Synthesis, antimicrobial and antibiofilm activities of silver(I) complexes with N-alkylbenzimidazole derivatives and their protein interaction modelling study
This study investigates the antimicrobial and antibiofilm potential of three silver(I) complexes derived from Nalkylbenzimidazole derivatives: bis[(Z)-1-styryl-benzimidazole]silver(I) nitrate (4), bis[(E)-1-styryl-benzimidazole]silver(I) nitrate (5), and bis(1-cinnamyl-benzimidazole)silver(I) nitrate (6). Detailed synthesis and characterization of these complexes are followed by assessments of their efficacy against Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa, Escherichia coli, and Candida albicans. Among the tested compounds, complex 6 exhibited the highest antimicrobial activity, with a minimal inhibitory concentration of 6 mu mol/L against P. aeruginosa, and significant antibiofilm activity, achieving 67.72 % inhibition at a concentration of 3 mu mol/L. These interesting findings led us to conduct a molecular docking study to understand the mechanisms of action by investigating the interactions between the silver(I) complexes and some key protein targets involved in bacterial and fungal biofilm formation, including (p)ppGpp synthetase/hydrolases, FtsZ proteins, and pyruvate kinases. This comprehensive approach, combining experimental and computational analyses derived from N-alkylbenzimidazole derivatives, particularly complex 6, which exhibits remarkable efficacy against various pathogens, reveals promising therapeutic applications of these silver(I) complexes and advances our understanding of their potential mode of action against biofilm-associated infections

Leptin/Melanocortin Pathway in Cholelithiasis Patients: A Diagnostic Perspective
Background: Cholelithiasis is the most prevalent inflammatory condition of the gallbladder. The regulation of biological processes, including energy homeostasis, and control of body weight are key mechanisms that the leptin and melanocortin pathways play a role in Cholelithiasis is the most prevalent inflammatory condition of the gallbladder. There are various risk factors for the development of gallstone disease, especially weight gain, and obesity is just one of them. This risk factor can be minimized by maintaining appetite and energy balance. Here, leptin and melanocortin pathways are the key mechanisms in maintaining appetite and energy homeostasis. Objectives: The aim of this study was to investigate the relationship between the levels of LEP, LEPR, TrkB, BDNF, POMC, and MC4R proteins in patients with Cholelithiasis. This study aims to determine the relationship between LEP, LEPR, TrkB, BDNF, POMC, and MC4R protein levels, which play a role in maintaining appetite and energy homeostasis, and cholelithiasis. Methods This study examined 44 patients diagnosed with Cholelithiasis and 44 healthy control subjects who had not previously been diagnosed with any form of Cholelithiasis. The levels of leptin (LEP), Leptin Binds To Leptin Receptors (LEPR), Tropomyosin Receptor Kinase B (TrkB), Brain-Derived Neurotrophic Factor (BDNF), Pro-OpioMelanoCortin (POMC), and Melanocortin-4 Receptors (MC4R) molecules were analyzed using the Enzyme-Linked Immunosorbent Assay (ELISA) method. The results were analyzed using the SPSS Software (Version 22.0) program and GraphPad Prism 8.0.1 software. Results: The study found a statistically significant decrease (p < 0.05) in MC4R, TrkB, BDNF, and POMC protein levels in Cholelithiasis patients compared to the control group. There was no statistically significant difference in LEP and LEPR concentration values between the two groups (p = 0.247, p = 0.674). Conclusion: The proteins MC4R, TrkB, BDNF, and POMC, which are involved in the leptin and melanocortin pathways may play a significant role in Cholelithiasis disease. However, more detailed research on the relevant proteins is needed. Nevertheless, this research will guide new studies

Mediator role of cognitive distortions in the relationship between posttraumatic stress disorder symptoms and peritraumatic stress levels
Background: There are many studies showing the relationships between posttraumatic stress disorder (PTSD), peritraumatic stress, and cognitive distortions. However, there is a very limited number of studies focusing on the relationship between PTSD and cognitive distortions in children and adolescents. This study aimed to investigate the relationship between PTSD symptoms and peritraumatic stress levels in adolescents who experienced the 6 February Turkey earthquakes and the mediator role of cognitive distortions in this relationship. Methods: The study sample consisted of 261 adolescents with a mean age of 15.51 +/- 1.07. The Child Posttraumatic Stress Disorder Reaction Index (CPTSD-RI), the Peritraumatic Distress Inventory (PDI), and the Cognitive Distortion Scale (CDS) were administered to the participants. Results: It was determined that there was a positive relationship between the peritraumatic stress levels and PTSD symptoms of the participants, and their peritraumatic stress levels significantly and positively predicted their PTSD symptoms and cognitive distortions. According to the results of the regression analyses, the self-perception, self-blame, helplessness, hopelessness, and perceiving life as dangerous dimensions of cognitive distortions and total CDS scores of the participants had partial mediator roles in the relationship between their peritraumatic stress levels and PTSD symptoms. Limitations: Participants' PTSD symptoms were assessed using self-report scales. Pre-traumatic factors were not questioned. Conclusion: Our results showed that the peritraumatic stress levels of adolescents who survived the 6 February Turkey earthquakes predicted their PTSD symptom levels, and their cognitive distortions played a mediator role in the relationship between their peritraumatic stress levels and PTSD symptoms. Implications and contribution: In psychological and psychiatric assessments of adolescents living in earthquake zones, peritraumatic stress levels should be kept in mind as a significant factor that can pose a risk in terms of PTSD. Furthermore, it is believed that cognitive distortions that can be causally related to PTSD should be taken into account in the treatment of PTSD

However, 68.9% reported that they worked in the same position for a long time and 52.4% reported that they did not do stretching or breathing exercises. NMQ results were found to be statistically significantly different according to gender and place of work (
Comparison of nutritional screening tools in pediatric oncology patients receiving chemotherapy treatment
46th Congress of the European-Society-for-Clinical-Nutrition-and-Metabolism (ESPEN) -- SEP 07-10, 2024 -- Milan, ITALYBackground: Malnutrition in pediatric oncology patients is a complex and multifactorial process, and the most appropriate screening tool to determine the risk of malnutrition is not clear. The study aimed to compare the Screening Tool for Risk on Nutritional Status and Growth (StrongKids), Pediatric Yorkhill Malnutrition Score (PYMS), Screening Tool for the Assessment of Malnutrition in Pediatrics (STAMP), which are commonly used in the general pediatric population, with the Nutrition Screening Tool for Childhood Cancer (SCAN), which was specifically validated for pediatric cancer, in determining the risk of malnutrition. Methods: This cross-sectional study included 92 children aged 2-18 years who were admitted to the hematology- oncology unit and were receiving chemotherapy. The prevalence of nutritional risk was determined using the SCAN, StrongKids, PYMS, STAMP, and anthropometric assessments were performed. Sensitivity and specificity values, and the degree of agreement between screening tools were calculated. Result: The SCAN identified nutritional risk in 69.6 % of the children. According to StrongKids, PYMS and STAMP, the high nutritional risk in children was 43.5 %, 56.5 % and 64.1 %, respectively. The sensitivity of STAMP (81.2 %) was higher than that of StrongKids (60.9 %) and PYMS (79.7 %). The specificity of StrongKids, PYMS and STAMP was 96.4 %, 96.4 %, and 75.0 %, respectively. There was substantial agreement between SCAN and PYMS. Conclusion: PYMS has the best specificity and sensitivity in identifying patients at risk of malnutrition as determined by SCAN. Implications for practice: In addition to anthropometric measurements, population-specific or highly sensitive nutritional screening tools should be used to determine the risk of malnutrition.
