Proliferation and Alkaline Phosphatase Activity of Osteoblast-like Cells on the Sintered Rutile Titanium Dioxide
application/pdfThe purpose of this study is creation of biomaterials from titanium dioxide (TiO2). This TiO2 has known for photocatalysis and osteogenesis. For the purpose of applying this function to orthodontic brackets and coating materials for implant, the relationship between surface of sintered and cell proliferation were examined. In addition, crystal structure and the surface property of sintering TiO2 were investigated. TiO2 were sintered at 1300°C for use as samples. We examined surface roughness, x-ray diffraction and scanning electron microscopy to make observations of the surface properties and texture. Moreover, mouse osteoblast-like cell line, MC3T3-E1 was cultured on sintered TiO2 in order to evaluate the cell proliferation and ALP. For the samples sintered at 1300°C, the crystalline phase of rutile-type TiO2 was confirmed.5000-fold magnified SEM
images of the surface of the unsintered samples, needle-like TiO2 crystals were pressure welded and showed mutual overlap, with pores occurring among the crystals. Sintering at 1300°C produced numerous small pores. Rutile TiO2 as a starting material was sintered at 1300°C and subjected to a cell culture experiment in which MC3T3-E1 cells were cultured on the sample, followed by viable cell counting and cell morphology observation
on days 7, 14, 21, and 28 of culture. In the test of cell proliferation, sintered at 1300°C samples was found to remarkable cell proliferation even after time had passed. ALP activity of cells on 1300°C TiO2 sample, the values were 110% and 126% on days 14 and 28 of culture, respectively. These changes were calculated using polystyrene dish as the reference condition. Thus, TiO2 sintered at 1300°C showed good compatibility and increase in the ALP activity in MC3T3-E1 cells.journal articl

Examination of the hearing, taste and smell function in patients with chronic dizziness following minor trauma
application/pdfDizziness and vertigo are common symptoms associated with minor head trauma or whiplash. Although these symptoms improve within a few weeks in many patients, the symptoms can last much longer in some patients. Furthermore, patients with minor trauma often suffer from other symptoms such as hearing loss, tinnitus, olfactory dysfunction, or taste disorder. The aim of the present study was to clarify the process by which dizziness following trauma arises. We investigated 254 patients (115 males and 139 females, with an average age of 44.2 years) who suffered from chronic dizziness following trauma but who did not have any abnormal head CT or MRI findings. In each of the patients, we evaluated the vestibular function and hearing, taste and smell by performing a spontaneous or gaze nystagmus test, an ENG evaluation (eye tracking test, optokinetic nystagmus test, caloric test), a stabilometer test, standard pure tone audiometry, a speech discrimination test, taste test (electrogustometry, filter-paper disk assay using taste solutions), and a smell test (intravenous olfaction test, odor-identification card test for Japanese). Accordingly, we investigated the relation between body sway and other sensory functions. We found a larger sway area than usual in 60% of the patients undergoing a stabilometer test with their eyes open and with their eyes closed. The results of the nystagmus and ENG evaluations suggested a vestibular disorder in 15% to 20% of the patients and a brainstem or cerebellum disorder in 10% or less of the patients. A significant correlation between the body sway area and smell identification was observed. However, smaller correlations were observed between body sway and the hearing threshold, the laterality of the hearing, the taste function, and the smell threshold. According to this study, we speculated that chronic dizziness following minor trauma might arise from related to central nervous system disorders, such as disorders of the hippocampus, hypothalamus, entorhinal cortex or other olfactoryrelated cognitive areas.journal articl

application/pdfRecently, the number of middle–aged and elderly orthognathic surgery patients with un-healthy oral conditions has been increasing. Although an interdisciplinary approach in-volving orthodontist, oral surgeon and prosthodontist is required in orthognathic surgery of these patients, deciding the jaw position after orthognathic surgery is difficult due to unhealthy oral conditions such as edentulous jaw and tooth loss. In the recently study, preoperative simulation methods for orthognathic surgery have been developed through the use of virtual reality computed technology. A male first examined at the age of 36 years and 7 months was diagnosed as having mandibular protrusion with multiple tooth loss and mandibular deviation. The patient was treatedinterdisciplinary by a team of specialists in surgical orthodontic, dental prosthetic and periodontal treatment. Using the interdisciplin-ary approach and haptic device with virtual tactile perception showed satisfactory results loss treated with orthognathic surgery.journal articl
HSP47 as a Possible Molecular Chaperone for the Collagen Synthesis in the Mouse Periodontal Ligament Cells due to Orthodontic Force
application/pdfOrthodontic mechanical stress induces various molecules, which cause structural changes in the proteins in periodontal tissue. The expression kinetics of HSP47 in mouse periodontal ligament (PDL) tissue after application of corrective mechanical stress and HSP47 expression during the PDL recovery were observed. Our examination data are as follows: HSP47 is constitutively expressed in the PDL and defends cells from different types of stress and maintains homeostasis under normal conditions. HSP47 is expressed in PDL fibroblasts on the pressure side damaged by application of mechanical stress and contributes to the repair of collagen tissue by activating PDL fibroblasts, supporting recovery from cell damage. These data suggests that, during bone addition to the PDL on the tension side, HSP47 also acts as molecular chaperone, which assists the maturation of bone morphogenetic proteins and aids osteoblast activation. On the other hand, it is possible that, when a mechanical stress is applied to the periodontal membrane on the tension side for a time too short for bone addition and abnormalities are caused in the collagen structure of the PDL fibroblasts, by functioning at the damage site, HSP47 inhibits extracellular secretion of abnormal collagen, stores the modified protein in the endoplasmic reticulum, thereby controlling decalcification of the PDL. Treatment of OPG-deficient mice with WP9QY, a RANKL-binding peptide, recovers alveolar bone loss by suppressing osteoclastogenesis and enhancing osteoblastogenesis.
application/pdfOsteoblasts express two key molecules for osteoclast differentiation, receptor activator of NF-κB ligand (RANKL) and osteoprotegerin (OPG), a soluble decoy receptor for RANKL. RANKL induces osteoclastogenesis, while OPG inhibits it by blocking the binding of RANKL to RANK, a cellular receptor of RANKL. OPG-deficient (OPG–/–) mice exhibit severe alveolar bone loss with enhanced bone resorption. WP9QY (W9) peptide binds to RANKL and blocks RANKL-induced osteoclastogenesis. W9 is also reported to stimulate bone formation in vivo. Here, we show that treatment with W9 restores alveolar bone loss in OPG–/–mice by suppressing osteoclastogenesis and enhancing osteoblastogenesis. Administration of W9 or risedronate, a bisphosphonate, to OPG–/–mice significantly decreased the osteoclast number in the alveolar bone. Interestingly, treatment with W9, but not risedronate, enhanced Wnt/β-catenin signaling and induced alveolar bone formation in OPG–/–mice. Expression of sclerostin, an inhibitor of Wnt/β-catenin signaling, was significantly lower in tibiae of OPG–/–mice than in wild-type mice. Treatment with risedronate recovered sclerostin expression in OPG–/–mice, while W9 treatment further suppressed sclerostin expression. Histomorphometric analysis confirmed that bone formation-related parameters in OPG–/–mice, such as osteoblast number, osteoblast surface and osteoid surface, were increased by W9 administration but not by risedronate administration. These results suggest that treatment of OPG–/–mice with W9 suppressed osteoclastogenesis by inhibiting RANKL signaling and enhanced osteoblastogenesis by attenuating sclerostin expression in the alveolar bone. Taken together, W9 may be a useful drug to prevent alveolar bone loss in periodontitis.journal articl
Reactions to Bioabsorbable Suture Thread Embedded in Rat Subcutaneous Tissue
application/pdfWe examined the subcutaneous tissue reactions in rats to bioabsorbable suture thread using histopathological methods. Using Wister rats, Vicryl®, a bioabsorbable suture thread, was embedded into the subcutaneous tissue and histopathological examination was carried out after 4 weeks. Cholesterin crystals were used for the control. Furthermore, immunohistochemistry for CD68 was done. Histopathological examination showed proliferation of granulation tissues in
both experimental and control groups. The majority of cells in the granulation tissues were macrophages and giant cells. Fibroblasts were also observed in the proliferating granulation tissues surrounding the embedded bioabsorbable suture
thread. Immunohistochemistry revealed that macrophages and giant cells were positive to CD68. The results suggest that the embedded bioabsorbable suture thread is not only fabricated to undergo absorption but also for phagocytosis by
macrophages and foreign body giant cells.journal articl
