Data on the positive synergic action of dimethylacetamide and trehalose on quality of cryopreserved chicken sperm
AbstractThis data article contains supporting information regarding the research article entitled “Combined effect of permeant and non-permeant cryoprotectants on the quality of frozen/thawed chicken sperm”(Mosca et. al., 2016) [1]. The combined effect of the permeant cryoprotectants agent dimethylacetamide and the non-permeant cryoprotectants agent trehalose on the quality of frozen-thawed chicken semen was assessed. In particular, the quantitative dimethylacetamide/trehalose ratio was investigated freezing semen samples according to the following treatments: trehalose 0.1M+0% dimethylacetamide (DMA-0), trehalose 0.1M+3% dimethylacetamide (DMA-3), trehalose 0.1M+6% dimethylacetamide (DMA-6)
Tão dúbio, tão coerente: forma e conteúdo nas crônicas jornalísticas de Sayed Kashua
Este artigo consiste na análise de duas crônicas jornalísticas do escritor árabe-israelense Sayed Kashua publicadas no jornal israelense Haaretz. Estes textos são selecionados devido à sua riqueza de conteúdo e pelo uso de estratégias retóricas, tais como a autoironia e o sarcasmo, para expor os conflitos pelos quais passa o “eu do cronista” retratado pelo autor. Este estudo tem como base os Estudos Culturais e os Novos Paradigmas Literários, ferramentas que permitem o estudo das crônicas jornalísticas como textos literários. Nestas crônicas, de uma maneira despretensiosa, partindo de experiências subjetivas e de temas familiares, com autoironia e tom satírico, Kashua reporta as experiências de um árabe israelense que vive num embate em torno de suas duplas lealdades e ideologias conflitantes, refletindo uma negociação entre sua etnicidade e sua nacionalidade. A crônica, neste contexto, é considerada o local apropriado para a escrita de Kashua: a ambiguidade do gênero é similar à necessidade de quebras de dualidades na sociedade israelense para entendermos e convivermos com o “eu do cronista” de Sayed Kashua. So dubious, so coherent: form and content in Sayed Kashua´s journalistic chronicles - Abstract: This paper consists of the literary analysis oftwo journalistic chronicles written in Hebrew by the Muslim Arab Israeli author Sayed Kashua published in the Israeli newspaper Haaretz. These texts are selected due to their richness of content and use of rhetorical strategies such as self-irony and sarcasm to expose the conflicts through which the “I of the chronicler” portrayed by the author goesthrough. This analysis will rely on theoretical assumptions of Cultural Studies and the “New literary paradigms”, tools that will enable us to study the journalistic chronicle as a literary text. In these chronicles, in a non-pretentious way, coming from the subjective experiences and family topics, with self-irony and in a satirical way, Kashua reports the experiences of an Israeli Arab who lives in a clash over his dual loyalty and conflicting ideologies, reflecting a negotiation between his ethnicity and his nationality. The chronicle, in this context, is considered the appropriate location for Kashua’s writings: the ambiguity of the literary genre is similar to the necessity of breaking dualities in Israeli society to understand and to be acquainted with the “I of the chronicler” of Sayed Kashua
Assisted Oocyte Activation Method (Ca ACTIVATION) Improves Fertilization in Patients with Recurrent Fertilization Failure
Abstract: Total fertilization failure (TFF) happens in 1–3% of total intracytoplasmic sperm injection (ICSI) cycles and may reoccur in sequent cycles. Regardless the high success rate with the applying of intracytoplasmic sperm injection (ICSI), there's still a little number of couples who cannot get fertilized eggs after using ICSI. Six couples experiencing recurrent TFF or low fertilization rate (<10%) after ICSI were recorded in this study. Compared with the regular ICSI group and the conventional ICSI-AOA group, the new AOA method using calcium ionophore, can seriously increase the fertilization rate from less than 10% up to approximately 50% in most cases. The normal distribution of sperm-related oocyte activation factor phospholipase C zeta (PLCz1) in the sperms of the cases indicated the absence of an aberrant Ca signaling activation.
AOA method using calcium (Calcium ionophore A23187) was able to effectively improve the fertilization rate in the majority of patients suffering from TFF. This AOA method had a potential therapeutic effect on those couples experiencing TFF, even after intracytoplasmic sperm injection (ICSI), which may overcome the severe fertilization deficiencies in patients with a repeated low fertilization or TFF.

Distributed chance-constrained model predictive control for condition-based maintenance planning for railway infrastructures
We develop a Model Predictive Control (MPC) approach for condition-based maintenance planning under uncertainty for railway infrastructure systems composed of multiple components. Piecewise-affine models with uncertain parameters are used to capture both the nonlinearity and uncertainties in the deterioration process. To keep a balance between robustness and optimality, we formulate the MPC optimization problem as a chance-constrained problem, which ensures that the constraints, e.g., bounds on the degradation level, are satisfied with a given probabilistic guarantee. Two distributed algorithms, one based on Dantzig-Wolfe decomposition and the other derived from a constraint-tightening technique, are proposed to improve the scalability of the MPC approach. Computational experiments show that the distributed method based on Dantzig-Wolfe decomposition performs the best in terms of computational time and convergence to global optimality. Effect of n-methylacetamide concentration and thawing rate on chicken sperm quality after cryopreservation
In seeking alternative cryoprotectants to glycerol for a reference chicken semen freezing procedure, the aim of the present study was to compare the effect of two concentrations of N-Methylacetamide (MA) and two thawing rates on the quality of frozen-thawed semen. Semen samples were diluted in Lake pre-freezing extender, including 0.1 M trehalose in presence of 6% or 9% MA, loaded into straws, frozen in nitrogen vapors, and stored in liquid nitrogen. The following thawing treatments were used: 5 °C for 100 s and 38 °C for 30 s. Sperm quality (cell membrane integrity, motility and kinetic parameters) was assessed before and after cryopreservation. The decrease of MA concentration from 9 to 6% improved sperm quality after freezing/thawing and this effect was dependent on thawing temperature. Decreasing the MA concentration from 9 to 6% improved the proportion of undamaged membrane, motile, and progressive motile sperm recovered after thawing at 5 °C for 100 s; in contrast, no effect of the MA concentration was observed thawing at 38 °C for 30 s. Therefore, the treatment with 6% MA and thawing at 5 °C for 100 s has given the best cryoprotective action. These results contribute to improve the efficacy of the current chicken semen cryopreservation procedures

Muhammad Hasan Askari initiated discussions on Pakistani Literature in 1949. He presented his theory of “Pakistani Adab”. By presenting this theory, Askari intended to form such a common and harmonized national literature which has all the historical, cultural and social features and characteristics of our golden Islamic values. Samad Shaheen, Mumtaz Sheerin, Saleem Ahmad, Sajjad Baqir Rizvi, Muzaffar Ali Sayed, Jamil Jalbi and many other literary figures warmly welcomed and supported Askari’s theory of “Pakistani Adab”. The reflection of these discussions can be seen in the literary journals like “Saqi”, “Naqoosh”, “Mah e Nau”, Humayon”, “Adab e Latif”, “Fanoon”, “Nizam”, “Urdu Adab” and “Naya Doar” of 50’s, 60’s and 70’s
Assessment of Sperm Viability and Computer-Assisted Motility Analysis in Budgerigars (Melopsittacus undulatus): Effect of Several In Vitro Processing Conditions
In order to preserve endangered psittacine species, more basic and applied research in reproductive biology is required. Assisted reproductive technologies such as artificial insemination play an important role in parrots species conservation programs to overcome the problem of infertile eggs and male infertility. The aim of this study was to define an effective in vitro protocol in order to standardize the sperm quality evaluation in psittacines, studying Melopsittacus undulatus as model species. Semen was collected from twenty adult males by massage technique from May to June. Sperm concentration was measured by the spectrophotometric method. Sperm quality (sperm membrane integrity (SMI), motility, and kinetic parameters) was assessed on fresh semen. Three different experimental protocols were performed to compare the effects of various processing conditions on SMI, motility, and kinetic parameters. In protocol 1, test was performed by Lake extender with three different pH, 7.4 versus 8.2 versus 8.4, and two different equilibration temperatures after dilution of fresh semen (4 degrees C versus 25 degrees C). In protocol 2, two dilution rates of semen after collection were valuated, 1 : 3 versus 1 : 4, as well as three different semen storage temperatures (4 degrees C versus 25 degrees C versus 38 degrees C) before sperm motility analysis with the computer-assisted sperm analysis (CASA). In protocol 3, two different Makler chamber temperatures (38 versus 41 degrees C) during motility analysis were tested. A significant progressive improvement in spermatozoa motility and kinetic parameters was registered with pH 8.4. Progressive motility and all kinetic parameters were higher at 4 degrees C equilibration temperature. Straightness (STR) kinetic parameter was better with 1 : 4 dilution rate. Total motile sperm was higher in 41 degrees C Makler chamber. In this study, for the first time, the effects of different processing protocols on psittacines seminal quality analysis were investigated. Effect of dimethylacetamide and N-methylacetamide on the quality and fertility of frozen/thawed chicken semen
The aim was to compare the effect of dimethylacetamide (DMA) and N-methylacetamide (NMA) concentrations on the quality and fertility of post-thaw chicken semen. Ejaculates were obtained from 30 Hi-Line White roosters and processed according to the following treatments: lake pre-freezing extender + 0.1 M trehalose (LPF-T) + 6% DMA (control treatment), LPF-T + 9% DMA, LPF-T + 6% NMA, and LPF-T + 9% NMA. Sperm quality (viability, motility, and kinetic traits) was assessed before and after cryopreservation. A total of 15 laying hens per treatment were inseminated to assess fertility and embryo viability. Sperm cryopreserved in presence of DMA had significantly better in vitro quality compared to NMA, showing the highest proportion of viable and progressive motile sperm recovered after thawing. Furthermore, proportion of progressive motile sperm and the VCL, LIN, ALH, and WOB mean values were significantly improved in semen samples frozen/thawed with 6% compared to 9% cryoprotectant concentration. However, the best cryoprotective action on sperm quality played by DMA and the lowest cryoprotectant concentration did not translate into a concomitant advantage in in vivo semen fertility that showed no differences between cryoprotectant and cryoprotectant concentration treatments. Finally, the cryoprotectant DMA and NMA showed an opposite effect on embryo viability in comparison with the effect played on in vitro semen quality, being NMA more efficient than DMA on preserving viable embryos. The present results suggest the urgency to further decrease the cryoprotectant concentration in poultry semen freezing procedures and to assess the specific toxic effect of cryoprotectant on sperm integrity, fertility, and embryo development
