In Vitro Shoot Formation on Sugarcane (Saccharum officinarum L.) Callus as Affected by Benzyladenine Concentrations
In vitro regeneration of sugarcane (Saccharum officinarum L.) clones can support breeding program of sugarcane. Thisresearch was conducted to study the effect of benzyladenine on in vitro shoot formation from sugarcane callus. Leafrolls werecultured for 8 weeks on callus induction medium containing MS salts, 30 g L-1 sucrose, 150 ml L-1 coconut water, 100 mg L-1myo-inositol, 0.1 mg L-1 thiamine-HCl, 0.5 mg L-1 pyridoxine-HCl, 0.5 mg L-1 nicotinic acid, 2 mg L-1 glycine, and 3 mg L-12,4-D. Callus was then subjected to different concentrations of benzyladenine (BA) (0, 0.5, 1, 2, and 2.5 mg L-1) containedin MS media. The experiment showed that after 8 weeks in culture 2 and 2.5 mg L-1 BA led to the highest percentage of shootformation (100%). The experiment also showed that addition of BA caused an increase in percentage of shoot formation,number of shoot per callus clumps, and average shoot length. In the range of 0-2.5 mg L-1, the higher the concentrationsof BA the more shoots and the longer shoots were produced. Highest number of shoots was recorded at BA 2.5 mg L-1(36.4 shoots per callus clump) and highest average shoot length was obtained at 2 and 2.5 mg L-1 BA, i.e. 2.25 and 2.3 cmrespectively. The shoot formation system was then applied to 12 sugarcane genotypes, resulting in statistically differentresponse and producing substantial number of shoots, ranging from 29 to 41.33 shoots per clump.Keywords: In vitro, sugarcane, leafrolls, benzyladenine, shoot formatio
Keragaman Waktu Tanam Tanaman Padi di Pulau Kalimantan
Rice planting time varied among farming sites. This research was aimed to study the variation in planting time especiallyin rainy season in Kalimantan. ‘Planting time’ was deterimined using assumption as the time when 8% of paddy fields ina sub district had been planted. Analysis was done by using mean ten-days of planting area of each sub-district during theperiod of 2000 to 2009 that was obtained from Statistics Indonesia. The result showed that the farmers in Kalimantan startedplanting rice during the first and second ten-days of September each year. Peak time of rice planting varied among provinces,i.e., on October II/III (West Kalimantan), January I/II and June II/III (East Kalimantan), and March III/April I (South andCentral Kalimantan). Evaluation of Fruit Characters, Xanthones Content, and Antioxidant Properties of Various Qualities of Mangosteens (Garcinia mangostana L.)
Xanthone in mangosteen fruit hull has antioxidant activities. The objective of this experiment was to determine xanthones content, physical and chemical characters, and antioxidant potentials of several groups of mangosteens qualities. The experiment was carried out from January 2008 to August 2008. The research used a randomized complete block design with four replications. The treatments were four physical fruit conditions: large size mangosteens (≥ 100 g), small size mangosteens (min. 50 g), yellow sap dotted fruits, and scabbed fruit. The parameters being observed were physical and chemical characteristics, xanthone content, and radical scavenging activity of mangosteen fruit hull extract. The results demonstrated that the physical condition of fruit significantly affected physical fruit hull characters and chemical fruit characters. Characters of fruit hull, thickness, fresh weight, and dry weight, differed among fruit qualities. The physical condition of fruit hull also significantly influenced total soluble solid and vitamin C content, but did not affect total titrable acid and degree of acidity of the fruits. Fruit hulls of all groups of mangosteen qualities had similar antioxidant properties with the IC50 value of 5.57-6.11 ppm. Keywords: benzophenone, scab, vitamin C, yellow sa
Senyawa Fenol dari Tajuk dan Umbi Teki (Cyperus rotundus L.) pada Berbagai Umur Pertumbuhan serta Pengaruhnya terhadap Perkecambahan Gulma Berdaun Lebar
Purple nutsedge (Cyperus rotundus L.) is noxious weed that contains allelochemicals which inhibit other plants germination, thereby it can be potentially used as bioherbicide. Laboratorium experiments were conducted to identify phenol compound in purple nutsedge’s shoot and tuber from three plant ages, and to study its effect on germination of Asystasia gangetica and Borreria alata. First experiment was single factor using descriptive method and the second experiment was arranged in completely randomized design with single factor. The first experiment treatments were shoot and tuber extracts of purple nutsedge from 1, 2, and 3 months after planting. The second experiment treatments were control, shoot, tuber, and all parts extracts of purple nutsedge from 1, 2, and 3 months after planting, with 3 replications. Data from the second experiment was analyzed using F test and followed by Tukey test 5%. Result showed that shoot extracts from 2 months after planting had the highest type of phenol compound namely 2-methoxy-4-vinylphenol; phenol,2,6-dimethoxy; 2-furanmethanol; and α-tocopherol. Tuber extract from 3 months after planting suppressed germination rate of A. gangetica about 54.72%. All parts extracts from 2 months after planting suppressed germination rate of B. alata about 60.98%. Purple nutsedge’s extracts inhibited growth of plumule and radicle of B. alata at 2 days after sowing. Functional Classification of Skinning Injury Responsive Genes in Storage Roots of Sweetpotato
Skinning injury in sweetpotato due to loss of skin or periderm which occurred during harvest is inevitable and account for financial loss due to dehydration, pests, and pathogens. Hence, studies on gene expression changed due to skinning injury can provide important information about this protective tissue and for improving the life of storage roots. New candidate genes involved in skinning injury were isolated with an Annealing Control Primer (ACP). Using 20 ACP primers, a total of 103 differentially expressed genes (DEGs) were retrieved. In this study, the functional annotation of these selected 15 up-regulated DEGs (10 contigs and 5 singletons) were characterized. The results showed that these 15 “DEG-unigenes” are mainly associated with defense and stress responses, regulation and signaling, protein synthesis and fate, and metabolism may play an important role in the primary responses to skinning injury in storage roots of sweetpotato. This study showed the importance of defense and stress responses genes to the formation of wound periderm. Furthermore, this results can be used for better understanding of the molecular mechanism of skinning/mechanical injury-related genes in the storage roots of sweetpotato as well as to all stems, fruits, and roots of all plants. Keragaan dan Kemampuan Meratun Lima Genotipe Sorgum
Ratooning can be used as an alternative cultivation technique for production of sorghum. This study was aimed to evaluate the growth and production of ratoon following the main crop. The study was conducted during August 2013 until November 2014, at Cikabayan Experimental Station, Bogor Agricultural University. The research was arranged in a randomized complete block design with four replications. The results showed that genotypes was significantly affected the vegetative and reproductive characters, physiologycal characters of the main crop, ratooning ability, and vegetative characters of ratoon crop. Plant height, stem diameter, fresh and dry weight, harvest time, and harvest index of main crops had significant correlation with ratooning ability. Plant height, total chlorophyll, fresh and dry weight, panicle weight, grain weight per panicle, 1,000 grain weight, harvest index, and transpiration rate of main crops had significant correlation with ratoon growth. Based on path analysis, the main crop growth directly affected ratoon height, and vegetative characters of ratoon directly affected the yield. The ratoon yielded 48.9-65.7% of the main crop yield. Produksi Benih Kentang Sistem Aeroponik dan Root Zone Cooling dengan Pembedaan Tekanan Pompa di Dataran Rendah
ABSTRACTNutrition pumps pressure is important in aeroponic. Optimal pump pressure produces well oxygenation, so that increases the dissolved oxygen content in the nutrition. It is good for plant roots. The purpose of this study was to determine pump pressure on the growth and yield potato seeds grown in aeroponics in lowland 125 m asl with root zone cooling. This study used a randomized block design non factorial. Data were analyzed using analysis of variance followed by a further test of orthogonal contrasts at the level of α = 5%. Results showed that different pump pressure on the provision of nutrient, temperature cooling in the root zone gave different results on the number of potato tubers per plant and weight per tuber in each variety. Root zone cooling temperature 15 °C, the pump pressure> 1.5 atm produce highest number of tubers per plant, i.e. 11.8 tuber of Granola variety and 8.2 tuber of Atlantic variety. The was no tuber produced from control (without referigeration). The highest weights per tuber (10.35 and 5.01 g for Atlantic and Granola variety, respectively) were reached with cooling temperature at 15 °C and the pump pressure > 1.5 atm.Keywords: evaporative cooling, hydroponics, potato, tuber, variety
Perilaku Biokimia Benih Kedelai Selama Penyimpanan dalam Kondisi Terkontrol
ABSTRACTPhysiological and biochemical changes in seeds indicates levels of viability and vigor during deterioration. The aims of the research were to study the patterns of biochemical changes in soybean seeds and its relationship to seed vigor during storage under controlled conditions. The experiment was arranged in a completely randomized design using two varieties as a factor which was nested into storage period and replicated four times. Two varieties were Gepak Kuning and Mallika, stored in controlled condition at temperature of 19-22 °C and 64-67% of relative humidity for six months. The seeds were evaluated for protein, peroxide value, electrical conductivity and seed viability. The exponential regression analysis was used to determine the patterns of biochemical changes of each soybean varieties during the storage period. The results showed that the biochemical behaviour of soybean seeds stored under controlled conditions for 24 weeks following the exponential equation of y = a + b expcx, which a, b, and c were constants that has different value for each variety. Electrical conductivity and peroxide value could be used as indicators of seed vigor during storage under controlled conditions for 24 weeks. Seed storage under controlled conditions are to maintain seed vigor >80% up to 12 weeks for Mallika and 24 weeks for Gepak Kuning with low oxidative process and less membrane damages.Keywords:deterioration, storability, vigo
Sensitivitas dan Keragaan Tanaman Coleus sp. terhadap Mutasi Induksi Kimia Menggunakan Ethyl Methane Sulfonate (EMS) Aplikasi Cara Rendam dan Tetes
Variant of Coleus sp. in Indonesia is very low. Induced mutation by Ethyl Methane Sulphonate (EMS) is able to increase variant of Coleus sp.. EMS is a mutagen that is effective and efficient in causing mutations. In mutation breeding, the high variant usually occurs around LC50 (Lethal Concentration). The objective of this study were (1) to obtain the LC50 value and sensivity of coleus plants, and (2) to evaluate the response of plant growth and to obtain new variance of coleus used soak and drops by EMS application. The experiment was arranged in randomized complete block design with three replications in each coleus. The materials used two Coleus sp. shoot cuttings i.e., C. amboinicus Lour. and C. blumei the purple/green color and the red. The concentration of EMS consisted of 0.00, 0.50, 0.75, 1.00, and 1.25% with application soak method (100 minutes) and drops (3 drops pipette). The results showed that the LC50 value of C. amboinicus Lour. 5.86% (soak). The LC50 value of C. blumei the purple/green color was 0.69% (soak), 0.82% (drops) and the red color 0.29% (drops), 0.89% (drops). C. blumei (the purple/green and red) was higher sensitivity rather than C. amboinicus Lour. There was an interaction between treatments on C. amboinicus Lour. whereas C. blumei (purple/green and red) had no interaction among treatments. Keragaan Agronomi Galur-galur Kedelai Potensial pada Dua Agroekologi Lahan Kering Masam
Performance of soybean growth and development is influenced by environmental conditions. On similar soil fertility conditions but different environmental conditions such as rainfall will also lead the changes in the performance of the plant. The research aimed to study the agronomic performance of the potential soybean lines in two different drought acid land agroecologies. The experiment was conducted at the South Lampung Tegineneng experimental station and East Lampung Taman Bogo experimental station from February until May 2013. The South Lampung Tegineneng experimental station has a pH of 5.50 and an average rainfall of 89 mm per month, and The East Lampung Taman Bogo experimental station has a pH of 5.12 and an average rainfall of 213 mm per month. The experiment consisted of 10 soybean lines adapted acid land and two check varieties. The experiment was arranged in randomized completely block design with four replications. The least significant different test (LSD) at α = 0.05 was used to calculate significant differences among all treatments. The results showed that rainfall influenced agronomic performance of soybean more than soil acidity. The yield performance in South Lampung was lower than in the East Lampung. Four genotypes produced higher grain yield than the Tanggamus variety in both locations, i.e. Tgm/Anj-862, Tgm/Anj-784, Tgm/Anj-888 Tgm/Anj-857, and Tgm/Anj-858. Tgm/Anj-862 was the most superior genotypes for having the highest number of branches, number of pods, and 100-seeds weight and seed yield and consistent at both locations. The four genotypes, especially Tgm/Anj-862, were potential to be developed in both locations or other locations with similar environmental conditions.Keywords: grain yield, ultisol, yield component
