Identifikasi Gen Myostatin (MTSN) pada Kambing Kacang dan Kambing Peranakan Etawah di Deli Serdang
Goats in Indonesia have potential that has not been utilized optimally, even though goat meat in demand by Indonesian people were high. Needs to be done an effort to improve carcass quality of local goat. One approach that can be done is with a molecular approach or used adopting molecular or marker assisted selection ( MAS ). A gene that can used as marker of carcass quality is myostatin genes (MSTN) exon 1. The purpose of this study was to obtain the polymorphisms of the MSTN gene in local goat in Deli Serdang District used PCR-RFLP and sequencing methode and find SNP in the fragmen target MSTN gene exon 1. The number of sample used were 60 heads of goats consisted of 30 Kacang goats and 30 PE goats from Galang, Hamparan Perak and Namorambe , Deli Serdang district. The MSTN gene forward primer 5’-AGAACAGCGAGCAGAAGGAA-3’ and reverse 5’-CGTCGTAACGTGGTAGTCAT-3’, the enzyme for restriction was HinIII. The polymorphisms of the MSTN gene were identified by PCR RFLP and direct sequnencing method. To determine Hardy-Weinberg equilibrium status, both Allele and genotype frequencies were analyzed. To alignment sequencing result have done analyzed using BioEdit and MEGA6 softwere. The result showed that there is MSTN gene exon 1 have found SNP in 53 point at Kacang goats and PE goats in Galang, Hamparan Perak and Namorambe, Deli Serdang district is monomorphic. Only one genotype from MSTN exon 1, there is AA. For Heterozigosity and HW equilibrium is 0, because only one genotype that found in MSTN gene in this research. Myostatin gene at Kacang goats and PE goats population in Deli Serdang District are monomorphic.Kambing di Indonesia memiliki potensi yang belum dimanfaatkan secara optimal, padahal daging kambing banyak diminati oleh masyarakat. Untuk itu perlu dilakukan upaya peningkatan kualitas kambing lokal kearah perdagingan. Salah satu pendekatan yang dapat dilakukan adalah dengan pendekatan molekuler atau dengan kata lain dengan menggunakan Marker Assisted Selection (MAS). Gen yang digunakan sebagai marker adalah gen Myostatin (MSTN). Tujuan dari penelitian ini adalah untuk mengidentifkasi keragaman gen MSTN pada kambing lokal di Kabupaten Deli Serdang dengan menggunakan metode PCR RFLP dan sekuensing, menentukan SNP pada frgamen target gen MSTN ekson 1. Jumlah sampel yang digunakan adalah 30 ekor kambing Kacang dan 30 ekor kambing PE yang berasal dari kecamatan Galang, Hamparan Perak dan Namorambe dengan jumlah masing-masing wilayah 10 ekor. Amplifikasi gen MSTN ekson 1 menggunakan primer forward 5’-AGAACAGCGAGCAGAAGGAA-3’ dan reverse 5’-CGTCGTAACGTGGTAGTCAT-3’, yang dipotong enzim restriksi HinIII. Keragaman genetik yang diamati adalah frekuensi alel, frekuensi genotipe, heterozigositas dan keseimbangan populasi. Dan penyejajaran hasil sekuensing dilakukan dengan menggunakan aplikasi BioEdit dan MEGA5. Hasil analisis Gen Myostatin ekson 1 pada titik mutasi ke-53 pada kambing Kacang dan kambing PE di daerah Galang, Hamparan Perak dan Nomorambe di Sumatera Utara bersiat monomorfik. Hanya satu genotipe dari gen MSTN ekson 1 yang ditemukan baik pada kambing Kacang maupun kambing PE yaitu genotipe AA. Untuk nilai heterozigositasdan keseimbangan populasi bernilai 0 dikarenakan hanya satu genotipe yang ditemukan dalam penelitian ini. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
Study of phenotypics and genetics diversity of some indonesian local goat sub populations and its strategies for sustainable use
The genetic resources of local goats are very important because it was well adapted to the local environment with low input management systems. Local goats are very potential for breeding stock program which suitable to the Indonesia local conditions. The study was carried out in order to describe the phenotypic and genetic of 6 sub-populations of Indonesian local goats based on the analysis of the morphometrics, mitochondrial DNA, Y Chromosome and GDF9 gene. The sampling was conducted in four of the Provinces in Indonesia, namely North Sumatra Province (Samosir goat n=42 in the Samosir District, Muara goat n=34 in the North Tapanuli district, Kacang goat n=217 in the Deli Serdang District), Central Java Province (Jawarandu goat n=94 in the Blora District); Province of South Sulawesi (Marica goat n=60 in the Maros District, Makassar City, Jeneponto District) and Nusa Tenggara Timur Province (Bengala goat n=96 in the Kupang area, Sikka District, Ende District). All of them were measured for morphometric and was bleeding to collect blood as DNA sources. The blood sample was preserve with absolute ethanol.
