    Cold Stress Response Genes of Lantiplantibacillus plantarum subsp. plantarum Mut-3 and Lantiplantibacillus plantarum subsp. plantarum Mut-7 Support the Ability to Survive in Low-Temperature Conditions

    Probiotics are widely consumed in various food matrices to provide health benefits to the host. The viability of probiotic cells is influenced by several factors, including exposure to high temperatures during the production process and low temperatures during storage. In this study, we report the response to cold stress of Lantiplantibacillus plantarum subsp. plantarum Mut-3 and Mut-7 after 24 h of storage at 4°C and -20℃. The cell number of Lantiplantibacillus plantarum subsp. plantarum Mut-3 and Mut-7 in low-temperature condition had no significant differences than their initial number: 11.88 log CFU/ml and 11.62 log CFU/ml at 4°C; 11.51 log CFU/ml and 11.47 log CFU/ml at -20°C for Mut-3 and Mut-7 respectively. The results indicated the survival capacity of Lantiplantibacillus plantarum subsp. plantarum Mut-3 and Mut-7 at low temperatures. The genes encoding cold shock proteins for the response to cold stress were evaluated by genome sequencing. The CspA/CspC genes of Lantiplantibacillus plantarum subsp. plantarum Mut-3 and Mut-7 possibly play a role in maintaining cell resistance at low temperatures, since the genes products predicted to have conserved motifs in the RNA binding protein (RNP) -1 and RNP-2 responsible for cold response stress which are similar to those in other bacteria

    Banana Bunchy Top Virus Molecular Confirmation and DNA-S Phylogenetic of some Banana Isolates from Indonesia

    This study was aimed to confirms the Banana Bunchy Top Virus (BBTV) infection of ten banana accessions from Indonesia through PCR assay using primer of BBTV coat protein (CP) gene, also the sequences characteristics and phylogeny. Preliminary morphological results showed five acessions were positively infected with slight to severe intensity symptoms i.e. Pisang Berlin, Candi, Billa, Morosebo, and Mas Kripik; and five accessions were symptomless i.e. M. acuminata var. rutilifes and M. balbisiana, Pisang Ebung, Madu and Moseng. However, PCR results qualitatively confirms that all accessions were positively infected. Two sizes of amplicons were identified. Pisang Candi isolate was showing ±500 bp amplicons, whereas the others were ±1000-1100 bp. The total aligned and selected BBTV CP sequences of 36 accessions (bananas and other host species) was 450 nt. It was considered highly conserved (85.55%), low G+C content (41%), and high genetic similarities (92.47 to 100%). Phylogenetic analysis delineates isolates into two large groups, i.e. the Asian group (East Asia and South-east Asia) and the South Pacific group (South Asia, Africa and the Pacific). The findings of this study are usefull for broader applicability of further banana breeding program particularly for mitigation, selection and evaluation of banana with BBTV resistance

    Diversity and Potential Carbon Services of In-Situ Conservation Reserve in Gunung Sepuluh Timur Forest at Balikpapan, East Kalimantan

    Establishing an in-situ conservation reserve is one of the strategic approaches to conserving an ecosystem and protecting it from immediate threats, including deforestation and land-use conversion. Kalimantan is one of the islands in Indonesia that is currently threatened by deforestation. To protect East Kalimantan diversity, mainly in Balikpapan, an in-situ conservation reserve has been established in Gunung Sepuluh Timur (GST) forest. In contrast, the information on tree diversity and how the presence of tree community within the forest can benefit the ecosystem are still limited. This research aims to assess the potential benefits and carbon storage of the GST conservation forest. The assessment method applied tree, pole, and sapling surveys combined with forest cover analysis using GIS and NDVI (Normalized Difference Vegetation Index) approaches. The GST contained 56 tree species, 25 pole species, and 16 sapling species. Diversity (H’) at tree stages was the highest (H’ = 0.759; 95%CI: 0.64-0.88) and H’ at sapling stages was the lowest (H’ = 0.719; 95%CI: 0.51-0.92). The carbon service potential of GST forest was 87.04 t C/ha on average. It was estimated that the economic benefits of GST in sequestering carbon based on the compliance market rate were equal to US$ 189,758/year

    Genetic Diversity and Connectivity of Sea Urchin Tripneustes gratilla in Region Surrounding Cenderawasih Bay, Papua-Indonesia and Indo-Pacific

    Cenderawasih Bay has extremely high biodiversity with an enormous amount of endemic species due to geography isolation. Tripneustes gratilla is one of the species that is abundant in the bay. The species has ecological value that is suitable for bio-indicators of the environmental condition. Since the Bay relatively isolated area, then we examined the impact of the geographical conditions on genetic diversity and connectivity of Tripneustes gratilla among populations in the region surrounding Cenderawasih Bay based on the mitochondrial cytochrome c-oxidase-1 gene. Results of the study showed that genetic variation of the species within the population is high; despite the genetic variation among populations was low. The data suggested that the entire population of T. gratilla were closed connected, homogeneous, and shared polymorphic profile. Then we assumed the gene flow occurred for a long time among populations without geographical barriers. This information is a warrant to develop an effective strategy to maintain biodiversity in the Cenderawasih Bay

    Implementation of Dimer-based Screening System in Escherichia coli BL21(DE3) for Selection of Actinomycetes Compounds as Anti-HIV Candidate

    Actinomycetes are reported to have inhibitory activity against several types of Human Immunodeficiency Virus proteases, enzyme with major role in the process of maturation of the virus thus it can infect new cells. Therefore, exploration of Indonesia’s actinomycetes species is expected to be a breakthrough for HIV treatment. In this study, selection of anti-HIV candidate compounds was conducted using a dimer-based screening system on recombinant Escherichia coli BL21(DE3). The construct includes the fusion of the AraC DNA binding domain + HIV-1 protease as the regulator and the green fluorescence protein as the reporter. Confirmation of the plasmid construct was carried out by PCR which showed size of ~1,076 bp. Sequencing analysis proved 100% similarity and identity between construct used in this study and one previously designed. SDS-PAGE showed the presence of band in the size of ~24 kDa equal to the size of the fusion protein. Compounds BLH 1-12 (2) EA, MAE 1-13 EA, BLH 1-1 EA, BLH 7-5 MetA, LC 98 (1) EA, exhibited consistent and significant protease-HIV inhibitory activity at certain concentrations. Thus, in this study, dimer-based screening system is considered to be able to detect actinomycetes as a new anti-HIV candidate for the protease inhibitor group

    Decolorization of Synthetic Dyes by Tropical Fungi Isolated from Taman Eden 100, Toba Samosir, North Sumatra, Indonesia

    Taman Eden 100 is one of the tourist parks located in Toba Samosir, North Sumatra, Indonesia that provides a wide range of biodiversity. The objective of this research was to obtain the potential fungal isolate with high laccase activity to decolorize synthetic dyes. The results show that six isolates of Perenniporia subtephropora EDN 050, Trametes hirsuta EDN 082, T. hirsuta EDN 084, T. hirsuta EDN 085, Deconica coprophila EDN 114, and T. pavonia EDN 134, which were confirmed by molecular identification using 5.8 rDNA/ITS analysis, exhibited decolorization activity. Six potential isolates showed the ability to decolorize textile dyes of Acid Blue 129, RBBR,Orange II, and Reactive Black 5. The highest decolorization rate of 100 ppm Acid Blue 129, RBBR, Orange II, and Reactive Black 5 dyes by using T. pavonia EDN 134 were 98.87%, 98.26%, 100%, and 98.11%, respectively after 96 h incubation. Almost all positive isolates also decolorized dyes at higher concentrations (1,000 ppm). This study offers the potential of Indonesian tropical fungal isolates for synthetic dyes waste treatment

    Protein Hydrolysate of Green Peas Bromelain Attenuates Kidney Fibrosis in Cisplatin-Induced Nephrotoxicity Rats: Emphasis on Anti-Inflammatory Activities

    Massive tubule fibrosis is a histopathology hallmark of chronic renal failure (CRF). The previous study of protein hydrolysate of green pea (Pisum sativum) bromelain (PHGPB) showed promising results as an antifibrosis in glucose-induced renal mesangial culture cells, by decreasing their TGF-β levels. In this experiment the anti-inflammatory, anti-fibrotic effect of PHGPB in Cisplatin-induced chronic renal failure (CRF) rats was measured. The purpose of this study is to evaluate the anti-inflammatory and antifibrosis effect of PHGPB in Cisplatin-induced chronic renal failure rats. Five groups consisted of five rats: negative control, Cisplatin control, and three groups of Cisplatin+PHGPB (dose of 100, 200, and 400 mg/kg BW/day) treated for 56 days. The examined parameters are Beta-2-microglobulin (β2MG), hs-CRP, histopathological observations of Masson\u27s trichrome stain, and IHC. The level of β2MG and hs-CRP in the Cisplatin+PHGPB group lowered and was highly different from Cisplatin control (p<0.05) depending on doses. In histopathology, the Cisplatin+PHGPB 400 mg/kg BW group showed less fibrosis and no significant difference with the negative control in Masson trichome staining. While in IHC histopathology, PHGPB treatment slightly ameliorated TGF-β expression and intensity of TGF-β values convincingly. In conclusion, PHGPB can relieve kidney fibrosis in cisplatin-induced nephrotoxicity rats: Emphasis on anti-inflammatory activities

    In Silico Analysis of Protein of Milk, Soybean, and Kefir as Anti- Thrombotic Bioactive Peptide

    Currently, Cardiovascular Diseases (CVDs) cases are increasing significantly in Indonesia and the world. WHO reported that in 2019-related CVD patients accounted for 85%, which increased to 87% in 2021. The cases increased significantly by 2%, which is a significant number as it covered the world population. Stroke is a disease affected by platelets undergoing thrombolysis by the α-thrombin (IIa) receptor with coagulation factors on the heart blood vessel. Chemical or synthesis drugs to treat thrombolytics such as heparin are already available, but they would be harmful if misused. In this study, we focused on the activity, interaction, and prediction of the physicochemical of anti-thrombotic peptides. We found that k-casein from kefir has the highest activity of 0.037 A based on BIOPEP analysis with 17 peptide libraries and molecular docking visualization analysis using PyMol, PyRx, and Discovery Studio for screening the best peptide. ToxinPred and AlgPred showed there are no potentially harmful peptides, but the protein variant of β-conglycinin subunit can be an allergen known to the immune system IgE. In conclusion, the GPR peptide has the highest binding affinity, with a total energy of -6.8 kcal/mol

    Paederia foetida Linn Leaves-Derived Extract Showed Antioxidant and Cytotoxic Properties Against Breast Carcinoma Cell

    Paederia foetida Linn. is a rich source of active compounds, yet its antioxidant and cytotoxic properties remain poorly studied. This current research aimed to investigate the chemical profiles of a crude extract derived from P. foetida leaves, its fractionated substances, and their antioxidant and cytotoxic properties on MCF-7 cells, a breast carcinoma cell. Six fractionated substances have been separated in thin layer chromatography (TLC) plate. Five fractions showed antioxidant properties, as indicated by the formation of the yellowish band on the TLC plate after spraying with the DPPH reagent. Furthermore, crude extract of P. foetida leaves and its fractions are also characterized by antioxidant properties against 2, 2\u27-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals. F1 fraction exhibited the most potent antioxidant activity with IC50 values of 10.94±2.67 and 50.04±0.48 µg/ml, respectively, toward DPPH and ABTS radicals. Crude extract and all six fractions also varied in their cytotoxic properties against MCF-7 cells, with IC50 values ranging from 410.24±0.30 to 831.57±6.91 µg/ml. F3 fractions showed the strongest cytotoxicity (IC50 value: 410.24±0.30 µg/ml). Methanolic extract and active plant leaf fractions contained flavonoid and phenolic compounds, as measured by aluminum chloride and Folin-Ciocalteu reagent, respectively. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis showed that phenol, flavonoid, alkaloid, limonoid, and steroid were the major compound found in the crude extract and fractions. In conclusion, the present study proved that crude extract of P. foetida leaves and its fractionated substances were potential as antioxidant and cytotoxic agents

    Anti-Inflammatory Effect of Isolate Compounds from Ethyl Acetate Fraction of Piper crocatum Ruiz and Pav Leaves on Lipopolysaccharide-induced RAW 264.7 Cells

    Red betel (Piper crocatum Ruiz and Pav) is a traditional Indonesian plant. The ethyl acetate fraction of red betel leaf has the potential as an anti-inflammatory. This research aimed to determine the anti-inflammatory effect of isolate compound from ethyl acetate fraction of Piper crocatum leaves on the production of cytokine pro-inflammatory TNF-α and ICAM-1 in lipopolysaccharide-induced RAW 264.7 cells. Cell viability was determined by the MTT method in the concentration range of 1.25; 2.5; 5; 10; 20, and 40 µg/ml. The TNF-α and ICAM-1 level was determined by the TNF-α and ICAM-1 assay kit as measured by an ELISA plate reader. Cell viability test of isolate compound from ethyl acetate fraction, the concentrations that gave viability percentages above 80% were 10; 5; 2.5 and 1.25 ug/ml. Isolate compound at concentrations of 10, 5, 2.5 and 1.25 ug/ml significantly inhibited TNF-α and ICAM-1 production with p<0.05 (p = 0.000)
