    Low temperatures could inhibit bacteria producing histamine during fish handling process. Longtail tuna (Thunnus tonggol) is kept in ice starting from catching process until delivering to retail market to maintain quality of fish. However, long period of this storage causes growth of bacteria and deformation of histidine into histamine. This study was aimed to determine the effect of different storage conditions to the quality of longtail tuna through sensory and histamine test. Samples of longtail tuna were kept for 18 days in room temperature storage (28±2°C), cold temperature (0±3°C) and freezing temperature (-10±4°C). The quality of tuna was evaluated using Quality Index Method (QIM). The formation of histamine was analysed on day 0, 1, 2, 3, 4, 8, 14, 18 using spectrofluorometry method. Sensory acceptability limit at room temperature was one day, while cold storage reached 10 days with QIM score  11. Longtail tuna was spoiled on day 2 at room temperature with QIM score >12 and concentration of histamin at 1713.88 mg/kg. The results of histamine in frozen and cold storage on the 18th day were 59.99 mg/kg and 77.98 mg/kg, these results exceeded the FDA limit (50 mg/kg) but still below the limit of FAO (100 mg/kg).The best storage condition is frozen storage, maintaining sensory acceptability and preventing histamine formation

    Chitosan is a white amorphous solid chitin, and of high biological and mechanical characteristics.Chitosan production is divided into two stages. First is chitin isolation (deproteination, demineralization,and depigmentation), and second is deacetylation of chitin to chitosan. This research was aimed to determinethe characteristic of chitosan isolated from the shells of the white leg shrimp using different temperatureand time in the deasetilation process. The FTIR test results showed that the chitin group was partiallybroken into chitosan in every treatment. The highest content of chitosan was obtained at the temperaturetreatment of 100oC for 2 hours, while the highest degree of deacetylation, viscosity, and molecular weightwere observed at the temperature treatment of 120oC for 3 hours. The sample were not soluble in aquades,methanol, or ethanol indicating the samples were chitosan

    DNA Mini-Barcodes as A Molecular Marker for Various Hairtail Fish Products Traceability

    Hairtail fishery is an important fishery commodity with high economic value as export commodity. Authentication based on DNA mini-barcodes can be applied to overcome seafood fraud and to support Indonesian traceability system. This study was aimed to design species-specific primers of DNA minibarcodes and authentication of various processed hairtail fish products such as smoked, baked, fried, canned, and boiled. Analytical method applied PCR-based of cytochrome c oxidase 1 (COI) full length andmini-barcodes. Mini-barcode primer specific for hairtail (Trichiurus lepturus) successfully amplified DNA of processed hairtail fish. The BLAST analysis showed the samples were identified as Trichiurus sp. and T. lepturus with homology level of 91% to 100%. Phylogenetic analysis showed the processed sample (LC dan LD) were in the same group with T. lepturus

    Characteristics of Daggertooth Pike Conger Fish (Muraenesox cinerus) as Raw Material for Diversification Products Development

    Daggertooth pike conger fish (Muraenesox cinerus) has elongated body shape similar to eels and distribute widely worldwide. The fish has edible portion of 50-60% but there has been less information regarding the chemical characteristics. The aim of this research was to determine chemical composition of daggertooth pike conger fish (proximate, amino acid, fatty acid and mineral) and textural properties from its kamaboko. Purposive sampling method applied in this research with length, freshness and distance from location as criteria. Analytical method performed including proximate, amino acids, fatty acids, and minerals (P, Na, Ca, K, Mg dan Fe). Chemical composition from daggertooth pike conger fish showed that on proximate levels of protein, fat, ash, carbohydrates and moisture content value were 12.273%, 4.96%, 1.17%, 1.12% and 80.49% respectively. Dominant macro minerals, namely potassium (K) 170.20 mg /100 g, Ca 90.75 mg /100 g, and Na 80.15 mg /100 g. Amino acid (AA), namely glutamic acid 2.68%, 1.57% lysine, leusine 1.25%, 1.54% aspartic acid and arginine 1:04%. Moreover, the essential fatty acids namely EPA (eicosa pentanoat acid) 0.6%, DHA (docosa hexanoat acid) 0.9%, 0.2% fatty acids linoleic, oleic and linolenic 1.4% 1.0%.     Supplementation of Skipjack Roe Protein Concentrate (RPC) Into Breastfeeding Complimentary Foods

    Protein malnutrition in toddler is a problematic issue in Indonesia. Inclusion of roe protein concentrate (RPC) from skipjack in the breastfeeding complimentary foods of or infant foods can be an alternative to solve the protein malnutrition. This research was aimed to analyze the organoleptic score, nutritional facts and functional properties of the infant food supplemented with the RPC. Six different formulations (F1-F6) were prepared. The F2 formula was considered as the best formula. The infant food prepared using this formula contained 19.43% of protein and was higher than the control and the commercial product. The food also contained nutrition as required by the Indonesian National Standard (SNI) and Food and Agriculture Organization (FAO) standard. Furthermore, the functional properties of the infant food was comparable to that of the commercial and control product

    Effectiveness of Sodium Bicarbonate (NaHCO3) on Physicochemical and Sensory Characteristic of Notopterus notopterus Bone Chips

    This study was aimed to determine the physicochemical and sensory characteristics of chips made from bones of Notopterus notopterus previously soaked in sodium bicarbonate. Different concentrations of sodium bicarbonate (0; 0.5; 1; 1.5; 2; and 2.5%) were used to soak the bones. The parameters observed were chemical analysis, physical analysis, and sensory analysis.  The soaking of  the fish bones in sodium bicarbonate  had significant effects (p<0.05) on the chemical characteristics including water content, ash content, protein content, NaHCO3 residue as well as physical characteristic of the chips. However, on the soaking did not affect phosphorus, calcium and sensory characteristics of the chips

    Optimization of Extraction Temperature on The Quality of Alginate from Seaweed Sargassum muticum

    Seaweed is one of Indonesia’s marine biological resources which has potential to produce alginate which can be used for food and non-food industries. Several studies had been carried out in an effort to improve the quality of alginate extraction. This study aims to determine the effect of extraction temperature on the quality of sodium alginate from Sargassum muticum. This study used variations in extraction temperatures, namely 30; 40; 50; 60 and 70°C. The parameters observed included yield (%), moisture content (%), ash content (%), pH, whiteness degree and functional groups of alginate. The results showed that the difference in extractiontemperature of sodium alginate had a significant effect on yield and viscosity, but did not significantly affect water content, ash content, and whiteness degree. Extraction temperature of 60°C produced the best yieldand viscosity, 14.90±2.26% and 236.10±36.61 cPs, respectively

    Characteristics of Combination Shark Liver Oil (Centrophorus sp.) And Spirulina Powder as Food Supplement

    The content of fish oil supplements is sufficient to fulfill the need of EPA and DHA, however adding nutritional value will be beneficial. This study was aimed to characterize the chemical content of the supplement from a combination of shark liver oil and Spirulina powder. The oil was extracted using dry rendering method at 50oC for 8 hours. The extracted oil was then combined with Spirulina powder with a concentration of 10; 30; and 50%. In general, the chemical characteristics of fish oil were affected by the addition of Spirulina plantesis. The fish oil with adddition of Spirulina 50% had the highest protein (24.3%) and essential amino acid content (but comparable DHA content. In conclusion, addition of Spirulina increased the chemical content of the fish oil, but reduced their fatty acid content

    Optimization of Biodegradable Film from Cellulosa of Seaweed Solid Waste Eucheuma cottonii with Addition of Glycerol, Chitosan, CMC and Tapioca

    Cellulose from the solid waste of seaweed Eucheuma cottonii is one of the wastes that can be used as raw material for making biodegradable films. The condition for making optimum conditions for making biodegradable film from E. cottonii with treatment of glycerol concentration (0; 0,25; 0,5; 0,75; and 1%), tapioca concentration (2; 4; 6; 8; and 10%) , carboxy methyl cellulose (CMC) concentration (1; 2; 3; 4and 5%) and chitosan concentration (1; 2; 3; 4; and 5 The experimental design uses a surface response method with 4 replications. The analysis carried out consisted of elongation percent, tensile strength, solubility and biodegradable film using surface response method. The optimum value occurs in the process conditions using a concentration of glycerol of 0.162%, tapioca concentration of 3.78%, CMC concentration of 2.5% and chitosan concentration of 1.62% with tensile strength 95.013 Mpa, percent elongation of 8.92%, and solubility 80.62%

    Using Oven Microwave for Synthesis ZnO Nps using Sargassum sp. and Padina sp. Extract

    An oven microwave was established for synthesis of zinc oxide nanoparticles using Sargassum sp. and Padina sp. extract and 0.1 M ZnCl2 solution as a precursor. The ZnO nanoparticles had been characterized such as chemical structure by Fourier transform infrared (FTIR) spectroscopy, particle size distribution with Particle Size Analyzer (PSA), Scanning Electron Microscopy-Energy Dispersive Spectroscopy (SEMEDS) and crystalline properties with X-ray diffraction (XRD). The result showed that FTIR spectra revealed the involvement of hydroxyl groups and sulfate polysaccharide in the formation of ZnO NPs and protein was involved in stabilizing. PSA analysis showed that the size ranged of ZnO NPs extract Sargassum sp. synthesized 351,14 and 1.336.19 nm for extract Padina sp. The particle size distribution was homogeneous but their size hadn’t met the size of nanometers. The range of %mass elements Zn and O ZnO extract Sargassum sp. and Padina sp. synthesized didn’t approach standard ZnO. The crystallinity properties showed only ZnO extract of Padina sp. synthesized which had a peak with an angle value of 2θ was almost equal to standard ZnO and after confirmed to Match program! 3 crystal structure ZnO synthesized wasn’t imperfect hexagonal wurtzite. Ovens microwave could be used to synthesize ZnO from Padina sp. extract but it was still necessary to develop a method so that ZnO could be obtained with nanoparticle size and ideal hexagonal wurtzit structure
