    ­­­­­­Kemiripan dan Evaluasi Produksi Aksesi Kenikir (Cosmos caudatus Kunth.) dari Jawa Barat

    ABSTRACTKenikir (Cosmos caudatus Kunth.) is an Indonesian indigenous vegetable which is potential to be developed. Exploration conducted in Bogor, Sukabumi, Bandung, Bandung Barat, Subang, Garut, Majalengka, Kuningan, and Tasikmalaya had succesfully collected 20 accessions of Cosmos sp. The objectives of this study were  to observe similarity and estimate the yield of Cosmos accessions from West Java. Clusters analysis grouped 20 Cosmos accessions into three clusters. Cluster I consisted of Pendeuy, Saribakti, Karang agung, Sindangbarang, Langensari, Perbawati, Sudajaya girang, Karang tengah, Argalingga, Warnasari, Sukaresmi, Ciwidey, Jalan cagak, Lebaksiuh, Tugu selatan, Ciwarak, Linggarjati and Babakan accessions. Cluster II and III consisted only one accession each which were Ciaruteun and Dramaga, respectively. Seven accessions from different sub cluster were evaluated to estimate the yield. A significant variability was found among the accessions. Results showed that those accessions were significantly different on plant height, stem girth, numbers of primary branches, number of leaf, leaves width, leaves length, days to flowering and yield. The results showed that Dramaga and Ciaruteun acessions were recomended to be developed because of its high yield. Keywords: characterization, cluster analysis, exploratio

    ABSTRACTMucuna bracteata DC. is a legume cover crop to prevent erosion and to suppress weed growth. M. bracteata can fix N2 from the air with the help of Rhizobium. M. bracteata get benefit from the symbiosis in the form of increased nodule weight, shoot dry weight, and leaf nitrogen content when the Rhizobium population in the soil is optimal. Application of inoculant is one method to increase the Rhizobium population in the soil. This study aimed to analyze the effects of inoculation times and inoculant rates on M. bracteata growth. The research was conducted in Boyolali, Central Jawa, from February to August 2014. The inoculant contained Bradyrhizobium japonicum and Aeromonas punctata. This experiment was arranged in a randomized block design with two factors and three replications. The first factor was two inoculation times, i.e at the nursery when the seedlings were 2 weeks old and in the field when the seedlings reached 5 weeks old. The second factor was 5 inoculant rates (0, 2, 4, 6, and 8 g per plant). The results showed that inoculation on 5-week-old seedlings was the best result compared to the 2-week-old seedlings.     Toleransi Galur Harapan Padi Sawah (Oryza sativa L.) pada Persaingan dengan Gulma Echinochloa crus-galli

    ABSTRACTBarnyard grass (Echinochloa crus-galli (L.) P. Beauv.) is a major weed competitor to rice production in Indonesia. In order to develop integrated weed management program, a research to select competitive rice lines to E. crus-galli was conducted in a green house of Indonesian Centre of Agricultural Biotechnology and Genetic Resource Research and Development Bogor. The research design was split plot with four replications, E. crus-galli was designed as the main plot (rice without E. crus-galli compared  rice with four E. crus-galli per pot), and the sub-plots were 25 genotypes (23 lines, 1 tolerant variety and 1 sensitive variety). Level of tolerance was determined by the reduction percentage of grains weight, the number of productive tillers and dry matter weight. The results showed that rice competition with E. crus-galli reduced plant height, productive tiller numbers, filled spikelet numbers per panicle, dry matter weight and dry grain weight. Three lines, i.e., IR10L-155, IR10L-133 and BIO-R84-1 were classified as tolerant, 19 lines were moderate and 3 lines were sensitive to E. crus-galli competition.Keywords: Barnyard grass, competition, sensitive line, yield reductio

    ABSTRACT Storage condition and pathogen infection of seed can cause seed deteriorates faster. The rate of deterioration during storage could be slowed by seed invigoration, and pathogen infection could be elimited by application of natural pesticide. The objective of this research was to determine the effect of seed invigoration on seed physiological quality and health of Intani-2 hybrid rice seed during storage. All research activities were done at Laboratory of Quality Control and Plant Protection, PT. BISI International, Kediri, East Java. Split plot design was used in this experiment with 4 replications. Three seed lots of Intani-2 rice seed were used as main plot and 5 invigoration treatments were used as sub plot. Seed lot 1, 2 and 3 were harvested on 30 June 2012, 29 September 2012, and 2 November 2012, respectively. Invigoration treatments consisted of untreated, priming with ascorbic acid 40 ppm, osmoconditioning with KNO3 2%, osmoconditioning with PEG -0.2 MPa, and hydropriming. All invigoration treatments were added with clove oil 0.3%. Osmoconditioning with KNO3 2% + clove oil 0.3% was effective to maintain vigour index of seed lots 2 and 3 for up to 3 months storage. All seed invigoration treatments increased speed of germination before storage. Priming with ascorbic acid 40 ppm + clove oil 0.3% and osmoconditioning with PEG -0.2 MPa + clove oil 0.3% were effective to reduce the growth of Xanthomonas sp. on rice seeds monitored at 0, 2, and 3 months after storage.Keywords: ascorbic acid, clove oil, KNO3, osmoconditioning, PE

    ABSTRACTOptimum fertilization is not only proposed for obtaining desireable quality and quantity of the plant, but also to avoid excess fertilizer causing toxicity and environmental damage. The objective of this research  was to study and to determine an optimum rate of organic and NPK compound fertilizers for immature oil palm. The experiment was carried out at IPB-Cargill  Oil palm Teaching Farm, Jonggol, from March 2013 to March 2014. The treatments was laid out in a factorial randomized complete block design with three replications. The first factor was organic fertilizer,  i.e. 0, 15, 30 and 45 kg organic fertilizer plant-1, and the second was NPK compound fertilizer, i.e. 0, 0.65, 1.3, 1.95 and  2.6 kg NPK compound fertilizer plant-1. The result  showed  that vegetative growth increases  with fertilizer application. The height and stem girth of  plant were significantly affected by interaction between organic and NPK compound fertilizers, however there was no significant effect on frond production, frond length and leaf area. N, P leaf content of  plant were affected by interaction between organic and NPK compound fertilizers, however there was no effect on K leaf content, chlorophyll and number of stomata. Based on the height and stem girth regression equation, the optimum recommendation rate of organic and NPK compound fertilizers for immature first year oil palm are 40.7 kg organic fertilizer plant-1 and 1.9 kg NPK compound plant-1 Keywords: inorganic fertilizers, nutrient level, stem girth, vegetative growt

    Induksi Mutasi Kalus Embriogenik Gandum (Triticum aestivum L.) melalui Iradiasi Sinar Gamma untuk Toleransi Suhu Tinggi

    Mutation techniques through gamma ray irradiation is useful to support breeding programs for genetic improvement of wheat. Genetic improvement on tolerance to high temperatures is necessary for development of wheat in Indonesia. The purpose of this study was to determine the level of radiosensitivity to be used as the basis for the induction of mutations by gamma ray irradiation on embryogenic callus to obtain putative mutants with high temperature tolerance. Explants used were embryogenic callus cultured on MS medium containing 30 g L-1 sukrosa, 2 g L-1 gelrite, 2 mg L-1 2.4D and 1 mg L-1 picloram. Culture incubated for 6 weeks with temperature 20±4 oC in the room culture. Five irradiation doses (10, 20, 30, 40 and 50 Gy) were used in radiosensitivity testing. A factorial, completely randomized design was applied to the experiment. The first factor was selection temperature with three levels (25, 30 and 35 oC), and the second factor was doses of gamma ray iradiation with three levels (10, 20, and 30 Gy). The result showed that radiosensitivity levels varied among varieties, LD20: 7.79 to 18.96 Gy and LD50: 24.29-33.63 Gy. Selayar variety which has highest sensitivity value compared with Dewata and Nias. Increasing doses of iradiation and temperature decrease survival of embryogenic callus, number of embryos, and percentage of germinated plantlets. Based on in vitro selection using high temperature (25, 30, and 35 oC), the obtained 19 putative mutants were derived from embryos that appear on the surface of embryogenic callus survival after irradiation and high temperature selection.Keywords: in vitro selection, putative mutant, radiosensitivity, somatic embry

    Seleksi Segregan Gandum (Triticum aestivum L.) pada Dataran Tinggi

    Pedigree selection is a method of selection used in segregating populations which takes the expertise of breeders in achieving the desired goal. This method is implemented on the early generations with a high degree of segregation and is generally performed on highly heritable traits. The purpose of this research was to study the performance of F3 generation segregants and to estimate the genetic parameters to be used for identification and selection of wheat segregants under optimum environmental condition. The research was carried out at the experiment field of  Research Institute for Ornamental Crops, in Cipanas from May to September 2012. Genetic material used were 58 F3 family and six check varieties, namely  Dewata, Selayar, Oasis, Rabe, Basribey, and HP 1744. The experiment used an augmented design. Results of analysis of variance showed that the family effect was significant for days to flowering, days to harvest, percentage of empty florets, number of grains per panicle, grain weight per panicle, number of seeds per plant and seed weight per plant. High heritability estimates were  found in days to flowering, days to harvest, seed filling period, percentage of empty florets, number of grains per panicle, grain weight per panicle, number of seeds per plant, and seed weight per plant. These characters are suitable for yield improvement of tropical highland wheat due to its high heritability values. The mean value of F3 generation changes subsequent to selection either based on grain weight per plant, or based on  number of productive tillers,  number of grain per panicle, and grain weight per plant. Keywords: selection criteria, segregation, tropical, yiel

    Pertumbuhan Tithonia diversifolia dengan Dosis Pupuk Kandang dan Jarak Tanam yang Berbeda

    Mexican sunflower (Tithonia diversifolia) is a invansive weed but it can be used as continuous and on-farm source of nutrient for organic farming system. The objective of this study was to investigate growth and biomass production of T. diversifolia with different rates of chicken manure and plant spacings. The experiment was conducted at Cikarawang, Bogor, on Inceptisol soil type, from October 2011 until January 2012. The experiment was arranged in a randomized block design with 2 factors and 3 replications. The rates of chicken manure were  0, 2.5, 5 and 10 ton ha-1, while the plant spacings were 50 cm x 50 cm and 75 cm x 75 cm. The interaction effect between manure rates and plant spacing was significant on nett assimilation rate at 4-8 weeks after planting, but it was not significant on biomass production. The highest dry weight of biomass (1,129.1 kg ha-1) was obtained from the addition of 10 ton chicken manure ha-1 with the potential contribution as much as 55.47 kg N, 4.54 kg P, 88.54 kg K per hectare. The highest dry weight of biomass produced with planting distance 50 cm x 50 cm (897 kg ha-1). Referring to the results, it can be recommended that the cultivation of T. diversifolia at the first planting needs 10 ton chicken manure ha-1 and the planting distance of 50 cm x 50 cm.Keywords: biomass production, Mexican sun flower, nett assimilation ratio, nutrient contributio, wee

    Ketahanan 25 Genotipe Tomat (Solanum lycopersicum Mill.) terhadap Pecah Buah dan Korelasinya dengan Karakter-karakter Lain

    ABSTRACTFruit cracking is one of the major constraints in tomato production in lowland areas. This reduces fruit quality and quantity both as freshmarket and processed fruits. The objective of this study was to determine the resistance of 25 tomato genotypes to fruit cracking, correlation to others characters and their genetic similarity. The experiment was conducted from April until August 2012 at Leuwikopo Field Station, Bogor Agricultural University. A randomized complete block design was used with three replications. Fruit crack index was used to evaluate tomato fruit crack resistance. Correlation and impacts of variables were analyzed by path analysis. The genetic similarity was analyzed by clustering analysis. Result of resistance evaluation showed that fruit crack index of 25 tomato genotypes ranged from 0.00 to 20.58. Fruit crack index was directly related to percentage of  fruit crack weight per plant, percentage of number of fruit crack per plant and total soluble solids.     Hubungan Metilasi DNA dengan Ekspresi Gen MADS-box pada Buah Mantel Tanaman Kelapa Sawit (Elaeis guineensis Jacq.)

    ABSTRACT The presence of mantled fruit on large-scale clonal production of oil palm had trully decreased the oil productivity. Mantled phenotype is likely to be that of an epigenetic change involving DNA methylation and the MADS-box transcription factor gene which encoded floral organ homeotic transformation. The objectives of this research were to quantify the degree of methylation which determined fruit abnormality through Ultra Performance Liquid Chromatography (UPLC) and to study its correlation with the MADS-box EgAGL6, EgAG2, and EgAGA genes expressed on mantled fruit derived from oil palm clonal plants which have been quantified using Quantitative Real-Time PCR (qPCR). This research was arranged in two replications for each gene and cDNA. The expression of the target genes were compared to EF1-α1 as the reference gene. Through the Least Significant Difference (LSD) Test at 95% confidence level of qPCR result, EgAGL6 expression was significantly lower in mantled fruit which decreased from 1.88 fold in Abn m to 0.46 fold in Abn. EgAG2 expression was increased non-significantly from 0.91 fold in Abn m to 1.13 fold in Abn, while EgAGA expression was higher in mantled which increased significantly from 1.48 fold in Abn m to 1.71 fold  in Abn. Nuclease S1 digestion and UPLC revealed the genome-wide increase in DNA methylation on mantled fruit (18.33-19.55%) compared to its normal counterparts (5.67%). This increased in global DNA methylation was showed by the significant decreased in EgAGL6 transcript level of mantled fruit.
