IDENTIFICATION OF CO-DOMINANT DNA MARKER TIGHTLY LINKED TO Tm-2 LOCUS IN TOMATO
Tm-2 adalah gen yang mengendalikan ketahanan terhadap tomato mosaic virus (ToMV) pada tomat (Lycopersicon esculentum) dan berasal dari Lycopersicon peruvianum. Pada penelitian ini digunakan dua galur tomat yang mempunyai latar belakang genetik yang sarna GCR26 dan GCR3 6, akan tetapi keduanya dapat dibedakan oleh adanya gen Tm-2 pada GCR36. Melalui analisis random amplified-polymorphic DNA (RAPD) terhadap dua galur tomat tersebut di atas diketahui adanya primer RAPD yang menghasilkan fragmen spesifik untuk GCR236. Selanjutnya fragmen tersebut sekuennya dianalisis, dan dari hasil analisis sekuen tersebut dibuat sepasang primer sequence characterized amplified region (SCAR) Yang merupakan marka DNA kodominan yang bisa membedakan baik genotipe homozigot gen Tm-2. Oleh karena fragmen dihasilkan terkait dengan Tm-2, maka primer SCAR tersebut merupakan sarana yang dapat digunakan untuk pemulian tanarnan tomat bagi pembentukan varietas tahan virus
PERBANYAKAN IN VITRO TANAMAN BAWANG PUTIH (Allium sativum L.) VARIETAS LUMBU PUTIH MELALUI INDUKSI TUNAS ADVENTIF
Two sets of experiments were conducted to determine the effect of growth hormones, argynine, and coconut water, on the adventitious shoot induction from garlic tissue, and also to find out the best medium for adventitious shoots proliferation. Both experiments could induce direct adventitious shoot and indirect adventitious shoot formation, from calli. Medium with 2 ppm Kinetin and 0.4ppm 2,4-Dproduce good quantity and quality of shoots. The number of shoot from this medium were 32.6 shoots per explant. Medium with 0.5 ppm Kinetin, 0.1 ppm 2.4-D, 25 ppm Argynine, and 10% coconut water produce the highest diameter and good quality of calli, while medium with 1 ppm 2iP and 25 ppm Argynine induced adventitious shoot from calli and produced the highest number of shoot per culture (33.9 shoots)
PENGARUH PENGATURAN POPULASI DAN UKURAN LEMPENGAN RUMPUT MANILA (Zoysia matrella (L.) Merr) TERHADAP PERTUMBUHAN DAN PERKEMBANGANNYA
Zoysia matrella (L.) Merr. is a widely used ornamental grass, particularly in home and landscape gardens, because of its soft appearance and high quality. One of main problem is its low growth rate. Therefore, specific cultural techniques are required to accelerate its growth while maintaining its attractive appearance. This experiment was aimed to study the effect of sod size and planting distance of Zoysia mairella. on grass growth and development. The experiment was conducted at the IPB experimental station at Darmaga IV, Bogor, for 8 months and was completed in August 1990. The trials used a split plot design with 2 factors and 3 replications. Treatment consisted of 2 planting distances i.e. (20 x 20) cm and (25 x 25) cm, and 4 sod sizes, i.e. (2 x 2) cm, (5 x 5) cm, (8 x 8) cm, and (10 x 10) cm. The variables observed were percentage of coverage, rate of coverage, quality of leaf color quality of appearance, and uniformity of growth. The results indicated that population density did not significantly affect variables observed except for uniformity of growth. Hence, the larger the sod size, the higher its rate of coverage and the more uniform its appearance, but the lower its quality of appearance. Transformasi Padi Indica Kultivar Batutegi dan Kasalath dengan Gen Regulator HD-Zip untuk Perakitan Varietas Toleran Kekeringan
water deficiency. Genetic engineering at the level of transcription factors (TF) is particulary a promising strategy in developing drought tolerant rice cultivar. HD-Zip genes are TF that function in plant adaptation to some environmental stresses including water deficit. The recombinant plasmid pC1301H Oshox6 which contained HD-Zip Oshox6 gene was placed under a drought inducible promoter called LEA promoter, gusA and hpt genes were driven with CaMV promoter. The aim of research was to obtain indica rice transgenic plants of Batutegi and Kasalath cultivars using pC1301H Oshox6 plasmid. Recombinant plasmid was transformed into immature rice embryos using Agrobacterium tumefaciens. Kasalath cultivar showed a better capacity to form embryogenic calli compared to Batutegi. Transformation efficiency of Batutegi is lower (1.5 - 0.3%) than Kasalath (2.2-28.3%). Regeneration efficiency is 25-83.3% and 7.7-100% for Batutegi and Kasalath, respectively. Number of putative transformant plantlets of Batutegi and Kasalath are 63 and 48 plantlets, respectively. Southern blot analysis (using hpt probe) on 12 independent lines of each Batutegi and Kasalath cultivars showed different gene copy number, ranging from one to four copies of gene. Efisiensi Beberapa Substrat dalam Pengujian Viabilitas Benih Berukuran Besar dan Kecil (Efficiency of Several Substrates for Seed Viability Testing of Large and Small Seeds)
The quality of paper substrate for seed viability testing is determined mainly by the variety and amount of the papers. A study was conducted to find alternative paper and the optimum amount of paper for testing the viability of large and small seed with the rolled paper method. Two experiments were carried out at the Seed Science and Technology Laboratory, Bogor Agricultural University during April-September 2004 and July-December 2007. First experiment: large and small seeds represented by 5 plants (angled loofah, pumpkin, garden pea, bitter gourd, yam bean) and 7 plants (sorghum, cucumber, mungbean, soybean, horenzo, radish and wheat), respectively, were tested on 4 different paper substrates (straw, stencil, CD, and HVS) following the rolled paper method. Second experiment: large seeds of maize and ground nut, and small seeds of rice and mungbean were tested on paper substrates selected from the first experiment, straw, stencil, and CD, with different number of sheet, i.e. 2, 3, 4, and 5 sheets. Each plant seed was considered as an experiment and a randomized block design was applied to all experiments. The result indicated that for the large seeds, stencil and CD papers had equal and high similarity with the reference straw paper substrate, 100% and 80% for germination percentage and dry weight of normal seedlings, respectively. For the small seeds, only stencil paper showed high similarity for germination percentage variable, 86%, with the reference paper, whereas the other paper showed lower similarity, 57% for both CD and HVS papers. The second experiment showed that 2 sheets of straw, stencil or CD papers were enough for testing viability of large and small seeds to obtain germination percentage, but ground nut needs 4 sheets of straw paper. For vigor index variable, efficiency of substrate paper on large and small seeds varied from 2 to 4 sheets. Studi Alternatif Substrat Kertas untuk Pengujian Viabilitas Benih dengan Metode Uji Diatas Kertas
Viability of small seed is usually tested with top of paper method using paper substrate, but filter and straw papers commonly used for substrate have become more expensive and scarce recently. The aim of this study was to find an alternative paper substrate for seed viability testing using top of paper method. The study consist of 12 experiments using 12 species i.e. pak choy, white mustard, chinese kale, cauliflower, cabbage, sesame, broccoli, green mustard, lettuce, carrot, red spinach and celery. The experiment was arranged in a Randomized Block Design with 10 replications. Sample of 50 seeds were tested on different paper substrate including straw paper, filter paper, stencil paper, CD paper and sterilized filter paper, to identify the best alternative substrate. Result of the study showed that stencil and CD papers as a substrate for seed viability testing performed 100% similarity compared to straw paper. When filter paper was used as standard substrate, there wasn\u27t any paper has 100% similarity, but straw paper showed the highest (97.22%) followed by stencil paper (88.87%). Based on less than 5% coefficient of variation criteria, stencil paper was the best with 61.11% uniformity of seed viability data followed by straw paper (58.33%). In higher tolerance level with less than 10% coefficient of variance criteria, stencil paper was still the highest with 100% uniformity, followed by straw paper (97.22%). Stencil paper as germination substrate performed similar seed viability to the control straw and filter papers also produced high uniformity data. Stencil paper could be used for substrate viability testing of small seed with top of paper method. Key words : Top of paper method, alternative, paper substrate, viability, coefficient of variance
Studi RegeDerasi daD Produksi Protoplas Mesofil Daun Beberapa KlOD TanamaD Kentang (Solanum tuberosum L.) .
The objectives of these experiments were to obtain medium composition to enlarge leaf size for protoplast production and to obtain medium composition for plant regeneration. The result showed that the best medium to produce the larger leaves was medium MS with double concentration of macronutrients without hormone supplemented with Morel vitamins, 3% (w/v) sucrose and. 7 g/l agar. This medium produced leaves with diameter of 1.44 cm comparing to control medium MS with 0.67 cm in diameter. Medium MS containing 0.1 mg/l 1AA, 0.5 mg/l Zeatin and 0.5 mg/l GA3 was able to regenerate vigorous shoots of 7 clones. Protoplast isolation of 5 clones using enzyme composition containing 0.5 % cellulase Onozuka RS, 0.05 %, pectolyase Y-23, 0.05 % MES, 9.1 % mannitol and pH 5.5, without CP W medium produced protoplast with variable yield from 10.50 x 1oJ protoplast/g leaves for Atlantic to 46.58x 1oJ protoplast/gforBF15. Transformasi Genetik Tanaman Kentang cv. Atlantik Dengan Mengintroduksikan Gen Hordothionin untuk Mendapatkan Ketahanan terhadap Penyakit Bakteri
Hordothionins are small anti-bacteria proteins present in barley endosperm, To reveal the potential of this proteins for engineering bacterial disease resistance into potato, a semi-synthetic hordothionin gene construct was introduced in potato cv. Atlantic via Agrobacterium tumefaciens strain LBA 4404, under the control of a cauliflower mosaic virus (CaMV) 358 promotor. The in vitro grown stem (internodus) was used in this research. After 6 weeks in regeneration medium and 2 weeks in rooting medium there were 22 regenerated plants that were screened in kanamycine containing medium. PCR analysis using spesific primer from CaMV 358 promotor showed the presence of amplified T-DNA in 4 transgenic lines from 22 putative transgenic plants were tested, The in vitro toxicity against Ralstonia solanacearum tested from transgenic lines showed variation in resistance level, There were only 2 of the transgenic lines were tolerant, while one of them was moderate tolerant even one of them was susceptible. Pertumbuhan 11 Aksesi Stroberi yang Dibudidayakan Secara Hidroponik
The objective of the experiment was to study the growth and fruit quality of several accessions of strawberry grown by hydroponics. The experiment was conducted at farmer farm in Pacet Cianjur, from March 1999 to June 2000. These accessions were collected from Cipanas Cianjur, Ciwidey and Pangalengan Bandung, all of those were unidentified its origin. Nutrition compound used was made by modify nutrition compound for tomato which was obtained from previous experiment. The result of this experiment showed that strawberry plant could produce runner with average one runner per month, whenever all fruit thinned. A strawberry plant produced one leave average per two week\\u27, J 1-18 fruit per year with 13-24 gram per fruit or 210-314 gram per plant per year. It has been found that five accessions tasteless, four accessions had strong sour taste, one accession (No.4) with sweet taste and one another accession (No. 10) with sour sweet taste. The last two accessions had a good taste with fruit weight around 17 gram in average per fruit, smoth texture and adequate firmness. These accessions are being propagated for further experiment. Pengaruh Pemberian CaCI2 Prapanen terhadap Perubahan Kualitas Tomat Segar Selama Penyimpanan
The objective of the research was to determine the effect of pre harvest CaCI2 application on quality changes of tomato fruits. Experiment consisted of 5 treatments: control, application of 0.1 M CaCI2 at 18 ± 2 days after anthesis (DAA), 0.1 M CaCI2 at 25 ± 2 DAA, 0.2 M CaCI2 at 18.t2 DAA and 0:2 M CaCI2 at 25 ± 2 DAA. Tomato fruits cv. Ratna harvested at breaker stage were used throughout the experiment. Variables to be observed were peel color, weight loss, fruit firmness. total soluble solids, titratable acidity, and respiration. Result of the experiment showed that application , of CaCl2 could not maintain quality and extend shelf life.
