Investigation of Antioxidant Effect of Origanum Hypericifolium Extracts in Different Solvents
Origanum hypericifolium is one of the most widely distributed endemic species of the family Lamiaceae. Origanium hypericifolium contains mostly monoterpenes. Monoterpenes are volatile compounds with 10 carbons and are widely found in the plant kingdom. Monoterpenes, which are used as fragrances in perfumes and foodstuffs, have anti-fungal, anti-bacterial, anti-oxidant and anti-cancer effects.Various in vitro assays can be used to determine the antioxidant activity of Origanium hypericifolium extracts. In our study, total polyphenolic, total flavonoid content, iron reducing power, cupric ion reducing capacity and radical scavenging activity were evaluated to the antioxidant properties of various extracts of Origanum hypericifolium. TAS and TOS levels were also analyzed. The highest DPPH radical scavenging acitiviy value was 3.09±0.01 µg/mL in ethanol extract, Total polyphenol 144.00±1.89 µg GAE /g in ethanol extract, Total polyphenol 17. 08±0.85 µg CE /g in ethanol extract, FRAP 942.46±1.89 µg TE /g ethyl acetate and CUPRAC 2.54±0.01 mmol TE/g ethyl acetate extract.[No abstract available
Synthesis, optimization, and characterization of β-cyclodextrin-based biological metal-organic frameworks as drug carriers
In this study, beta-cyclodextrin (beta-CD)-based metal-organic frameworks (MOFs) were synthesized using the vapor diffusion method, a cost-effective and straightforward approach, to evaluate their potential as drug delivery systems. Ketoprofen (KP), a poorly water-soluble nonsteroidal anti-inflammatory drug (NSAID), was used as a model drug. The effects of reaction time, temperature, ligand-to-metal salt molar ratio and cetrimonium bromide (CTAB) surfactant addition on the properties of beta-CD-Bio-MOFs were systematically investigated. Optimal synthesis conditions-1 day, 30 degrees C, and a 1:4 beta-CD:KOH molar ratio-produced quadrilateral crystals averaging 10-20 mu m in size. Using KCl as the metal source yielded uniform crystals (10-40 mu m), whereas NaOH and FeCl3 did not result in consistent structures. Adding CTAB as a surfactant further reduced crystal sizes to 5-20 mu m. Also, KP loading experiments were conducted to optimize parameters such as temperature, agitation speed, loading time, solvent medium, and mass ratio for achieving maximum loading efficiency. The optimal conditions were determined to be 25 degrees C, 200 rpm, and 3 h in ethanol. Under these conditions, loading efficiencies of 13.4%, 9.6%, and 15.8% were achieved for beta-CD-Bio-MOF, KCl-beta-CD-Bio-MOF, and CTAB-beta-CD-Bio-MOF samples, respectively, with corresponding surface areas of 11.8, 9.7, and 18.4 m2 g-1. In comparison, pure beta-CD, with a specific surface area below 5 m2 g(-)1, exhibited a lower loading efficiency of 4.1%
In vitro cytotoxicity, gene expression, bioinformatics, biochemical analysis, and in silico analysis of synthesized carbonitrile derivatives
In this work, an efficient single-step green synthesis protocol of 2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitriles was applied with the purpose of their biological activity studies. It was found that the cytotoxic activities of 2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitriles in the SHSY-5Y cancer cell line were most active after 72 h of incubation. It was determined that 1,6-diamino-4a-methyl-3-oxo-2,8-diphenyl-2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitrile showed higher anticancer activity on SHSY-5Y cancer cells compared to other molecules. The expression levels of Myc-N, Casp2, Tp53, RAD51, BRCA2, MDM2, BAX and NF-kappa B1 genes were analyzed by RT-PCR method by applying 2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitriles to the SHSY-5Y neuroblastoma cancer cell line. It was observed that there were differences in expression levels between study groups and all differences were found to be statistically significant (p < 0.000). Bioinformatics analysis was performed using the STRING v 11 Protein interaction tool. When the molecules in the SHSY-5Y cell line were compared to the control group, an 80% increase in GSH levels was observed in 1,6-diamino-4a-methyl-3-oxo-2,8-diphenyl-2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitrile and 1,6-diamino-8-(4-chlorophenyl)-4a-methyl-3-oxo-2-phenyl-2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitrile groups. SOD enzyme activity increased significantly in the former group compared to the control and other groups. LDH activity was detected at a higher rate in this cell line compared to the control group. Calculations were made for 2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitriles using the Gaussian software package on B3LYP, HF, and M06-2 x levels with the 6-31++g(d,p) basis sets. The activities of the 2,3,4,4a-tetrahydroisoquinoline-5,7-dicarbonitriles found in the study against SH-SY5Y protein (2F37, 3PBL and 5WIV), comparison has been made. MM/GBSA methods are calculated binding free energy for all molecule and 3PBL protein. Novel thiohydantoin derivatives: design, synthesis, spectroscopic characterization, crystal structure, SAR, DFT, molecular docking, pharmacological and toxicological activities
In a sustained search for novel drug candidates with multifunctional therapeutic potential, a serie of novel thiohydantoins was designed and synthesized through an alkylation reaction. The series of synthesized compounds (AM0-3) was confirmed through X-ray diffraction studies, as well as by standard spectroscopic methods (1H and 13C NMR, IR), and high-resolution mass spectrometry. The obtained heterocycles were evaluated for their antibacterial, antidiabetic and antioxidant activities both in vitro and in vivo. In vitro antibacterial screening revealed that derivatives AM0, AM1, and AM2 exhibit inhibition comparable to that of Chloramphenicol against the three strains: Rhodococcus equi, Rhodococcus sp. GK1, and Rhodococcus sp. GK3. However, compound AM3 shows superior inhibition compared to the reference, with an inhibition zone diameter of 24.00 mm, compared to 21.75 mm for Chloramphenicol. Compound AM3 exhibits the highest alpha-amylase inhibitory activity, with an IC50 of 22.09 mu M, which is significantly lower than that of acarbose (IC50=124.32 mu M). Similarly, compound AM2 shows greater alpha-amylase inhibitory activity compared to the reference, with an IC50 of 46.97 mu M. In antioxidant tests (DPPH, ABTS, and FRAP), AM3 demonstrates superior properties compared to ascorbic acid, the reference drug. Additionally, toxicity studies revealed that compounds AM2 and AM3 are nontoxic at a dose of 2 g/kg body weight. Furthermore, the compounds AM0-AM3 were analyzed using Gaussian calculations at the B3LYP, HF, and M062X level on the 6-31++g(d,p) basis set. Effects of the size and applied electric field on the photoionization cross-section of elliptical cylindrical CdS/ZnS core-shell quantum dots immersed in various dielectric matrices
In this study, the effects of the electric field in different directions on hydrogenic donor impurity (HDI) confined to a CdS/ZnS elliptical core/shell cylindrical quantum dot (ECSCQD) immersed in a dielectric matrix (DM) were investigated. The binding energy (BE) and their corresponding photoionization cross-section (PICS) are computed by solving the Schrodinger equation in cylindrical coordinates in the framework of the effective mass approximation (EMA) using a variational and perturbation approach. The effects of different dielectric matrices, lateral and normal electric field (EF) force, geometric factors such as the size of ECSCQDs and ellipticity constants on BE and PICS were presented. It has been indicated that weak lateral EF strength compared to normal EF strength, dielectric matrix, and geometric factors have a significant effect on the BE and PICS. In addition, it was observed that dielectric matrices and different ellipticity constants were quite effective on the Stark shift (S.S.) in the hydrogenic donor impurity ground state energy due to the electric field. Timing and Adherence Matter for Sodium-Glucose Cotransporter-2 Inhibitors in Heart Failure
Background It is imperative to maintain the use of sodium-glucose cotransporter-2 inhibitors (SGLT-2is) in patients with diabetes both after the index diagnosis of heart failure (HF) and even prior to the index diagnosis of HF. We aimed to investigate whether timing of SGLT-2 is before the index diagnosis of HF, and second, adherence to SGLT-2is in the form of the proportion of days covered metric matter in patients with HF and diabetes. Methods and Results All-cause death up to 7 years were evaluated in HF with diabetes from the subgroup analysis of TRends-HF (TRends in Heart Failure in T & uuml;rkiye). Patients with HF and diabetes, who were prescribed an SGLT-2i either before or after the index diagnosis of HF were identified, categorized according to duration of exposure before the index HF diagnosis and according to proportion of days covered after the index diagnosis of HF, and compared with nonusers. There were 1 229 833 patients with HF and diabetes in the cohort. A total of 247 987 were on an SGLT-2i and had available timing data, and 14.06% had SGLT-2i on board before the index HF diagnosis. Median duration of SGLT-2i exposure before the index HF diagnosis was 417 days. Prognosis was the best among patients with diabetes who were prescribed an SGLT-2i before the index diagnosis of HF with an exposure more than median duration. Of note, among patients who were prescribed an SGLT-2i after the index HF diagnosis; there was a numerically graded increase in all-cause mortality rate such that a >10% decrease in SGLT-2i proportion of days covered was associated with a 59% increase in all-cause death (hazard ratio, 1.21-2.09). Conclusions Regardless of time or adherence, SGLT-2is offer a remarkable all-cause death benefit to patients with HF and diabetes. SGLT-2is' all-cause death benefit for patients with HF and diabetes was greatest when it was prescribed before the HF index diagnosis. Poor adherence to SGLT-2is was associated with worsening survival in patients with HF and diabetes following the diagnosis of index HF
A cross-sectional study on the comparison of serum SIRT-1 and MMP-9 levels of patients with bronchiectasis and healthy controls
Background & Objectives: Bronchiectasis is the permanent enlargement of the bronchi following damage to the respiratory tract (bronchi) in the lungs. Bronchiectasis not associated with cystic fibrosis is gaining an increasing place among chronic respiratory diseases worldwide. The purpose of this study was to identify the levels of MMP-9, known to cause bronchial damage in chronic pulmonary illness, and SIRT-1, an anti-aging and anti-infective regulatory protein, in patients with bronchiectasis and to evaluate the importance of these biomarkers in diagnosis. Methods: This cross-sectional study was conducted in the Chest Diseases Clinic of Sivas Cumhuriyet University Medical Faculty Hospital between November 2020 and September 2022. We recruited 30 patients with bronchiectasis whose diagnosis was verified by high-resolution chest CT scan and 30 healthy controls. SIRT-1 and MMP-9 levels in the serum of the study group were determined by the ELISA method. Results: SIRT-1 and MMP-9 concentrations were found to be statistically significant. In comparison to the control group, it was observed that the bronchiectasis group had a lower serum SIRT-1 levels (p<0.001). The bronchiectasis group had higher serum MMP-9 values than the control group (p<0.001). Age-related differences in SIRT-1 and MMP-9 concentrations were not observed. No correlation was found between MMP-9 and SIRT-1. The results of Receiver Operating Characteristic (ROC) analysis indicated that MMP-9 has relatively high sensitivities. Conclusions: We concluded that, higher inflammation elevates MMP-9 levels while decreasing SIRT-1 levels. MMP-9 and SIRT-1 may be potential biomarkers in the diagnosis of bronchiectasis. © 2025, Professional Medical Publications. All rights reserved
The impact of malignancy on death anxiety and psychological well-being in middle-aged and older patients undergoing abdominal surgery: a quasi-experimental study
Background: Malignancy is a critical factor affecting death anxiety and psychological well-being. This study examined the impact of malignancy on death anxiety and psychological well-being in middle-aged and older adult patients undergoing abdominal surgery and projected it along with sociodemographic profiles. Methods: This study is quasi-experimental in design. It was conducted with patients undergoing abdominal surgery at a university hospital due to suspected malignancy. A total of 93 patients participated in the study: 57 patients diagnosed with malignancy were included in the experimental group, while 36 patients without a malignancy diagnosis were included in the control group. Data were collected using a questionnaire developed by the researchers based on the literature, the 'Turkish Death Anxiety Scale', and the 'Psychological Well-being Scale'. The analyses revealed that the data were normally distributed, and parametric tests were used to analyze differences between demographic variables and groups. Cohen's d and r values were examined to assess the effect size in the tests. Results: The study found a significant difference in death anxiety between the experimental and control groups (P < 0.05), with higher mean scores in the experimental group. Malignancy had a large effect on death anxiety (d = 1.42). Additionally, the control group had significantly higher mean psychological well-being scores compared to the experimental group (P < 0.05). Conclusions: The study found that malignancy significantly affects death anxiety and psychological well-being. Increased death anxiety in patients with malignancy was associated with reduced psychological well-being. Thus, assessing death anxiety and providing targeted nursing care are essential for supporting the psychological well-being of these patients
Destination image's destination effect on satisfaction (Iran example)
Çalışmanın amacı, İran'ın destinasyon imajının belirlenmesi ve destinasyon imajının üç boyutuyla (Deneyim Kalitesi, Çekicilik, Değer/Çevre), destinasyon memnuniyeti üzerinde etkisinin incelenmesidir. Bu çalışmayla beraber; imaj ve destinasyon imajı kavramlarını tanımlayarak bu kavramlarla ilgili yapılan bilimsel çalışmalar ışığında bir bütünlük sağlamak ve bu destinasyon imajının turistlerin destinasyon memnuniyetinin üzerindeki etkilerini ortaya koymaktır. Aynı zamanda destinasyon imajına olan tutumlarının destinasyon memnuniyetini ne derecede etkilediğini ortaya koymaktır. Bu amaçlar doğrultusunda İran'da bulunan 376 turiste anket gerçekleştirmiştir. Araştırmada nicel yöntem kullanılmıştır. Anketten elde edilen bulgular SPSS 25.0 programı kullanarak değerlendirilmiştir. Araştırmada turistlerin destinasyon imajının destinasyon memnuniyetine etkisini belirlemek amacıyla Regresyon analizi yapılmıştır. Analiz sonucunda katılımcıların Destinasyon memnuniyetine "Deneyim Kalitesi", "Çekicilik" ve "Değer / Çevre" faktörlerinin etkilediği sonucuna varılmıştır. Destinasyon imajı yönelik tutumların alt boyutu olan Deneyim Kalitesi, Çekicilik Destinasyon memnuniyetini etkilediği, Değer / Çevre, tutumun Destinasyon memnuniyetini etkilemediği tespit edilmiştir.The aim of the research is to determine the destination image of Iran and to examine the effect of destination image on destination satisfaction through its three dimensions (Quality of Experience, Attractiones, Value/Environment). This study aims to provide a comprehensive understanding by defining the concepts of image and destination image, supported by scientific studies on these topics, and to reveal the effects of this destination image on tourists' destination satisfaction. Additionally, it seeks to explore the extent to which attitudes towards destination image influence destination satisfaction. To achieve these objectives, a survey was conducted with 376 tourists in Iran. A quantitative research method was employed in the study. The findings obtained from the questionnaire were evaluated using the SPSS 25.0 program. Regression analysis was performed to determine the effect of tourists' destination image on destination satisfaction. The results indicated that the factors of " Quality of Experience," "Attractions," and "Value/Environment" influence destination satisfaction. It was found that while the sub-dimensions of destination image attitudes, namely Quality of Experience and Attractiones, affect destination satisfaction, Value/Environment does not have a significant impact on destination satisfaction
SAĞLIK ÇALIŞANLARININ AFETLERE HAZIRLIK ALGILARI VE PSİKOLOJİK SAĞLAMLIKLARININ İŞ TATMİNLERİ ÜZERİNE ETKİSİ
Bu araştırma, sağlık çalışanlarının afetlere hazırlık algıları ve psikolojik sağlamlıklarının iş tatminleri üzerine etkisini incelemeyi amaçlamaktadır. Çalışmada ilişkisel tarama modeli kullanılmış olup 107 sağlık çalışanıyla araştırma gerçekleştirilmiştir. “Kişisel Bilgi Formu”, Hastane Öncesi Acil Sağlık Hizmetlerinde Çalışan Personelin Afetlere Hazırlık Algısı Ölçeği (HASPAHA), Kısa Psikolojik Sağlamlık Ölçeği (KPSÖ) ve İş Tatmini Ölçeği(İTÖ)’nin kullanıldığı araştırmada verilerin analizinde çoklu doğrusal regresyon analizinden faydalanılmıştır. Bulgulara göre araştırmaya katılanların 36’si erkek (%33,6) 71’i kadındır (%66,4). Sağlık çalışanlarında yüksek düzeyde afet hazırlık algısı, ortalamanın üzerinde psikolojik sağlamlık ve iş tatmini saptanmıştır. HASPAHA Ölçeğinin istek ve özyeterlilik alt boyutları İTÖ’yü pozitif yönde etkilerden diğer alt boyutların bir etkisi saptanamamıştır. Benzer şekilde KPSÖ’de İTÖ’yü pozitif yönde etkilemektedir. Bulgulardan sağlık çalışanlarının afetlere hazırlıklı olmaya istekli olmaları ve afet anı ve sonrasında ne yapacakları konusunda kendilerini yeterli görmeleri ve psikolojik açıdan sağlam hissetmelerinin iş tatminlerini yükselttiği söylenebilir. Sonuçta sağlık çalışanlarının iş tatminlerinin yükseltilebilmesi için afetlere hazırlık düzeylerinin artırılması ve psikolojik sağlamlıklarının desteklenmesi gerektiği vurgulanmaktadır.[No abstract available
