    ACCURACY IMPROVEMENT ON SEA SURFACE HEIGHT ESTIMATION BASED ON WAVEFORM RETRACKING ANALYSES OF JASON-2 SATELLITE IN JAVA SEA

    A waveform created by the reflected signal from altimeter satellite in offshore is generally in ideal shape (Brown-waveform) and produces an accurate sea surface height (SSH) estimation. However, over coastal waters, the waveform shape becomes complex due to a disruption by reflected signal from land, resulting inaccurate SSH estimation. The objective of this research was to improve the accuracy of SSH estimation employing waveform retracking analyses of Jason-2 altimeter satellite data in the Java Sea during the years of 2012-2014. This study used data from the Sensor Geophysical Data Record type D (SGDR-D) from Jason-2 satellite (cycle 129 - 239) and global geoid undulation data of Earth Gravitational Model 2008 (EGM08). Waveform retracking analyses were conducted using several retracker methods. The performance of the all retrackers were examined using a world reference undulation geoid of EGM08. The results showed that the waveform retracking analyses were able to improve the accuracy of SSH estimation approximately 29.7% in the north coast and 56.4% in the south coast of total non-Brown-waveform in each region. Higher improvement percentage (IMP) of SSH estimation found in the southern coastal areas was due to a relatively smooth coastline formation in this region than in northern coastal region. There was no specific retracker that produce dominant IMP of SSH estimation. However, the  threshold 10% retracker produced better SSH estimation than the other retrackers with dominant IMP values of 57.1% (pass 051), 48.1% (pass 064), and 25.7% (pass 127).     WIRELESS SENSOR NETWORKS BUOY FOR COASTAL WATERS OBSERVATION

    The availability of data in real time and continuous is important to monitor in environmental change as early as possible. Wireless sensor networks (WSN) offer a new paradigm in the field of oceanography that can measure the parameters of complex marine environment using a moored buoy. This paper described design of a data transmission system with a moored buoy and tested the performance of WSN instrument based on ZigBee protocol radio module for monitoring coastal water environment in real time. Instruments were divided into two i.e., (1) five sensors served to measure sea surface temperature, stored the data, and transmitted the data to the base station, and (2) a coordinating instrument that placed on the bases station served to receive and record all measurement results of each sensor. The testing was done by deploying the instrument sensors in waters with depths of 2 to 5 meters and a coordinating instrument was located on the ground as a base station. Each instrument\u27s sensor measure sea surface temperature, store, and transmit it to other nearby sensors and forward data to the next sensor and then to the next sensor send it to the base station. The Packet Delivery Ratio (PDR) value wa used as an indicator to determine the instrument performance and the values were from 89.69% up to 100% with transmission range up to 430 meter and battery endurance was up to 26 hours. The result showed that a buoy moored instrument based on WSN ZigBee radio module protocol has the potential for monitoring coastal water environment in a real time.     THE ADDITION OF ZEOLITE, ACTIVATED CARBON, CLOVE OIL, AND DIFFERENT SALINITY FOR GLUCOSE RESPONSE AND OSMOTIC PRESSURE ON JUVENILE GIANT PRAWN IN CLOSED TRANSPORT SIMULATION SYSTEM WITH HIGH DENSITY

    Long distance transportation of prawn juvenile usually use a closed system. Indonesian farmers often face  problem in this juvenile transportation such as a low survival rate due to shrimp stress and changes in water quality during transportation. This study was aimed to determine the effect of salt, cloves oil, zeolite, and activated carbon for maintaining water quality in 24 hours juvenile giant prawn simulation transportation at high density. The study was conducted at laboratory scale with a complete randomized design with 4 treatments and 3 replicates. For each treatments, it was added 4.67 µl/L clove oil, 20 g of zeolite, 10 g of activated carbon within 4 (four) different salt concentration i.e., 0 g/L (A), 4.7 g/L (B), 9.4 g/L(C), 14.1 g/L(D) and K (without salt, clove oil, zeolites and activated carbon). The results showed that the addition of 9.4 g/L of salt, 4.67 µl/L clove oil, 20 g of zeolite, and 10 g of activated carbon produced the best results with the highest survival rate by 88±2% at the end of simulation transportation and 82±2.83% after rearing, water quality and physiological respons of shrimp remains in good condition at the simulation transportation and at rearing. Keywords: juvenile giant prawn, salt, high density, physiological response

    EVALUATION OF MICROBIAL FLOC AND MICROALGAE Spirulina platensis COMBINATION FOR JUVENILE COBIA Rachycentron canadum DIETS ON GROWTH AND PHYSIOLOGICAL RESPONSES AFTER IMMERSION IN FRESHWATER

    In marine aquaculture, immersing marine fish species in fresh water can remove ectoparasite that adhere to all over the fish body. This study aimed to evaluate the impacts of combining microbial floc and microalgae Spirulina platensis in juvenile cobia diet on growth performance and stress responses after immersion in aerated fresh water for 15 minutes. The fishes were reared in concrete tanks for 40 days before collecting data on their growth performance. The stress response was determined by mea-suring both glucose and cortisol levels before (0 h) and after (1, 2, 4, 6, 24 hours) immersion. The fish-es fed on the 15% of combining microbial flock and microalgae Spirulina platensis diet showed the highest growth rate with the lowest feed conversion ratio compared to other treatments. The cortisol level of juvenile cobia in both the 15% and 30% combination of microbial floc and microalgae Spiru-lina platensis treatments did not increase during the first hour following the immersion compared to the control treatment. The glucose level also increased after one hour immersion in freshwater of all treatments. This indicated that feeding juvenile cobia on microbial flocs and microalgae diets had a retarding effect on the physiological responses (cortisol and glucose) after immersion in fresh water.Keywords: microbial, microalga, Spirulina, glucose, cortisol, stress, cobi

    BINARY VECTOR CONSTRUCTION OF KAPPA(κ)-CARRAGEENASE GENE AND TRANSFORMATION TO Agrobacterium tumefaciens AS MEDIATOR FOR SEAWEED TRANSGENIC GENERATION

    Increasing of kappa (κ)-carrageenan content in Kappaphycus alvarezii seaweed is potentially be achieved by applying transgenesis technology. This study was performed to obtain a construction of  κ-Carrageenase gene and Agrobacterium tumefaciens to carry those construction genes.  The κ-Carrageenase (κ-Car) gene was involved in κ-carrageenan biosynthesis. The κ-Car gene sequence was ligated between the 35S CaMV promoter and tNos terminator sequences to generate pMSH/κ-Car expression vector. Transformation of pMSH/κ-Car plasmid to Escherichia coli was performed by heat-shock method, and to Agrobacterium tumefaciens by tri-parental mating method. The results showed that several colonies of E. coli and A. tumefaciens grew in the selective culture mediums containing antibiotic. PCR analysis using primers 35S-Forward and tNos-Reverse with DNA template from those bacterial colonies resulted DNA fragment of about 2,000 bp, the same as the total length of 35S CaMV promoter, κ-Car gene and tNos terminator sequences. Therefore, the construction of pMSH/κ-Car gene was succeeded and a colony of A. tumefaciens transformant carrying pMSH/κ-Car plasmid was successfully produced.                                                                                   Keywords:  Agrobacterium tumefaciens, kappa(κ)-Carrageenase gene, transgenesis, vecto

    HABITAT QUALITY MUD CRAB (Scylla serrata) IN MANGROVE ECOSYSTEM OF BINTAN BAY, BINTAN DISTRIC, RIAU ISLANDS

    Mud crab (Scylla serrata) is one of small scale fisheries commodity that have high economic value and typically associated with good mangrove ecosystem. A habitat degradation will cause a serious impact on the existence of mud crab population. The puropse of this study was to assess the ecological quality of mud crab habitat condition on Bintan Bay’s mangrove ecosystem. Ecological data collection was conducted by using plots line transect for mangrove and the mud carb data collection by using fishermen catch (fisher based survey). The results showed the value of habitat quality index (HQI) of mud crab (Scylla serrata) ranged from 52-82 which mean in the “moderate” category (index value 43-66) and "good" (index value 67-90). It showed that Bintan Bay’s mangrove ecosystems was good enough to support the viability of mud crab. Habitat quality had a linier impact on body weight gain (R2 = 99.78%) and carapace width (R2= 99.21%).     SUSTAINABILITY STATUS OF MANGROVE ECOSYSTEM MANAGEMENT IN SEMBILANG NATIONAL PARK, BANYUASIN REGENCY, SOUTH SUMATERA PROVINCE

    he utilization of mangrove in Sembilang National Park area has increased lately so that it is necessary to assess the sustainability of the mangrove ecosystem management in the region. The study was conducted in March-April 2015. The research location was in the National Park Sembilang, Banyuasin Regency, South Sumatra. The study used primary data and secondary data collections. The primary data were collected from direct observation of mangrove vegetation and via directs interviews of 86 respondents using purposive sampling method. The sustainability status and recommendations for management strategies were determined using Multi Dimensional Scaling (MDS) method approaching by RAPFISH (Rapid Assessment Technique for Fisheries).Current sustainabilitystatus of mangrove management in Sembilang National Park, Banyuasin Regency, South Sumatra was "less sustainable" with multidimensional sustainability index of 49.81. Priority attributes that need to be fixed in order to improve the status of sustainable management of mangrove ecosystems in the region were area changes, local knowledge, conflict of interest, and local wisdom.     COMMUNITY STRUCTURE OF GROUPER FISH (Serranidae) LANDED IN PEUKAN BADA SUBDISTRICT, ACEH PROVINCE

    ABSTRACTInformation on important and economic grouper fish (Serranidae family) in Peukan Bada waters, Province of Aceh is very limited. This objecives of this research were to determine spatial and temporal variabilities in species composition and community structure of grouper landed in Peukan Bada, Aceh Besar District. The research was conducted using survey method during February to June 2015 on three fish landing sites i.e., Ujong Pancu, Lamtengoh, and Lamteh. The groupers were caught by using handline. The landed fishes were therefore counted and identified up to species level. The data were also analyzed based on taxonomic information and individual number of each species, community structure using the diversity index (H’), eveness (E), and dominance (D). The results obtained 835 individual, 21 species of grouper belong to 4 genera (Aethaloperca, Cephalopholis, Epinephelus dan Variola). Based on species, Epinephelus fasciatus and Cephalopholis sonnerati were the most grouper fishes in the region.  Based on community structure, diversity value (H’) in Lam-tengoh was higher than that in Ujong Pancu and Lamteh. Total grouper fish catches were not significantly different (P>0,05) among the three region. Meanwhile, total grouper fish cacthes were significantly different between seasons (p<0,05). The diversity species variation during five months of observation was affected by the differences in catches area, the condition of aquatic enviroment, and oceanographic condition.Keywords: Groupers, Peukan Bada, species composition, community structur

    PLANKTON COMMUNITY STRUCTURE OF TRADITIONAL AND INTENSIVE BRACKISHWATER PONDS IN PROBOLINGGO REGENCY, EAST JAVA PROVINCE

    This experiment aimed to determine the plankton community structure in intensive and traditional ponds of  Probolinggo Regency, East Java Province. Plankton collected with plankton net size of 60 μm and preserved with lugol 1%. Water qualities such as temperature, transparancy, salinity, dis-solved oxygen, pH, total organic matter, NO2-N, NO3-N, NH3-N, PO4-P, and total suspended solids were measured and analysed in this study. Planktons were identified using microscopy. Cells were counted using cell counting method. The results showed that in intensive pond we found 16 species of phytoplankton and 7 species of zooplankton with abundance 570-1.808 ind./L, while in traditional ponds, we found 10 species of phytoplankton and 3 species of  zooplankton with abundance  134-776 ind./L. The dominant species of phytoplankton in the intensive and traditional ponds were Navicula sp andNitzschia sp, each of Bacillariophyceae class with abundance of 423 ind./L and 198 ind./L, res-pectively. Zooplankton species in intensive pond was copepod sp, while in traditional  one was Oitho-na sp, each of Crustaceae class with abundance of  66 ind./L and 37 ind./L, respectively. Diversity and abundance of plankton in intensive pond was higher than in the traditional ponds. The water quality in intensive pond was optimal so that the response to the absorption of  N and P inorganic by phyto-plankton was quicker and more effective than in traditional pond producing an increase in plankton communities. The increase value of NO3-N and total suspended solids in intensive pond caused the in-crease abundance of Navicula sp.  The high value of NH3-N caused the abundance of Navicula sp to decline. The high values of dissolved oxygen, pH, total suspended solid, and NH3-N in traditional pond can decrease the Nitzschia sp abundance.  Keywords: plankton communities, intensive and traditional ponds, Probolinggo, East Jav

    THE EFFECT OF STOCK DENSITY OF GOLDEN-MOUTH TURBAN (Turbo chysostomus, L.) ON THEIR SURVIVAL AND GROWTH UNDER AIR WATER LIFT SYSTEM

    Golden-mouth turban (Turbo chrysostumus, L.) belongs to the phylum of molluscs that inhabits coral reef ecosystem in group. Golden-mouth turban normally uses algae for their food. Study on survival and growth of the juvenile golden-mouth turban focussing on different density under air water lift sys-tem is still limited. This study was conducted on 7 January – 6 May,  2015, in a laboratory to observe the effect of different density on the survival and growth of golden-mouth turban under air water lift system. Based on the analyses of variance (ANOVA), there was no significant different (P>0.05) on mortality rate for different stock density treatment (5 ind., 10 ind., 15 ind., and 20 ind. each in 10 liter water volume). However, based on growth rate, the density of 5 individuals produced the highest monthly growth rate of 1.88 mm/month of shell height, 1.18 mm/month of mouth opening, and 1.1 gr/month of wet weight with the highest daily mean food absorption of 91.55 %. For all treatment, the correlation of shell height and wet body weight exhibited a similar growth pattrern i.e., allometry minor (b<3).
