New benzimidazole derivatives containing hydrazone group as anticancer agents: Inhibition of carbonic anhydrase IX and molecular docking studies
In this study, we propose identifying potential novel compounds targeting carbonic anhydrase (CA) IX and anticancer activity. To study the impact of these synthesized compounds on CA IX and anticancer activity, we have developed and synthesized novel benzimidazole-hydrazone derivatives (3a-3j). The target compounds' H-1 NMR (nuclear magnetic resonance), C-13 NMR, and high resolution mass spectrometry spectra were used to confirm their chemical structures. L929 (healthy mouse fibroblast cell line) used as control healthy cell line and MCF-7 (breast cancer), C6 (rat glioblastoma), HT-29 (colon cancer), cells were used in cell culture studies. As a result of cell culture studies, it was determined that the newly synthesized compounds 3d and 3j had cytotoxic effects on colon cancer. Again, it was determined that the compound 3d had a more toxic effect than cisplatin on both breast cancer and glioma cells. According to the CA IX activity results, compounds 3d and 3j were found to have the highest activity. Compounds 3d and 3j are essential for having anti-cancer properties and inhibiting the carbonic anhydrase IX enzyme. Molecular docking of these compounds was carried out in the active site of CA IX. Flow cytometry and immunofluorescence microscope analyses also confirmed that these compounds had cytotoxic effects on cancer cells.Anadolu University Medicinal Plant Drug and Scientific Research and Application Center (AUBIBAM)The authors thank Anadolu University Medicinal Plant Drug and Scientific Research and Application Center (AUBIBAM) for their support and contributions

Effect of Reaction Parameters on the Synthesis of Cyclodextrin-Based Nanostructured Polymers for Drug Delivery
In this study, cyclodextrin-based nanostructures (CDNSs) were synthesized through the cross-linking of cyclodextrin (CD) with epichlorohydrin (ECH) as a cross-linker. Two types of CDNSs, alpha-CDNS and beta-CDNS, were prepared to systematically investigate the influence of reaction parameters-such as the solubilization time of alpha-CD and beta-CD, the molar ratio of ECH to CD, and NaOH concentration-on the physicochemical properties of the final product. Naproxen (NAP), a poorly water-soluble drug, was selected as a model compound to assess the drug-loading capacity of the synthesized CDNSs. The effect of each reaction parameter on NAP integration into the CDNSs was examined at varying weight ratios. The optimal reaction conditions were determined to be a solubilization time of 6 h, an ECH/CD molar ratio of 8/1, and an NaOH concentration of 33%. Under these conditions, the NAP loading efficiency of alpha-CDNSs was calculated as 67.12%. Comparative analysis revealed that alpha-CDNSs outperformed beta-CDNSs in terms of drug-loading capacity. Additionally, the synthesized CDNSs and NAP-loaded CDNSs were characterized using FTIR, DSC, XRD, SEM, and Zetasizer analyses, while the NAP concentration was determined by HPLC.Sivas Cumhuriyet University Scientific Research Projects (CUBAP); [M-561]This study was financially supported by Sivas Cumhuriyet University Scientific Research Projects (CUBAP) under project number M-561
Comprehensive bibliometric analysis and perspectives on therapies targeting colon cancer stem cells over a 40-year period
The presence of cancer stem cells (CSCs) is one of the primary causes of recurring therapy resistance because they have two main capacities: self-renewal and avoiding apoptotic pathways. Despite their relevance, no full bibliometric analysis has yet been done in this topic. The goal of this work is to use bibliometric analysis to map the fundamental and emergent areas in therapeutics targeting colon cancer stem cells. To perform bibliometric analysis on colon cancer stem cells (CCSCs) literature, spanning roughly the last 40 years, in order to establish a firm base for future projections by emphasizing the findings of the most notable research. All information pertinent to CCSCs was accessed from Web of Science Core Collection database. In order to identify and analyze the research hotspots and trends related to this topic, Biblioshiny (RStudio) and VOSviewer were utilized to ascertain the countries/regions, institutions, journals, authors, references, and keywords involved. The targeted time span covered 1735 research-, and review articles. The most frequent keywords were colorectal cancer, cancer stem cells, and colon cancer, while the most trending keywords in the last few years were protein stability, spheroid formation, ubiquitination, exosomes, patient-derived organoids, and gut microbiota. Over the past 40 years, there has been a significant advancement in researchers' understanding of colon cancer stem cells. In addition, the cluster map of co-cited literature showed that colon cancer stem cell research has emerged as a research hotspot. It was also anticipated that the main focus of the future efforts appears to involve clinical applications of cell-targeted colon cancer therapy. These results provide researchers with a comprehensive understanding of this field and provide insightful ideas for further research. (c) 2025 The Author(s). Our thesis on Ebubekir Kani Efendi, born in Tokat in 1712, a member of the Tokat Mevlevihanesi, an Ottoman poet and scribe, and one of the Ottoman State Divan secretaries, and his Sufi understanding, consists of three sections. The first section includes explanations about the topic, scope, method, and format of the thesis. The second section provides information about the life of Ebubekir Kani Efendi. The third section examines Ebubekir Kani Efendi's Sufi views within the context of his Divan, analyzed under Sufi concepts. Ebubekir Kani Efendi, who lived in the 18th century, continued his spiritual and moral training at the Tokat Mevlevihanesi until he was forty. When Hekimoğlu Ali Pasha became Grand Vizier for the third time, he passed through Tokat on his way from Trabzon to Istanbul and took Ebubekir Kani Efendi to Istanbul in 1755. In Istanbul, Kani Efendi completed his education at the Divan-ı Hümayun office and was given the rank of civilian captain. As a Divan Secretary, Ebubekir Kani Efendi was often someone who was bored with bureaucratic routines and did not enjoy being registered. He traveled to places like Bucharest, Wallachia, and Ruschuk, worked as a scribe for the local beys, and wrote down his observations about these places. His writings were compiled by his followers and admirers after his death. While fulfilling his official duties, Kani Efendi did not refrain from writing his feelings and thoughts. He appears as an outspoken, affable Ottoman intellectual. In his poems, he explored themes such as the truth of existence, the importance of servitude consciousness, love, divine and metaphorical love, as well as social issues and moral shortcomings, using subtle metaphors and witticisms. He especially emphasized the significance and meaning of human existence by employing Sufi discourse and concepts. In this thesis, we aimed to introduce Ebubekir Kani Efendi and shed light on his spiritual and Sufi aspects within the context of the poems in his Divan
Covalently Bonded 1D Chains and 2D Networks From Si-Doped CL-20: Computational Study
To discover high-energy-density materials with characteristics superior to current models, it is necessary to study a wide range of potential structures. A promising representative of new derivatives of the class of high-energy compounds is silicon-substituted molecules CL-20, which have a reactivity and kinetic stability close to pure CL-20 but have a higher density and energy release. Low-dimensional covalent SiCL-20 nanostructures based on silicon analogue of the classical CL-20 high-energy molecule are considered in this work. Covalent nanostructures may have advantages over molecular crystals due to their special properties, such as higher packing density and kinetic stability. It has been established that silicon-substituted CL-20 molecules can connect through CH2 molecular bridges into covalent structures. Geometrical parameters, energy characteristics, electronic properties, and quantum chemical reactivity descriptors for several representatives of 1D and 2D systems based on Si5CL-20 have been calculated using density functional theory. The skeleton of each silicon fragment of the CL-20 system undergoes small changes when combined into covalent chains and networks. Still, the systems retain their consistency, and the effective diameter of the silicon frameworks in the nanostructure takes average values from 4.300 to 4.462 & Aring;. The binding energy of nanostructures increases with the number of silicon CL-20 fragments in the system. The binding energies for a single silicon molecule CL-20 and a double chain SiCL-20 consisting of 12 fragments are 3.846 and 4.077 eV/atom, respectively. Thus, the silicon nanostructures become more thermodynamically stable with increasing the size and dimension of the compound. The study of electronic characteristics made it possible to establish that the value of the HOMO-LUMO gap decreases with an increasing number of fragments in the system, and the considered SiCL-20 covalent molecules can be classified as wide-gap semiconductors, like their classical CL-20 analogues. For example, the values of the HOMO-LUMO gaps for silicon derivatives of CL-20 with dimensions 1 x 1, 6 x 1, 6 x 2, and 4 x 3L are 5.601, 4.378, 4.004, and 3.882 eV respectively. Despite their highly stressed skeleton, they are stable enough to be considered for energy applications and are promising candidates for building blocks of high-energy materials and fuels.Ministry of Science and Higher Education of the Russian Federation [FSWU-2024-0014]; Ministry of Science and Higher Education of the Russian FederationThe work was supported by the Ministry of Science and Higher Education of the Russian Federation, project FSWU-2024-0014
