    Mekanisme Adaptasi Genotipe Baru Kedelai dalam Mendapatkan Hara Fosfor dari Tanah Mineral Masam

    Phosphate defi ciency is one of the main constraints to increase soybean yield in acid mineral soil. An experiment was conducted with an objective to discover scientifi c base of fertilizer selection based on soybean adaptation mechanism to mitigate P defi ciency in acid mineral soils in Bengkulu Province. A factorial experiment was arranged in a randomized complete block design with four replications. The fi rst factor was soybean genotypes, i.e. Slamet and three new soybean genotypes (NSGs) i.e. 19BE, 25EC and 13ED, and second factor was P fertilization dosages (0 and 400 mg P2O5 kg-1 soil). Soybean was grown in a greenhouse using double pot system. Research results showed that the NSGs had different mechanism in P uptake from soil with high P absorbing capacity compared to Slamet genotype. NSGs translocated more carbon to root, and decreasing of shoot-root ratio, about 66-70%. NSGs 25EC produce more oxalic and malic acid, and 13ED produce more citric acid to obtain same amount of P absorbed by Slamet variety. Under low P, NSGs developed symbioses with mycorrhizal arbuscular fungus, however, only 19BE which increase microbe population in rhizosfer. Phosphosrus fertilization supressed arbuscular mycorrhizal fungus development (15-100%), alkaline phosphatase activity (20-88%), and microbial C biomass (40-71%) in rhizosphere of new soybean genotype

    ABSTRACTHybrid rice varieties  is an alternative technology to improve  productivity of low land rice. The results of previous studies have identified and found the male sterile lines Wild Abortive type and Kalinga are resistant to bacterial leaf blight pathotype III, IV and VIII. The objectives of the research were to obtain information on agronomic characters, yield evaluation, genetic parameters, and repeatability information. The experiment was conducted in two locations namely  Muara and Indramayu experimental stations. The design used was a Randomized Complete Block Design with three replication at each site.  Each replicate consisted of 17 hybrid rice and three check varieties. The results from locations showed that genotype BI485A/BP1 (IR53942) has the highest yield of 5.8 ton ha-1. The coefficient of genetic and phenotypic diversity of six agronomic characters was low. The repeatability for seven character observed were low, except for days to flowering. The  scoring value  of bacterial leaf blight disease in Indramayu showed that nine genotypes exhibit resistance.Keywords: cytoplasmic male sterile lines, disease resistanc

    Characterization of Full Length Sequence of JJ3 Isolated from Shade Tolerant Soybean

    The aim of this study was to characterize full length sequence of JJ3 isolated from shade tolerant soybean. The characterization of full length sequence involved analyses of structure of full length sequence, open reading frame (ORF) or coding sequence (CDS), and homology. The analyses of JJ3 full length sequence structure were done using GeneScanW program, coding sequence (CDS) or open reading frame (ORF) using ORF Finder program, and the homology of JJ3 full length sequence were done using BLAST and clustalW programs. Results showed that the full length sequence of JJ3 consisted of single exon and polyA. Single exon was a coding sequence (CDS) 633 bp produced open reading frame (ORF) with 210 amino acid deduction comprised of 136 conserved amino acids and 77 varied amino acids. The conserved domain of JJ3 produced functional protein, PsaD photosystem I (PSI) subunit, related to photosynthetic transport electron in PSI reaction center. The full length of JJ3 homolog to psaD, a gene encoding protein subunit photosystem I (PSI) related to mechanism of plant adaptation to shade stress, in wood tobacco (Nicotiana sylvestris), rice (Oryza sativa), tomato (Lycopersicon esculentum), barley (Hordeum vulgare), spinach (Spinacia oleracea), Arabidopsis (Arabidopsis thaliana), and potato (Solanum tuberosum).Key words: full length sequence, gene, homology, shade tolerant, soybea

    Iron (Fe) toxicity is a major constraint in rice production that decreases yield due to high level of soluble Fe. The aims of this experiement were to study the effect of particular Fe concentrations in nutrient solution on rice growth, and to determine Fe concentration in nutrient solution that caused light, moderate, and severe Fe toxiciy symptom. The experiment was conducted in a greenhouse, Bogor Agricultural University from May to July 2010. A randomized block design with two factors and three replications were used in this study. The first factor was Fe concentration in the medium solution (2, 50, 100, 200, 400, 600 ppm Fe), and second factor was rice genotypes (IR64, Margasari). The results showed the higher Fe concentration in the solution resulted in higher scores of iron toxicity symptoms, higher Fe levels in the plant, and caused stunted growth of rice plants. Levels of Fe in a solution of ≥ 200 ppm Fe inhibited plant growth. Iron toxicity symptom in Margasari was lower than the IR64 varieties, especially at concentrations of 200 and 400 ppm. According to the regression equation Y = 0.022X+ 1849, Fe concentration in the solution which caused light Fe toxicity symptom (score ≤ 3) was ≤ 52 ppm Fe, moderate (score = 5) was 143 ppm Fe, heavy (score = 7) was 234 ppm Fe, and severe (score ≥ 9) was ≥ 325 ppm Fe.     Keragaman Karakter Komponen Hasil dan Hasil pada Genotipe Kedelai Hitam

    The objective of this research was to determine the trait variance in black soybean lines in terms of yield and yield components. The materials used were F8 lines generated from hybridization of two local varieties, i.e. black seeded Ceneng and greenish yellow seeded Godek, and then selected under single seed descent method, except  SC-39-1, SC-62-2 and GC-74-7 that were generated through bulk selection method. The results showed that the traits between plant genotypes were significantly different, except number of productive node and number of unfilled pod. Total variance was contributed more by number of filled pod, seed weight per plant and 100 seed weight.  Seed weight per plant and 100 seed weight had high heritability with genetic variance and broad-sense heritability which were significantly different from zero.     ABSTRACTThe breeding of rainfed rice tolerant to drought can be accomplished using anther culture. The objectives of this research were to determine regeneration abilities of six F1 anther culture and its acclimatization ability. The experiment was arranged in completely randomized design with 14 replications. The treatments consisted of six F1 derived from crossing:  INPARI 18 x IR83140-B-11-B (G1), INPARI 18 x B12825E-TB-1-25 (G2), INPARI 18 x IR87705-14-11-B-SKI-12 (G3), INPARI 22 x IR83140-B-11-B (G4), Bio-R81 x O18b-1 (G5), Bio-R82-2 x O18b-1 (G6). Media for callus induction was based on N6 medium + 2.0 mg L-1 NAA + 0.5 mg L-1 kinetin + 1.0 mM putresin + 60 g L-1 sucrosa, media for regeneration was based on MS + 0.5 mg L-1 NAA + 2.0 mg L-1 kinetin + 1.0 mM  putresin, and media for rooting was based on  MS + 0.5 mg L-1 IBA + 30 g L-1 sucrosa. The result indicated that all six F1 had different ability in anther culture. Bio-R82-2 x O18-b1 (G6) and  Bio-R81 x O18-b1 (G5) F1 genotype had good response both of callus induction and plant regeneration. These two F1 genotypes also gave the highest ratio of green planlet production to number of anther inoculated (GP:AI) were 5.50% and 4.65%,  respectively.     ABSTRACTFusaric acid produced by Fusarium spp. played a major role in potato dry rot development. Using fusaric acid as a selection agent may be useful to identify resistant clones. The aim of this experiment was to evaluate the morphological responses of 10 potato clones (Granola, Atlantic, Cipanas, DTO 28, DTO 33, Russet Burbank, IPB 1, CIP 801040, CIP 801045, dan CIP 801050) and their resistance level to fusaric acid and Fusarium solani. The research was conducted in Plant Breeding Laboratory and Tissue Culture Laboratory 3, Departement of Agronomy and Horticulture from April 2014-February 2015. The in vitro experiment was arranged in a randomized block design with 4 fusaric acid concentrations and 4 replications, while the F. solani infection experiment was arranged in a completely randomized design with 5 replications. The result showed that fusaric acid inhibits growth, reduce microtubers production, and caused planlets death. Correlation analysis between in vitro resistance to fusaric acid and F. solani infection on tuber showed positive and notable result.     Uji Ketahanan Anggrek Hibrida Phalaenopsis terhadap Penyakit Busuk Lunak yang Disebabkan oleh Dickeya dadantii

    ABSTRACTOne of the most popular orchids and cultivated by Indonesia and other countries is Phalaenopsis. The main disease of Phalaenopsis orchids in Indonesia is soft rot caused by bacteria Dickeya dadantii. The purpose of this study was to know the resistanctcy of Phalaenopsis hybrid to soft rot disease. The experiment was conducted at the Bacterial Plant Laboratory, Department of Plant Protection, the Greenhouse of Leuwikopo, Bogor Agricultural University, and the plastic house of Alam Sinar Sari Dramaga, Bogor from June 2014 to February 2015. The experimental design was randomized block design with three replications. Five genotypes of hybrid Phalaenopsis were tested, namely Phal. KHM 205, Phal. KHM 1126, Phal. KHM 1318, Phal. AMP 17, and Phal. KHM 2249. Phal. amabilis, Phal. esmeralda, Phal. amboinensis, and Phal. cornu-cervi were used as controls. The resistance testing was performed by inoculating bacteria D. dadantii on leaves of the orchids. The results showed all Phalaenopsis hybrid showed disease symptoms after inoculation. Phal. KHM 2249 had the lowest number of fallen leaves and the highest number of survive plants compared to the other hybrid Phalaenopsis.     Morfologi dan Pertumbuhan Planlet Hasil Induksi Poliploidi melalui Perlakuan Kolkisin pada Kuncup Bunga Anggrek Bulan (Phalaenopsis amabilis (L.) Blume)

    Induction of polyploid gametes is one of useful plant polyploidization methods. Some of its benefits are to obtain triploid and tetraploid progenies at the same time by cross and self pollination. Previous research showed that some morphological characters which could be the indications of polyploidy plantlets before the analysis of chromosome number. Colchicine treatment on flower bud of diploid Phalaenopsis amabilis was conducted to determine the effect of colchicine on flower bud development, plantlets morphology and growth, and potential of polyploidy induction based on plantlets morphology. Colchicine concentrations in the experiment were 0, 50, 500, 1,000, and 2,000 mg L-1, with three days duration of treatments with aluminium foil wraps on flower buds. The results showed that high colchicine concentrations (2,000 mg L-1) inhibited flowers blooming of treated flower buds. Based on morphological characters, plantlets were classified into normal and putative polyploid plantlets. Putative polyploid plantlets from colchicine with the concentration of 50, 500, and 1,000 mg L-1 were 71.2, 86.4, and 100.0% respectively.Keywords: colchicine concentration, morphological characters, normal plantlets, putative polyploidy, reproductive orga

    Interaksi Genotipe x Lingkungan dan Stabilitas Hasil Biji Kedelai Toleran Naungan

    ABSTRACTTwelve soybean shade tolerant promising lines and two check varieties, Pangrango and Argomulyo, were evaluated in eight locations covered varying degree of shades. The objective of this study was to assess the effect of genotype x environment interaction on seed yield and yield components, as well as on adaptation and yield stability of the lines  under those environments. The trial, in each location, was arranged in randomized block design repeated four times. The traits evaluated were days of flowering, maturity days, plant height, pod number, 100 seed weight, and seed yield. Light intensity was measured during generative phase, started at plants’ 30 days old,  two week interval. The results showed that genotype x environment interaction significantly affected those evaluated traits. Stability analysis revealed that four lines, i.e., IBK5-173-5-372, IBM22-861-2-22, IBM22-862-4-1, and IBM22-867-4-7 poorly adapted to the environments as indicated by coefficient regressions approximating 1.0 and low yield average. Eight lines, i.e., IBK5-143-3-7, IBK5-147-2-11, IBK5-172-4-36, IBK5-173-5-371, IIj9-299-1-4, IBM22-873-1-13, IBIj11-431-2-20, and AI26-1114-8-28, and the two check varieties, Pangrango and Argomulyo, were unstable. Of the 13 unstable genotypes, two lines, AI26-1114-8-28  and IBM22-873-1-13, gave higher average yield under shade condition (35%-70% shading level), 1.68 t/ha and 1.36 ton ha-1, respectively, than the two check varieties.Keywords: adaptation, Glycine ma
