Abstract 3686: Generation of an off-the-shelf cell-based platform (PDC-vac) for active antitumor immunotherapy
Abstract
The development of efficient vaccines for the treatment of cancers in combination or not with other agents such as checkpoint inhibitors represents a major public health issue but remains a challenge. A dozen of vaccine drug products is expected to be marketed in the next few years. In this context, off-the-shelf and potent platforms are more attractive than individualized vaccines that are tailored to each patient in many aspects. We have recently developed a new cell-based vaccine approach named PDC*vac (formerly GeniusVac). This cell-based vaccine was developed thanks to the establishment, from leukemic PDC, of unique human PDC line (PDC*line). We have previously shown the potential of peptide-loaded PDC*line to induce strong and fully functional HLA-A*02:01-restricted anti-tumor immunity in vitro and in vivo (Aspord et al, 2010, 2012). PDC*mel (GenusVac-Mel4), our first product is currently developed for the treatment of melanoma patients (NCT01863108). The high efficiency of PDC*vac renders the strategy very attractive for further clinical developments in active immunotherapy field. Our aim is to optimize PDC*vac technology by passing use peptides and render the vaccine easier to manufacture. We engineered our PDC*line using retrovirus to force the endogenous expression of one or more tumor or viral antigenic peptides or whole tumor/viral proteins (gp100, tyrosinase, Melan-A, Mage-A3, CMVpp65, flu, EBV) that broaden the panel of antigens presented. Data obtaining with different polypeptide and protein gene coding constructions and the expansion of multi-specific antigen-specific T-cells at the same time will be presented. Moreover, we will present how we succeed to obtain 100% of efficacy in antigen presentation by the PDC*line engineered to express tumor proteins or polyepitopes. We have validated the use of retrovirus constructs to create a new generation of an “off-the-shelf” cancer vaccines highly potent to stimulate antitumor-specific T-cells.

Use of e-government solutions in local government units

i ich zastosowanie w administracji publicznej. Podjęto próbę zdefiniowania terminu
e-Government, ukazując sposoby pomiaru jego efektywności oraz dokonując oceny poziomu
rozwoju takich wdrożeń w Polsce. Przedstawiono cele, korzyści i bariery związane
z wdrażaniem i użytkowaniem tego typu rozwiązań. Następnie skupiono się na ich wykorzystaniu
w jednostkach samorządu terytorialnego.
Celem artykułu jest zwrócenie uwagi na możliwość zastosowania technologii ICT
w jednostkach administracji publicznej oraz ukazanie korzyści, jakie z takich wdrożeń
wynikają. Przeprowadzony przegląd literatury oraz analiza danych statystycznych pozwoliły
na ocenę dostępnej wiedzy oraz poziomu rozwoju opisywanego zjawiska w Polsce na
tle innych krajów członkowskich Unii Europejskiej.The paper discusses possibilities offered by information and communication
technologies (ICT) and their use in public administration. It defines the “e-Government”
term and shows ways of measuring its efficiency, attempting to analyse its development
in Poland at the same time. Then it presents purposes, advantages and barriers associated
with e-Government implementations. In the last part of this paper, the author focuses on
usage of the ICT in local government units. The main objective of the paper is to emphasize possibility of applying ICT in public
administration units and advantages which result from this kind of implementations.
The review of literature enables assessment of the existing knowledge. Statistical data
analysis let the author evaluate the level of development of described phenomenon in

Diagnosis and Voice Therapy: A Holistic Approach. Methodological Guidelines and a Case Study
Voice is the subject of interest of several scientific and practical disciplines, including speech therapy, phoniatrics, vocal studies, choral conducting, acoustics. This study presents a speech therapy approach of the voice, emphasising the importance of the holistic approach. The author will introduce it as a methodologist and voice therapist – practitioner, conducting various types of classes in the field of voice: prevention, diagnosis and therapy. Inter-laboratory assessment of flow cytometric monocyte HLA-DR expression in clinical samples
International audienceBackground: Diminished expression of human leukocyte antigen DR on circulating monocytes (mHLA-DR) is a reliable indicator of immunosuppression in critically ill patients, predictive of both adverse outcome and septic complications. The objective of the present work was to test, in an interlaboratory clinical study, a standardized protocol for mHLA-DR measurement by flow cytometry.Methods: mHLA-DR was assessed in fresh whole blood according to a standardized staining protocol. Cells were analyzed on different flow cytometers (FC500, Navios, FACS Canto II) in different laboratories (Lyon and Grenoble). Results were expressed as numbers of antibodies bound per cell (AB/C).Results: Correlations between results were excellent (Pearson and interclass correlation coefficients > 0.98). Coefficients of variations for intra-assay precision ranged from 1.9 to 3.2%. Conclusion: The present report highlights the robustness of this standardized flow cytometric protocol for mHLA-DR measurement in multicentric clinical studies
