Morphologic and functional correlates of synaptic pathology in the cathepsin D knockout mouse model of congenital neuronal ceroid lipofuscinosis
Mutations in the cathepsin D (CTSD) gene cause an aggressive neurodegenerative disease (congenital neuronal ceroid lipofuscinosis) that leads to early death. Recent evidence suggests that presynaptic abnormalities play a major role in the pathogenesis of CTSD deficiencies. To identify the early events that lead to synaptic alterations, we investigated synaptic ultrastructure and function in presymptomatic CTSD knockout (Ctsd) mice. Electron microscopy revealed that there were significantly greater numbers of readily releasable synaptic vesicles present in Ctsd mice than in wild-type control mice as early as postnatal day 16. The size of this synaptic vesicle pool continued to increase with disease progression in the hippocampus and thalamus of the Ctsd mice. Electrophysiology revealed a markedly decreased frequency of miniature excitatory postsynaptic currents (mEPSCs) with no effect on paired-pulse modulation of the evoked excitatory post synaptic potentials in the hippocampus of Ctsd mice. The reduced mEPSCs frequency was observed before the appearance of epilepsy or any morphologic sign of synaptic degeneration. Taken together, these data indicate that CTSD is required for normal synaptic function and that a failure in synaptic trafficking or recycling may bean early and important pathologic mechanism in Ctsd mice; these presynaptic abnormalities may initiate synaptic degeneration in advance of subsequent neuronal loss
Large-scale phenotyping of an accurate genetic mouse model of JNCL identifies novel early pathology outside the central nervous system.
Cln3(Δex7/8) mice harbor the most common genetic defect causing juvenile neuronal ceroid lipofuscinosis (JNCL), an autosomal recessive disease involving seizures, visual, motor and cognitive decline, and premature death. Here, to more thoroughly investigate the manifestations of the common JNCL mutation, we performed a broad phenotyping study of Cln3(Δex7/8) mice. Homozygous Cln3(Δex7/8) mice, congenic on a C57BL/6N background, displayed subtle deficits in sensory and motor tasks at 10-14 weeks of age. Homozygous Cln3(Δex7/8) mice also displayed electroretinographic changes reflecting cone function deficits past 5 months of age and a progressive decline of retinal post-receptoral function. Metabolic analysis revealed increases in rectal body temperature and minimum oxygen consumption in 12-13 week old homozygous Cln3(Δex7/8) mice, which were also seen to a lesser extent in heterozygous Cln3(Δex7/8) mice. Heart weight was slightly increased at 20 weeks of age, but no significant differences were observed in cardiac function in young adults. In a comprehensive blood analysis at 15-16 weeks of age, serum ferritin concentrations, mean corpuscular volume of red blood cells (MCV), and reticulocyte counts were reproducibly increased in homozygous Cln3(Δ) (ex7/8) mice, and male homozygotes had a relative T-cell deficiency, suggesting alterations in hematopoiesis. Finally, consistent with findings in JNCL patients, vacuolated peripheral blood lymphocytes were observed in homozygous Cln3(Δ) (ex7/8) neonates, and to a greater extent in older animals. Early onset, severe vacuolation in clear cells of the epididymis of male homozygous Cln3(Δ) (ex7/8) mice was also observed. These data highlight additional organ systems in which to study CLN3 function, and early phenotypes have been established in homozygous Cln3(Δ) (ex7/8) mice that merit further study for JNCL biomarker development
Cellular models of batten disease
The Neuronal Ceroid Lipofuscinoses (NCL), otherwise known as Batten disease, are a group of neurodegenerative diseases caused by mutations in 13 known genes. All except one NCL is autosomal recessive in inheritance, with similar aetiology and characterised by the accumulation of autofluorescent storage material in the lysosomes of cells. Age of onset and the rate of progression vary between the NCLs. They are collectively one of the most common lysosomal storage diseases, but the enigma remains of how genetically distinct diseases result in such remarkably similar pathogenesis. Much has been learnt from cellular studies about the function of the proteins encoded by the affected genes. Such research has utilised primitive unicellular models such as yeast and amoeba containing gene orthologues, cells derived from naturally occurring (sheep) and genetically engineered (mouse) animal models or patient-derived cells. Most recently, patient-derived induced pluripotent stem cell (iPSC) lines have been differentiated into neural cell-types to study molecular pathogenesis in the cells most profoundly affected by disease. Here, we review how cell models have informed much of the biochemical understanding of the NCLs and how more complex models are being used to further this understanding and potentially act as platforms for therapeutic efficacy studies in the future
Markets Equilibrium: The Is-Lm Model
. The purpose of this study is to analyze how the concept of markets equilibrium: the IS-LM Model. This research uses library research method by using reference sources from books and journals according to the theme. The author uses a qualitative method which is explained graphically, namely the market balance of the IS-LM model where the focus is on money and goods markets associated with macroeconomics where researchers take the side of investors. The results of this study are that the balance in the economy is the point where the IS and LM curves intersect. This point provides an interest rate (r) and income level (Y) that satisfies the equilibrium conditions that occur in the goods market and money market. In other words, planned spending equals actual spending, and the demand for real money balances equals the supply. So that the IS-LM balance, it is stated that IS=LM

Possibilities and testing of CPRNG in block cipher mode of operation PM-DC-LM
This paper discusses the chaotic pseudo-random number generator (CPRNG), which is used in block cipher mode of operation called PM-DC-LM. PM-DC-LM is one of possible subversions of general PM mode. In this paper is not discussed the design of PM-DC-LM, but only CPRNG as a part of it because designing is written in other papers. Possibilities, how to change or to improve CPRNG are mentioned. CF/LM-PAEK: Characterisation and sensitivity to critical process parameters for automated fibre placement
This paper presents an investigation into Automated Fibre Placement process parameters for a novel carbon-fibre-reinforced thermoplastic material based on a low-melt polyaryl ether ketone polymer matrix (CF/LM-PAEK). The primary focus of this investigation is in-situ consolidation, with parameter sets for sample manufacturing selected based on a Design of Experiment method and analysed by means of mechanical and thermal testing. For in-situ consolidation, a maximum shear strength of 33.6 MPa and a crystallinity of 27.3% is achieved. Subsequent tempering increases shear strength up to 46.5 MPa and crystallinity up to 32%. The material was found to be insensitive to layup speed within the tested parameter range (1–15 m/min), achieving similar results for high layup speeds

Tenaga listrik merupakan tenaga yang dimanfaatkan manusia dalam kehidupannya sehari-hari. Tenaga listrik dimanfaatkan untuk menghidupkan atau menjalankan alat-alat elektronika yang mempermudah aktivitas manusia. Agar dapat menggunakan tenaga listrik pada alat-alat elektronik diperlukan pengubah daya dari arus listrik arus kuat ke arus listrik arus lemah. Konsep pengubahan arus kuat menjadi arus lemah diterapkan dalam bentuk adaptor. Dalam pembelajarannya adaptor memanfaatkan konsep dioda sebagai penyearah arus listrik. Pada program studi Pendidikan Teknik Elektronika, khususnya di Universitas Negeri Jakarta, penerapan konsep dioda sebagai penyearah dituangkan dalam pembuatan adaptor DC oleh mahasiswa. Menggunakan IC LM 7805, penulis membuat adaptor DC dengan keluaran 5V sebagai penerapan konsep dioda sebagai penyarah. Hasil dari perancangan adaptor menunjukkan hasil pengukuran output adaptor bernilai 5V. Hal ini berbanding lurus dengan sifat IC LM 7805 yang meregulasi tegangan output yang masuk ke IC menjadi 5V.

Electrical power is energy that humans use in their daily lives. Electrical power is used to turn on or run electronic devices that make human activities easier. In order to use electrical power in electronic devices, it is necessary to convert the power from strong current to weak current. The concept of converting strong current to weak current is implemented in the form of an adapter. In the lesson, the adapter utilizes the concept of a diode as an electric current rectifier. In the Electronics Engineering Education study program, especially at Jakarta State University, the application of the concept of diodes as rectifiers is outlined in the manufacture of DC adapters by students. Using IC LM 7805, the author made a DC adapter with 5V output as an application of the diode concept as a rectifier. The results of the adapter design show that the adapter output measurement results are 5V. This is directly proportional to the nature of the LM 7805 IC which regulates the output voltage entering the IC to 5V.

Generalized LM tests for functional form and heteroscedasticity
We present a generalized LM test of heteroscedasticity allowing the presence of data transformation and a generalized LM test of functional form allowing the presence of heteroscedasticity. Both generalizations are meaningful as non-normality and heteroscedasticity are common in economic data. A joint test of functional form and heteroscedasticity is also given. These tests are further "studentized" to account for possible excess skewness and kurtosis of the errors in the model. All tests are easy to implement. They are based on the expected information and are shown to possess excellent finite sample properties. Several related tests are also discussed and their finite sample performances assessed. We found that our newly proposed tests significantly outperform the others, in particular in the cases where the errors are non-normal. Copyright � 2008 The Author(s).
