania (urabiania węgla) nazwanego przez autora POU-BW/01-WAP. Jest jedynym na świecie przyrządem za pomocą którego istnieje możliwość bezpośredniego wyznaczenia wartości dwu składowych sił biorących udział w procesie skrawania: siły skrawania Fs oraz siły docisku noża Fd do urabianej calizny. Wyznaczenie wartości sił biorących udział w procesie skrawania jest możliwe za pomocą dwu niezależnych bloków pomiarowych, który stanowią tensometryczne czujniki siły. Do rejestracji sił, zastosowano nóż stosowany w ścianowych kombajnach bębnowych – styczno-obrotowy. Opisany został proces wzorcowania tensometrycznych czujników siły oraz przedstawiono wyniki badań weryfikujących. Przyrząd posiada certyfikat ATEX, umożliwiający pracę w warunkach rzeczywistych, jako urządzenia przeznaczonego do użytku w przestrzeniach zagrożonych wybuchem – zgodnie z dyrektywą 94/9/EC.This article presents the structure and operating principle of a globally unique device which allows for the measurement of forces involved in the process of cutting (coal breaking), named POU-BW/01-WAP by the author. It is the only device in the world which makes it possible to directly determine the value of the two component forces involved in the coal cutting process: the cutting force Fs and the contact force between the pick and the coal solid Fd. Determining the value of the forces involved in the cutting process is possible with the use of two independent measurement blocks, each consisting of strain gauges. The forces were measured using a tangential-rotary pick utilised in drum shearers. The calibration process of force strain gauges has been described and the results of verification research presented. The device has been approved with the ATEX certificate, which allows it to be operated in real environment as a device intended for use in explosive atmospheres in accordance with the 94/9/EC directive

The result is particularly interesting on noting that the underlying topological semigroup is not assumed to support an absolutely continuous measure—a theme studied by the author in a previous paper.</p

    The mobility scale for lower limb amputees: The SIGAM/WAP mobility scale

    Purpose. To translate the SIGAM mobility scale into the Dutch language and to test and validate its properties in everyday practice. Method. The SIGAM mobility scale as published by Ryall et al. was translated into the Dutch language with the local used verbs for prosthetic use. The translated Dutch text was reviewed by several authors and a panel of professionals. The retranslation by a native speaker was reviewed by the original author who suggested modifications. The Dutch trial version of the mobility scale was presented to a panel of prosthetic users and therapists who advised slight modifications for better understanding of the questionnaire. IN training sessions prosthetic teams across The Netherlands were trained in the use of the translated SIGAM/WAP mobility scale. Results. During the translation there were problems with slang words and the use of specific words in the care of amputee patients. The instruction of team members and the test scoring of the questionnaire and the algorithm showed no difficulties. There was good to perfect agreement between scores in case training sessions with perfect inter observer reliability. Conclusions. With this instrument we have a specific measurement tool in the English and Dutch language to measure mobility in lower limb amputees

    Determination of Coal Cutting Forces Using the Cutting Head of POU-BW/01-WAP Device

    The paper presents a method for measuring and recording the forces involved in the coal cutting process. Moreover, a method for visualization of all forces involved in the cutting process was described. In the following part, the construction and principle of operation of a device for determination of forces involved in the cutting process (coal mining), referred to by the author as POU-BW/01-WAP, are presented. Resistance extensometry was used to measure the forces. This is the only device in the world that determines two of three force components that take part in the cutting process. For this purpose, two independent measuring blocks were used, which are strain gauges of force: cutting (Fs) and knife pressure (Fd). In order to register these forces, a real mining knife used in longwall shearer drums was applied – i.e. tangential-rotary. The equipment has the ATEX certificate allowing for operation in real conditions as a device intended for use in potentially explosive atmospheres – in accordance with the directive 94/9/EC. It has received many awards at world fairs for inventions and innovative solutions
