The Effect of Curcuma domestica Val Extract on Phagocytic Activity of Macrophage in Listeria monocytogenes Inoculated BALB/C Mice
Background: Cellular imunological respond is started by macrophage activity as Antigen Presenting Cell, processing antigen in such a way that emerge interactions among immune cells. Listeria monocytogenes infection often used as an example to demonstrate the immune response to the intracellular bacteria. Curcuma has been proven in vitro to have immunomodulatory effect to improve the function of T cell, NKcell, macrophage, and splenocyte. Thus, it is presumed that curcuma extract may influence macrophage phagocytic activity. The aim of this research is to find out whether there is a difference between macrophage of mice inoculated with Listeria monocytogenes after treatment of curcuma extract in stepped dose compared with control.Design and Method: This laboratory experimental research used a Post-Test Only Control Group Design method. The samples were 20 males of balb/c mice. The mice were divided into 4 groups adopted for a week before the treatments for 20 days. The groups consist of control group, group I (receiving 0.13mg/day of curcuma extract), group II (receiving 0.26 mg/day of curcuma extract, group III (receiving 0.52mg/day curcuma extract). Mice were inoculated with Listeria monocytogenes on day 15. After isolation, the macrophage was desensitized using latex beads and observed under the microscope. The data were analyzed using One way Anova test was followed by Post hoc test using SPSS 15 programme.Result: Index of macrophage phagosytosis in control group, group I, II,II were 0.914±0,27, 1.940±0,42, 2.914±0.56, 1.30±0.31 respectively. One Way Anova test showed a difference among each of groups P<0.05 except for group I and II.Conclusion: Curcuma treatment can increase macrophage phagocytic activity on balb/c mice inoculated with Listeria monocytogenes (Sains Medika, 4(1):63-70)
Pengaruh Lendir Bekicot (Achatina fulica) terhadap Jumlah Sel Fibroblas pada Penyembuhan Luka Sayat Studi Eksperimental pada Kulit Mencit (Mus musculus)
Background: Wound healing is very important to return skin as soon as possible and the integration is a complex and dynamic process. Proliferation phase involving fibroblasts is one of important step-on wound healing. Use of drugs such as povidone iodine on cuts aims to accelerate wound healing is still being debated. Healing by land snails’ mucus can be an alternative because it has many benefits. This study wants to know about the influence of land snails’ mucus on the number of fibroblasts in healing cuts.Design and methods: The study is an experimental research with post test only randomized control group design. The subjects were 18 Mus musculus strain mice that were divided into 3 groups. 1st group as a control group of untreated, 2nd group was given povidone iodine 10%, and 3rd group was given land snails’ mucus during 5th days. On the 6th day the mices were killed for histological study.Results: The data showed that average number of fibroblast in 1st group is 93,85; 2nd group is 126,37; and 3rd is group 217,5. The results of one way anova statistical test showed significant difference among the three groups p = 0,000. The results of post hoc test showed significant difference in all groups (p < 0,05).Conclusion: The mucus of land snail is influence the number of fibroblast in healing cuts (Sains Medika, 4(2):195-203)
The Effect of Exercise on Elderly Body Balance which Measured By Romberg Test Study in Plamongansari Pedurungan Semarang
Background: Balance disorders is a major cause which often lead elders to fall easily. Body balance reduction of elders can be helped by many variety of balance exercises, one of them is elders exercise. There have been many researches on the assessment of body balance. But a research carrying a Romberg Test to assesses static balance by closing and opening eyes as a visual organ has not been done. This study aims to determine the effect of exercise on elderly body balance which measured by Romberg Test.Design and Method: Analytic observational research with cross-sectional approach was conducted to elderly who routinely do exercises and to them who don’t. There were 82 samples, consisted of 41 elderly do exercises and 42 elderly who don’t. Body balances of both groups were measured using the Romberg test. The data were analyzed by chi-square.Result: Among the 41 elderly who do exercise, consisted of 12 men and 29 women, showed 97.56% balanced and 2.44% unbalanced. While 41 elderly aged over 60 years who don’t do exercise, consisted of 15 men and 26 women, showed 46.34% balanced and 53.66% unbalanced. The results of chi-square test showed p = 0.000; r = 0.495.Conclusion: The elderly exercises influence the body balance as measured using the Romberg Test to healthy elderly, with a moderate degree of correlation (Sains Medika, 4(2):142-146)
Pengaruh Konsentrasi Ekstrak Etanolik Daun Teh Hijau (Camellia sinesis L.) dalam Sediaan Krim terhadap Sifat Fisik dan Aktivitas Antibakteri
Background: Green tea, is one of the herbs that have been proven to be effective as an anti-acne, among others.Green tea leaves contain 30-40% polyphenols known as catechins most of which are antimicrobial. For ease of use, the ethanolic leaf extract of green tea is made in cream. This study aimed to determine the effect of variations in the concentration of ethanolic extract of green tea leaves in cream on the physical properties and antibacterial activity of acne bacteria in particular.Design and Method: In this study, green tea leaf extract condensed obtained by maceration using 50% ethanol solution. Cream formula that is made in five concentration ethanolic extract of green tea leaves 1%, 3%, 5%, 7%, and 9% use a modified formula antiacne cream. Cream tested physical properties include homogenity, percent separation, dispersive power and adhesion. During the antibacterial activity was also tested. The data obtained were analyzed statistically using the non-parametric Kruskal-Wallis test followed by Mann Whitney test with a level of 95%.Results: The preparation cream ethanolic leaf green tea extract at various concentrations have good homogenity and not separate, the greater concentration of cream ethanolic extract of green tea leaves get smaller power and energy dispersive adhesion, whereas the inhibitory against Staphylococcus aureus bacteria is getting biger. Conclusion: Cream ethanolic extract of green tea leaves that are comparable with the positive control (Ristra acne creaming) the physical properties and the antibacterial activity at a concentration of 7% (Sains Medika, 4(2):147-156)
Problem Based Learning (PBL) in Competence Based Curriculum and The Accademic Achievement: Evaluation of PBL Implementation
Background: Medical Faculties in Indonesia has implemented problem based learning (PBL) in competence based curriculum as a learning strategy. The aim of this study is to know the effect of PBL method on academic achievement and factors affecting successfull PBL implementation.Design and Method: This study used quantitative and qualitative method. Data collection method is by distributing questionnaires, focus group discussion (FGD) and document analysis of students’ academic achievement. This study included 3 year level students (2004, 2005, 2006) using PBL in school of medicine faculty of medicine and health sciences UMY (FMHS UMY).Result: GPA average > 2.5 is 32.47 %, MCQ > 60 is 33.1% and OSCE is 92.8%. The role of tutor and also practical instructor to facilitate learning objective was not optimal. The factors for the poor implementation of PBL includes curriculum planning (understanding of PBL concept and dissemination of curriculum), implementation of curriculum (various of block load, opportunity for self directed learning, e-learning effectivity, etc) and assessment system (the number of students who reach minimal standar of MCQ is low, validity of MCQ is also low, etc).Conclusion: This study suggest the need to improve planning, implementation and curriculum assessment to support implementation of PBL. The role of Lecturer, tutor and practical instructor should be improved through faculty development to faciliate teaching learning proccess in PBL method (Sains Medika, 4(1):30-38)
Improvements Glutation Levels in the Hippocampus of Aged and Oxidative-Stressed Rats by Supplementation of Alanine-Glutamine Dipeptide
Background: Physiological aging or aging due to oxidative stress causes to disruption of glutathione homeostasis of the hippocampus. Disruption of glutathione homeostasis characterized by a decrease in hippocampus glutathione levels below the normal threshold. Decreased levels of glutathione in the hippocampus was demonstrated decreased hippocampus function. One way to increase glutathione levels in the hippocampus and the function of hippocampus is to increase the concentrations of glutathione precursor. One of the glutathione precursors is alanine-glutamine dipeptide. This research was designed to obtain the most effective level of alanine-glutamine dipeptide supplementation to improve glutathione synthesis in the hippocampus of aged and oxidative-stressed rats. The highest glutathione concentrations in the hippocampus showed the best improvement on the hippocampus function in physiological aging or oxidative-stress aging rats.Design and Methods: The experimental rats were assigned into a completely randomized design with 2x2x4 factorial arrangement. The first factor was the age of the experimental rats, consisted of two levels i.e., 12 and 24 months. The second factor was oxidative stress consisted of two levels, i.e., without or with oxidative stress. The third factor was the level of alanine-glutamine dipeptide administration consisted of 4 concentrations, i.e. 0%, 3%, 5%, and 7%.Results: The results showed that administration of 7% alanine-glutamine dipeptide has resulted the highest levels of glutathione hippocampus in younger (0.0154 mg/mg tissue) or aged (0.0140 mg/mg tissue) rats or in normal (0.0150 mg/mg tissue) and in oxidative-stressed (0.0144 mg/mg tissue) rats. The increased hippocampus glutathione levels were associated to the improved functions of the hippocampus. Conclusion: alanine-glutamine dipeptide administration of 7% consentrations gave the best results on repair function of the hippocampus and has the potential to slow aging, both physiological aging or oxidative-stress aging rats (Sains Medika, 4(1):1-12)
Effect of Tempe Flour Supplementation During Pregnancy on Blood Sugar and Lipid Profile Study on Wistar Rats which Gets Restricted Diet during Pregnancy
Background: Low birth weight (<2500 grams) is representation of a nutritional deficiency during pregnancy, this condition is associated with increased incidence of degenerative diseases. Tempe as traditional Indonesian food has the potential to improve of DNA damage and to inhibit the progression of degenerative diseases. This study aimed to determine the effect of tempe flour supplementation during pregnancy on blood sugar and blood lipid profiles of offsringMethods: An experimental study with a sample of 20 female Wistar rats were pregnant, were divided into 4 groups. Treatment during pregnancy: the first group received a diet 50% of the normal diet (10 g / day), group II get a normal diet and 50% tempe flour 1 g / day, group III received normal diet and 50% tempe flour 2 g / day, group IV, 50% received a normal diet and 1 g egg chicken / day. Provision of drinking with distilled water ad libitum. During lactation are given a normal diet, after weaning are given a diet high in saturated fat for 4 weeks in each group.Results: The mean number of children in group 1: 9.33 with a mean BB: 4.93 g, group II: with a mean of 6.17 BB: 5.32, group III: 5.17 with a mean BB: 5.33, group IV : BB with a mean of 6.33: 5.01. There are significant differences in the levels of blood sugar, total cholesterol, triglycerides, LDL, and HDL in each treatment group (p= 0,00). Conclusion: Supplementation of tempe flour effect on blood sugar levels and lipid profiles rat pups that received a restricted diet during pregnancy (Sains Medika, 4(2):182-194)
The Study of Mouth Washing Duration with Boiled Uncaria gambir on the Dental Plague Formation
Background: Dental plague has been known to be the major agent causing dental carries and periodontal disease which are the most common dental and soft tissue diseases with the prevalence of 70-80%. The preventive measure including the use of chemical substance of catechin having anti-plague activity in the form of mouth wash has been shown to have effects on the dental plague formation. The objective of this study was to find out the duration of mouth washing with boiled gambir (Uncaria ambir) on the plague formation in Pondok Pesantren Al-Badriyah Mranggen, Demak.Design and Method: In this study using Pretest-Posttest Control Group Design, 44 samples of subjects was divided into 4 groups of 11 each. Group I, II, II were treated with the mouth wash of Uncaria gambir boiled 2, 3, 4 minutes respectively. Group IV serving as control was treated with mouth wash of water for 3 minutes. The data was analyzed with non parametric test using Kruskal-Wallis followed by Mann-Whitney test with p 0.05) and significant difference for the rest of the groups (p < 0.05). Conclusion: The mouth rinse duration using boiled Gambir has been shown to have an effect on the dental plague formation. 3 minute mouth washing shows the optimum effect (Sains Medika, 4(1):57-62)
