Penapisan Genotipe Jarak Pagar (Jatropha curcas L.) untuk Toleransi terhadap Kekeringan
The selection of Jatropha curcasfor drought tolerance is one of the key points prior to growing the species extensively on marginal lands. The objective of this study was to determine drought tolerant genotypes based on morphological and physiological characters. The research consisted of two experiments. The first experiment was aimed at determining the moisture content of the media to generate drought stress. The experiment was arranged in a split-plot design with moisture content as the main plot and genotypes as sub-plot. Four level of moisture contents were tested, i.e. 22-23%, 27-28%, 32-33%, and 37-38%. The genotypes tested (9) were Dompu-1, Gunung Tambora, Bima (representing genotypes from dry areas); Aceh Besar, IP-2P, Komering (representing genotypes from wet areas); and IP-1M, Papua, Yogyakarta (representing genotypes from moderately dry areas). The result showed that 22-23% moisture content of the media was suitable for drought tolerance test in Jatropha curcas. The second experiment was screening of genotypes using the suitable media moisture content from first experiment. Twenty three genotypes were screened for drought tolerance under 22-23% media moisture content. The result show that Dompu-2, Indralaya and China were tolerant to drought; Sukabumi-1, Sukabumi-2, Pidi, Lahat, Kupang, Lampung-2, Lampung-3, Sumba, IP-2M, and IP-2A genotypes were moderately tolerant; Curup, Bogor-1, Bogor-2, Bogor-3, Pontianak, Pagar Alam, Palembang, Saweli, Jeneponto, and Medan genotypes were sensitive to drought. Plant height, leaf water content, leaf area and percentage of plant survival can be used as indicators of drought tolerance for seedling selection using a non-destructive method.Keywords: drought tolerance, genotypes, Jatropha curcas, screenin
Black pod disease (BPD) of cacao caused by Phytophthora palmivora(Butl.) is one of the major diseases on cacao plantation worldwide. Endophytic fungi (EF), fungi that live asymptomatically inside healthy plants, were examined to study their potentials as biocontrol agent of the disease. Six of EF selected from 37 species (from 2843 isolates), isolated from healthy pods of cacao from Marena in Central Sulawesi were tested for their abilities to control BPD. Pods on living trees in the field were sprayed with each EF propagules. The inoculated pods were harvested two weeks later and subsequently inoculated with P. palmivora. Scoring of disease development was performed and quantified as area under disease progress curve (AUDPC). Latent period, infection rate and effectiveness were also recorded. In vitro growth inhibition of pathogen and induced plant defense mechanisms due to EF were also investigated. Xylariaceaeand Calocybe gambosatreatment generated the highest effectiveness control level, i.e. 38.8% and 33.8% respectively, followed by Resinicium friabileand Aschersoniatreatment, i.e. 17.4% dan 12.7% respectively. Pestalotiopsisand Fusariumwere not effective to control BPD. There was a strong connection between disease severity of BPD with the latent period of pathogen. Growth inhibition of pathogen and induced resistance of plant were partially responsible for disease suppression by Xylariaceae, C. gambosa, R. friabileand Aschersonia. Penggunaan Pot Berbahan Dasar Organik untuk PembibitanGmelina arborea Roxb. di Persemaian
The objectve of this research was to obtain the best materials composition and adhesive of organic pot for forest trees seedling production. Organic pot strength was evaluated by testing the strength of rupture elasticity and elasticity stiffness of each composition. The bioassay testing used Completely Randomized Design with factorial pattern, consisting of two factors, (1) basic raw materials of the pot (used newspaper, litter and compost) and (2) the type of adhesive (control, tanin and starch). The results showed that the adhesive starch gave higher strength, whereas adhesive tannin gave higher stiffness as compared to control. Results of bioassay showed that the mixture (50:50 v/v) of basic materials of used newspaper and compost (KKK) which were glued with tannin, produced the best results for height and diameter increament with the value of 35.85 cm, 0.31 cm respectively and biomass value of 0.99 g after 12 weeks of planting. The highest level of damage in organic pot with basic raw materials KKK without adhesive was 47.6%, whereas lowest level of damage in organic pot with basic raw material used newspaper (KK) with tannin addhesive was 2.0% after 12 weeks in the nursery.Keywords: adhesive, Gmelina arborea,organic pot, organic matter, nursery
Perbanyakan In Vitro Sansevieria trifasciata ‘Lorentii’: Regenerasi Tunas, Pengakaran, dan Aklimatisasi Planlet
This research aimed to study effects of benzyladenine (BA) on in vitro shoot formation and effects of indolebutyric acid(IBA) and acclimatization media on ex vitro rooting and acclimatization of Sansevieria trifasciata ‘Lorentii’. Leaf segmentswere taken from young fully-expanded leaves, surface sterilized and cultured on Murashige and Skoog (MS) basal mediumcontaining 0.25 mg L-1 2,4-dichlorophenoxyacetic acid (2,4-D) for 2 weeks, transferred to medium without growth regulatorfor 2 weeks, and then subcultured on MS medium containing BA (0, 0.5, 1 and 2 and 5 mg L-1). The results showed thatadventitious shoot regeneration occured after callus formation. The best BA concentration was 2 mg L-1, producing 4.5 shootsper explants in 3 months and 11.1 shoots per explant in 4 months. Application of 2000 ppm IBA and the use of acclimatization medium consisting rice husk charcoal and compost (1:1) produced the highest number of primary roots, length of roots and root fresh weight. Selection method is one of the most important factors in determining the success of pepper breeding programs. Selection method will be effective if it is supported by a complete knowledge of genetic character inheritance. The aim of this research was to investigate the inheritance patterns to quantitative characters using large pepper (P1 (IPB C5)), curly pepper (P2 (IPB C105)), F1, F1R, BCP, BCP2 and F2. The result showed that dichotomous height, days to harvest and weight per fruit characters were not maternally inherited. The additive-dominant genetic model was the most suitable for dichotomous height character. The additive-dominant with influence of interaction additive-dominant and interaction dominant-dominant genetic model was suitable for days to harvest character. The additive-dominant with influence of interaction additive-additive and interaction dominant-dominant genetic model was suitable for the weight of fruit character. Broad-sense and narrow-sense heritabilities range medium for dichotomous height and weight per fruit characters. Heritability of the days to harvest character was high in a broad-sense but was low in the narrow-sense. Keywords: genetic model, heritability, inheritance

Efektivitas Amelioran pada Lahan Gambut Terdegradasi untuk Meningkatkan Pertumbuhan dan Serapan NPK Tanaman Jagung Manis (Zea mays L. var. saccharata)
This study was aimed to determine the effectiveness of several of ameliorant formulas to increase plant growthand NPK uptake on sweet corn (Zea mays L. var. saccharata) on degraded peatlands. The research was conducted in thegreenhouse of Balittra during May to July 2011. Sweet corn was planted in pot with soil taken from previously burnt peatin Kalampangan, Palangkaraya. The treatment consisted of two factors, i.e., combination of ameliorant (A1=80% chicken manure + 20% dolomite, A2=80% agricultural weeds + 20% dolomite, A3=80% mineral soil (Spodosol) + 20% dolomite,A4=20% chicken manure + 20% agricultural weeds + 20% Eleocharis dulcis + 20% mineral soil + 20% dolomite, andA5=19% chicken manure + 71.5% mineral soil + 9.5% dolomite) and application rates at 5, 10, 15, 20 and 25 ton ha-1, arranged in a randomized complete block design, with 3 replications. The results showed that ameliorants and application rates affected growth and NPK uptake of sweet corn. Application of 20 ton ameliorant (80% chicken manure + 20% dolomite) ha-1 produced the highest dry weight of root and shoot, and NPK uptake.Keywords: ameliorants, degraded peatland, nitrogen, phosphate, potassiu
Hubungan Karakter Daun dengan Hasil Padi Varietas Unggul
Penelitian ditujukan untuk mempelajari hubungan karakter daun dengan hasil padi varietas unggul, dan dilaksanakandi kebun percobaan Balai Besar Penelitian Tanaman Padi, Muara, Bogor pada bulan Desember 2010 sampai Juni 2011.Percobaan menggunakan rancangan kelompok lengkap teracak dengan empat ulangan dan menggunakan 12 padi varietasunggul sebagai perlakuan. Varietas yang digunakan adalah Rojolele dan Pandan Wangi (varietas unggul lokal/VUL); IR64 dan Ciherang (varietas unggul baru/VUB); Fatmawati, Cimelati, galur BP360 dan B11143 (padi tipe baru/PTB);serta Maro, Rokan, SL-8 SHS, dan PP1 (hibrida). Hasil penelitian menunjukkan Galur B11143, Maro, Cimelati, dan Rokanmemberikan hasil gabah tertinggi. Tingginya hasil berhubungan dengan karakter sudut tiga daun bagian atas, luas daunbendera, kandungan klorofil, dan kandungan gula daun bendera. Karakter sudut tiga daun bagian atas dan luas daunbendera berkorelasi negatif dengan hasil. Kandungan klorofil dan gula daun bendera tahap berbunga dan pengisian bijiberkorelasi positif dengan hasil.Kata kunci: hasil, karakter daun, padi varietas unggu
Pengaruh Kandungan Cu dalam Air Irigasi terhadap Pertumbuhan dan Produksi Padi (Oryza sativa L.)
The research was conducted to investigate the effect of copper (Cu) levels in irrigation water on the growth and production of rice variety IR64. The experiment was arranged in randomized complete block design with six replications. The Cu treatments were: 0, 300, 600 and 900 mg Cu L-1of irrigation water. The results showed that Cu content significantly decreased rice growth and production. Copper level of 600 mg L-1 decreased plant height, tiller number and leaf number. Copper level of 300 mg L-1 decreased panicle length, number of grains per panicle, empty grain weight, 100 grains weight, milled grain weight, and shoot dry weight. This experiment implies that remediation treatments should be considered when Cu level in irrigation water was more than 300 mg Cu L-1. Copper level of 300 mg L-1 could be used as a critical level for rice screening for Cu tolerant varieties in the plant breeding program. Variasi Genetik Mutan Anggrek Spathoglottis plicataBlume. Berdasarkan Marker ISSR
Mutants of Spathoglottis plicataBlume were obtained from plants treated with Gamma Irradiation at 30-100 Gray. The mutants showed variations in the flower morphology. The objective of this experiment was to identify genetic variations of orchids S. plicataand its mutants using inter-simple sequent repeat (ISSR) markers. The amplified product of 10 ISSR primers produced 360 bands and 71 ISSR of the loci (90.14%) were polymorphic. The coefficient of similarity and principal component analysis produced five major groups with similarity coefficient of 0.68. The goodness of fit correlation matrix value reached 0.91. Pengaruh Batang Bawah dan Jenis Tunas pada Mikrografting Manggis (Garcinia mangostana) secara In Vitro
The aim of this study was to investigate the effect of rootstock and shoot types on in vitro mangosteen micrografting.The experiment was arranged in completely randomized design (CRD) with two factors. The first factor was the rootstocktype, i.e. rooted planlet from the germination of quartered seed, and rooted planlet from the germination of undivided seeds.The second factor was the developmental phase of scion, i.e. dormant buds, and flush (had new leaf more than 2-4 mm). Theresults showed that rootstock derived from the germination of undivided seed had a higher success rate than other treatmentson all variables, except for number of new leaves. The use of flush as scion was better than dormant buds; flush resulted in ahigher percentage of successful micrograft and longer shoots. In vitro micrografting had a better growth rate than grafting at the same age. The results of anatomical observation conducted at four months after micrografting demonstrated that there was a good graft union, indicated by excellent fusion between rootstock and scion xylem tissues.Keywords: flush, in vitro, micrografting, rootstock, scio
